    Effects of astaxanthin on accommodation, critical flicker fusion, and pattern visual evoked potential in visual display terminal workers

    We evaluated the effects of astaxanthin, a red carotenoid, on accommodation, critical flicker fusion (CFF), and pattern visual evoked potential (PVEP) in visual display terminal (VDT) workers. As controls, 13 non-VDT workers received no supplementation (Group A). Twenty-six VDT workers were randomized into 2 groups: Group B consisted of 13 subjects who received oral astaxanthin, 5 mg/day, for 4 weeks, and Group C consisted of 13 subjects who received an oral placebo, 5 mg/day, for 4 weeks. No significant difference in age was noted among the 3 groups. A double-masked study was designed in Groups B and C. Accommodation amplitude in Group A was 3.7± 1.5 diopters. Accommodation amplitudes (2.3±1.4 and 2.2±1.0 diopters) in Groups B and C before supplementation were significantly (p<0.05) lower than in Group A. Accommodation amplitude (2.8±1.6 diopters) in Group B after astaxanthin treatment was significantly (p<0.01) larger than before supplementation, while accommodation amplitude (2.3±1.1 diopters) in Group C after placebo supplementation was unchanged. The CFFs and amplitude and latency of P100 in PVEP in Group A were 45.0±4.2 Hz, 6.5±1.8μV, and 101.3±6.5 msec, respectively. The CFFs in Groups B and C before supplementation were significantly (p<0.05) lower than in Group A. The CCFs in Groups B and C did not change after supplementation. Amplitudes and latencies of P100 in PVEP in Groups B and C before supplementation were similar to those in Group A and did not change after supplementation. Findings of the present study indicated that accommodation amplitude improved after astaxanthin supplementation in VDT workers. 赤色カロチノイドの一種であるアスタキサンチンのvisual display terminal(VDT)作業者の調節力,中心フリッカー値,パターン視覚誘発電位に及ぼす影響を調べた。VDT作業を行わない13人をコントロールとした(Group A)。26人のVDT作業者を2群に無作為に分けた。Group Bはアスタキサンチン一日5mg 4週間内服した13人で,Group Cはアスタキサンチンを含有しないカプセルを4週間内服した13人とした。外見上同じカプセルでの内服投与を行った。結果:Group AはGroup B及びGroup Cと比較して,調節力,中心フリッカー値は有意に高い値であったが,パターン視覚誘発電位検査結果は,Group B,Cと有意差はなかった。Group Bでは,アスタキサンチンの投与前後で有意な調節力の改善がみられた(p<0.01)。しかし,中心フリッカー値,パターン視覚誘発電位に変化はみられなかった。Group Cでは,投与前後で,調節力,中心フリッカー値,パターン視覚誘発電位に変化はみられなかった。考察:VDT作業者では,非作業者と比べ調節力,中心フリッカー値が低下していることは以前より報告されており,今回の我々の研究でも同様の結果であった。VDT作業者で,アスタキサンチン非内服群では,調節力は投与前後で変化がなかったが,アスタキサンチンの内服群で,有意に調節力が改善した。VDT作業者の調節力の改善には,アスタキサンチンの内服が有効と考えられた

    The truncated disk from Suzaku data of GX 339-4 in the extreme very high state

    We report on the geometry of accretion disk and high energy coronae in the strong Comptonization state (the very high/steep power law/hard intermediate state) based on a Suzaku observation of the famous Galactic black hole GX 339-4. These data were taken just before the peak of the 2006-2007 outburst, and the average X-ray luminosity in the 0.7-200 keV band is estimated to be 2.9E38 erg/s for a distance of 8 kpc. We fit the spectrum with both simple (independent disk and corona) and sophisticated (energetically coupled disk and corona) models, but all fits imply that the underlying optically thick disk is truncated significantly before the innermost stable circular orbit around the black hole. We show this directly by a comparison with similarly broadband data from a disk dominated spectrum at almost the same luminosity observed by XMM-Newton and RXTE 3 days after the Suzaku observation. During the Suzaku observation, the QPO frequency changes from 4.3 Hz to 5.5 Hz, while the spectrum softens. The energetically coupled model gives a corresponding 5+/- 8 % decrease in derived inner radius of the disk. While this is not significant, it is consistent with the predicted change in QPO frequency from Lense-Thirring precession of the hot flow interior to the disk and/or a deformation mode of this flow, as a higher QPO frequency implies a smaller size scale for the corona.     Atypical Lipomatous Tumor of the Tongue:Report of a Case

