    An Unsuccessful Urban Deep Excavation in Soft Soils

    This case history presents, from a geotechnical engineer\u27s viewpoint, various technical, budgetary, coordination, staffing and hidden agenda issues that contributed to poor performance of a shored deep excavation 10 soft soils. The anticipated and actual construction procedures are discussed and compared. In addition, approaches to stabilize significant movements of the shoring, and the methodology needed to justify a satisfactory future performance of a pile foundation system that was speculated to be damaged are discussed. By highlighting these issues, it is hoped geotechnical engineers will anticipate and better deal with the issues under their influence, understand issues over which they may not have control, and appreciate the need for mutual understanding and cooperation by members of the design team

    Unconventional Drilled Pier Underpinning

    An “unconventional” earth retaining system consisting of drilled underpinning piers, was used successfully to underpin a four- story building in Redwood City, California. Considerations to other conventional underpinning systems and their feasibility at the subject site are also examined. An in-depth discussion of the unconventional underpinning system selected for the project is presented with details of the construction sequence and difficulties encountered. Post-construction performance of the underpinning system and the relative costs of the “unconventional” underpinning system are also presented

    Integrative Solutions in Online Crowdsourcing Innovation Challenges

    Online crowdsourcing challenges are widely used for problem-solving and innovation. Existing theory has characterized such challenges as tools for tapping distant knowledge. By building on information processing theory we move beyond this characterization and present a perspective that describes innovation challenges as virtual places in which ideas are not simply submitted or commented upon but knowledge is integrated. This perspective shifts the role of crowdsourcing challenges from mere tools for gathering ideas to representing the locus of innovation. Our perspective suggests that three types of knowledge affect the quality of integrative solutions: elementary ideas, facts, and analogical examples. Based on a large dataset, we find that elementary ideas and analogical examples are related to increased solution quality, while facts are related to decreased solution quality. We expand the research on online crowdsourcing innovation challenges to include how crowd participants influence the quality of solutions through the content of their postings

    Settlement of Shallow Foundations Constructed over Reinforced Soil: Design Estimates vs. Measurements

    Faced with difficult soil conditions for the support of two 6-story office towers in Dublin, CA and of a 6-story parking garage in Sacramento, CA, engineers recommended the use of Geopier Rammed Aggregate Piers to reinforce the soil for the support of high bearing capacity spread footings instead of deep foundations. Foundation selection for both sites was influenced by long-term settlement performance, schedule and cost savings, and seismic uplift resistance. Rammed aggregate piers were installed to strengthen upper weak and compressible soil layers resulting in a substantially stiffer soil layer on which shallow, high bearing capacity spread footings were constructed. Several rammed aggregate piers were also installed with steel anchors to resist seismically induced overturning forces. Design parameter values were confirmed by full scale aggregate pier modulus tests and uplift tests and a 24 hour load test at the Dublin site. Total settlements were estimated to be less than 1œ inches for the Sacramento site and less than 1 inch for the Dublin site. Measured settlements are less than 1 inches total for both projects with differential settlements less than œ inches, confirming the design approaches and soil properties used for design. Site selection, rammed aggregate pier design methodology, modulus and uplift load test results, and measured settlement performance are presented for two projects in California

    As BPM (Business Process Management) is gaining acceptance among academics and practitioners, questions remain about its role in Information Systems (IS) teaching and research. This panel continues and expands the dialogue within the IS community on how BPM is and should be integrated within IS. Initially proposed by Donald Chand, Alina Chircu and M. Lynne Markus (Information and Process Management, Bentley University, USA), the panel is chaired by Alina Chircu and features Varun Grover (Clemson University, USA), Ann Majchrzak (University of Southern California, USA), and Michael Rosemann (Queensland University of Technology, Australia). The panelists will address the current status, the IS field capabilities, and the future opportunities related to the integration of BPM in IS research and teaching. Each panelist will be able to interpret the definition of BPM in their own way and will be taking pro and con positions for the purposes of stimulating the debate

