    The pan-genome of Lactobacillus reuteri strains originating from the pig gastrointestinal tract

    Background Lactobacillus reuteri is a gut symbiont of a wide variety of vertebrate species that has diversified into distinct phylogenetic clades which are to a large degree host-specific. Previous work demonstrated host specificity in mice and begun to determine the mechanisms by which gut colonisation and host restriction is achieved. However, how L. reuteri strains colonise the gastrointestinal (GI) tract of pigs is unknown. Results To gain insight into the ecology of L. reuteri in the pig gut, the genome sequence of the porcine small intestinal isolate L. reuteri ATCC 53608 was completed and consisted of a chromosome of 1.94 Mbp and two plasmids of 138.5 kbp and 9.09 kbp, respectively. Furthermore, we generated draft genomes of four additional L. reuteri strains isolated from pig faeces or lower GI tract, lp167-67, pg-3b, 20-2 and 3c6, and subjected all five genomes to a comparative genomic analysis together with the previously completed genome of strain I5007. A phylogenetic analysis based on whole genomes showed that porcine L. reuteri strains fall into two distinct clades, as previously suggested by multi-locus sequence analysis. These six pig L. reuteri genomes contained a core set of 1364 orthologous gene clusters, as determined by OrthoMCL analysis, that contributed to a pan-genome totalling 3373 gene clusters. Genome comparisons of the six pig L. reuteri strains with 14 L. reuteri strains from other host origins gave a total pan-genome of 5225 gene clusters that included a core genome of 851 gene clusters but revealed that there were no pig-specific genes per se. However, genes specific for and conserved among strains of the two pig phylogenetic lineages were detected, some of which encoded cell surface proteins that could contribute to the diversification of the two lineages and their observed host specificity. Conclusions This study extends the phylogenetic analysis of L. reuteri strains at a genome-wide level, pointing to distinct evolutionary trajectories of porcine L. reuteri lineages, and providing new insights into the genomic events in L. reuteri that occurred during specialisation to their hosts. The occurrence of two distinct pig-derived clades may reflect differences in host genotype, environmental factors such as dietary components or to evolution from ancestral strains of human and rodent origin following contact with pig populations

    Generalized Lie bialgebroids and Jacobi structures

    The notion of a generalized Lie bialgebroid (a generalization of the notion of a Lie bialgebroid) is introduced in such a way that a Jacobi manifold has associated a canonical generalized Lie bialgebroid. As a kind of converse, we prove that a Jacobi structure can be defined on the base space of a generalized Lie bialgebroid. We also show that it is possible to construct a Lie bialgebroid from a generalized Lie bialgebroid and, as a consequence, we deduce a duality theorem. Finally, some special classes of generalized Lie bialgebroids are considered: triangular generalized Lie bialgebroids and generalized Lie bialgebras.Comment: 32 page

    Health System Characteristics and Rates of Readmission After Acute Myocardial Infarction in the United States

    Background: Interventions to reduce early readmissions have focused on patient characteristics and the importance of early follow‐up; however, less is known about the characteristics of health systems, including quality, capacity, and intensity, and their influence on readmission rates in the United States. Therefore, we examined the association of hospital patterns of medical care with rates of 30‐day readmission. Methods and Results: Medicare beneficiaries hospitalized for an AMI (n=188 611) between 2008 and 2009 in 1088 hospitals in the United States were included in our cohort. We tested the association between hospital patterns of medical care quality (discharge planning care quality), capacity (hospital size measured as the number of beds, hospital‐level Medicare all medical admission rates, supply of primary care physicians and cardiologists), and intensity (measures of care during the last 6 months of life) on CMS risk‐adjusted rates of 30‐day readmission using Poisson multilevel mixed‐effects models adjusting for patient‐ and hospital‐level covariates. There were 38 350 readmissions at 30‐days (20.3%) AMI discharges. Controlling for patient characteristics, measures of hospital care associated with higher rates of readmission included higher hospital‐level rates for all medical admissions, per capita primary care physicians and cardiologists, and last 6 months of life care intensity measures including increased number of hospital days, number of ICU days, number of physician visits, and 10 or more different physicians seen during the last 6 months of life. Better discharge quality and larger hospitals were associated with lower rates of readmission. Conclusions: In addition to quality of care, high 30‐day readmission rates are associated with hospital‐level measures of capacity and intensity. Efforts to reduce readmission rates may need to address these broader patterns of medical care

