    Overexpression of proinflammatory cytokines in dental pulp tissue and distinct bacterial microbiota in carious teeth of Mexican Individuals

    The prevalence of dental caries in the Mexican adult population aged 20 to 85 years is around 93.3%, and 50% in Mexican children and adolescents. Worldwide, it is the most common non-communicable disease. One of the main etiological factors for dental caries is the oral microbiome and changes in its structure and function, with an expansion of pathogenic bacteria like Streptococcus mutans. The exposed dental pulp tissue triggers an innate immune response to counteract this bacterial invasion. The relation between oral dysbiosis and innate immune responses remains unclear. We aimed to understand the relationship between innate immune response and the oral microbiota by quantifying the expression of Toll-like receptors (TLRs) and proinflammatory markers (cytokines and a chemokine) in dental pulp tissue, either exposed or not to carious dentin, and to correlate this information with the oral microbiome found in healthy teeth and those with moderate caries. RNA was purified from pulp tissue, subjected to RT-qPCR and analysed with the ΔΔCt method. Supragingival dental plaque of non-carious teeth and dentin of carious teeth were subjected to 16S targeted sequencing. Principal coordinate analysis, permutational multivariate ANOVA, and linear discriminant analysis were used to assess differences between non-carious and carious teeth. Correlations were assessed with Spearman´s test and corrected for multiple comparisons using the FDR method. The relative abundance (RA) of Lactobacillus, Actinomyces, Prevotella, and Mitsuokella was increased in carious teeth; while the RA of Haemophilus and Porphyromonas decreased. Olsenella and Parascardovia were only detected in carious teeth. Significant overexpression of interleukin 1 beta (IL1 β), IL6, and CXCL8 was detected in pulp tissue exposed to carious dentin. IL1β correlated positively with TLR2 and Actinomyces; yet negatively with Porphyromonas. These findings suggest that immune response of pulp tissue chronically exposed to cariogenic microbiome is triggered by proinflammatory cytokines IL1β and IL6 and the chemokine CXCL8

    A systematic sampling of the flora was carried out between 1993 and 1999 in several evergreen tropical rainforest stands, “acahuales” (local name for old agricultural fields) of various ages and agricultural fields belonging to the Lacanhá Chansayab community, located at the Lacandonian tropical rainforest. Collected individuals were identified and processed as herbarium specimens, including information on life form, phenology and status of conservation. Four hundred and eighty five vascular plant species were recorded, corresponding to 355 genera from 104 families, which represent 11.23% of all species estimated for the area. Fabaceae, Asteraceae and Rubiaceae were the best represented families, as well as the genera Piper, Eupatorium, Encyclia, Chamaedorea and Solanum.Entre 1993 y 1999 se realizaron herborizaciones sistemáticas en rodales de selva alta perennifolia, acahuales de diferentes edades y milpas pertenecientes a la comunidad de Lacanhá Chansayab, en la Selva Lacandona. Los especímenes recolectados se identificaron y procesaron como ejemplares de herbario, acompañados de información sobre su forma de vida, fenología y estado de conservación. Se registraron 485 especies de plantas vasculares, pertenecientes a 355 géneros de 104 familias, las cuales representan 11.23% del total de especies estimado para la zona. Fabaceae, Asteraceae y Rubiaceae fueron las familias mejor representadas, así como los géneros Piper, Eupatorium, Encyclia, Chamaedorea y Solanum

    The association between the size of adipocyte-derived extracellular vesicles and fasting serum triglyceride-glucose index as proxy measures of adipose tissue insulin resistance in a rat model of early-stage obesity

    IntroductionObesity is a complex disease that predisposes individuals to cardiometabolic alterations. It leads to adipose tissue (AT) dysfunction, which triggers insulin resistance (IR). This suggests that people with obesity develop local IR first and systemic IR later. AT secretes extracellular vesicles, which may be physiopathologically associated with the development of IR. Our aim was to evaluate the effect of a high-fat diet on different parameters of adiposity in a rat model of early-stage obesity and to determine if these parameters are associated with markers of systemic IR. In addition, we sought to explore the relationship between fasting blood measures of IR (Triglycerides/High Density Lipoprotein-cholesterol [TAG/HDL-c] and Triglycerides-Glucose Index [TyG Index]) with the size of adipocyte-derived extracellular vesicles (adEV).MethodsWe used a model of diet-induced obesity for ten weeks in Wistar rats exposed to a high-fat diet. Final weight gain was analyzed by Dual X-ray absorptiometry. Visceral obesity was measured as epididymal AT weight. IR was evaluated with fasting TyG Index & TAG/HDL-c, and adEV were isolated from mature adipocytes on ceiling culture.ResultsIn the high-fat diet group, glucose and triglyceride blood concentrations were higher in comparison to the control group (Log2FC, 0.5 and 1.5 times higher, respectively). The values for TyG Index and adEV size were different between the control animals and the high-fat diet group. Multiple linear regression analyses showed that adEV size can be significantly associated with the TyG Index value, when controlling for epididymal AT weight.ConclusionOur results show that lipid and glucose metabolism, as well as the size and zeta potential of adEV are already altered in early-stage obesity and that adEV size can be significantly associated with liver and systemic IR, estimated by TyG Index

    O objetivo foi apresentar a trajetória da pesquisa qualitativa realizada no Programa Regional de Capacitação em Pesquisa para Enfermeiros no Estudo do Fenômeno das Drogas na América Latina. Foram revisados 35 artigos publicados na Revista Latino-Americana de Enfermagem no período 2004-2008. Os estudos analisados fornecem bases para o desenvolvimento de atividades de promoção da saúde e a concepção de políticas públicas setoriais.The study aimed to present the trajectory of the qualitative research in the Regional Research Training Program for nurses to study the Drug Phenomenon in Latin America. In total, 35 articles published in the Latin American Journal of Nursing between 2004 and 2008 were reviewed.     Cytokine and microbiota profiles in obesity-related hypertension patients

    BackgroundSystemic arterial hypertension is linked to a heightened risk of cardiovascular diseases on a global scale. In Mexico, nearly half of adults in vulnerable conditions experience hypertension. Imbalance in the oral and intestinal microbiota composition has been observed in patients with hypertension, documented by a decrease of bacteria producing short-chain fatty acids, which play a critical role in blood pressure regulation.AimTo examine the cytokines’ profile and assess the characteristics of oral and gut microbiota in obesity-related hypertension in Mexican patients.MethodsA cross-sectional, observational, and analytical study was carried out. Twenty-two patients were categorized by their body mass index (BMI) as overweight and obese, and the diagnosis of primary hypertension. DNA from supragingival dental plaque and feces samples was used to carry out 16S rRNA sequencing. Additionally, 13 cytokines were quantified.ResultsIn the oral microbiota, Kluyvera was found to be significantly enriched in obese compared to overweight patients. Instead, the gut microbiota was dominated by Firmicutes. However, the correlation between certain genera and proinflammatory cytokines was noted.ConclusionThis exploratory study provides insights into the complex relationship between the oral and gut microbiota and their association with systemic inflammation in obesity-related hypertension
