    Spatio-temporal changes of Munida Rutllanti Zariquiey-Alvarez, 1952 (Decapoda: Galatheidae) in the North-Western Ionian Sea (Central Mediterranean)

    The spatio-temporal pattern of Munida rutllanti distribution in the north-western Ionian Sea has been studied. Data were collected during 14 experimental trawl surveys conducted from 1997 to 2010 as part of the international MEDITS project. The hauls were carried out during day-light hours between depths of 10 and 800 m in the spring season. A progressive increase in the abundance index (N/km2) of M. rutllanti was observed from 2000 to 2008, then a sharp decrease was shown in the last two years. The greatest and lowest abundance indices were observed in the Apulian and central Calabrian sub-areas, respectively. The species was collected between 107 and 795 m in depth, with a significant increase and decrease over time in the maximum and minimum depth of finding, respectively. A highly significant increase over time in the mean carapace length was also observed in the whole study area. The widespread occurrence and increasing abundance of this species in the Ionian Sea could be related to the increase in temperature and the variation in hydrographic conditions which occurred in the Ionian basin during the EMT-BiOS phenomenon

    Disorder chaos in spin glasses

    We investigate numerically disorder chaos in spin glasses, i.e. the sensitivity of the ground state to small changes of the random couplings. Our study focuses on the Edwards-Anderson model in d=1,2,3 and in mean-field. We find that in all cases, simple scaling laws, involving the size of the system and the strength of the perturbation, are obeyed. We characterize in detail the distribution of overlap between ground states and the geometrical properties of flipped spin clusters in both the weak and strong chaos regime.     Rock magnetism and palaeomagnetism of the Montalbano Jonico section (Italy): evidence for late diagenetic growth of greigite and implications for magnetostratigraphy

    The Montalbano Jonico (MJ) section, cropping out in Southern Italy, represents a potential candidate to define the Lower/Middle Pleistocene boundary and it has been proposed as a suitable Global Stratotype Section and Point (GSSP) of the Ionian Stage (Middle Pleistocene). The MJ section is the only continuous benthic and planktonic δ18O on-land reference in the Mediterranean area for the Mid-Pleistocene transition, spanning an interval between about 1240 and 645 ka. Combined biostratigraphy and sapropel chronology, tephra stratigraphy and complete high-resolution benthic and planktonic foraminiferal stable oxygen isotope records already provide a firm chronostratigraphic framework for the MJ section. However, magnetostratigraphy was still required to precisely locate the Brunhes-Matuyama transition and to mark the GSSP for the Ionian stage. We carried out a palaeomagnetic study of a subsection (Ideale section) of the MJ composite section, sampling 61 oriented cores from 56 stratigraphic levels spread over a ca. 80-m-thick stratigraphic interval that correlates to the oxygen isotopic stage 19 and should therefore include the Brunhes-Matuyama reversal. The palaeomagnetic data indicate a stable and almost single-component natural remanent magnetization (NRM). A characteristic remanent magnetization (ChRM) was clearly identified by stepwise demagnetization of the NRM. The ChRM declination values vary around 0◦ and the ChRM inclination around the expected value (59◦) for a geocentric axial dipole field at the sampling locality. This result indicates that the section has been remagnetized during the Brunhes Chron. A preliminary study of 27 additional not azimuthally oriented hand samples, collected at various levels from other parts of the MJ composite section, indicates that all the samples are of normal polarity and demonstrates that the remagnetization is widespread across the whole exposed stratigraphic sequence. A series of specific rock magnetic techniques were then applied to investigate the nature of the main magnetic carrier in the study sediments, and they suggest that the main magnetic mineral in the MJ section is the iron sulphide greigite (Fe3S4). Scanning electron microscope observations and elemental microanalysis reveal that greigite occurs both as individual euhedral crystals and in iron sulphides aggregates filling voids in the clay matrix. Therefore, we infer that the remagnetization of the section is due to the late-diagenetic growth of greigite under reducing conditions, most likely resulting in the almost complete dissolution of the original magnetic minerals. Iron sulphide formation in the MJ section can be linked to migration of mineralized fluids. Our inferred timing of the remagnetization associated with greigite growth represents the longest remanence acquisition delay documented in greigite-bearing clays of the Italian peninsula so far

