    Oportunidades de la tecnología blockchain

    La industria cinematográfica ha comenzado a generar desde hace x tiempo nuevos procesos en la cadena de valor gracias a la irrupción de nuevas tecnologías como el blockchain. La tecnología blockchain procede del ámbito informático y es analizada en este artículo con el objetivo de identificar y reflexionar sobre los nuevos modelos de negocio que conlleva dicha tecnología. En este sentido, se analiza cómo los NFTs (Non Fungible Tokens), soportes únicos digitales regulados precisamente a través de la tecnología blockchain, irrumpen en formatos de creación artística y cinematográfica, logrando así alcanzar nuevos públicos. Esto supone controversias respecto a la propiedad intelectual y su consiguiente gestión. Como resultado de la investigación realizada, con una valoración positiva y una apertura a las nuevas formas de producción, se descubre que la irrupción de nuevos procesos requiere de ciertos tiempos de asimilación, en tanto la transparencia en las gestiones, el grado de conocimiento y manejo de la tecnología y la desconfianza se presentan como aspectos observados entre los usuarios

    A Slow-Digesting Carbohydrate Diet during Rat Pregnancy Protects O spring from Non-Alcoholic Fatty Liver Disease Risk through the Modulation of the Carbohydrate-Response Element and Sterol Regulatory Element Binding Proteins

    High-fat (HF) and rapid digestive (RD) carbohydrate diets during pregnancy promote excessive adipogenesis in o spring. This e ect can be corrected by diets with similar glycemic loads, but low rates of carbohydrate digestion. However, the e ects of these diets on metabolic programming in the livers of o spring, and the liver metabolism contributions to adipogenesis, remain to be addressed. In this study, pregnant insulin-resistant rats were fed high-fat diets with similar glycemic loads but di erent rates of carbohydrate digestion, High Fat-Rapid Digestive (HF–RD) diet or High Fat-Slow Digestive (HF–SD) diet. O spring were fed a standard diet for 10 weeks, and the impact of these diets on the metabolic and signaling pathways involved in liver fat synthesis and storage of o spring were analyzed, including liver lipidomics, glycogen and carbohydrate and lipid metabolism key enzymes and signaling pathways. Livers from animals whose mothers were fed an HF–RD diet showed higher saturated triacylglycerol deposits with lower carbon numbers and double bond contents compared with the HF–SD group. Moreover, the HF–RD group exhibited enhanced glucose transporter 2, pyruvate kinase (PK), acetyl coenzyme A carboxylase (ACC) and fatty acid (FA) synthase expression, and a decrease in pyruvate carboxylase (PyC) expression leading to an altered liver lipid profile. These parameters were normalized in the HF–SD group. The changes in lipogenic enzyme expression were parallel to changes in AktPKB phosphorylation status and nuclear expression in carbohydrate-response element and sterol regulatory element binding proteins. In conclusion, an HF–RD diet during pregnancy translates to changes in liver signaling and metabolic pathways in o spring, enhancing liver lipid storage and synthesis, and therefore non-alcoholic fatty liver disease (NAFLD) risk.     Prevalencia de la esteatosis hepática no alcohólica en población con hipertransaminasemia y grado de adecuación del diagnóstico registrado en atención primaria

    La esteatosis hepática no alcohólica (EHNA) es la causa más frecuente de elevación de transaminasas en el adulto. Determinar la prevalencia de EHNA en pacientes con hipertransaminasemia mantenida, y conocer el grado de adecuación del diagnóstico registrado en Atención Primaria (AP). 1) Estudio descriptivo transversal con muestra aleatoria de pacientes con elevación de la enzima alanina aminotransferasa (ALT) mantenida (ALT > 32 durante ≥ 6 meses), descartadas otras causas de hepatopatía, siguiendo criterios clínicos, analíticos y ecográficos en AP, y 2) descriptivo transversal sobre todos los casos con diagnóstico de EHNA registrado (K76 - CIE10) con análisis de adecuación diagnóstica según criterios estándar. Se analizaron 290 pacientes: 76 fueron diagnosticados de EHNA (26,1%), 44 mujeres (57,9%). El análisis multivariado ajustado por edad y sexo mostró asociación entre EHNA y sexo masculino (OR: 0,5; IC 95%: 0,3-0,9), diabetes mellitus (DM) (OR: 2,42; IC 95%: 1,2-4,9) e hipertensión arterial (HTA) (OR: 3,07; IC 95%: 1,6-5,6). De los 209 con registro diagnóstico de EHNA: 51 (24,4%) cumplían criterios de EHNA. El resto carecían de registros suficientes. Destacan: 53,1% carecían de hipertransaminasemia mantenida; 48% de serologías víricas; 11% de ecografía abdominal compatible y 53,1% de registro del consumo de alcohol. La EHNA severa es frecuente entre los pacientes con hipertransaminasemia mantenida. La DM y la HTA incrementan significativamente el riesgo de EHNA. El diagnóstico de EHNA se registra sin considerar todos los criterios de EHNA y se hace mayoritariamente por ecografía. Se deberían unificar criterios diagnósticos en el registro de la EHNA

