    Fatty acid metabolism complements glycolysis in th selective regulatory t cell expansion during tumor growth

    The tumor microenvironment restrains conventional T cell (Tconv) activation while facilitating the expansion of Tregs. Here we showed that Tregs’ advantage in the tumor milieu relies on supplemental energetic routes involving lipid metabolism. In murine models, tumor-infiltrating Tregs displayed intracellular lipid accumulation, which was attributable to an increased rate of fatty acid (FA) synthesis. Since the relative advantage in glucose uptake may fuel FA synthesis in intratumoral Tregs, we demonstrated that both glycolytic and oxidative metabolism contribute to Tregs’ expansion. We corroborated our data in human tumors showing that Tregs displayed a gene signature oriented toward glycolysis and lipid synthesis. Our data support a model in which signals from the tumor microenvironment induce a circuitry of glycolysis, FA synthesis, and oxidation that confers a preferential proliferative advantage to Tregs, whose targeting might represent a strategy for cancer treatment

    Several anesthetics are used in aquaculture to reduce the stress caused by management. In the field, the most common anesthetic is benzocaine, but several authors are researching about eugenol. The objective of this study were to compare the action of these drugs on the anesthetics stages and measure the effects of successive administration of eugenol on the anesthetics stages in Oreochromis niloticus. In the first test, fish (47.73 +/- 8.73 g and 14.23 +/- 0.81 cm) were subjected to five concentrations of eugenol (40, 60, 80, 100 and 120 mg L-1) and one of benzocaine (100 mg L-1). All the anesthetic concentrations tested were able to induce fish to all stages of anesthesia, and revealed an inverse relationship between the induction times and increased concentrations. The minimum concentration of eugenol to induce anesthesia was 100 mg L-1. In the second test, the juveniles (38.07 +/- 5.00 g and 12.70 +/- 0.54 cm) were exposed to concentrations of 60, 80, 100 mg L-1 of eugenol, as determined in the first test. Evaluating the differences between successive days, there was no significant difference (P > 0.05) between anesthetics stages in the concentrations of eugenol tested, suggesting that the residue of eugenol was removed from the fish in less than 24 hours

    BACKGROUND: The ocular vestibular myogenic potentials (oVEMP) can be elicited by monaural air-conducted sound stimulation, and are usually recorded from the contralateral eye. In clinical setting a binaural stimulation would save time and require less effort from the subjects. OBJECTIVE: We evaluated the differences between monaural and binaural stimulation, and the possible effect of age and gender on oVEMP parameters. METHODS: Air-conducted oVEMP were recorded by binaural and by monaural stimulation in a group of 54 normal subjects, aged from 12 to 83 years, and in 50 vestibular patients. From each side, we measured the latency of the N1 component, and the peak-to-peak N1-P1 amplitude. For both parameters we also computed the asymmetry ratio. RESULTS: In normal subjects binaural stimulation produced slightly larger responses than monaural stimulation; detectability, latency and amplitude ratio were the same for the two techniques. We found no differences related to gender, and the age-induced amplitude decline was likely to be negligible.oVEMP recorded not in an acute phase of their disorder, proved to be abnormal in about 20% of the patients, and the normal or abnormal findings obtained either with monaural or with binaural stimulation were always concordant. CONCLUSIONS: The oVEMP obtained after binaural and monaural stimulation are very similar, and they are largely independent from age and gender

