    IDENTIFICATION OF CANINE VISCERAL LEISHMANIASIS IN A PREVIOUSLY UNAFFECTED AREA BY CONVENTIONAL DIAGNOSTIC TECHNIQUES AND CELL-BLOCK FIXATION

    After the report of a second case of canine visceral leishmaniasis (CVL) in São Bento da Lagoa, Itaipuaçu, in the municipality of Maricá, Rio de Janeiro State, an epidemiological survey was carried out, through active search, totaling 145 dogs. Indirect immunofluorescence assay (IFA), enzyme-linked immunosorbent assay (ELISA), and rapid chromatographic immunoassay based on dual-path platform (DPP(r)) were used to perform the serological examinations. The parasitological diagnosis of cutaneous fragments was performed by parasitological culture, histopathology, and immunohistochemistry. In the serological assessment, 21 dogs were seropositive by IFA, 17 by ELISA, and 11 by DPP(r), with sensitivity of 66.7%, 66.7% and 50%, and specificity of 87.2%, 90.2% and 94%, respectively for each technique. The immunohistochemistry of bone marrow using the cell-block technique presented the best results, with six positive dogs found, three of which tested negative by the other parasitological techniques. Leishmania sp. was isolated by parasitological culture in three dogs. The detection of autochthonous Leishmania infantum in Itaipuaçu, and the high prevalence of seropositive dogs confirm the circulation of this parasite in the study area and alert for the risk of expansion in the State of Rio de Janeiro

    POTENCIAL DE USO DE ESPÉCIES VEGETAIS DE ÁREAS DE DUNAS EM SÃO LUÍS, MARANHÃO, BRASIL

    O avanço das cidades em todo litoral brasileiro tem aumentado a ameaça aos ecossistemas costeiros, fazendo-se necessários estudos florísticos para obter-se conhecimento da diversidade de determinadas áreas. Esses estudos, ultimamente, estão sendo associados à etnobiologia ampliando o conhecimento e as estratégias usadas pelos humanos e suas relações com os recursos biológicos. Nesse contexto, o objetivo do presente estudo foi indicar as formas de uso e nome popular das espécies vegetais de áreas de dunas do litoral da Ilha do Maranhão a fim de compilar essas informações como forma de documentar quais espécies de dunas podem ser utilizadas pela população. O levantamento das espécies vegetais foi realizado em três áreas de dunas situadas na Ilha do Maranhão, São Luís. A lista de espécies da Praia de São Marcos foi compilada com base no estudo florístico publicado e as espécies das praias de Caolho e Calhau foram compiladas no banco de dados do INCT e das exsicatas do Herbário MAR. A classificação quanto às formas de uso e os nomes populares foi realizada com base em consultas bibliográficas. Para as três áreas foram compiladas 148 espécies, 114 gêneros e 48 famílias. Foram categorizadas seis categorias de uso: medicinal, alimentícia, ornamental, madeireira, artesanal e ritualística. As partes da planta mais utilizadas para diferentes fins foram folhas, caule, raiz e fruto. O estudo destacou a importância dos conhecimentos tradicionais, pontuando a necessidade de conduzir esse conhecimento para o meio cientifico com responsabilidade, ampliando assim as possibilidades de pesquisas

    Predicting the Proteins of Angomonas deanei, Strigomonas culicis and Their Respective Endosymbionts Reveals New Aspects of the Trypanosomatidae Family

    Endosymbiont-bearing trypanosomatids have been considered excellent models for the study of cell evolution because the host protozoan co-evolves with an intracellular bacterium in a mutualistic relationship. Such protozoa inhabit a single invertebrate host during their entire life cycle and exhibit special characteristics that group them in a particular phylogenetic cluster of the Trypanosomatidae family, thus classified as monoxenics. in an effort to better understand such symbiotic association, we used DNA pyrosequencing and a reference-guided assembly to generate reads that predicted 16,960 and 12,162 open reading frames (ORFs) in two symbiont-bearing trypanosomatids, Angomonas deanei (previously named as Crithidia deanei) and Strigomonas culicis (first known as Blastocrithidia culicis), respectively. Identification of each ORF was based primarily on TriTrypDB using tblastn, and each ORF was confirmed by employing getorf from EMBOSS and Newbler 2.6 when necessary. the monoxenic organisms revealed conserved housekeeping functions when compared to other trypanosomatids, especially compared with Leishmania major. However, major differences were found in ORFs corresponding to the cytoskeleton, the kinetoplast, and the paraflagellar structure. the monoxenic organisms also contain a large number of genes for cytosolic calpain-like and surface gp63 metalloproteases and a reduced number of compartmentalized cysteine proteases in comparison to other TriTryp organisms, reflecting adaptations to the presence of the symbiont. the assembled bacterial endosymbiont sequences exhibit a high A+T content with a total of 787 and 769 ORFs for the Angomonas deanei and Strigomonas culicis endosymbionts, respectively, and indicate that these organisms hold a common ancestor related to the Alcaligenaceae family. Importantly, both symbionts contain enzymes that complement essential host cell biosynthetic pathways, such as those for amino acid, lipid and purine/pyrimidine metabolism.     Hypothalamic S1P/S1PR1 axis controls energy homeostasis

    Sphingosine 1-phosphate receptor 1 (S1PR1) is a G-protein-coupled receptor for sphingosine-1-phosphate (S1P) that has a role in many physiological and pathophysiological processes. Here we show that the S1P/S1PR1 signalling pathway in hypothalamic neurons regulates energy homeostasis in rodents. We demonstrate that S1PR1 protein is highly enriched in hypothalamic POMC neurons of rats. Intracerebroventricular injections of the bioactive lipid, S1P, reduce food consumption and increase rat energy expenditure through persistent activation of STAT3 and the melanocortin system. Similarly, the selective disruption of hypothalamic S1PR1 increases food intake and reduces the respiratory exchange ratio. We further show that STAT3 controls S1PR1 expression in neurons via a positive feedback mechanism. Interestingly, several models of obesity and cancer anorexia display an imbalance of hypothalamic S1P/S1PR1/STAT3 axis, whereas pharmacological intervention ameliorates these phenotypes. Taken together, our data demonstrate that the neuronal S1P/S1PR1/STAT3 signalling axis plays a critical role in the control of energy homeostasis in rats

    Multidisciplinary approach in breast cancer.

    Breast cancer is the most common neoplasm among women. Worldwide, there will be about 2.1 million newly diagnosed female breast cancer cases in 2018, accounting for almost 1 in 4 cancer cases among women. The disease is the most frequently diagnosed cancer in the vast majority of the countries. The purpose of this article is to report the positive experience of a multidisciplinary team in the care of women with breast cancer and their family members. Our approach that is part of the Cancer Patient Support Center (CPSC) at the public health (IPSEMG) in Brazil has been developed taking into account a broader concept of health care. We value not only individual dimensions in patient care, but also common ones, we recognize the importance of dealing with non-biological aspects of the disease, such as socioeconomic, political and cultural facets and our service is focused on health promotion rather than merely on curative treatment. Among the advantages of our approach, we highlight the facilitated accessibility to health services, the patient-centered communication and shared decision making, and the strong bonds between health professionals, patients, and family members. As part of CPSC`s activities, we emphasize the services provided by ?Aconchego? (?Warmth?), that is our breast cancer support group at public health in Brazil
