    “Welcome to my side of town” – Teaching and learning by means of service learning in translator education

    This article investigates Service Learning (SL) as pedagogy for educating translator students in higher education. Situating translator education within new theories of knowledge, this article provides a description of a first-year module presented in translation practice. It makes use of Bringle and Hatcher’s conceptualisation of SL, with a focus on the application thereof at the UFS. The aim of the article is to argue for more attention to the curriculum in translator education, rather than mere pedagogy, as well as for attention to philosophical and ideological issues in designing the curriculum of translator education. It also focuses on the transforming role that SL could play in translator education in particular and higher education in general by exposing students to various cultural and ideological spaces.Keywords: service learning, translator education, teaching, learning, philosophical and ideological issue

    The implementation of a rapid sample preparation method for the detection of SARS-CoV-2 in a diagnostic laboratory in South Africa

    The SARS-CoV-2 pandemic has resulted in shortages of both critical reagents for nucleic acid purification and highly trained staff as supply chains are strained by high demand, public health measures and frequent quarantining and isolation of staff. This created the need for alternate workflows with limited reliance on specialised reagents, equipment and staff. We present here the validation and implementation of such a workflow for preparing samples for downstream SARS-CoV-2 RT-PCR using liquid handling robots. The rapid sample preparation and inactivation technique evaluated, which included sample centrifugation and heating prior to RT-PCR, showed a 97.37% (95% CI: 92.55-99.28%) positive percent agreement and 97.30% (95% CI: 90.67-99.52%) negative percent agreement compared to nucleic acid purification-based testing. A total of 195 samples were tested as part of the validation. This method was subsequently adopted as the primary sample preparation method in the Groote Schuur Hospital Virology Diagnostic Laboratory in Cape Town, South Africa

    A personalised medicine approach for ponatinib-resistant chronic myeloid leukaemia.

    BACKGROUND: Chronic myeloid leukaemia (CML) is characterised by the presence of a fusion driver oncogene, BCR-ABL1, which is a constitutive tyrosine kinase. Tyrosine kinase inhibitors (TKIs) are the central treatment strategy for CML patients and have significantly improved survival rates, but the T315I mutation in the kinase domain of BCR-ABL1 confers resistance to all clinically approved TKIs, except ponatinib. However, compound mutations can mediate resistance even to ponatinib and remain a clinical challenge in CML therapy. Here, we investigated a ponatinib-resistant CML patient through whole-genome sequencing (WGS) to identify the cause of resistance and to find alternative therapeutic targets. PATIENTS AND METHODS: We carried out WGS on a ponatinib-resistant CML patient and demonstrated an effective combination therapy against the primary CML cells derived from this patient in vitro. RESULTS: Our findings demonstrate the emergence of compound mutations in the BCR-ABL1 kinase domain following ponatinib treatment, and chromosomal structural variation data predicted amplification of BCL2. The primary CD34(+) CML cells from this patient showed increased sensitivity to the combination of ponatinib and ABT-263, a BCL2 inhibitor with a negligible effect against the normal CD34(+) cells. CONCLUSION: Our results show the potential of personalised medicine approaches in TKI-resistant CML patients and provide a strategy that could improve clinical outcomes for these patients

    The Effect of Micro-oxygenation on the Phenolic Composition, Quality and Aerobic Wine-Spoilage Microorganisms of Different South African Red Wines

    The effect of micro-oxygenation treatments on the composition of different South African red wines was evaluated.  In some wines, micro-oxygenation led to an increase in colour density, with a corresponding change in colour, due to the formation of polymeric pigments. This increase also led to the red colour becoming more resistant to the bleaching effect of SO2. Micro-oxygenation also seemed to be more effective in increasing the colour densities of younger red wines than the older ones, although total red pigments were found to be highest in an older red wine that had received micro-oxygenation. Differences in the gelatine index were also observed over time. The micro-oxygenated and barrel-matured wines also had lower concentrations of catechin and the procyanidin B1, and showed a corresponding increase in polymeric pigment and polymeric phenols. Micro-oxygenation led to higher acetic acid bacteria counts, although no increase in volatile acidity was observed in these wines. A tasting panel preferredyounger red wines that had undergone micro-oxygenation. In an older red wine prolonged micro-oxygenation led to the wine becoming over-aged, with an increase in the barnyard/medicinal character, which corresponded with an increase in Brettanomyces counts. Micro-oxygenation can be used to increase the quality of young red wines, but further research is needed in this area

    Natural variation in abiotic stress responsive gene expression and local adaptation to climate in Arabidopsis thaliana.

