T Lymphocytes in Histiocytic Sarcomas of Flat-Coated Retriever Dogs.
Flat-Coated Retriever dogs are predisposed to the development of histiocytic sarcoma (HS), a poorly differentiated, highly malignant neoplasm. The authors have previously documented a significant lymphocytic infiltrate in such tumors. The objective of this study was to examine the presence and expression of regulatory T cells in HS tumor samples. Forty tumors were included in this study. All tumors were immunolabeled for CD3, CD79a, CD25, CD45RA, and FOXP3. The proportion of positive cells was compared between tumors presenting as a localized primary soft tissue mass (soft tissue origin HS) and disseminated HS affecting viscera, especially the spleen (splenic origin HS). By immunohistochemistry, 95% of infiltrating T cells were positive for Foxp3 in all sections, suggesting the presence of regulatory T cells. The proportion of cells positive for FOXP3 was higher in the tumors arising in soft tissues, whereas the proportion of CD45RA-positive cells was higher in the splenic origin HS. Canine HS has an aggressive clinical behavior and is uniformly fatal. The difference in the proportion of tumor-infiltrating lymphocytes positive for these 2 markers in the 2 locations may represent differences in tumor microenvironment between the 2 sites
Influence of washing and quenching in profiling the metabolome of adherent mammalian cells: A case study with the metastatic breast cancer cell line MDA-MB-231
Metabolome characterisation is a powerful tool in oncology. To obtain a valid description of the intracellular
metabolome, two of the preparatory steps are crucial, namely washing and quenching. Washing
must effectively remove the extracellular media components and quenching should stop the metabolic
activities within the cell, without altering the membrane integrity of the cell. Therefore, it is important to
evaluate the efficiency of the washing and quenching solvents. In this study, we employed two previously
optimised protocols for simultaneous quenching and extraction, and investigated the effects of a number
of washing steps/solvents and quenching solvent additives, on metabolite leakage from the adherent
metastatic breast cancer cell line MDA-MB-231. We explored five washing protocols and five quenching
protocols (including a control for each), and assessed for effectiveness by detecting ATP in the medium
and cell morphology changes through scanning electron microscopy (SEM) analyses. Furthermore, we
studied the overall recovery of eleven different metabolite classes using the GC-MS technique and compared
the results with those obtained from the ATP assay and SEM analysis. Our data demonstrate that a
single washing step with PBS and quenching with 60% methanol supplemented with 70 mM HEPES
(−50 °C) results in minimum leakage of intracellular metabolites. Little or no interference of PBS (used in
washing) and methanol/HEPES (used in quenching) on the subsequent GC-MS analysis step was noted.
Together, these findings provide for the first time a systematic study into the washing and quenching
steps of the metabolomics workflow for studying adherent mammalian cells, which we believe will
improve reliability in the application of metabolomics technology to study adherent mammalian cell
metabolism
Vitamin D Levels in Asymptomatic Adults-A Population Survey in Karachi, Pakistan
Background: It is well established that low levels of 25(OH) Vitamin D (/dL) are a common finding world over, affecting over a billion of the global population. Our primary objective was to determine the prevalence of vitamin D deficiency and insufficiency in the asymptomatic adult population of Karachi, Pakistan and the demographic, nutritional and co-morbidity characteristics associated with serum vitamin D levels. Methods: A cross-sectional population survey was conducted at two spaced out densely populated areas of the city. Serum levels of 25OH vitamin D were measured and GFR as renal function was assessed by using 4 variable MDRD formula. Results: Our sample of 300 had a median age of 48(interquartile range 38-55) years. The median level of serum vitamin D was 18.8 (IQ range 12.65-24.62) ng/dL. A total of 253 (84.3%) respondents had low levels (/dL) of 25OH vitamin D. Serum PTH and vitamin D were negatively correlated (r = -0.176, p = 0.001). The median PTH in the vitamin D sufficiency group was 38.4 (IQ range28.0-48.8)pg/mL compared with 44.4 (IQ range 34.3-56.8) pg/mL in the deficiency group (p = 0.011).The median serum calcium level in the sample was 9.46(IQ range 9.18-9.68) ng/dL. Low serum levels of vitamin D were not associated with hypertension (p = 0.771) or with an elevated spot blood pressure (p = 0.164).In our sample 75(26%) respondents had an eGFR corresponding to stage 2 and stage 3 CKD. There was no significant correlation between levels of vitamin D and eGFR (r = -0.127, p-value = 0.277). Respondents using daily vitamin D supplements had higher 25 OH vitamin D levels (p-value = 0.021). Conclusion: We observed a high proportion of the asymptomatic adult population having low levels of vitamin D and subclinical deterioration of eGFR. Practical guidelines for the supplementation of vitamin D and the treatment of deficits in Central Europe — recommended vitamin D intakes in the general population and groups at risk of vitamin D deficiency