    The term atypical lipomatous tumor (ALT) is synonymous with well-differentiated liposarcoma (WDL). This tumor occurs very rarely in the tongue. Thus, it is difficult to predict its prognosis. Although recurrence of ALT/WDL is thought to be unlikely after complete excision, long-term follow-up is necessary when considering the pathologic conditions of this tumor at other sites. Here, we report a case of an ALT of the tongue, with a review of the literature. A 68-year-old man was referred to our hospital because of a tumor on the left side of his tongue. Upon palpation, the tumor was 12mm in diameter, circumscribed, elastic and hard, well demarcated, movable, and painless. We diagnosed the lesion as a lipoma and extirpated the tumor under local anesthesia. Because the specimen was histopathologically diagnosed as an ALT, as a precaution, we excised an additional 5mm from the area surrounding the original tumor under general anesthesia. Three years after the operation, the tongue demonstrated good healing without paresthesia or dysfunction, and to date there has been no evidence of recurrence

    Protection by Exogenously Added Coenzyme Q9 against Free Radical-Induced Injuries in Human Liver Cells

    Reduced coenzyme Q10 (CoQ10H2) is known as a potent antioxidant in biological systems. However, it is not yet known whether CoQ9H2 could act as an antioxidant in human cells. The aim of this study is to assess whether exogenously added CoQ9 can protect human liver cells against injuries induced by a water-soluble radical initiator, 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) and a lipid-soluble radical initiator, 2,2'-azobis(2,4-dimethylvaleronitrile) (AMVN). CoQ9-enriched cells were obtained by treatment of HepG2 cells with 10 µM CoQ9 liposomes for 24 h. CoQ9-enriched cells were exposed to 10 mM AAPH and 500 µM AMVN over 4 h and 24 h, respectively. The loss of viability after treatment with AAPH or AMVN was much less in CoQ9-enriched cells than in naive HepG2 cells. The decrease in glutathione and the increase in thiobarbituric acid-reactive substance after treatment with AAPH or AMVN were also suppressed in CoQ9-enriched cells. The incubation of CoQ9-enriched cells with AAPH or AMVN led to a decrease in cellular CoQ9H2 and reciprocal increase in cellular CoQ9 resulting from its antioxidant function. Taken together, it was demonstrated for the first time that exogenously added CoQ9 could prevent oxidative stress-mediated damage to human cells by virtue of its antioxidant activity

    Elimination of Specific miRNAs by Naked 14-nt sgRNAs

    tRNase ZL-utilizing efficacious gene silencing (TRUE gene silencing) is a newly developed technology to suppress mammalian gene expression. TRUE gene silencing works on the basis of a unique enzymatic property of mammalian tRNase ZL, which is that it can recognize a pre-tRNA-like or micro-pre-tRNA-like complex formed between target RNA and artificial small guide RNA (sgRNA) and can cleave any target RNA at any desired site. There are four types of sgRNA, 5′-half-tRNA, RNA heptamer, hook RNA, and ∼14-nt linear RNA. Here we show that a 14-nt linear-type sgRNA against human miR-16 can guide tRNase ZL cleavage of miR-16 in vitro and can downregulate the miR-16 level in HEK293 cells. We also demonstrate that the 14-nt sgRNA can be efficiently taken up without any transfection reagents by living cells and can exist stably in there for at least 24 hours. The naked 14-nt sgRNA significantly reduced the miR-16 level in HEK293 and HL60 cells. Three other naked 14-nt sgRNAs against miR-142-3p, miR-206, and miR-19a/b are also shown to downregulate the respective miRNA levels in various mammalian cell lines. Our observations suggest that in general we can eliminate a specific cellular miRNA at least by ∼50% by using a naked 14-nt sgRNA on the basis of TRUE gene silencing

    Modelling the high mass accretion rate spectra of GX 339-4: Black hole spin from reflection?