    <span style="font-family: Times New Roman; font-size: small;"> </span><p>The objective of this study was to assess the growth performance and the histomorphometric characterization of the middle intestine of GIFT strain Nile tilapia, <em>Oreochromis niloticus</em> juveniles fed probiotics, added either before or after the feed processing. The experimental design was completely randomized, with five treatments and five replications: pelleted feed without any probiotic; pelleted feed with inclusion of probiotic before the processing; pelleted feed with inclusion of probiotic after the processing; extruded feed without any probiotic and extruded feed with inclusion of probiotic after the processing. Two hundred and fifty juveniles were distributed in 25 aquaria (20L) and fed for 63 days. Differences in the mean values of total weight were found at the end of the experiment. After 42 days of feed intake, significant differences in feed conversion were verified for treatments with extruded diets when compared to the pelleted ones. The fishes fed diets supplemented with probiotic presented increase in the thickness of the epithelium of the intestine. The best zootechnical performance was observed in the extruded diet supplemented with probiotic added after the feed processing. The inclusion of 4.0 x 10<sup>8 </sup>CFU g<sup>-1</sup> of <em>Bacillus subtilis</em> and 4.0 x 10<sup>8 </sup>CFU g<sup>-1</sup> of <em>Bacillus toyoi</em> in both the pelleted and the extruded diets promoted an increase in the thickness of the epithelial layer of the middle intestine.</p> <p class="akeyword"> </p><p class="MsoNormal" style="margin: 0cm 0cm 0pt; text-align: justify; line-height: 150%;"> </p><span style="font-family: Times New Roman; font-size: small;"> </span

    Incorporação de probióticos na dieta para juvenis de tilápias-do-nilo: Parâmetros hematológicos, imunológicos e microbiológicos

    Probiotics are generally considered as live microorganisms which, when administered in adequate amounts, confer a health benefit to the host. The processing method of diet and the form of inclusion of probiotic can interfere in hematological, immunological and microbiological parameters in fish. The aim was to estimate the hematological, immunological and microbiological parameters in juveniles of Nile tilapia, fed probiotic, included before and after the process of pelletization and extrusion. The experimental design was completely randomized, with five treatments: pelleted diet without probiotic, pelleted diet with inclusion of probiotic before and after processing, extruded feed without probiotic and extruded feed with inclusion of probiotic after processing and five replications. Two hundred and fifty fish were distributed in 25 aquaria (20 L) and fed for 63 days. The blood composition (red and white) showed no significant differences except mean corpuscular hemoglobin concentration of control when compared to other treatments. The phagocytic capacity of the animals that had received the extruded diet supplemented with probiotic was significantly higher when compared to the other treatments. However, there were no differences between the treatments regarding to the phagocytic index. Fish fed the extruded diet exhibited significant improvement in the nonspecific immunity. The probiotic bacteria colonized the intestine, since it was possible to recover them. We can affirm that these fishes remained healthy, because the hematological parameters were not altered during the experimental. The study shows that any form type of inclusion in the feed tested (before or after and after pelletizing extrusion) may be easily used by the fish farmer

    Neurophysiological evaluation of visual function in iRBD: potential role in stratifying RBD conversion risk

    Study objectives: To evaluate neurophysiological alterations of visual function in idiopathic REM sleep Behavior Disorder (iRBD) both as markers and predictors of neurodegenerative disorders.Methods: In a longitudinal follow-up study of 46 consecutive iRBD patients (follow-up duration 8.4 +/- 3.4 years), the baseline parameters in luminance-contrast pattern (VEPp), red-green color (VEPc) and motion-onset (VEPm) Visual Evoked Potentials in iRBD were compared to early (ePD) and advanced (aPD) Parkinson's Disease subjects. Parameters of latency and amplitude of iRBD converters to neurodegenerative disease were compared with those of the non-converters.Results: The VEP P100 mean latency values for both eyes and for both stimulation checks (30' and 15') were significantly longer in all the three groups of patients as compared to controls; moreover latencies were longer in aPD than in the iRBD group who did not differ from the ePD group. The same held true when we analyzed the number of abnormal subjects belonging to each diagnostic group with a higher number of abnormal subjects in the aPD group compared to both the ePD and in iRBD groups. Chromatic and motion potentials were not different from controls and did not differ in the 3 diagnostic groups. The iRBD subjects who converted to a neurodegenerative disorder showed longer P100 latencies and a higher occurrence of VEPp abnormalities than those who did not convert. Again chromatic and motion VEPs were not different depending on conversion.Conclusions: In iRBD patients the detection of an abnormal VEPp should be considered as a red flag for possible synnucleinopathy, eventually contributing in stratifying the risk of phenoconversion.
