    Physical and chemical constraints on core - mantle differentiation in terrestrial planets.

    In this study a physical mechanism and geochemical parameters have been examined in high pressure and high temperature experiments in order to place constraints on the conditions and the manner by which core-mantle differentiation occurred on Earth and terrestrial planets. The wetting characteristics of liquid Fe-Si alloys in a matrix of the respective predominating stable silicate mantle mineral (forsterite or silicate perovskite) at pressures of 2 - 5 and 25 GPa and temperatures of 1600 - 2000 ʿC were studied by determining the liquid metal-solid silicate contact angles. The median angle values from texturally-equilibrated samples were found to be independent of pressure, temperature, silicate mineralogy and the Si content in the metal fraction and range between 130ʿ and 140ʿ which is far above the critical wetting boundary of 60ʿ. This shows that within the studied range of conditions dissolved Si does not lower the surface energies between Fe-rich liquids and silicate mantle grains.     Mutation in the V2 vasopressin receptor gene, AVPR2, causes nephrogenic syndrome of inappropriate diuresis

    Nephrogenic syndrome of inappropriate antidiuresis (NSIAD) is a recently discovered rare disease caused by gain-of-function mutations of the V2 vasopressin receptor gene, AVPR2. To date, mutations of Phe229 and Arg137 have been identified as gain-of-function in the V2 vasopressin receptor (V2R). These receptor mutations lead to hyponatremia, which may lead to clinical symptoms in infants. Here we present a newly identified I130N substitution in exon 2 of the V2R gene in a family, causing NSIAD. This I130N mutation resulted in constitutive activity of the V2R with constitutive cyclic adenosine monophosphate (cAMP) generation in HEK293 cells. This basal activity could be blocked by the inverse agonist tolvaptan and arginine-vasopressin stimulation enhanced the cAMP production of I130N-V2R. The mutation causes a biased receptor conformation as the basal cAMP generation activity of I130N does not lead to interaction with β-arrestin. The constitutive activity of the mutant receptor caused constitutive dynamin-dependent and β-arrestin-independent internalization. The inhibition of basal internalization using dominant-negative dynamin resulted in an increased cell surface expression. In contrast to the constitutive internalization, agonist-induced endocytosis was β-arrestin dependent.     POLR3A variants with striatal involvement and extrapyramidal movement disorder

    Biallelic variants in POLR3A cause 4H leukodystrophy, characterized by hypomyelination in combination with cerebellar and pyramidal signs and variable non-neurological manifestations. Basal ganglia are spared in 4H leukodystrophy, and dystonia is not prominent. Three patients with variants in POLR3A, an atypical presentation with dystonia, and MR involvement of putamen and caudate nucleus (striatum) and red nucleus have previously been reported. Genetic, clinical findings and 18 MRI scans from nine patients with homozygous or compound heterozygous POLR3A variants and predominant striatal changes were retrospectively reviewed in order to characterize the striatal variant of POLR3A-associated disease. Prominent extrapyramidal involvement was the predominant clinical sign in all patients. The three youngest children were severely affected with muscle hypotonia, impaired head control, and choreic movements. Presentation of the six older patients was milder. Two brothers diagnosed with juvenile parkinsonism were homozygous for the c.1771-6C > G variant in POLR3A; the other seven either carried c.1771-6C > G (n = 1) or c.1771-7C > G (n = 7) together with another variant (missense, synonymous, or intronic). Striatal T2-hyperintensity and atrophy together with involvement of the superior cerebellar peduncles were characteristic. Additional MRI findings were involvement of dentate nuclei, hila, or peridentate white matter (3, 6, and 4/9), inferior cerebellar peduncles (6/9), red nuclei (2/9), and abnormal myelination of pyramidal and visual tracts (6/9) but no frank hypomyelination.     The FANCM:p.Arg658* truncating variant is associated with risk of triple-negative breast cancer

    Abstract: Breast cancer is a common disease partially caused by genetic risk factors. Germline pathogenic variants in DNA repair genes BRCA1, BRCA2, PALB2, ATM, and CHEK2 are associated with breast cancer risk. FANCM, which encodes for a DNA translocase, has been proposed as a breast cancer predisposition gene, with greater effects for the ER-negative and triple-negative breast cancer (TNBC) subtypes. We tested the three recurrent protein-truncating variants FANCM:p.Arg658*, p.Gln1701*, and p.Arg1931* for association with breast cancer risk in 67,112 cases, 53,766 controls, and 26,662 carriers of pathogenic variants of BRCA1 or BRCA2. These three variants were also studied functionally by measuring survival and chromosome fragility in FANCM−/− patient-derived immortalized fibroblasts treated with diepoxybutane or olaparib. We observed that FANCM:p.Arg658* was associated with increased risk of ER-negative disease and TNBC (OR = 2.44, P = 0.034 and OR = 3.79; P = 0.009, respectively). In a country-restricted analysis, we confirmed the associations detected for FANCM:p.Arg658* and found that also FANCM:p.Arg1931* was associated with ER-negative breast cancer risk (OR = 1.96; P = 0.006). The functional results indicated that all three variants were deleterious affecting cell survival and chromosome stability with FANCM:p.Arg658* causing more severe phenotypes. In conclusion, we confirmed that the two rare FANCM deleterious variants p.Arg658* and p.Arg1931* are risk factors for ER-negative and TNBC subtypes. Overall our data suggest that the effect of truncating variants on breast cancer risk may depend on their position in the gene.     Organic-rich sediments were recognized in early Miocene strata from the Norwegian-Greenland Sea during ODP Leg 151. Three organic-geochemical subunits were distinguished in Hole 909C using detailed organic-geochemical and microscopic analyses. TOC (up to 3 wt%), HI (up to 200 mg HC/g TOC), and d13Corg values (~24–26 per mil) indicate the predominance of terrestrial type III organic matter in Subunit 3 (~18–16.2 Ma). Biomarker and vitrinite reflectance values (R0 ~0.5%) point to rather fresh immature terrestrial organic matter supplied by river discharge from adjacent vegetated coastal areas. Although the sediments of Subunit 3 have a fair (to good) source rock potential they are insufficiently mature to generate significant amounts of oil or gas. To test if the moderate generation potential and source rock quality of Subunit 3 are applicable to the entire area, the computer software OF-Mod was applied. Several modelling runs testing the most probable depositional scenarios result, even under conservative assumptions, in the formation of good to excellent source rocks towards the basin margins
