    Sediment structural properties mediating dominant feeding types patterns in soft-bottom macrobenthos of the Northern Adriatic Sea

    In sediments sampled in Northern Adriatic Sea invertebrate benthic macrofauna was examined in comparison with both substratum bulk parameters (average grain size, sorting, total organic matter content) and attributes of grain size classes (specific organic matter content of dimensional fractions). The aims were: 1) to determine whether variations of community overall parameters (total spatial density, number of taxa) and in numerically dominant feeding types (deposit-feeders and suspension-feeders) patterns were related to changes in sediment general properties; 2) to evaluate whether the observed patterns of abundance for deposit- and filter-feeders had more valid correlates in specific properties of substratum dimensional fractions. Number of taxa and animal spatial density resulted conventionally related to substratum bulk parameters while feeding types patterns in comparison with sediment total organic matter content exibited a reciprocal negative effect; only when both feeding types and sediment structure were resolved to their specific attributes in terms of prevalence of tubicolous taxa among deposit-feeders and organic matter richness of single dimensional fractions, the analysis emphasized patterns unequally affected by specific size fractions abundance and organic matter content: deposit feeders demonstrated to be related to the presence in the substrate of intermediate grain size classes necessary for tube-building. Those intermediate fractions proved to be more adequate descriptors than average grain size or other overall sediment parameters per se of relations actually established between feeding types and the complex nature of bottom sediments. Our results emphasize that for Northern Adriatic Sea benthic communities, sediment organic matter content may represent a factor of minor importance in comparison with other substrate attributes, for which it may be necessary a detailed analysis of sediment structure. Thus, we concluded that the complexity of soft-bottom communities may defy any simple paradigm relating macrobenthic patterns to any single sediment bulk attribute, and we propose a shift in focus towards an higher resolution of both functional groups in macrobenthic associations las already suggested in other investigations) and of substratum structural description

    Mars rover sample return: An exobiology science scenario

    A mission designed to collect and return samples from Mars will provide information regarding its composition, history, and evolution. At the same time, a sample return mission generates a technical challenge. Sophisticated, semi-autonomous, robotic spacecraft systems must be developed in order to carry out complex operations at the surface of a very distant planet. An interdisciplinary effort was conducted to consider how much a Mars mission can be realistically structured to maximize the planetary science return. The focus was to concentrate on a particular set of scientific objectives (exobiology), to determine the instrumentation and analyses required to search for biological signatures, and to evaluate what analyses and decision making can be effectively performed by the rover in order to minimize the overhead of constant communication between Mars and the Earth. Investigations were also begun in the area of machine vision to determine whether layered sedimentary structures can be recognized autonomously, and preliminary results are encouraging

    The Atlantic blue crab Callinectes sapidus spreading in the Tyrrhenian sea: evidence of an established population in the Stagnone di Marsala (Sicily, southern Italy)

    The blue crab Callinectes sapidus is a portunid brachyuran native of the Atlantic coasts of America. In the last decades, the species has greatly expanded its range in invaded areas, currently including the Atlantic coasts of Morocco, the Mediterranean Sea, the Black Sea as well as the eastern Atlantic coasts of Europe from Portugal to France, Belgium, and Germany. In the present study, several specimens of C. sapidus were collected in May, June and July 2021 in the Stagnone di Marsala, a semi-enclosed basin located in the north-western coast of Sicily (southern Italy, Mediterranean Sea) characterized by low hydrodynamics and water exchange with the adjacent open sea and high salinity conditions. Juveniles at diff erent instar stages and ovigerous females were repeatedly captured, suggesting the presence of an established population of the blue crab, despite the peculiar environmental conditions characterizing the basin. These fi ndings are discussed in the context of the current expansion of the species along the African coasts of the Mediterranean Sea

    H3 histamine receptor-mediated activation of protein kinase calpha inhibits the growth of cholangiocarcinoma in vitro and in vivo