    Programming Skeletal Muscle Metabolic Flexibility in Offspring of Male Rats in Response to Maternal Consumption of Slow Digesting Carbohydrates during Pregnancy

    Skeletal muscle plays a relevant role in metabolic flexibility and fuel usage and the associated muscle metabolic inflexibility due to high-fat diets contributing to obesity and type 2 diabetes. Previous research from our group indicates that a high-fat and rapid-digesting carbohydrate diet during pregnancy promotes an excessive adipogenesis and also increases the risk of non-alcoholic fatty liver disease in the offspring. This effect can be counteracted by diets containing carbohydrates with similar glycemic load but lower digestion rates. To address the role of the skeletal muscle in these experimental settings, pregnant rats were fed high-fat diets containing carbohydrates with similar glycemic load but different digestion rates, a high fat containing rapid-digesting carbohydrates diet (HF/RD diet) or a high fat containing slow-digesting carbohydrates diet (HF/SD diet). After weaning, male offspring were fed a standard diet for 3 weeks (weaning) or 10 weeks (adolescence) and the impact of the maternal HF/RD and HF/SD diets on the metabolism, signaling pathways and muscle transcriptome was analyzed. The HF/SD offspring displayed better muscle features compared with the HF/RD group, showing a higher muscle mass, myosin content and differentiation markers that translated into a greater grip strength. In the HF/SD group, metabolic changes such as a higher expression of fatty acids (FAT/CD36) and glucose (GLUT4) transporters, an enhanced glycogen content, as well as changes in regulatory enzymes such as muscle pyruvate kinase and pyruvate dehydrogenase kinase 4 were found, supporting an increased muscle metabolic flexibility and improved muscle performance.     Intake of slow-digesting carbohydrates is related to changes in the microbiome and its functional pathways in growing rats with obesity induced by diet

    Introduction: The main cause of insulin resistance in childhood is obesity, which contributes to future comorbidities as in adults. Although high-calorie diets and lack of exercise contribute to metabolic disease development, food quality rather than the quantity of macronutrients is more important than food density. The purpose of the present study was to examine the effects of changing the quality of carbohydrates from rapidly to slowly digestible carbohydrates on the composition of the gut microbiota and the profiles of the functional pathways in growing rats with obesity due to a high-fat diet (HFD). Methods: During the course of 4 weeks, rats growing on an HFD-containing carbohydrates with different digestive rates were fed either HFD-containing carbohydrates with a rapid digestion rate (OBE group) or HFD-containing carbohydrates with a slow digestion rate (OBE-ISR group). A non-obese group (NOB) was included as a reference, and rats were fed on a rodent standard diet (AIN93G). An analysis of gut microbiota was conducted using 16S rRNA-based metagenomics; a linear mixed-effects model (LMM) was used to determine changes in abundance between baseline and 4 weeks of treatment, and functional pathways were identified. Gut microbiota composition at bacterial diversity and relative abundance, at phylum and genus levels, and functional profiles were analyzed by integrating the Integrated Microbial Genomes (IMG) database. Results: The groups showed comparable gut microbiota at baseline. At the end of the treatment, animals from the ISR group exhibited differences at the phylum levels by decreasing the diversity of Fisher’s index and Firmicutes (newly named as Bacillota), and increasing the Pielou’s evenness and Bacteroidetes (newly named as Bacteroidota); at the genus level by increasing Alistipes, Bifidobacterium, Bacteroides, Butyricimonas, Lachnoclostridium, Flavonifractor, Ruminiclostridium 5, and Faecalibaculum and decreasing Muribaculum, Blautia, and Ruminiclostridium 9. Remarkably, relative abundances of genera Tyzzerella and Angelakisella were higher in the OBE group compared to NOB and OBE-ISR groups. In addition, some microbiota carbohydrate metabolism pathways such as glycolysis, glucuronic acid degradation, pentose phosphate pathway, methanogenesis, and fatty acid biosynthesis exhibited increased activity in the OBE-ISR group after the treatment. Higher levels of acetate and propionate were found in the feces of the ISR group compared with the NOB and OBE groups. Conclusion: The results of this study demonstrate that replacing rapidly digestible carbohydrates with slowly digestible carbohydrates within an HFD improve the composition of the gut microbiota.     Relationship of NDVI and oak (Quercus)pollen including a predictive model in the SW Mediterranean region