    Gene expression varies widely in natural populations, yet the proximate and ultimate causes of this variation are poorly known. Understanding how variation in gene expression affects abiotic stress tolerance, fitness, and adaptation is central to the field of evolutionary genetics. We tested the hypothesis that genes with natural genetic variation in their expression responses to abiotic stress are likely to be involved in local adaptation to climate in Arabidopsis thaliana. Specifically, we compared genes with consistent expression responses to environmental stress (expression stress responsive, "eSR") to genes with genetically variable responses to abiotic stress (expression genotype-by-environment interaction, "eGEI"). We found that on average genes that exhibited eGEI in response to drought or cold had greater polymorphism in promoter regions and stronger associations with climate than those of eSR genes or genomic controls. We also found that transcription factor binding sites known to respond to environmental stressors, especially abscisic acid responsive elements, showed significantly higher polymorphism in drought eGEI genes in comparison to eSR genes. By contrast, eSR genes tended to exhibit relatively greater pairwise haplotype sharing, lower promoter diversity, and fewer nonsynonymous polymorphisms, suggesting purifying selection or selective sweeps. Our results indicate that cis-regulatory evolution and genetic variation in stress responsive gene expression may be important mechanisms of local adaptation to climatic selective gradients

    A Geostationary Air Quality Monitoring Platform for Africa

    African populations and economies are growing rapidly, but there are few surface observations to monitor the effects on air quality. Trend analysis of the 19-year record of space-based observations from remote sensors onboard low Earth orbit (LEO) satellites shows that anthropogenic pollution is on the rise. Conversely, biomass burning, the largest contributor to surface ozone, is declining. UVvisible instruments on LEO satellites with daily resolution have provided invaluable constraints on sources, evolution, and transport of air pollution in Africa. Sensors in geostationary orbit (GEO) with hourly resolution and a smaller ground pixel than current and past fleets of LEO satellites would further our understanding of air quality in Africa and address the dearth of surface monitoring sites on the continent. Africa has successfully launched Earth observation platforms to retrieve satellite imagery and should expand its remote sensing capabilities by joining the northern hemisphere constellation of GEO Earth observation satellites

    Global mortality from outdoor fine particle pollution generated by fossil fuel combustion: Results from GEOS-Chem

    The burning of fossil fuels – especially coal, petrol, and diesel – is a major source of airborne fine particulate matter (PM2.5), and a key contributor to the global burden of mortality and disease. Previous risk assessments have examined the health response to total PM2.5, not just PM2.5 from fossil fuel combustion, and have used a concentration-response function with limited support from the literature and data at both high and low concentrations. This assessment examines mortality associated with PM2.5 from only fossil fuel combustion, making use of a recent meta-analysis of newer studies with a wider range of exposure. We also estimated mortality due to lower respiratory infections (LRI) among children under the age of five in the Americas and Europe, regions for which we have reliable data on the relative risk of this health outcome from PM2.5 exposure. We used the chemical transport model GEOS-Chem to estimate global exposure levels to fossil-fuel related PM2.5 in 2012. Relative risks of mortality were modeled using functions that link long-term exposure to PM2.5 and mortality, incorporating nonlinearity in the concentration response. We estimate a global total of 10.2 (95% CI: -47.1 to 17.0) million premature deaths annually attributable to the fossil-fuel component of PM2.5. The greatest mortality impact is estimated over regions with substantial fossil fuel related PM2.5, notably China (3.9 million), India (2.5 million) and parts of eastern US, Europe and Southeast Asia. The estimate for China predates substantial decline in fossil fuel emissions and decreases to 2.4 million premature deaths due to 43.7% reduction in fossil fuel PM2.5 from 2012 to 2018 bringing the global total to 8.7 (95% CI: -1.8 to 14.0) million premature deaths. We also estimated excess annual deaths due to LRI in children (0-4 years old) of 876 in North America, 747 in South America, and 605 in Europe. This study demonstrates that the fossil fuel component of PM2.5 contributes a large mortality burden. The steeper concentration-response function slope at lower concentrations leads to larger estimates than previously found in Europe and North America, and the slower drop-off in slope at higher concentrations results in larger estimates in Asia. Fossil fuel combustion can be more readily controlled than other sources and precursors of PM2.5 such as dust or wildfire smoke, so this is a clear message to policymakers and stakeholders to further incentivize a shift to clean sources of energy

    Critical Values for Yen’s Q3: Identification of Local Dependence in the Rasch model using Residual Correlations

    The assumption of local independence is central to all IRT models. Violations can lead to inflated estimates of reliability and problems with construct validity. For the most widely used fit statistic Q3 there are currently no well-documented suggestions of the critical values which should be used to indicate local dependence, and for this reason a variety of arbitrary rules of thumb are used. In this study, we used an empirical data example and Monte Carlo simulation to investigate the different factors that can influence the null distribution of residual correlations, with the objective of proposing guidelines that researchers and practitioners can follow when making decisions about local dependence during scale development and validation. We propose that a parametric bootstrapping procedure should be implemented in each separate situation in order to obtain the critical value of local dependence applicable to the data set, and provide example critical values for a number of data structure situations. The results show that for the Q3 fit statistic no single critical value is appropriate for all situations, as the percentiles in the empirical null distribution are influenced by the number of items, the sample size, and the number of response categories. Furthermore, our results show that local dependence should be considered relative to the average observed residual correlation, rather than to a uniform value, as this results in more stable percentiles for the null distribution of an adjusted fit statistic
