    Protease activity of some mesophilic streptomycetes isolated from Egyptian habitats

    Different streptomycetes (317 isolates) were obtained from several sources and areas in Egypt and were screened for proteolytic activity. Thirty nine of them produced proteases and were subjected to identification. Streptomyces anulatus formed the most abundant portion of the isolates. This species deserves special attention because it is a good candidate for biotechnological applications

    Body Composition Changes after Weight-Loss Interventions among Obese Females: A Comparison of Three Protocols

    AIM: To evaluate body composition changes after use of three different types of obesity management protocols: dietary measures and physical activity; acupuncture or laser acupuncture with healthy diet; aiming at achieving stable weight loss among obese Egyptian females.METHODS:  A randomized longitudinal prospective study included 76 obese adult females; aged 26 up to 55 years. Anthropometric, body composition, ultrasonographic and biochemical assessments were done.RESULTS: The three types of obesity management protocols showed significant improvement in body composition (decrease in fat% and increases in FFM and TBW) and visceral fat by US. However, nutritional intervention showed highly significant improvement in the skin fold thickness at triceps and biceps sites and peripheral adiposity index.  Acupuncture intervention showed highly significant improvement in fasting blood glucose (decreased) and lipid profile (decreased triglycerides, total cholesterol and LDL, and increased HDL). Laser intervention showed highly significant improvement in all the skin fold thickness and some parameters of lipid profile (decreased total cholesterol and LDL). CONCLUSIONS: The three obesity management protocols have significant effect on body composition, but acupuncture has the best effect in improving the lipid profile and fasting blood sugar.     Impact of opioid-free analgesia on pain severity and patient satisfaction after discharge from surgery: multispecialty, prospective cohort study in 25 countries

    Background: Balancing opioid stewardship and the need for adequate analgesia following discharge after surgery is challenging. This study aimed to compare the outcomes for patients discharged with opioid versus opioid-free analgesia after common surgical procedures.Methods: This international, multicentre, prospective cohort study collected data from patients undergoing common acute and elective general surgical, urological, gynaecological, and orthopaedic procedures. The primary outcomes were patient-reported time in severe pain measured on a numerical analogue scale from 0 to 100% and patient-reported satisfaction with pain relief during the first week following discharge. Data were collected by in-hospital chart review and patient telephone interview 1 week after discharge.Results: The study recruited 4273 patients from 144 centres in 25 countries; 1311 patients (30.7%) were prescribed opioid analgesia at discharge. Patients reported being in severe pain for 10 (i.q.r. 1-30)% of the first week after discharge and rated satisfaction with analgesia as 90 (i.q.r. 80-100) of 100. After adjustment for confounders, opioid analgesia on discharge was independently associated with increased pain severity (risk ratio 1.52, 95% c.i. 1.31 to 1.76; P < 0.001) and re-presentation to healthcare providers owing to side-effects of medication (OR 2.38, 95% c.i. 1.36 to 4.17; P = 0.004), but not with satisfaction with analgesia (beta coefficient 0.92, 95% c.i. -1.52 to 3.36; P = 0.468) compared with opioid-free analgesia. Although opioid prescribing varied greatly between high-income and low- and middle-income countries, patient-reported outcomes did not.Conclusion: Opioid analgesia prescription on surgical discharge is associated with a higher risk of re-presentation owing to side-effects of medication and increased patient-reported pain, but not with changes in patient-reported satisfaction. Opioid-free discharge analgesia should be adopted routinely

    Production and characterisation of exopolysaccharide from Streptomyces carpaticus isolated from marine sediments in Egypt and its effect on breast and colon cell lines

    Twenty streptomycete strains were isolated from marine sediment samples collected from Nabq area, Sharm El-Sheikh, Red Sea Coast, Egypt. Four of them produce exopolysaccharides (EPS) showing marked in vitro antitumor activities. Morphological and cultural characteristics of the most significant strain (No. 3) were shown. Moreover, the sequence of this strain showed similarity with Streptomyces carpaticus. The results reveal that EPS produced by Streptomyces carpaticus No. 3 had high cytotoxicity reaching 51.7% and 59.1% against human tumor cells of breast and colon lines respectively. A chemical analysis of EPS indicated that the composing monosaccharides were galactouronic acid, glucose, xylose, galactose, mannose, and fructose with relative ratio of 3:1:1:2:2:1 respectively, with an average molecular weight (Mw) 1.180 × 105 g/mol and of a number average molecular weight (Mn) 1.052 × 105 g/mol.     In order to produce neutral protease from Streptomyces microflavus, it was cultivated in basal medium containing soluble starch, potassium nitrate, dipotassium hydrogen phosphate, magnesium sulphate, sodium chloride, calcium carbonate and ferrous sulphate. Protease production increased with decreasing the ratio of culture broth to vessel volume 1:5 and shaking at 150 rpm. Protease production was low when pH was 9. The productivity of protease decreased sharply when the incubation temperature increased from 30 to 450C. The maximum yield of protease was obtained at the third day of incubation. Soluble starch and yeast extract were the best carbon and nitrogen sources, respectively. Supplementation with calcium carbonate enhanced protease production. In addition, the dipotassium hydrogen phosphate was the best phosphorous source. Para chloromercuribenzoic acid and phenyl methylsulfonylfloride had significant inhibitory effect on protease production

    Abstract Background L-arginase, is a powerful anticancer that hydrolyzes L-arginine to L-ornithine and urea. This enzyme is widely distributed and expressed in organisms like plants, fungi, however very scarce from bacteria. Our study is based on isolating, purifying, and screening the marine bacteria that can produce arginase. Results The highest arginase producing bacteria will be identified by using microbiological and molecular biology methods as Bacillus licheniformis OF2. Characterization of arginase is the objective of this study. The activity of enzyme was screened, and estimated beside partial sequencing of arginase gene was analyzed. In silico homology modeling was applied to generate the protein's 3D structure, and COACH and COFACTOR were applied to determine the protein's binding sites and biological annotations based on the I-TASSER structure prediction. The purified enzyme was undergone an in vitro anticancer test. Conclusions L-arginase demonstrated more strong anti-cancer cells with an IC50 of 21.4 ug/ml in a dose-dependent manner. L-arginase underwent another investigation for its impact on the caspase 7 and BCL2 family of proteins (BCL2, Bax, and Bax/Bcl2). Through cell arrest in the G1/S phase, L-arginase signals the apoptotic cascade, which is supported by a flow cytometry analysis of cell cycle phases

    Parametricism vs Materialism : Evolution of digital technologies for development

    We build on previous technological developments in CAAD by looking into parametric design exploration and the development of the concept of parametricism. We use the phenomenological backdrop to account for our physical experiences and encounters as well as our mental ones; both evident in the link between parametric design as a process and an outcome. In specific, we previously examined two particular metaphors. The first metaphor addressed aspects of virtual environments that resemble our physical world; In other words, computer model as physical model and digital world as material world. In this volume, we extend the exploration into aspects of virtual environments and their resemblance to physical environments by looking at ‘performance’ aspects: the way in which environments are sensed, measured, tracked and visualised. Moreover, we reflect on matters and materiality in both virtual and physical space philosophically, theoretically, practically and reflectively. The second metaphor looked into the modes and means of interaction between our bodies and such virtual environment. Here we extend the investigation to look at the ways in which measures of environmental performance influence human interaction in real environments. The exploration takes us further to look into the area of design fabrication of the built environment, and methods in which developed processes meet environmental performance requirements, and the innovative outcomes that lead to disruptive technologies getting introduced into design and we revisit parametric design under this focus area