    Exploring differential transcriptome between jejunal and cecal tissue of broiler chickens

    Simple Summary: Intestinal main functions and different gut tract peculiarities in poultry are known. However, a wider view at the molecular level in terms of functional genes may contribute to deepening knowledge on less considered biological aspects, with possible differentiation in gene expression and functionality between gut tracts. This experiment aimed to extend the exploration of chicken gut functional aspects by scouting differential gene expression in the jejunum and cecum, which may help in the detection of new interesting functions from a biological point of view. The work identified key aspects linked to gut biological processes that may be worthy of further investigations in experimental studies considering factors which may specifically address peculiarities of the different chicken gut tracts at the molecular level. Abstract: The study proposed an exploratory functional analysis on differential gene expression of the jejunum and of cecum in chickens. For this study, 150 Ross 308 male chickens were randomly allotted in six pens (25 birds/pen) and fed the same commercial diet. From 19 birds of 42 days of age, jejunum and cecum mucosae were collected for RNA extraction for transcriptome microarray analysis. Differentially expressed genes (DEGs) submitted to DAVID (Database for Annotation, Visualization, and Integrated Discovery) and Gene Set Enrichment Analysis (GSEA) software evidenced enriched gene clusters for biological functions differentiated in the tissues. DAVID analysis in the jejunum showed enriched annotations for cell membrane integral components, PPAR (peroxisome proliferator-activated receptor) signaling pathway, and peroxisome and lipid metabolism, and showed DEGs for gluconeogenesis, not previously reported in chicken jejunum. The cecum showed enriched annotations for disulfide bond category, cysteine and methionine metabolism, glycoprotein category, cell cycle, and extracellular matrix (ECM). GSEA analysis in the jejunum showed peroxisome and PPAR signaling pathway-related gene sets, as found with DAVID, and gene sets for immune regulation, tryptophan and histidine metabolism, and renin\u2013angiotensin system, like in mammals. The cecum showed cell cycle and regulation processes, as well as ECM receptor interaction and focal adhesion-related gene sets. Typical intestinal functions specific for the gut site and interesting functional genes groups emerged, revealing tissue-related key aspects which future studies might take advantage of

    Weak first order transition in the three-dimensional site-diluted Ising antiferromagnet in a magnetic field

    We perform intensive numerical simulations of the three-dimensional site-diluted Ising antiferromagnet in a magnetic field at high values of the external applied field. Even if data for small lattice sizes are compatible with second-order criticality, the critical behavior of the system shows a crossover from second-order to first-order behavior for large system sizes, where signals of latent heat appear. We propose "apparent" critical exponents for the dependence of some observables with the lattice size for a generic (disordered) first-order phase transition.Comment: Final version, accepted for publicatio

    Physical assessment of toxicology at nanoscale: nano dose-metrics and toxicity factor

    In this work, we propose a systematic and reproducible evaluation of nanoparticles (NPs) toxicology in living systems, based on a physical assessment and quantification of the toxic effects of NPs by the experimental determination of the key parameter affecting the toxicity outcome (i.e., the number of NPs) and of the NPs "toxicity factor". Such a strategy was applied to a well determined scenario, i.e., the ingestion of citrate-capped gold NPs (AuNPs) of different sizes by the model system Drosophila melanogaster. Using these AuNPs as a reference toxicity standard, we were able to define different regions in the multiparametric space of toxicity, enabling the classification of the toxic levels of other nanomaterials, such as quantum dots and pegylated AuNPs. This approach may pave the way to a systematic classification of nanomaterials, leading to important developments in risk assessment and regulatory approval, as well as in a wide range of nanomedicine applications