    Towards Routinely Using Virtual Reality in Higher Education

    Virtual reality promises to be a tool that can improve higher education. Immersive virtual environments offer the chance to enrich courses with experiential learning experiences. The technological possibilities evolve rapidly and more and more researchers report on adopting virtual reality for learning – albeit such work often has a more or less experimental character. However, the base of knowledge on using virtual reality in higher education is growing; educators who want to employ virtual reality to amend courses, to extend the curriculum with experiential learning, or who want to offer new content enabled through virtual reality, find increasingly rich advice. With this article, we contribute to this advice by providing insights from three research cases. Although these were experimental, their embedding into a larger project enables us to propose recommendations for educators. The ultimate aim of our work is the routine use of virtual reality in higher education

    Multicentre Performance Evaluation of the Elecsys Anti-SARS-CoV-2 Immunoassay as an Aid in Determining Previous Exposure to SARS-CoV-2

    Introduction We performed a multicentre evaluation of the Elecsys¼ Anti-SARS-CoV-2 immunoassay (Roche Diagnostics), an assay utilising a recombinant protein representing the nucleocapsid (N) antigen, for the in vitro qualitative detection of antibodies to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Methods Specificity was evaluated using serum/plasma samples from blood donors and routine diagnostic specimens collected before September 2019 (i.e., presumed negative for SARS-CoV-2-specific antibodies); sensitivity was evaluated using samples from patients with polymerase chain reaction (PCR)-confirmed SARS-CoV-2 infection. Point estimates and 95% confidence intervals (CIs) were calculated. Method comparison was performed versus commercially available assays. Results Overall specificity for the Elecsys Anti-SARS-CoV-2 immunoassay (n = 9575) was 99.85% (95% CI 99.75–99.92): blood donors (n = 6714; 99.82%), routine diagnostic specimens (n = 2861; 99.93%), pregnant women (n = 2256; 99.91%), paediatric samples (n = 205; 100.00%). The Elecsys Anti-SARS-CoV-2 immunoassay demonstrated significantly higher specificity versus LIAISON SARS-CoV-2 S1/S2 IgG (99.71% vs. 98.48%), EUROIMMUN Anti-SARS-CoV-2 IgG (100.00% vs. 94.87%), ADVIA Centaur SARS-CoV-2 Total (100.00% vs. 87.32%) and iFlash SARS-CoV-2 IgM (100.00% vs. 99.58%) assays, and comparable specificity to ARCHITECT SARS-CoV-2 IgG (99.75% vs. 99.65%) and iFlash SARS-CoV-2 IgG (100.00% vs. 100.00%) assays. Overall sensitivity for Elecsys Anti-SARS-CoV-2 immunoassay samples drawn at least 14 days post-PCR confirmation (n = 219) was 93.61% (95% CI 89.51–96.46). No statistically significant differences in sensitivity were observed between the Elecsys Anti-SARS-CoV-2 immunoassay versus EUROIMMUN Anti-SARS-CoV-2 IgG (90.32% vs. 95.16%) and ARCHITECT SARS-CoV-2 IgG (84.81% vs. 87.34%) assays. The Elecsys Anti-SARS-CoV-2 immunoassay showed significantly lower sensitivity versus ADVIA Centaur SARS-CoV-2 Total (85.19% vs. 95.06%) and iFlash SARS-CoV-2 IgG (86.25% vs. 93.75%) assays, but significantly higher sensitivity versus the iFlash SARS-CoV-2 IgM assay (86.25% vs. 33.75%). Conclusion The Elecsys Anti-SARS-CoV-2 immunoassay demonstrated very high specificity and high sensitivity in samples collected at least 14 days post-PCR confirmation of SARS-CoV-2 infection, supporting its use to aid in determination of previous exposure to SARS-CoV-2

    A systems approach for discovering linoleic acid derivatives that potentially mediate pain and itch

