    Formulation and Evaluation of Mucoadhesive Buccal Tablet of Repaglinide

    The aim of present investigation was formulation and evaluation of mucoadhesive buccal tablet of Repaglinide to study the effect of different polymers on release profile of drug for prolonged release. In this study mucoadhesive buccal tablet were prepared by direct compression method. Various rheological characteristics of the powder bed like bulk density, compressibility index, and angle of repose were evaluated and studied. Mucoadhesive buccal tablets were compressed on a 8 station mini press using 10 mm flat faced punches and were all assessed for weight variation, hardness, thickness, percent swelling index, mucoadhesive strength and in vitro release of the drug by using USP TDT 08L dissolution testing apparatus method II using a paddle at 50 rpm. Data was optimized by using 32 full factorial design by using software named as design expert and with the help of kinetic study. The stability studies showed that there is no decrease in the drug content of all formulations for the period of 2 months. Keywords: Buccal tablet, Repaglinide, HPMC K100M, Xanthan gum

    Formulation and evaluation of sustained release matrix tablet of captopril

    The objective of the present study was to study the effect of polymers on sustained release of Captopril from tablets. Compatibility was studied by Fourier transform infrared spectroscopy and DSC. The tablets were prepared by direct compression technique using Xanthan gum and Ethyl Cellulose. The prepared matrix tablets were evaluated for their physicochemical parameters such as weight variation, hardness, friability, content uniformity and in-vitro dissolution. Pre and post compression parameters were evaluated and all the parameters were found within the limit. The drug release data were subjected to different models in order to evaluate release kinetics and mechanism of drug release. Formulation F4 was selected as best formulation.     Antioxidant and anti-inflammatory activity of extract obtained from <i style="">Aspergillus candidus </i>MTCC 2202 broth filtrate

    468-473Antioxidant potential of Aspergillus candidus MTCC 2202 broth filtrate extract was studied using different antioxidant models, whereas anti-inflammatory potential was studied using carrageenan-induced rat paw oedema model. The ethyl acetate extract at 1000 µg/ml showed maximum scavenging activity of the stable radical 1,1-diphenyl,2-picryl hydrazyl upto 96.65% (IC50=430.36 µg/ml) and scavenging of the radical cation, 2,2-azinobis-(3-ethylbenzothiazoline-6-sulphonate) upto 92.25% (IC50=606.29 µg/ml) at the same concentration. The extract had good reducing power, however showed moderate inhibition for conjugated dienes and thiobarbituric acid reactive acid substances (59.56 and 51.45%). The total phenolic content of various extracts of A. candidus broth filtrate was measured and a correlation between radical scavenging activities of extracts with total phenolic content was observed. The ethyl acetate extract (125 mg/kg ip) showed significant anti-inflammatory activity in carrageenan-induced rat paw oedema model. The exhibited antioxidant activity of ethyl acetate extract of A. candidus broth filtrate was comparable with BHA and ascorbic acid, while anti-inflammatory activity was comparable with standard diclofenac sodium

    Using an HIV-1 Reverse Transcriptase (RT)-associated RNase H inhibition assay as lead, bioguided fractionation of the dichloromethane extract of the Ocimum sanctum leaves led to the isolation of five triterpenes (1–5) along with three 3-methoxy-4-hydroxy phenyl derivatives (6–8). The structure of this isolates were determined by 1D and 2D NMR experiments as well as ESI-MS. Tetradecyl ferulate (8) showed an interesting RNase H IC50 value of 12.4 μM and due to the synthetic accessibility of this secondary metabolite, a structure-activity relationship study was carried out. A series of esters and amides of ferulic and caffeic acids were synthesized and, among all, the most active was N-oleylcaffeamide displaying a strong inhibitory activity towards both RT-associated functions, ribonuclease H and DNA polymerase. Molecular modeling studies together with Yonetani-Theorell analysis, demonstrated that N-oleylcaffeamide is able to bind both two allosteric site located one close to the NNRTI binding pocket and the other close to RNase H catalytic site
