    Biofilm formation and Biocide resistance in Listeria

    La presente attività di ricerca ha avuto come oggetto di studio la formazione di biofilm in condizioni dinamiche: per raggiungere tale obiettivo, è stato costruito un idoneo apparato che permettesse di ottenere una valutazione quantitativa sia delle cellule presenti nella soluzione di crescita sia di quelle adese su tre materiali di prova(acciaio inossidabile, PET e rame). Oltre ad esprimere una valutazione dell’adesione batterica alle superfici, si è proceduto ad indagare sui possibili parametri sperimentali che potessero influenzare la formazione di biofilmdi Listeria su superfici abiotiche. Allo stesso tempo l’efficacia battericida di alcuni biocidi è stata valutata sulle cellule planctoniche così come su biofilm batterici sviluppati su superfici abiotiche. Il ruolo del sistema di quorum sensing basato su luxS nella formazione di biofilm batterici: un ceppo di L. innocua con buone capacità adesive è stato confrontato con il suo ceppo deficiente di luxS per la sua suscettibilità ad agenti biocidi così come per la sua capacità di adesione superficiale L’effetto della precendentemente menzionata inattivazione genetica sulla cinetica di crescita batterica e, quindi, sull’efficienza metabolica complessiva è stata esaminate mediante lo strumento BioscreenC. L’effetto inibitorio di due composti polifenolici è stato valutato sul ceppo deficiente di luxS così come sul ceppo originale.The current research activity was focused on biofilm formation in dynamic conditions. A suitable device was thus constructed, in order to have a quantitative evaluation of the cells freely suspended in the flowing nutrient solution used as well as the sessile cells which adhered on the three tested surfaces (stainless steel, polyethylene therephthalate and copper). During this project, other than of a quantitative assessment of bacterial adhesion, we investigated also on the parameters which could affect significantly the biofilm formation of Listeria on abiotic surfaces. At the same time antimicrobial efficiency of selected biocides was investigated on planktonic cells as well as on bacterial biofilms grown on abiotic surfaces. The role of luxS-based quorum sensing system in the biofilm development process was evaluated: a luxS-null mutant of an adhesive strain of L. innocua was obtained through Campbell-like genetic inactivation and then evaluated for its sensitivity to biocidal agents and adhesion to surfaces, in comparison with its parental strains. Furthermore the effect of genetic inactivation on bacterial growth kinetic and thus on overall bacterial metabolic efficiency was assessed through the apparatus of BioscreenC.     PN fast winds: Temporal structure and stellar rotation

    To diagnose the time-variable structure in the fast winds of central stars of planetary nebulae (CSPN), we present an analysis of P Cygni line profiles in FUSE satellite far-UV spectroscopic data. Archival spectra are retrieved to form time-series datasets for the H-rich CSPN NGC 6826, IC 418, IC 2149, IC 4593 and NGC 6543. Despite limitations due to the fragmented sampling of the time-series, we demonstrate that in all 5 CSPN the UV resonance lines are variable primarily due to the occurrence of blueward migrating discrete absorption components (DACs). Empirical (SEI) line-synthesis modelling is used to determine the range of fluctuations in radial optical depth, which are assigned to the temporal changes in large-scale wind structures. We argue that DACs are common in CSPN winds, and their empirical properties are akin to those of similar structures seen in the absorption troughs of massive OB stars. Constraints on PN central star rotation velocities are derived from Fast-Fourier Transform analysis of photospheric lines for our target stars. Favouring the causal role of co-rotating interaction regions, we explore connections between normalised DAC accelerations and rotation rates of PN central stars and O stars. The comparative properties suggest that the same physical mechanism is acting to generate large-scale structure in the line-driven winds in the two different settings.Comment: Accepted for publication in MNRAS; 10 pages, 5 figure

    Experimental and numerical evaluations on palm microwave heating for Red Palm Weevil pest control

    The invasive Red Palm Weevil is the major pest of palms. Several control methods have been applied, however concern is raised regarding the treatments that can cause significant environmental pollution. In this context the use of microwaves is particularly attractive. Microwave heating applications are increasingly proposed in the management of a wide range of agricultural and wood pests, exploiting the thermal death induced in the insects that have a thermal tolerance lower than that of the host matrices. This paper describes research aiming to combat the Red Palm pest using microwave heating systems. An electromagnetic-thermal model was developed to better control the temperature profile inside the palm tissues. In this process both electromagnetic and thermal parameters are involved, the latter being particularly critical depending on plant physiology. Their evaluation was carried out by fitting experimental data and the thermal model with few free parameters. The results obtained by the simplified model well match with both that of a commercial software 3D model and measurements on treated Phoenix canariensis palms with a ring microwave applicator.     Far-UV FUSE spectroscopy of the OVI resonance doublet in Sand2 (WO)

    We present Far-Ultraviolet Spectroscopic Explorer (FUSE) spectroscopy of Sand 2, a LMC WO-type Wolf-Rayet star, revealing the OVI resonance P Cygni doublet at 1032-38A. These data are combined with HST/FOS ultraviolet and Mt Stromlo 2.3m optical spectroscopy, and analysed using a spherical, non-LTE, line-blanketed code. Our study reveals exceptional stellar parameters: T*=150,000K, v_inf=4100 km/s, log (L/Lo)=5.3, and Mdot=10^-5 Mo/yr if we adopt a volume filling factor of 10%. Elemental abundances of C/He=0.7+-0.2 and O/He=0.15(-0.05+0.10) by number qualitatively support previous recombination line studies. We confirm that Sand 2 is more chemically enriched in carbon than LMC WC stars, and is expected to undergo a supernova explosion within the next 50,000 yr.Comment: 17 pages, 4 figures, AASTeX preprint format.     Impacto da vacina pneumocócica conjugada 10-valente na meningite pneumocócica em crianças com até dois anos de idade no Brasil

    O objetivo deste trabalho foi analisar o impacto da vacinação contra o Streptococcus pneumoniae na morbidade e mortalidade por meningite pneumocócica em crianças ≤ 2 anos, no Brasil, entre 2007-2012. Este é um estudo descritivo com análise ecológica, utilizando dados do Sistema de Informação de Agravos de Notificação. Foram definidos os períodos pré-vacinal (2007-2009) e pós-vacinal (2011-2012) para comparar as taxas de incidência e mortalidade. Foram identificados 1.311 casos e 430 óbitos no período do estudo. A taxa de incidência diminuiu de 3,70/100.000 no ano de 2007 para 1,84/100.000 em 2012, e a mortalidade reduziu de 1,30/100.000 para 0,40/100.000, o que significa uma redução de 50% e 69%, respectivamente, com maior impacto identificado na faixa etária de 6 a 11 meses. Os resultados indicam uma diminuição nos indicadores de morbidade e mortalidade de meningite pneumocócica, observados dois anos após a introdução da vacina pneumocócica conjugada 10-valente, sugerindo sua efetividade.The objective of this study was to analyze the impact of vaccination against Streptococcus pneumoniae on the morbidity and mortality from pneumococcal meningitis in children ≤ 2 years in Brazil, from 2007 to 2012. This is a descriptive study and ecological analysis using data from the Information System on Notifiable Diseases. Pre-vaccination (2007-2009) and post-vaccination (2011-2012) periods were defined to compare incidence rates and mortality. A total of 1,311 cases and 430 deaths were reported during the study period. Incidence decreased from 3.70/100,000 in 2007 to 1.84/100,000 in 2012, and mortality decreased from 1.30/100,000 to 0.40/100,000, or 50% and 69% respectively, with the greatest impact in the 6-11 month age group.