    Dural sinus thrombosis (DST), or cerebral venous thrombosis, is an uncommon cause of stroke. It has a variable presentation, and the symptoms and signs can be non-specific. The diagnosis of DST can be difficult to make and is often delayed or missed. Computed tomography venography or magnetic resonance venography are the typical imaging modalities used to diagnose DST. However, computed tomography venography and magnetic resonance venography both have limitation for emergency department patients.     Analysis by Surface Plasmon Resonance of the Influence of Valence on the Ligand Binding Affinity and Kinetics of an Anti-carbohydrate Antibody

    The kinetics of ligand binding by Se155-4, an antibody specific for the Salmonella serogroup B O-polysaccharide, were studied by surface plasmon resonance. Because trace amounts of oligomers in Fab and single-chain antibody variable domain (scFv) preparations resulted in biphasic binding profiles that were difficult to analyze, all kinetic measurements were performed on purified monomeric fragments and, for certain mutant scFv, dimeric forms. Results obtained with monomeric forms indicated that the relatively low affinity of the antibody was due to rapid dissociation (koff approximately 0.25 s-1). Dimeric forms generally showed off-rates that were approximately 20-fold slower and a 5-fold increase in association rate constants to approximately 2 x 10(5) M-1 s-1. Although the association phases for scFv dimers showed good curve fitting to a one component interaction model, the dissociation phases were biphasic, presumably because the availability and accessibility of sites on the antigen always leads to some monovalent attachment. The fast off-rate for dimers was the same as the monomer off-rate. Se155-4 IgG off-rates were very similar to those observed for scFv dimer, whereas the onrate was the same as that obtained with Fab and scFv monomer

    Online interventions for problem gamblers with and without co-occurring mental health symptoms: protocol for a randomized controlled trial

    BACKGROUND Comorbidity between problem gambling and depression or anxiety is common. Further, the treatment needs of people with co-occurring gambling and mental health symptoms may be different from those of problem gamblers who do not have a co-occurring mental health concern. The current randomized controlled trial (RCT) will evaluate whether there is a benefit to providing access to mental health Internet interventions (G + MH intervention) in addition to an Internet intervention for problem gambling (G-only intervention) in participants with gambling problems who do or do not have co-occurring mental health symptoms. METHODS Potential participants will be screened using an online survey to identify participants meeting criteria for problem gambling. As part of the baseline screening process, measures of current depression and anxiety will be assessed. Eligible participants agreeing (N = 280) to take part in the study will be randomized to one of two versions of an online intervention for gamblers – an intervention that just targets gambling issues (G-only) versus a website that contains interventions for depression and anxiety in addition to an intervention for gamblers (G + MH). It is predicted that problem gamblers who do not have co-occurring mental health symptoms will display no significant difference between intervention conditions at a six-month follow-up. However, for those with co-occurring mental health symptoms, it is predicted that participants receiving access to the G + MH website will display significantly reduced gambling outcomes at six-month follow-up as compared to those provided with G-only website. DISCUSSION The trial will produce information on the best means of providing online help to gamblers with and without co-occurring mental health symptoms.This research was funded by the Manitoba Gambling Research Program of Manitoba Liquor and Lotteries

    Spatial attention affects the early processing of neutral versus fearful faces when they are task-irrelevant: a classifier study of the EEG C1 component

    EEG studies suggest that the emotional content of visual stimuli is processed rapidly. In particular, the C1 component, which occurs up to 100 ms after stimulus onset and likely reflects activity in primary visual cortex V1, has been reported to be sensitive to emotional faces. However, difficulties replicating these results have been reported. We hypothesized that the nature of the task and attentional condition are key to reconcile the conflicting findings. We report three experiments of EEG activity during the C1 time range elicited by peripherally presented neutral and fearful faces under various attentional conditions: the faces were spatially attended or unattended and were either task-relevant or not. Using traditional event-related potential analysis, we found that the early activity changed depending on facial expression, attentional condition, and task. In addition, we trained classifiers to discriminate the different conditions from the EEG signals. Although the classifiers were not able to discriminate between facial expressions in any condition, they uncovered differences between spatially attended and unattended faces but solely when these were task-irrelevant. In addition, this effect was only present for neutral faces. Our study provides further indication that attention and task are key parameters when measuring early differences between emotional and neutral visual stimuli
