    La extirpación de la grasa de Hoffa y de la porción central del tendón rotuliano: Estudio experimental en la rodilla del conejo

    Se estudia la repercusión de la extirpación parcial de la grada de Hoffa, y de ésta junto con el tercio central del tendón rotuliano en la rodilla de 16 conejos machos adultos, seguidos durante 6 meses. En los 6 casos que completan el estudio con extirpación de la grasa aislada no se detectó sobre el tendón o las superficies articulares cambios macro o microscópicos. Cuando se extirpó además de la grada el tercio central del ligamento rotuliano, se observó adelgazamiento y elongación del tendón rotuliano con erosiones mínimas en la articulación femororotuliana en 4 rodillas examinadas. En ninguno de los dos grupos se observó neoformación de la grasa extirpada.The repercussion on the knee after partial excision of the Hoffa pad, isolated or in combination with the central portion of the patella tendon, was studied in 16 adult male rabbits, with a follow-up of 6 months. In the 6 surviving cases of isolated Hoffa pad excision, no changes were observed on the tendon neither the articular surfaces on macro and microscopic. In the 6 cases of Hoffa pa and central portion of the patella tendon excision, thinning and elongation of the tendon with some erosion of the articular surface of the femoropatellar joint was observed. The fat pad was nor regenerated in neither group

    Sex differences in the inflammatory response of primary astrocytes to lipopolysaccharide

    <p>Abstract</p> <p>Background</p> <p>Numerous neurological and psychiatric disorders show sex differences in incidence, age of onset, symptomatology or outcome. Astrocytes, one of the glial cell types of the brain, show sex differences in number, differentiation and function. Since astrocytes are involved in the response of neural tissue to injury and inflammation, these cells may participate in the generation of sex differences in the response of the brain to pathological insults. To explore this hypothesis, we have examined whether male and female astrocytes show a different response to an inflammatory challenge and whether perinatal testosterone influences this response.</p> <p>Methods</p> <p>Cortical astrocyte cultures were prepared from postnatal day 1 (one day after birth) male or female CD1 mice pups. In addition, cortical astrocyte cultures were also prepared from female pups that were injected at birth with 100 μg of testosterone propionate or vehicle. Cultures were treated for 5 hours with medium containing lipopolysaccharide (LPS) or with control medium. The mRNA levels of IL6, interferon-inducible protein 10 (IP10), TNFα, IL1β, Toll-like receptor 4 (TLR4), steroidogenic acute regulatory protein and translocator protein were assessed by quantitative real-time polymerase chain reaction. Statistical significance was assessed by unpaired <it>t</it>-test or by one-way analysis of variance followed by the Tukey <it>post hoc </it>test.</p> <p>Results</p> <p>The mRNA levels of IL6, TNFα and IL1β after LPS treatment were significantly higher in astrocytes derived from male or androgenized females compared to astrocytes derived from control or vehicle-injected females. In contrast, IP10 mRNA levels after LPS treatment were higher in astrocytes derived from control or vehicle-injected females than in those obtained from males or androgenized females. The different response of male and female astrocytes to LPS was due neither to differences in the basal expression of the inflammatory molecules nor to differences in the expression of the LPS receptor TLR4. In contrast, the different inflammatory response was associated with increased mRNA levels of translocator protein, a key steroidogenic regulator, in female astrocytes that were treated with LPS.</p> <p>Conclusions</p> <p>Male and female cortical astrocytes respond differentially to an inflammatory challenge and this may be predetermined by perinatal testosterone exposure.</p

Esto facilita la condiloplastia, la visión de los puntos de inserción del ligamento cruzado anterior y la realización de intervenciones asociadas con meniscectomía o suturas meniscales. Los resultados clínicos son semejantes a los de otras técnicas a cielo abierto o por atroscopia.The reconstruction of the anterior cruciate ligament by bone-tendon-bone through a transpatellar tendon approach facilitates notchplasty, allows visualization of the insertion points of the ligament, and the performance of other associated surgery like meniscectomy and meniscal sutures.     A Meiotic Checkpoint Alters Repair Partner Bias to Permit Inter-sister Repair of Persistent DSBs

    Accurate meiotic chromosome segregation critically depends on the formation of inter-homolog crossovers initiated by double-strand breaks (DSBs). Inaccuracies in this process can drive aneuploidy and developmental defects, but how meiotic cells are protected from unscheduled DNA breaks remains unexplored. Here we define a checkpoint response to persistent meiotic DSBs in C. elegans that phosphorylates the synaptonemal complex (SC) to switch repair partner from the homolog to the sister chromatid. A key target of this response is the core SC component SYP-1, which is phosphorylated in response to ionizing radiation (IR) or unrepaired meiotic DSBs. Failure to phosphorylate (syp-16A) or dephosphorylate (syp-16D) SYP-1 in response to DNA damage results in chromosome non-dysjunction, hyper-sensitivity to IR-induced DSBs, and synthetic lethality with loss of brc-1BRCA1.     Healthcare Experience and their Relationship with Demographic, Disease and Healthcare-Related Variables: A Cross-Sectional Survey of Patients with Chronic Diseases Using the IEXPAC Scale

