    EFEITO DE DOSES DE NITROGÊNIO NA SEVERIDADE DA QUEIMA DAS PONTAS DAS FOLHAS DA CEBOLA

    A queima das pontas das folhas da cebola causada por Botrytis squamosa tem comprometido a produção de mudas e consequente produtividade de cebola. Seus danos podem ser severos se as plantas estiverem desequilibradas com adubação nitrogenada e mediante isso, o objetivo deste trabalho foi avaliar o efeito de doses de nitrogênio (N) na severidade da doença. Sementes de cebola da cultivar Bola Precoce foram semeadas em blocos casualizados e quatro repetições com os tratamentos contendo doses de 0, 0,5, 1, 1,5 e 2 vezes a dose recomendada de nitrogênio na forma de ureia (45%) dividindo no plantio e na cobertura aos 30 dias. A cada 7 dias após 15 dias da semeadura foi avaliada a severidade da doença. O teor de clorofila no momento da colheita aos 60 dias foi feito em dois pontos centrais da antepenúltima folha pelo Clorofilômetro portátil Modelo CFL 1030 ClorofiLOG da marca Falker. Os dados de severidade foram calculados e integralizados pela área abaixo da curva de progresso da doença (AACPD) e as plantas avaliadas para a doença foram pesadas para expressar a biomassa. As médias da AACPD, da severidade final, teor de clorofila e biomassa (g/planta) das mudas foram submetidos à análise de variância pelo teste de F e se significativos comparadas pelo teste de Tukey 5%. Não houve significância para os teores de clorofila nas folhas de cebola com a presença da doença em função das doses de N. Os valores tiveram diferença de 24% entre o maior valor (34,68 na dose 0) e o menor (26,17 na dose 0,5). Possivelmente isso reflete que a concentração do teor de clorofila também está relacionado com a presença da doença do qual obtém os solutos nitrogenados para seu desenvolvimento. Esses valores de teor de clorofila indicam que o teor de clorofila diminui em função do aumento da dose de N até 1,5 vezes, e posteriormente ocorre um leve acréscimo de apenas 0,73 pontos em relação ao dobro da dose. Houve diferença estatística na AACPD e severidade final, onde as doses de 0 e 0,5 apresentaram os maiores valores, no entanto a o teor de clorofila e biomassa não diferiram entre as doses. Independente disso, a adubação equilibrada é um importante fator a ser integrado no manejo da queima das pontas das folhas da cebola causada por Botrytis squamosa

    A vivens ex vivo study on the synergistic effect of electrolysis and freezing on the cell nucleus

    Freezing-cryosurgery, and electrolysis-electrochemical therapy (EChT), are two important minimally invasive surgery tissue ablation technologies. Despite major advantages they also have some disadvantages. Cryosurgery cannot induce cell death at high subzero freezing temperatures and requires multiple freeze thaw cycles, while EChT requires high concentrations of electrolytic products-which makes it a lengthy procedure. Based on the observation that freezing increases the concentration of solutes (including products of electrolysis) in the frozen region and permeabilizes the cell membrane to these products, this study examines the hypothesis that there could be a synergistic effect between freezing and electrolysis in their use together for tissue ablation. Using an animal model we refer to as vivens ex vivo, which may be of value in reducing the use of animals for experiments, combined with a Hematoxylin stain of the nucleus, we show that there are clinically relevant protocols in which the cell nucleus appears intact when electrolysis and freezing are used separately but is affected by certain combinations of electrolysis and freezing. \ua9 2015 Lugnani et al.     Promotion of plasma membrane repair by vitamin E

    Severe vitamin E deficiency results in lethal myopathy in animal models. Membrane repair is an important myocyte response to plasma membrane disruption injury as when repair fails, myocytes die and muscular dystrophy ensues. Here we show that supplementation of cultured cells with α-tocopherol, the most common form of vitamin E, promotes plasma membrane repair. Conversely, in the absence of α-tocopherol supplementation, exposure of cultured cells to an oxidant challenge strikingly inhibits repair. Comparative measurements reveal that, to promote repair, an anti-oxidant must associate with membranes, as α-tocopherol does, or be capable of α-tocopherol regeneration. Finally, we show that myocytes in intact muscle cannot repair membranes when exposed to an oxidant challenge, but show enhanced repair when supplemented with vitamin E. Our work suggests a novel biological function for vitamin E in promoting myocyte plasma membrane repair. We propose that this function is essential for maintenance of skeletal muscle homeostasis

    "Delirium Day": A nationwide point prevalence study of delirium in older hospitalized patients using an easy standardized diagnostic tool

