The hydrolysis of proteins by microwave energy
Microwave energy, at manually-adjusted, partial power settings
has been used to hydrolyse bovine serum albumin at 125 °C. Hydrolysis was complete within 2 h, except for valine and isoleucine which were completely liberated within 4 h. The aminoacid destruction was less than that observed at similar hydrolysis
conditions with other methods and complete hydrolysis was achieved more rapidly. These results provide a basis for automating the process of amino-acid hydrolysis
Photoionization Broadening of the 1S-2S Transition in a Beam of Atomic Hydrogen
We consider the excitation dynamics of the two-photon \sts transition in a
beam of atomic hydrogen by 243 nm laser radiation. Specifically, we study the
impact of ionization damping on the transition line shape, caused by the
possibility of ionization of the 2S level by the same laser field. Using a
Monte-Carlo simulation, we calculate the line shape of the \sts transition for
the experimental geometry used in the two latest absolute frequency
measurements (M. Niering {\it et al.}, PRL 84, 5496 (2000) and M. Fischer {\it
et al.}, PRL 92, 230802 (2004)). The calculated line shift and line width are
in excellent agreement with the experimentally observed values. From this
comparison we can verify the values of the dynamic Stark shift coefficient for
the \sts transition for the first time on a level of 15%. We show that the
ionization modifies the velocity distribution of the metastable atoms, the line
shape of the \sts transition, and has an influence on the derivation of its

Dopamine neurons create Pavlovian conditioned stimuli with circuit-defined motivational properties.
Environmental cues, through Pavlovian learning, become conditioned stimuli that guide animals toward the acquisition of rewards (for example, food) that are necessary for survival. We tested the fundamental role of midbrain dopamine neurons in conferring predictive and motivational properties to cues, independent of external rewards. We found that brief phasic optogenetic excitation of dopamine neurons, when presented in temporal association with discrete sensory cues, was sufficient to instantiate those cues as conditioned stimuli that subsequently both evoked dopamine neuron activity on their own and elicited cue-locked conditioned behavior. Notably, we identified highly parcellated functions for dopamine neuron subpopulations projecting to different regions of striatum, revealing dissociable dopamine systems for the generation of incentive value and conditioned movement invigoration. Our results indicate that dopamine neurons orchestrate Pavlovian conditioning via functionally heterogeneous, circuit-specific motivational signals to create, gate, and shape cue-controlled behaviors
Disease-modifying drug initiation patterns in commercially insured multiple sclerosis patients: a retrospective cohort study
<p>Abstract</p> <p>Background</p> <p>The goal of this research was to compare the demographics, clinical characteristics and treatment patterns for newly diagnosed multiple sclerosis (MS) patients in a commercial managed care population who received disease-modifying drug (DMD) therapy versus those not receiving DMD therapy.</p> <p>Methods</p> <p>A retrospective cohort study using US administrative healthcare claims identified individuals newly diagnosed with MS (no prior MS diagnosis 12 months prior using ICD-9-CM 340) and ℠18 years old during 2001-2007 to characterize them based on demographics, clinical characteristics, and pharmacologic therapy for one year prior to and a minimum of one year post-index. The index date was the first MS diagnosis occurring in the study period. Follow-up of subjects was done by ICD-9-CM code identification and not by actual chart review. Multivariate analyses were conducted to adjust for confounding variables.</p> <p>Results</p> <p>Patients were followed for an average of 35.7 ± 17.5 months after their index diagnosis. Forty-three percent (n = 4,462) of incident patients received treatment with at least one of the DMDs during the post-index period. Treated patients were primarily in the younger age categories of 18-44 years of age, with DMD therapy initiated an average of 5.3 ± 9.1 months after the index diagnosis. Once treatment was initiated, 27.7% discontinued DMD therapy after an average of 17.6 ± 14.6 months, and 16.5% had treatment gaps in excess of 60 days.</p> <p>Conclusions</p> <p>Nearly 60% of newly-diagnosed MS patients in this commercial managed care population remained untreated while over a quarter of treated patients stopped therapy and one-sixth experienced treatment gaps despite the risk of disease progression or a return of pre-treatment disease activity.</p
MODIS Tree Cover Validation for the Circumpolar Taiga-Tundra Transition Zone
A validation of the 2005 500m MODIS vegetation continuous fields (VCF) tree cover product in the circumpolar taiga-tundra ecotone was performed using high resolution Quickbird imagery. Assessing the VCF's performance near the northern limits of the boreal forest can help quantify the accuracy of the product within this vegetation transition area. The circumpolar region was divided into longitudinal zones and validation sites were selected in areas of varying tree cover where Quickbird imagery is available in Google Earth. Each site was linked to the corresponding VCF pixel and overlaid with a regular dot grid within the VCF pixel's boundary to estimate percent tree crown cover in the area. Percent tree crown cover was estimated using Quickbird imagery for 396 sites throughout the circumpolar region and related to the VCF's estimates of canopy cover for 2000-2005. Regression results of VCF inter-annual comparisons (2000-2005) and VCF-Quickbird image-interpreted estimates indicate that: (1) Pixel-level, inter-annual comparisons of VCF estimates of percent canopy cover were linearly related (mean R(sup 2) = 0.77) and exhibited an average root mean square error (RMSE) of 10.1 % and an average root mean square difference (RMSD) of 7.3%. (2) A comparison of image-interpreted percent tree crown cover estimates based on dot counts on Quickbird color images by two different interpreters were more variable (R(sup 2) = 0.73, RMSE = 14.8%, RMSD = 18.7%) than VCF inter-annual comparisons. (3) Across the circumpolar boreal region, 2005 VCF-Quickbird comparisons were linearly related, with an R(sup 2) = 0.57, a RMSE = 13.4% and a RMSD = 21.3%, with a tendency to over-estimate areas of low percent tree cover and anomalous VCF results in Scandinavia. The relationship of the VCF estimates and ground reference indicate to potential users that the VCF's tree cover values for individual pixels, particularly those below 20% tree cover, may not be precise enough to monitor 500m pixel-level tree cover in the taiga-tundra transition zone

With the advent and stunning success of combination antiretroviral therapy (ART) to prolong and improve quality of life for persons with HIV infection, HIV research has been afforded the opportunity to pivot towards studies aimed at finding "a cure." The mere idea that cure of HIV might be possible has energized researchers and the community towards achieving this goal. Funding agencies, both governmental and private, have targeted HIV cure as a high priority; many in the field have responded to these initiatives and the cure research agenda is robust. In this "salon" two editors of Pathogens and Immunity, Michael Lederman and Daniel Douek ask whether curing HIV is a realistic, scalable objective. We start with an overview perspective and have asked a number of prominent HIV researchers to add to the discussion