    Techniques of remote sensing are being used to develop phenological studies. Our goal is to study the correlation among the Normalized Difference Vegetation Index (NDVI) related with oak trees included in three set data polygons (15, 25 and 50 km to aerobiological sampling point as NDVI-15, 25 and 50), and oak (Quercus) daily average pollen counts from 1994 to 2013. The study was developed in the SW Mediterranean region with continuous pollen recording within the mean pollen season of each studied year. These pollen concentrations were compared with NDVI values in the locations containing the vegetation under a study based on two cartographic sources: the Extremadura Forest Map (MFEx) of Spain and the Fifth National Forest Inventory (IFN5) from Portugal. The importance of this work is to propose the relationship among data related in space and time by Spearman and Granger causality tests. 9 out of 20 studied years have shown significant results with the Granger causality test between NDVI and pollen concentration, and in 12 years, significant values were obtained by Spearman test. The distances of influence on the contribution of Quercus pollen to the sampler showed statistically significant results depending on the year. Moreover, a predictive model by using Artificial Neural Network (ANN) was applied with better results in NDVI25 than for NDVI15 or NDVI50. The addition of NDVI25 with the lag of 5 days and some weather parameters in the model was applied with a RMSE of 4.26 (Spearman coefficient r = 0.77) between observed and predicted values.     Tumor recurrence, metastatic spread and progressive gain of chemo-resistance of advanced cancers are sustained by the presence of cancer stem cells (CSCs) within the tumor. Targeted therapies with the aim to eradicate these cells are thus highly regarded. However, often the use of new anti-cancer therapies is hampered by pharmacokinetic demands. Drug delivery through nanoparticles has great potential to increase efficacy and reduce toxicity and adverse effects. However, its production has to be based on intelligent design. Likewise, we developed polymeric nanoparticles loaded with Zileuton™, a potent inhibitor of cancer stem cells (CSCs), which was chosen based on high throughput screening. Its great potential for CSCs treatment was subsequently demonstrated in in vitro and in in vivo CSC fluorescent models.     Quality More Than Quantity: The Use of Carbohydrates in High-Fat Diets to Tackle Obesity in Growing Rats

Although the energy density of food has been considered a main obesogenic factor, a focus on food quality rather that the quantity of the different macronutrients is needed. Therefore, this study investigates the effects of changing the quality of carbohydrates from rapidly to slowly digestible carbohydrates on metabolic abnormalities and its impact on obesity in growing rats fed a high-fat diet (HFD). Growing rats were fed on HFD containing carbohydrates with different digestion rates: a HFD containing rapid-digesting carbohydrates (OBE group) or slow-digesting carbohydrates (ISR group), for 4 weeks and the effect on the metabolism and signaling pathways were analyzed in different tissues. Animals from OBE group presented an overweight/obese phenotype with a higher body weight gain and greater accumulation of fat in adipose tissue and liver. This state was associated with an increase of HOMA index, serum diacylglycerols and triacylglycerides, insulin, leptin, and pro-inflammatory cytokines. In contrast, the change of carbohydrate profile in the diet to one based on slow digestible prevented the obesity-related adverse effects. In adipose tissue, GLUT4 was increased and UCPs and PPARg were decreased in ISR group respect to OBE group. In liver, GLUT2, FAS, and SRBP1 were lower in ISR group than OBE group. In muscle, an increase of glycogen, GLUT4, AMPK, and Akt were observed in comparison to OBE group. In conclusion, this study demonstrates that the replacement of rapidly digestible carbohydrates for slowly digestible carbohydrates within a highfat diet promoted a protective effect against the development of obesity and its associated comorbidities.Abbott Laboratories S.A