Objectives Our objective was to evaluate the healthcare experience of patients with chronic diseases to identify patient-perceived healthcare gaps and to assess the influence of demographic and healthcare-related variables on patient experiences. Methods A cross-sectional survey was delivered to adult patients with chronic diseases: diabetes mellitus (DM), human immunodeficiency virus (HIV) infection, inflammatory bowel disease (IBD) or rheumatic diseases. Patient experiences were assessed with the Instrument for Evaluation of the Experience of Chronic Patients (IEXPAC) questionnaire, with possible scores ranging from 0 (worst) to 10 (best experience). Results Of the 2474 patients handed the survey, 1618 returned it (response rate 65.4%). Patients identified gaps in healthcare related mainly to access to reliable information and services, interaction with other patients and continuity of healthcare after hospital discharge. The mean ± standard deviation (SD) IEXPAC score was 6.0 ± 1.9 and was higher for patients with HIV (6.6 ± 1.7) than for those with rheumatic disease (5.5 ± 2.0), IBD (5.9 ± 2.0) or DM (5.9 ± 1.9) (p < 0.001). In multivariate models, better overall IEXPAC experience was associated with follow-up by the same physician, follow-up by a nurse, receiving healthcare support from others and treatment with subcutaneous or intravenous drugs. The multivariate model that confirmed patients with HIV or DM had better experience than did those with rheumatic diseases. Conclusions Through IEXPAC, patients identified aspects for healthcare quality improvements and circumstances associated with better experience, which may permit greater redirection of healthcare toward patient-centered goals while facilitating improvements in social care and long-term healthcare quality.This project was funded by Merck Sharp and Dohme, Spain

    Protein profiling and classification of commercial quinoa grains by MALDI-TOF-MS and chemometrics

    Quinoa is an Andean grain that is attracting attention worldwide as a high-quality protein-rich food. Nowadays, quinoa foodstuffs are susceptible to adulteration with cheaper cereals. Therefore, there is a need to develop novel methodologies for protein characterization of quinoa. Here, we first developed a matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) method to obtain characteristic mass spectra of protein extracts from 4 different commercial quinoa grains, which group different varieties marketed as black, red, white (from Peru) and royal (white from Bolivia). Then, data preprocessing and peak detection with MALDIquant allowed detecting 47 proteins (being 30 tentatively identified), the intensities of which were considered as fingerprints for multivariate data analysis. Finally, classification by partial least squares discriminant analysis (PLS-DA) was excellent, and 34 out of the 47 proteins were critical for differentiation, confirming the potential of the methodology to obtain a reliable classification of quinoa grains based on protein fingerprinting

    Comparative Baseline Levels of Heavy Metals and Histopathological Notes in Fish From two Coastal Ecosystems of South-West of Spain

    This work reports the comparative study of heavy metal water concentrations from two coastal ecosystems of Cadiz Gulf (SWSpain): Bay of Cadiz and Ria ofHuelva estuary in samples taken on years 1999-2000. Sampling zones showed different heavymetal levels. Statistical analysis of dissolved trace metal concentrations (Cu, Zn, Cd and Pb) showed significant differences between Huelva and Cadiz zones based on the Cu level, with a marked seasonal factor and different metal sources (Industrial, geological, urban,...), observing a higher Cu concentration in Ria de Huelva respect to Bay of Cadiz. On the other hand, to evaluate the pollutant effects on aquatic biota, a histopathological analysis have been conducted in specimens of flat fish, Solea senegalensis from both zones. Lesions weremore severe in fish fromRia ofHuelva, being themost significant alterations: gill hyperplasia, fusion of secondary lamellae, disorganization of the hepatic parenchyma, cellular hypertrophy and vascular congestion in liver and tubular occlusion, loss of interstitial tissue and lipid-like vacuoles in kidney. Obtained results are useful as reference data for future environmental monitoring studies in these zones where to establish a heavy metal concentration temporal trend would be necessary

    A Proteomics Data Mining Strategy for the Identification of Quinoa Grain Proteins with Potential Immunonutritional Bioactivities

    Quinoa proteins are attracting global interest for their wide amino acid profile and as a promising source for the development of biomedical treatments, including those against immunemediated diseases. However, information about the bioactivity of quinoa proteins is scarce. In this study, a quinoa grain proteome map obtained by label-free mass spectrometry-based shotgun proteomics was investigated for the identification of quinoa grain proteins with potential immunonutritional bioactivities, including those related to cancer. After carefully examining the sequence similarities of the 1211 identified quinoa grain proteins against already described bioactive proteins from other plant organisms, 71, 48, and 3 of them were classified as antimicrobial peptides (AMPs), oxidative stress induced peptides (OSIPs), and serine-type protease inhibitors (STPIs), respectively, suggesting their potential as immunomodulatory, anti-inflammatory, and anticancer agents. In addition, data interpretation using Venn diagrams, heat maps, and scatterplots revealed proteome similarities and differences with respect to the AMPs, OSIPs, and STPIs, and the most relevant bioactive proteins in the predominant commercial quinoa grains (i.e., black, red, white (from Peru), and royal (white from Bolivia)). The presented proteomics data mining strategy allows easy screening for potentially relevant quinoa grain proteins and commercial classes for immunonutrition, as a basis for future bioactivity testing

    Classification of quinoa varieties based on protein fingerprinting by capillary electrophoresis with ultraviolet absorption diode array detection and advanced chemometrics

    Quinoa (Chenopodium quinoa Willd.) is an andean grain with exceptional nutritional properties that has been progressively introduced in western countries as a protein-rich super food with a broad amino acid spectrum. Quinoa is consumed as whole grain, but it is also milled to produce high-value flour, which is susceptible to adulteration. Therefore, there is a growing interest in developing novel analytical methods to get further information about quinoa at the chemical level. In this study, we developed a rapid and simple capillary electrophoresis-ultraviolet absorption diode array detection (CE-UV-DAD) method to obtain characteristic multiwavelength electrophoretic profiles of soluble protein extracts from different quinoa grain varieties. Then, advanced chemometric methods (i.e. multivariate curve resolution alternating least squares, MCR-ALS, followed by principal component analysis, PCA, and partial least squares discriminant analysis, PLS-DA) were applied to deconvolute the components present in the electropherograms and classify the quinoa varieties according to their differential protein composition