    Histamine regulates functions via four receptors (HRH1, HRH2, HRH3, and HRH4). The D-myo-inositol 1,4,5-trisphosphate (IP(3))/Ca(2+)/protein kinase C (PKC)/mitogen-activated protein kinase pathway regulates cholangiocarcinoma growth. We evaluated the role of HRH3 in the regulation of cholangiocarcinoma growth. Expression of HRH3 in intrahepatic and extrahepatic cell lines, normal cholangiocytes, and human tissue arrays was measured. In Mz-ChA-1 cells stimulated with (R)-(alpha)-(-)-methylhistamine dihydrobromide (RAMH), we measured (a) cell growth, (b) IP(3) and cyclic AMP levels, and (c) phosphorylation of PKC and mitogen-activated protein kinase isoforms. Localization of PKC alpha was visualized by immunofluorescence in cell smears and immunoblotting for PKC alpha in cytosol and membrane fractions. Following knockdown of PKC alpha, Mz-ChA-1 cells were stimulated with RAMH before evaluating cell growth and extracellular signal-regulated kinase (ERK)-1/2 phosphorylation. In vivo experiments were done in BALB/c nude mice. Mice were treated with saline or RAMH for 44 days and tumor volume was measured. Tumors were excised and evaluated for proliferation, apoptosis, and expression of PKC alpha, vascular endothelial growth factor (VEGF)-A, VEGF-C, VEGF receptor 2, and VEGF receptor 3. HRH3 expression was found in all cells. RAMH inhibited the growth of cholangiocarcinoma cells. RAMH increased IP(3) levels and PKC alpha phosphorylation and decreased ERK1/2 phosphorylation. RAMH induced a shift in the localization of PKC alpha expression from the cytosolic domain into the membrane region of Mz-ChA-1 cells. Silencing of PKC alpha prevented RAMH inhibition of Mz-ChA-1 cell growth and ablated RAMH effects on ERK1/2 phosphorylation. In vivo, RAMH decreased tumor growth and expression of VEGF and its receptors; PKC alpha expression was increased. RAMH inhibits cholangiocarcinoma growth by PKC alpha-dependent ERK1/2 dephosphorylation. Modulation of PKC alpha by histamine receptors may be important in regulating cholangiocarcinoma growth.     Ontogenetic shift in the trophic role of the invasive killer shrimp Dikerogammarus villosus: a stable isotope study

    none5noThe introduction of the amphipod Dikerogammarus villosus in European fresh waters is to date recognized as a threat to the integrity of invaded communities. Predation by D. villosus on native benthic invertebrates is assumed as the key determinant of its ecological impact, yet available information describe the species as a primary consumer as well as a carnivore depending on local conditions. Here, we assessed the trophic position (TP) of D. villosus in Lake Trasimeno, a recently invaded lentic system in central Italy, using the CN isotopic signatures of individuals captured in winter spanning two orders of magnitude in body size. TP estimations were compared with those characterizing the native amphipod Echinogammarus veneris and other representative invertebrate predators. On average, D. villosus showed a trophic position higher than E. veneris, and comparable with that of odonate nymphs. An in-depth analysis revealed that large-sized individuals had a trophic position of 3.07, higher than odonates and close to that of the hirudinean predator Erpobdella octoculata, while small-sized specimens had a trophic position of 2.57, similar to that of E. veneris (2.41). These findings indicate that size-related ontogenetic shifts in dietary habits may per se vary the nature of the interaction between Dikerogammarus villosus and native invertebrates from competition to predation. Information collated from published isotopic studies corroborated the generality of our results.     Mars Rover Sample Return: A sample collection and analysis strategy for exobiology

    For reasons defined elsewhere it is reasonable to search for biological signatures, both chemical and morphological, of extinct life on Mars. Life on Earth requries the presence of liquid water, therefore, it is important to explore sites on Mars where standing bodies of water may have once existed. Outcrops of layered deposits within the Valles Marineris appear to be ancient lake beds. Because the outcrops are well exposed, relatively shallow core samples would be very informative. The most important biological signature to detect would be organics, microfossils, or larger stromato-like structures, although the presence of cherts, carbonates, clays, and shales would be significant. In spite of the limitations of current robotics and pattern recognition, and the limitations of rover power, computation, Earth communication bandwidth, and time delays, a partial scenario was developed to implement such a scientific investigation. The rover instrumentation and the procedures and decisions and IR spectrometer are described in detail. Preliminary results from a collaborative effort are described, which indicate the rover will be able to autonomously detect stratification, and hence will ease the interpretation burden and lead to greater scientific productivity during the rover's lifetime

    Performance and egg quality of laying hens fed flaxseed: highlights on n-3 fatty acids, cholesterol, lignans and isoflavones

