    Youth deviance in group and prevention in educational-training contexts: Biodanza SRT as a strategy for the development of personal skills and talents

    Youth deviance in Italy is a phenomenon that is involving an ever-increasing number of young people and, unfortunately, is contributing strongly to crime, with a corresponding increase in frequent recidivism. It is urgent to undertake actions aimed at prevention through effective intervention proposals reflecting a high educational value and a transformative value of deviant behaviour and social maladaptation, aimed at the development of psycho-social and crossmodal (body-emotional-empathetic-cognitive-relational) skills useful to counter the onset of criminal behaviour. Being a valid educational-training strategy to be implemented in compulsory schools, though actively involving experiences, Biodanza SRT focuses body and corporeity as paths of knowledge, restoring human dignity, positive identification, empowerment, consciousness, talent enhancement and awareness of oneself, of one's own abilities and skills

    The exposure to high stress conditions has led to an increasing prevalence of Burnout among teachers; this is due to emotional exhaustion, depersonalization and reduced personal accomplishment. Soft Skills play an important role in working performance, representing the set of emotional skills capable of promoting personal well-being. Giving priority to emotional support enables the development of protective factors and positive skills to counteract work-related stress and the onset of Burnout. Biodanza SRT is a body-mediated system that, through an Embodiment Training, promotes the development of personal empowerment by strengthening emotional skills, Soft Skills and the acquisition of Best Practices making individuals able, capable and competent in the personal management of tools and strategies to improve their emotional, affective, relational, social and professional life

    Movement education is effective when sport-specific skills are successfully achieved. The variety of theoretical frameworks, of individual teaching and learning styles, lay the foundations to identify new practical teaching strategies, improve the mastery of motor skills, promote self-esteem, foster selfefficacy and positive self-image. The experimental educational and training strategy, proposed in this contribution, facilitates new motor experiences precisely through the ballistic action with the Kettlebell: the Swing. This movement develops and improves not only the conditional skills, but also the coordination ones. A didactic programming supports innovative and inclusive paths, and includes a new and innovative motor education based onfunctional movements, as a necessary strategy for developing learning processes. The knowledge development process will be transactional, within a reality in which the final product will be the relationship between subject and environment expressed through the body. For this reason, the movement holds the whole truth, made up not only of objective and subjective reality, external world and body, but more comprehensively, of relationships, emotions, symbols and meanings, all necessarily useful to build a knowledge of the vision of reality, of the real world.L’educazione al movimento è efficace quando le abilità specifiche di uno sport vengono raggiunte con successo. La varietà di quadri teorici, di stili individuali di insegnamento e apprendimento rappresentano la base per individuare nuove strategie pratiche di insegnamento, per migliorare la padronanza delle abilità motorie, per promuovere l’autostima, per favorire l’autoefficacia e l’immagine positiva di sé. La strategia educativa e formativa sperimentale, proposta in questo contributo, agevola nuove esperienze motorie proprio attraverso l’azione balistica con il Kettlebell: lo Swing. Questo movimento sviluppa e migliora non solo le capacità condizionali, ma anche quelle coordinative. Una programmazione didattica, a favore dei percorsi innovativi ed inclusivi che considera l’educazione motoria, nuova ed innovativa basata sui movimenti funzionali, strategia necessaria per lo sviluppo dei processi formativi di apprendimento. Il processo di sviluppo della conoscenza sarà transazionale, all’interno di una realtà il cui prodotto finale sarà costituito dal rapporto tra soggetto e ambiente attraverso il corpo. Per questo motivo, nel movimento c’è tutta la verità, fatta non solo dalla realtà oggettiva e soggettiva, esterno e corpo, ma, più complessivamente, dalle relazioni, dalle emozioni, dai simboli, dai significati, tutto necessariamente utile a costruire una conoscenza della visione della realtà, del reale.

    The RAS/MAP kinase pathway has attracted attention because activating mutations of the BRAF serine/threonine kinase was described in over 50% of melanomas. Very recently, selective and potent BRAF inhibitors have been developed. Several other signal transduction pathways have been found to be constitutively active or mutated in other subsets of melanoma tumors that are potentially targetable with new agents. Among these, NFκB is another pathway that melanoma tumors use to achieve survival, proliferation and resistance to apoptosis. Inhibition of NF-κB activation appears to be a very promising option for anti-cancer therapies

    Permissive role for mGlu1 metabotropic glutamate receptors in excitotoxic retinal degeneration