Wstęp: Wyniki badań z ostatnich lat dokumentują wiele korzyści wynikających z działania witaminy D na organizm człowieka na wszystkichetapach jego życia. Większość badań epidemiologicznych sugeruje, że niedobór witaminy D jest powszechny wśród mieszkańców EuropyŚrodkowej. Naturalną konsekwencją tej sytuacji jest konieczność ciągłego uświadamiania społeczeństwu oraz środowisku medycznemu,jaką rolę odgrywa witamina D w rozwoju i funkcjonowaniu organizmu ludzkiego.Metody: Na podstawie przeglądu danych literaturowych Polski Zespół Wielodyscyplinarny opracował tezy dotyczące zasad suplementacjiwitaminą D, które przesłano do członków Komitetu Naukowego konferencji „Witamina D — minimum, maksimum, optimum”,19–20 Październik, 2012, Warszawa. W trakcie powyższej konferencji z udziałem 550 delegatów oraz Ekspertów różnych dziedzin medycynyomówiono i przedyskutowano propozycje wytycznych suplementacji witaminą D populacji Europy Środkowej.Wyniki: W efekcie przeprowadzonych dyskusji Zespół Ekspertów opracował wytyczne suplementacji witaminą D dla wszystkich grupwiekowych populacji Europy Środkowej. Określono również kryteria diagnostyczne charakteryzujące stan zaopatrzenia organizmu w witaminę D: deficyt witaminy D ustalono jako stężenie 25(OH)D < 20 ng/mL (< 50 nmol/L)], suboptymalne zaopatrzenie jako stężenie25(OH)D wynoszące 20–30 ng/mL (50–75 nmol/L), a stężenie 30–50 ng/mL (75–125 nmol/L) uznano za docelowe dla zapewnienia efektuplejotropowego witaminy D.Wnioski: Poprawa obecnego stanu zaopatrzenia witaminy D w grupach dzieci, młodzieży, osób aktywnych zawodowo i seniorówpowinna zostać włączona do priorytetów polityki zdrowotnej społeczeństw Europy Środkowej.Introduction: Adequate Vitamin D intake and its concentration in serum are important for bone health and calcium–phosphate metabolismas well as for optimal function of many organs and tissues. Documented trends in lifestyle, nutritional habits and physical activityappear to be associated with moderate or severe Vitamin D deficits resulting in health problems. Most epidemiological studies suggest thatVitamin D deficiency is prevalent among Central European populations. Concern about this problem led to the organising of a conferencefocused on overcoming Vitamin D deficiency.Methods: After reviewing the epidemiological evidence and relevant literature, a Polish multidisciplinary group formulated theses onrecommendations for Vitamin D screening and supplementation in the general population. These theses were subsequently sent to ScientificCommittee members of the ‘Vitamin D — minimum, maximum, optimum’ conference for evaluation based on a ten-point scale.With 550 international attendees, the meeting ‘Vitamin D — minimum, maximum, optimum’ was held on October 19–20, 2012 in Warsaw(Poland). Most recent scientific evidence of both skeletal and non-skeletal effects of Vitamin D as well as the results of panellists’ votingwere reviewed and discussed during eight plenary sessions and two workshops.Results: Based on many polemical discussions, including post-conference networking, the key opinion leaders established ranges ofserum 25-hydroxyVitamin D concentration indicating Vitamin D deficiency [< 20 ng/mL (< 50 nmol/L)], suboptimal status [20–30 ng/mL(50–75 nmol/L)], and target concentration for optimal Vitamin D effects [30–50 ng/mL (75–125 nmol/L)]. General practical guidelines regardingsupplementation and updated recommendations for prophylactic Vitamin D intakes in Central European neonates, infants, childrenand adolescents as well as in adults (including recommendations for pregnant and breastfeeding women and the elderly) were developed.Conclusions: Improving the Vitamin D status of children, adolescents, adults and the elderly must be included in the priorities of physicians,healthcare professionals and healthcare regulating bodies. The present paper offers elaborated consensus on supplementationguidance and population strategies for Vitamin D in Central Europe
Teaching geology in Poland : an opinion in the discussion
The paper is an opinion in the discussion on the reconversion of uniform 5-year studies in geology. Our teaching experience, coupled with the opinions of our students, indicate that both models of teaching (2-level studies within the Bologna system and uniform 5-year studies) contain elements favouring the development offuture graduates. We postulate putting more emphasis on practical courses and individualized teaching methods
Magmatic and post-magmatic phenomena in the Karkonosze granite and its metamorphic envelope (West Sudetes, SW Poland)