    Background: To date, delirium prevalence in adult acute hospital populations has been estimated generally from pooled findings of single-center studies and/or among specific patient populations. Furthermore, the number of participants in these studies has not exceeded a few hundred. To overcome these limitations, we have determined, in a multicenter study, the prevalence of delirium over a single day among a large population of patients admitted to acute and rehabilitation hospital wards in Italy. Methods: This is a point prevalence study (called "Delirium Day") including 1867 older patients (aged 65 years or more) across 108 acute and 12 rehabilitation wards in Italian hospitals. Delirium was assessed on the same day in all patients using the 4AT, a validated and briefly administered tool which does not require training. We also collected data regarding motoric subtypes of delirium, functional and nutritional status, dementia, comorbidity, medications, feeding tubes, peripheral venous and urinary catheters, and physical restraints. Results: The mean sample age was 82.0 \ub1 7.5 years (58 % female). Overall, 429 patients (22.9 %) had delirium. Hypoactive was the commonest subtype (132/344 patients, 38.5 %), followed by mixed, hyperactive, and nonmotoric delirium. The prevalence was highest in Neurology (28.5 %) and Geriatrics (24.7 %), lowest in Rehabilitation (14.0 %), and intermediate in Orthopedic (20.6 %) and Internal Medicine wards (21.4 %). In a multivariable logistic regression, age (odds ratio [OR] 1.03, 95 % confidence interval [CI] 1.01-1.05), Activities of Daily Living dependence (OR 1.19, 95 % CI 1.12-1.27), dementia (OR 3.25, 95 % CI 2.41-4.38), malnutrition (OR 2.01, 95 % CI 1.29-3.14), and use of antipsychotics (OR 2.03, 95 % CI 1.45-2.82), feeding tubes (OR 2.51, 95 % CI 1.11-5.66), peripheral venous catheters (OR 1.41, 95 % CI 1.06-1.87), urinary catheters (OR 1.73, 95 % CI 1.30-2.29), and physical restraints (OR 1.84, 95 % CI 1.40-2.40) were associated with delirium. Admission to Neurology wards was also associated with delirium (OR 2.00, 95 % CI 1.29-3.14), while admission to other settings was not. Conclusions: Delirium occurred in more than one out of five patients in acute and rehabilitation hospital wards. Prevalence was highest in Neurology and lowest in Rehabilitation divisions. The "Delirium Day" project might become a useful method to assess delirium across hospital settings and a benchmarking platform for future surveys

    Ferruginous bodies resolved by synchrotron XRF in a dog with peritoneal malignant mesothelioma

    Mesothelioma is a malignant tumor mainly correlated to occupational asbestos exposure. Rare reports describe its occurrence also in animals, mainly linked to asbestos in the environment. Asbestos exposure is demonstrated by the appearance of characteristic histological hallmarks: asbestos containing ferruginous bodies that are iron-based structures forming around fibers and also other dust particles. Here we present a clinical case of a suspect of mesothelioma in the peritoneum of a dog with parallel histological observation of ferruginous bodies. To possibly correlate the dog tumor to environmental exposure, we performed Xray fluorescence (XRF) analyses at two different synchrotrons to resolve the ferruginous bodies\u2019 composition. While the histological examination diagnoses a tubulo-papillary mesothelioma, the XRF analyses show that ferruginous bodies contain Si particles, resembling formations of exogenous origin; however, the morphology is unlikely that of asbestos fibers. We speculate that the peritoneal mesothelioma of this dog could be related to environmental exposure to non-asbestos material

    Histone acetylation as a new mechanism for bilirubin-induced encephalopathy in the Gunn rat

    Abstract Bilirubin neurotoxicity has been studied for decades and has been shown to affect various mechanisms via significant modulation of gene expression. This suggests that vital regulatory mechanisms of gene expression, such as epigenetic mechanisms, could play a role in bilirubin neurotoxicity. Histone acetylation has recently received attention in the CNS due to its role in gene modulation for numerous biological processes, such as synaptic plasticity, learning, memory, development and differentiation. Aberrant epigenetic regulation of gene expression in psychiatric and neurodegenerative disorders has also been described. In this work, we followed the levels of histone 3 lysine 14 acetylation (H3K14Ac) in the cerebellum (Cll) of the developing (2, 9, 17 days after the birth) and adult Gunn rat, the natural model for neonatal hyperbilirubinemia and kernicterus. We observed an age-specific alteration of the H3K14Ac in the hyperbilirubinemic animals. The GeneOntology analysis of the H3K14Ac linked chromatin revealed that almost 45% of H3K14Ac ChiP-Seq TSS-promoter genes were involved in CNS development including maturation and differentiation, morphogenesis, dendritogenesis, and migration. These data suggest that the hallmark Cll hypoplasia in the Gunn rat occurs also via epigenetically controlled mechanisms during the maturation of this brain structure, unraveling a novel aspect of the bilirubin-induced neurotoxicity

    Effect of the state of water and relative humidity on ageing of PLA films

    International audienceVarious types of food are now commercialized in packaging materials based on poly(lactic acid) (PLA) due to its eco-friendly nature. However, one of the main limitations related to PLA is its reactivity with water. For food applications, it is of critical importance to better understand the hydrolysis of PLA driven by water molecules either in liquid or in vapour state. This work focuses on the modifications of PLA induced by water when simulating contact with semi-dry foods (aw ≈ 0.5), high moisture foods (aw ≈ 1) and liquid foods (aw ≈ 1). This study undoubtedly shows that both the chemical potential of water and its physical state influence the hydrolytic degradation of PLA films. From a practical point of view, PLA packaging is very well suited for semi-dry foods, but is highly sensitive to high moisture and liquid foods