    We extract all the XMM-Newton EPIC pn burst mode spectra of GX 339-4, together with simultaneous/contemporaneous RXTE data. These include three disc dominated and two soft intermediate spectra, and the combination of broad bandpass/moderate spectral resolution gives some of the best data on these bright soft states in black hole binaries. The disc dominated spectra span a factor three in luminosity, and all show that the disc emission is broader than the simplest multicolour disc model. This is consistent with the expected relativistic smearing and changing colour temperature correction produced by atomic features in the newest disc models. However, these models do not match the data at the 5 per cent level as the predicted atomic features are not present in the data, perhaps indicating that irradiation is important even when the high energy tail is weak. Whatever the reason, this means that the data have smaller errors than the best physical disc models, forcing use of more phenomenological models for the disc emission. We use these for the soft intermediate state data, where previous analysis using a simple disc continuum found an extremely broad residual, identified as the red wing of the iron line from reflection around a highly spinning black hole. However, the iron line energy is close to where the disc and tail have equal fluxes, so using a broader disc continuum changes the residual 'iron line' profile dramatically. With a broader disc continuum model, the inferred line is formed outside of 30 Rg{\rm{R_g}}, so cannot constrain black hole spin. We caution that a robust determination of black hole spin from the iron line profile is very difficult where the disc makes a significant contribution at the iron line energy i.e. in most bright black hole states.Comment: 12 pages, 16 figures, published in MNRA

    Pressure-induced phase transition of Bi2Te3 into the bcc structure

    The pressure-induced phase transition of bismuth telluride, Bi2Te3, has been studied by synchrotron x-ray diffraction measurements at room temperature using a diamond-anvil cell (DAC) with loading pressures up to 29.8 GPa. We found a high-pressure body-centered cubic (bcc) phase in Bi2Te3 at 25.2 GPa, which is denoted as phase IV, and this phase apperars above 14.5 GPa. Upon releasing the pressure from 29.8 GPa, the diffraction pattern changes with pressure hysteresis. The original rhombohedral phase is recovered at 2.43 GPa. The bcc structure can explain the phase IV peaks. We assumed that the structural model of phase IV is analogous to a substitutional binary alloy; the Bi and Te atoms are distributed in the bcc-lattice sites with space group Im-3m. The results of Rietveld analysis based on this model agree well with both the experimental data and calculated results. Therefore, the structure of phase IV in Bi2Te3 can be explained by a solid solution with a bcc lattice in the Bi-Te (60 atomic% tellurium) binary system.Comment: 12 pages, 5 figure

    Dioxins and Fatty Acids in Breast Milk of Primiparas in Yonago District, Tottori Prefecture, Japan

    We analyzed the concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs) in breast milk collected 1 week after childbirth from 8 primiparas in Yonago district, Tottori Prefecture, and investigated the relationship between PCDDs or PCDFs and fatty acids in concentration, and the relationship between dioxin concentration and kind of daily foods. The mean total content of dioxins (PCDDs and PCDFs) was 0.48 pg-toxic equivalent (TEQ)/g (range 0.12?1.04 pg-TEQ/g) in breast milk, and 16.7 pg-TEQ/g-fat (range 9.6?32.7 pg-TEQ/g-fat) in total lipids of breast milk. The 8 primiparas showed a low mean dioxin concentration: the levels were lower in 6 of them and higher in 2 of them than in primiparas living in other cities in Japan. For 1 of the 2 mothers, the reason for the high level was thought to be her poor intake of vegetables in the diet. The total dioxin content was well correlated with the total lipid content ranging from 1% to 3%. Fatty acids with C16:0 and C18:1 dominated those with C12:0, C14:0, C16:1, C18: 2 and C18:0, which were commonly detected. The amount of fatty acids with C10:0, C20:1, C20:2, C20:3, C20:4, C22:5 and C22:6 was small. Gas chromatograms of these fatty acids generally showed similar distributions in breast milk of the 8 primiparas. The contents of fatty acids ranged from 17.1 to 31.3 mg/g (average 24.0 mg/g) in bulk breast milk. No clear correlation was found in concentration between PCDDs or PCDFs and specific fatty acids in breast milk