Mineralogical studies of the Karkonosze granite (ca. 322–312 Ma) and its surroundings in West Sudetes (SW Poland) have provided data on Nb-Ta-REE minerals from pegmatites in the NE part of the pluton and several new finds of Ag minerals and 15 oxygenic Bi phases, hitherto not reported from the massif. The Karkonosze pegmatites are enriched in HREE as fergusonite-(Y) or xenotime-(Y) appear in almost every studied pegmatite, together with a subordinate assemblage of the aeschynite, euxenite or columbite group. The abundance of LREE minerals such as allanite-(Ce) and the monazite group, correlates inversely with the Nb-Ta-Ti minerals, whilst an early generation of monazite-(Ce) revealed an exceptionally high amount of Nd (up to 22 wt.% of Nd2O3). The physical and chemical conditions during the magmatic and post-magmatic processes were reconstructed and the effects of contact metamorphism in amphibolites from hornfelsed zones examined. Changes in solution composition and concentration at the early magmatic stage (825–920ºC), pegmatitic stage overlapping with hydrothermal (560°C which ended at 160–90°C) and clearly hydrothermal stage (400 to 110°C) were studied in detail by means of melt and fluid inclusions in quartz. Furthermore, post-magmatic fluids, including some enriched in Li and B, were identified in rock-forming quartz from the whole pluton. In turn, study of the amphibolites indicates that the pair cummingtonite + anorthite or the presence of Ca-rich plagioclase with actinolite seem to be reliable mineral proxies of the thermal impact of the granitoid body on amphibolites in its envelope. The inferred conditions of the contact processes (450–550°C, 2.5–4.8 kbar) point to an elevated geothermal gradient (ca. 32–45°C/km) probably reflecting the heat flow induced by the Karkonosze intrusion. Moreover, despite the textural and mineral changes imposed by regional and contact metamorphism, the amphibolites have their pre-metamorphic (magmatic) geochemical features undisturbed
Sea-floor massive sulphides from the Galápagos Rift Zone – mineralogy, geochemistry and economic importance
The largest reserves of the Earth's mineral resources are located on the ocean floor. Amongst these, hydrocarbon and metallic resources are of the greatest importance. In 2010, the International Seabed Authority (ISA) adopted rules for documenting new metal resources contained with polymetallic sea-floor massive sulphides (SMS). Russia and China were the first countries to apply to the ISA to acquire the right to commence initial investigation and documentation in the Indian and Atlantic oceans. Apart from copper, cobalt and nickel, modern SMS ores contain inferred significant amounts of gold, silver and PGE. The results described here from Galápagos Rift samples indicate a high concentration of gold (up to 7.24 ppm) and platinum (up to 350 ppb), which are mainly concentrated in low temperature parts of the sulphide mounds. The high content of Au and Pt revealed by this study provides further motivation towards attempting the exploration of marine massive sulphides. This is particularly important in the light of the high prices of metal commodities, and especially the unprecedented price of gold. This paper is the first such detailed scientific account in the Polish scientific literature focusing on the genesis and economic significance of the modern sea-floor sulphides
Modyfikacja tkanin lnianych preparatami pro-zdrowotnymi i jej wpływ na wybrane właściwości użytkowe i środowisko naturalne człowieka
The study involved the determination of the effect of the modification of linen fabrics with health promoting products containing different substances such as vitamin E, beeswax and natural silk protein on the use features of the fabrics and their biodegradation. It was found that enriching linen fabrics with health promoting products contributes to the improvement of their touch, does not have a negative effect on the biodegradation process nor the whiteness degree and the modification effect is resistant to 10 cycles of care processes. The modification causes a decrease in the breaking force and thread slippage index in the stitches. However, the values obtained are in line with the requirements for linen fabrics used for clothing. The modification process also lowers the hygroscopicity, air permeability and, in some cases, water absorption as compared with fabrics before the modification.W ramach prowadzonych prac badawczych określono wpływ modyfikacji tkanin lnianych produktami pro-zdrowotnymi zawierającymi alternatywnie substancje typu: witaminę E, aloes, wosk pszczeli, białko z jedwabiu naturalnego, na ich właściwości użytkowe i biodegradację. Stwierdzono, że uszlachetnianie tkanin lnianych produktami pro-zdrowotnymi przyczynia się do ich korzystniejszego chwytu, nie wpływa negatywnie na przebieg procesu biodegradacji oraz stopień bieli, a efekt modyfikacji jest odporny na 10-cio krotny proces konserwacji. Proces modyfikacji powoduje obniżenie wartości siły zrywającej i pogorszenie przesuwalności nitek w szwie. Uzyskane wartości są jednak zgodne z wymaganymi dla tkanin lnianych odzieżowych. Proces modyfikacji wpływa również na obniżenie parametrów higroskopijności i przepuszczalności powietrza oraz w niektórych wariantach na obniżenie zdolności wchłaniania wody w porównaniu z tkaninami przed modyfikacją