    Flaxseed is a rich source of \u3b1-linolenic acid and phytoestrogens, mainly lignans, whose metabolites (enterodiol and enterolactone) can affect estrogen functions. The present study evaluated the influence of dietary flaxseed supplementation on reproductive performance and egg characteristics (fatty acids, cholesterol, lignans and isoflavones) of 40 Hy-Line hens (20/group) fed for 23 weeks a control diet or the same diet supplemented with 10% of extruded flaxseed. The flaxseed diet had approximately three times the content of lignans (2608.54 ng/g) as the control diet, mainly secoisolariciresinol diglucoside (1534.24 v. 494.72 ng/g). When compared with the control group, hens fed flaxseed showed a similar deposition rate (72.0% v. 73.9%) and egg yield. Furthermore, there was no effect of flaxseed on the main chemical composition of the egg and on its cholesterol content. Estradiol was higher in the plasma of the control group (1419.00 v. 1077.01 pg/ml) probably due to the effect of flaxseed on phytoestrogen metabolites. The plasma lignans were higher in hens fed flaxseed, whereas isoflavones were lower, mainly due to the lower equol value (50.52 v. 71.01 ng/ml). A similar trend was shown in eggs: the flaxseed group had higher level of enterodiol and enterolactone, whereas the equol was lower (198.31 v. 142.02 ng/g yolk). Secoisolariciresinol was the main lignan in eggs of the flaxseed group and its concentration was three times higher then control eggs. Flaxseed also improved the n-3 long-chain polyunsaturated fatty acids of eggs (3.25 v. 0.92 mg/g egg), mainly DHA, however, its oxidative status (thiobarbituric reactive substances) was negatively affected. In conclusion, 10% dietary flaxseed did not affect the productive performance of hens or the yolk cholesterol concentration, whereas the lignans and n-3 polyunsaturated fatty acid content of eggs improved. Further details on the competition between the different dietary phytoestrogens and their metabolites (estrogen, equol, enterodiol and enterolactone) should be investigated

    On the Atlantic blue crab (Callinectes sapidus Rathbun 1896) in southern European coastal waters: Time to turn a threat into a resource?

    [EN] The blue crab Callinectes sapidus is native to the coastal waters of the western Atlantic Ocean, and along the US coasts the species supports an important fishery. The crab was introduced to Europe at the beginning of the 20th century. To date, the species is considered invasive and it has been extensively recorded in southern European waters (SEW), where it is starting to penetrate the shellfish market. Here, an integrated management strategy is proposed for the blue crab in SEW, including the Mediterranean and Black Sea and the eastern Atlantic coasts of the Iberian Peninsula. Taking as introductory examples two case studies represented by the red king crab Paralithodes camtschaticus and the green crab Carcinus maenas, a framework of key issues is reviewed, considering the double nature of the species as invaders and shellfish products. A SWOT analysis is eventually presented for C. sapidus, in order to perform a state-of-the-art synthesis of the proposed scenario, highlighting the potential opportunities as well as the weaknesses related with the limited knowledge of the ecological and economic impact of the species in invaded habitats. The review is concluded by an appraisal of the current trends in global and European crustacean fisheries. The ongoing expansion of C. sapidus might represent a useful management case study, where the need to control an invasive species and mitigate its ecological impact can be harmonized with the opportunity to value it as a fishery resource.Funding from FUR 2013-2014 and 2015-2016 to G.     Tiny percutaneous needle biopsy: An efficient methodfor studying cellular and molecular aspectsof skeletal muscle in humans

    Needle biopsy is widely used to obtain specimens for physiological, anatomical and biochemical studies of skeletal muscle (SM). We optimized a procedure which we termed tiny percutaneous needle biopsy (TPNB), to efficiently gather good numbers of human satellite cells and single dissociated fibers for the functional study of skeletal muscle; these samples permit isolation of high-quality RNA and sufficient amounts of proteins to allow molecular analysis. Moreover, TPNB showed a clear advantage in that the technique was easier than other procedures used on healthy volunteers in human trials. TPNB is a very safe minor surgical procedure. It is less traumatic than needle aspiration biopsy, and significant complications are improbable. TPNB should become established as an important tool in the investigation of SM and may be employed to study various physiological aspects of SM in human subjects. We suggest that TPNB should also be used in the study of muscle diseases and disorders including muscular dystrophy, congenital myopathy, and metabolic defects