    Saúde ambiental de parques espanhóis: Abordagem ao potencial alergénico dos espaços verdes urbanos

    Los parques urbanos son elementos de la infraestructura verde que deben contribuir a mejorar la calidad de vida y el bienestar ciudadano. En este trabajo se presentan los resultados de la aplicación de un novedoso índice que estima la alergenicidad potencial de las zonas verdes urbanas. Este índice, que contempla parámetros biológicos y biométricos intrínsecos a las especies arbóreas existentes en los parques, genera un resultado cuyo valor está comprendido entre 0 y 1 según el potencial alergénico del parque sea nulo o de riesgo alto para la población. En una primera fase el índice se ha aplicado a parques de diferente tipología, diseño, tamaño, riqueza específica y biodiversidad ubicados en 20 ciudades españolas. Los resultados han mostrado que algunos de los parques estudiados registran un valor de índice superior a 0,30, umbral suficiente para causar síntomas de alergia a la población expuesta, y por tanto, de riesgo moderado o alto. Por el contrario, en la mayoría de los parques se obtuvo un valor inferior a este umbral. También es posible conocer cuáles son las especies que más contribuyen al valor resultante, que son aquellas con estrategia de polinización anemófila, periodos de floración extensos y alta alergenicidad referenciada. Estos requisitos los cumplen todas las especies de las familias Betuláceas, Cupresáceas y Moráceas, y en menor extensión, Oleáceas y Platanáceas. Puede concluirse que el desarrollo de un índice de estimación de alergenicidad de espacios verdes urbanos constituye una herramienta de utilidad para minimizar el impacto de la alergia polínica en la población.Urban parks are green infrastructure elements that should contribute to improving the quality of life and well-being of citizens. In this work there are presented the results of applying a new index to estimate the potential allergenicity of parks located in 20 Spanish cities. This index, which considers intrinsic biological and biometric parameters of existing plant species in parks, allows the allergenic risk thereof to be calculated on a scale ranging from 0 to 1, depending on whether to the park’s allergenicity is zero or has a high risk for the population. The parks selected for this study have different typologies, sizes, species richness and biodiversities, which has yielded highly variable index values. Almost half of the analysed parks have an index value higher than 0.30, a threshold considered having a moderate to high risk, and therefore, enough to cause allergy symptoms in the population. Conversely, most of the parks had an index value below this threshold, so that the risk of suffering allergies is low or very low. The formula also allows the species that most contribute to the resulting value for allergenicity to be known, which are those having an anemophillous strategy of pollination, extended periods of flowering, and a referenced high allergenicity. These requirements are met by all species of the Betulaceae, Cupressaceae and Moraceae families, and to a lesser extent by Oleaceae and Platanaceae. It can be concluded that the development of an index to estimate the allergenicity of urban green spaces constitutes a useful tool to minimize the impact of pollen allergy on the population.Os parques urbanos são elementos da infraestrutura verde que devem contribuir para a melhoria da qualidade de vida dos cidadãos. Neste trabalho apresentam-se os resultados da aplicação de um índice inovador, que permite estimar a alergenicidade potencial dos espaços verdes urbanos. Este índice, que inclui parâmetros biológicos e biométricos, intrínsecos às próprias espécies existentes nos parques, tem como resultado um valor numa escala entre 0 e 1, de acordo com o potencial alergénico do parque, caso este seja, respetivamente e nos seus extremos, nulo ou de máximo risco para as populações. O índice foi aplicado em parques de diferentes tipologias, desenho, tamanho, riqueza específica e biodiversidade, situados em 20 cidades espanholas. Os resultados demonstram que alguns dos parques estudados registam um valor de índice superior a 0,30, limite suficiente para causar sintomas de alergia na população exposta, e por tanto, risco moderado a alto. No entanto, a maioria dos parques apresenta um valor inferior a este limite. Também é possível conhecer quais as espécies que mais contribuem para o valor do índice, que correspondem aquelas com a estratégia de polinização anemófila, períodos de floração extensos e potencial alergénico referenciado. Estes requisitos são aplicáveis a todas as espécies das famílias Betuláceas, Cupressáceas e Moráceas, e em menor medida, Oleáceas e Platanáceas. Pode assim concluir-se que a aplicação de um índice de previsão dos níveis de alergenicidade dos espaços verdes urbanos constitui uma ferramenta útil para minimizar o impacto da alergia polínica sobre a população.peerReviewe