    PIP degron proteins, substrates of CRL4Cdt2, and not PIP boxes, interfere with DNA polymerase η and κ focus formation on UV damage

    International audienceProliferating cell nuclear antigen (PCNA) is a well-known scaffold for many DNA replication and repair proteins, but how the switch between partners is regulated is currently unclear. Interaction with PCNA occurs via a domain known as a PCNA-Interacting Protein motif (PIP box). More recently, an additional specialized PIP box has been described, the « PIP degron », that targets PCNA-interacting proteins for proteasomal degradation via the E3 ubiquitin ligase CRL4(Cdt2). Here we provide evidence that CRL4(Cdt2)-dependent degradation of PIP degron proteins plays a role in the switch of PCNA partners during the DNA damage response by facilitating accumulation of translesion synthesis DNA polymerases into nuclear foci. We show that expression of a nondegradable PIP degron (Cdt1) impairs both Pol η and Pol κ focus formation on ultraviolet irradiation and reduces cell viability, while canonical PIP box-containing proteins have no effect. Furthermore, we identify PIP degron-containing peptides from several substrates of CRL4(Cdt2) as efficient inhibitors of Pol η foci formation. By site-directed mutagenesis we show that inhibition depends on a conserved threonine residue that confers high affinity for PCNA-binding. Altogether these findings reveal an important regulative role for the CRL4(Cdt2) pathway in the switch of PCNA partners on DNA damage

    Dataset and species aggregation method applied to food-web models in the Northern Ionian Sea (Central Mediterranean Sea)

    The ecological roles of the species in the food web are studied through the Ecopath with Ecosim modelling approach. In this modelling approach, the food web is described by means of functional groups, each representing a species, a life stage of a species, or a group of species with similar trophic, ecological and physiological features. Links between the groups are formally described by a set of linear equations, informed with ecological and fishing data. Here, the data input collected to implement 3 Ecopath models in the Northern Ionian Sea (Central Mediterranean Sea) from 1995 to 2015 are reported. This dataset applied to study the ecological roles of the demersal Chondrichthyes in the study area could be useful to explore different fishing management scenarios. A large dataset of over 300 taxa is shown detailing the ecological inputs, such as Biomass (kg km−2), Production and Consumption rates (y−1), Diet information (weight in %), and fishing data represented by Landings and Discards (t km−2 y−1). In particular, the fishery data described the catches of trawls, longlines, passive nets, other gears and purse seine. In addition, a description of the aggregation method of the species is shown

    Exploring spatio-temporal changes in the demersal and benthopelagic assemblages of the northwestern Ionian Sea (central Mediterranean Sea)

    An exploration of the structure of demersal and benthopelagic assemblages in the north-western Ionian Sea was carried out by means of a set of statistical analyses. Self-organising map (SOM) and clustering methods applied to 183 taxa and their biomass (kg km-2) provided the classification of 1288 experimental hauls exploring the bathymetric range 10-800 m from 1995 to 2012. Six clusters were identified according to their similarities in species abundances (biomass), confirming the depth gradient as the main structuring agent. In order to identify key representative species in each cluster, the taxa were ranked by means of an indicator value index (IndVal) and the contribution of species to beta diversity (BD). Furthermore, the clusters were described by means of environmental and fishing characteristics. Particular habitat type, distance to canyon and fishing effort segregated the assemblages on the coastal shelf and slope. Temporal differences were detected in 2 bathyal groups, which were most likely affected by the 1990s environmental change in the deepwater circulation known as the Eastern Mediterranean Transient. The overall total BD in the study area was calculated as 0.79, with a temporal decrease observed at a rate of 0.7% yr-1. The approaches used are useful to identify and characterize the species aggregations inside complex faunal assemblages, without a priori assumptions about data distribution. These results can be a starting point for defining functional groups for Mediterranean food web modelling approaches, as well as for identifying indicator species to assess the environmental status in the context of the Marine Strategy Framework Directive