    Neuroprotection is an unmet need in eye disorders characterized by retinal ganglion cell (RGC) death, such as prematurity-induced retinal degeneration, glaucoma, and age-related macular degeneration. In all these disorders excitotoxicity is a prominent component of neuronal damage, but clinical data discourage the development of NMDA receptor antagonists as neuroprotectants. Here, we show that activation of mGlu1 metabotropic glutamate receptors largely contributes to excitotoxic degeneration of RGCs. Mice at postnatal day 9 were challenged with a toxic dose of monosodium glutamate (MSG, 3g/kg), which caused the death of >70% of Brn-3a+ RGCs. Systemic administration of the mGlu1 receptor negative allosteric modulator (NAM), JNJ16259685 (2.5mg/kg, s.c.), was largely protective against MSG-induced RGC death. This treatment did not cause changes in motor behavior in the pups. We also injected MSG to crv4 mice, which lack mGlu1 receptors because of a recessive mutation of the gene encoding the mGlu1 receptor. MSG did not cause retinal degeneration in crv4 mice, whereas it retained its toxic activity in their wild-type littermates. These findings demonstrate that mGlu1 receptors play a key role in excitotoxic degeneration of RGCs, and encourage the study of mGlu1 receptor NAMs in models of retinal neurodegeneration

    Support children's training through experimental pedagogical proposals of motor and sports education

    Motor and sports education supports a fundamental element in the educational growth of each child, and guarantees the acquisition of motor and pedagogical-relational skills, essential for an effective social integration of children in the surrounding environment. Therefore, school should be the main place where these skills are learned, especially at primary level; in this regard, this research contribution aims at evaluating the progresses of a sample class, which is given an experimental motor and sports design in order to obtain a pedagogical progress. The pursued outcomes are proposed both as a starting point for future scientific research in the educational field, and as a valid support for requesting pedagogical proposals from the decision makers of the education and training system

    Stimulation of S1PR5 with A-971432, a selective agonist, preserves blood-brain barrier integrity and exerts therapeutic effect in an animal model of Huntington's disease

    Huntington's disease (HD) is themost common neurodegenerative disorder for which no effective cure is yet available. Although several agents have been identified to provide benefits so far, the number of therapeutic options remains limited with only symptomatic treatment available. Over the past few years, we have demonstrated that sphingolipid-based approachesmay open the door to newandmore targeted treatments for the disease. In this study, we investigated the therapeutic potential of stimulating sphingosine-1-phosphate (S1P) receptor 5 by the new selective agonist A-971432 (provided by AbbVie) in R6/2mice, a widely used HD animalmodel. Chronic administration of low-dose (0.1mg/kg) A-971432 slowed down the progression of the disease and significantly prolonged lifespan in symptomatic R6/2mice. Such beneficial effects were associated with activation of pro-survival pathways (BDNF, AKT and ERK) and with reduction of mutant huntingtin aggregation. A-971432 also protected blood-brain barrier (BBB) homeostasis in the same mice. Interestingly, when administered early in the disease, before any overt symptoms, A-971432 completely protected HDmice fromthe classic progressivemotor deficit and preserved BBB integrity. Beside representing a promising strategy to take into consideration for the development of alternative therapeutic options for HD, selective stimulation of S1P receptor 5may be also seen as an effective approach to target brain vasculature defects in the disease

    mGlu1 Receptors Monopolize the Synaptic Control of Cerebellar Purkinje Cells by Epigenetically Down-Regulating mGlu5 Receptors

    In cerebellar Purkinje cells (PCs) type-1 metabotropic glutamate (mGlu1) receptors play a key role in motor learning and drive the refinement of synaptic innervation during postnatal development. The cognate mGlu5 receptor is absent in mature PCs and shows low expression levels in the adult cerebellar cortex. Here we found that mGlu5 receptors were heavily expressed by PCs in the early postnatal life, when mGlu1α receptors were barely detectable. The developmental decline of mGlu5 receptors coincided with the appearance of mGlu1α receptors in PCs, and both processes were associated with specular changes in CpG methylation in the corresponding gene promoters. It was the mGlu1 receptor that drove the elimination of mGlu5 receptors from PCs, as shown by data obtained with conditional mGlu1α receptor knockout mice and with targeted pharmacological treatments during critical developmental time windows. The suppressing activity of mGlu1 receptors on mGlu5 receptor was maintained in mature PCs, suggesting that expression of mGlu1α and mGlu5 receptors is mutually exclusive in PCs. These findings add complexity to the the finely tuned mechanisms that regulate PC biology during development and in the adult life and lay the groundwork for an in-depth analysis of the role played by mGlu5 receptors in PC maturation