    Chronic pain and itch are common hypersensitivity syndromes that are affected by endogenous mediators. We applied a systems-based, translational approach to predict, discover, and characterize mediators of pain and itch that are regulated by diet and inflammation. Profiling of tissue-specific precursor abundance and biosynthetic gene expression predicted that inflamed skin would be abundant in four previously unknown 11-hydroxy-epoxy- or 11-keto-epoxy-octadecenoate linoleic acid derivatives and four previously identified 9- or 13-hydroxy-epoxy- or 9- or 13-keto-epoxy-octadecenoate linoleic acid derivatives. All of these mediators were confirmed to be abundant in rat and human skin by mass spectrometry. However, only the two 11-hydroxy-epoxy-octadecenoates sensitized rat dorsal root ganglion neurons to release more calcitonin gene-related peptide (CGRP), which is involved in pain transmission, in response to low pH (which mimics an inflammatory state) or capsaicin (which activates ion channels involved in nociception). The two 11-hydroxy-epoxy-octadecenoates share a 3-hydroxy-Z-pentenyl-E-epoxide moiety, thus suggesting that this substructure could mediate nociceptor sensitization. In rats, intradermal hind paw injection of 11-hydroxy-12,13-trans-epoxy-(9Z)-octadecenoate elicited C-fiber-mediated sensitivity to thermal pain. In a randomized trial testing adjunctive strategies to manage refractory chronic headaches, reducing the dietary intake of linoleic acid was associated with decreases in plasma 11-hydroxy-12,13-trans-epoxy-(9Z)-octadecenoate, which correlated with clinical pain reduction. Human psoriatic skin had 30-fold higher 9-keto-12,13-trans-epoxy-(10E)-octadecenoate compared to control skin, and intradermal injection of this compound induced itch-related scratching behavior in mice. Collectively, these findings define a family of endogenous mediators with potential roles in pain and itch

    Use of Acceleration and Acoustics to Classify Behavior, Generate Time Budgets, and Evaluate Responses to Moonlight in Free-Ranging Snowshoe Hares

    Technological miniaturization is driving a biologging revolution that is producing detailed and sophisticated techniques of assessing individual behavioral responses to environmental conditions. Among the many advancements this revolution has brought is an ability to record behavioral responses of nocturnal, free-ranging species. Here, we combine captive validations of acceleration signatures with acoustic recordings from free-ranging individuals to classify behavior at two resolutions. Combining these classifications with ~2 month-long recordings, we describe winter time budgets, and responses of free-ranging snowshoe hares to changing moonlight. We successfully classified snowshoe hare behavior into four categories (not moving, foraging, hopping, and sprinting) using low frequency accelerometry, with an overall model accuracy of 88%, and acoustic recordings to three categories (silence, hopping, and chewing) with an accuracy of 94%. Broad-scale accelerometer-classified categories were composed of multiple fine-scale behavioral states with the composition varying between individuals and across the day. Time budgets revealed that hares spent ~50% of their time foraging and ~50% not moving, with most foraging and feeding occurring at night. We found that hares adjusted timing of activity in response to moon phase, with a 6% reduction in foraging and 30% reduction in traveling during the night when the moon was full. Hares compensated for this lost foraging time by extending foraging into the morning hours of the following day. Using two biologging technologies to identify behavior, we demonstrate the possibility of combining multiple devices when documenting behavior of cryptic species

    Identification of human CD4+ T cell populations with distinct antitumor activity

    How naturally arising human CD4+ T helper subsets affect cancer immunotherapy is unclear. We reported that human CD4+CD26high T cells elicit potent immunity against solid tumors. As CD26high T cells are often categorized as TH17 cells for their IL-17 production and high CD26 expression, we posited these populations would have similar molecular properties. Here, we reveal that CD26high T cells are epigenetically and transcriptionally distinct from TH17 cells. Of clinical importance, CD26high and TH17 cells engineered with a chimeric antigen receptor (CAR) regressed large human tumors to a greater extent than enriched TH1 or TH2 cells. Only human CD26high T cells mediated curative responses, even when redirected with a suboptimal CAR and without aid by CD8+ CAR T cells. CD26high T cells cosecreted effector cytokines, produced cytotoxic molecules, and persisted long term. Collectively, our work underscores the promise of CD4+ T cell populations to improve durability of solid tumor therapies
