    Monitoring Durability of Limestone Cement Paste Stored at Conditions Promoting Thaumasite Formation

    The durability of Portland-limestone cement with high limestone content was monitored at conditions promoting thaumasite formation. Pore structure and deterioration characteristics were assessed with X-ray micro-computed tomography and correlated with material's strength. Changes in crystalline and amorphous phases of the cement paste were investigated with X-ray powder diffraction and solid state nuclear magnetic resonance spectroscopy. Rapid deterioration was observed, evolving as a front causing concentric crack patterns followed by detachment of the part of specimen in contact with the corrosive solution. This ultimately led to loss of structural integrity after 4 months of exposure. During sulfate attack, thaumasite, ettringite and gypsum formed at the expense of portlandite, calcite and monocarboaluminate hydrate. Furthermore, polymerization of silicate chains in C-S-H and deterioration of C-S-H also occurred

    Examining the Relationship between Innovation Activity and Corporate Cooperation among Small and Medium-sized Business Operating in the Slovak ICT Sector

    The innovation process is a key factor in developing and maintaining competitiveness; therefore, the SME sector should focus on innovation as well. Examining the impact of corporate cooperation on innovation activity is a crucial element. The main goal of the research is to assess the innovative activity and corporate cooperation of micro enterprises and SMEs operating in the ICT sector. In accordance with the main goal of our research, a hypothesis was formulated regarding the relationship between participating in formal networks and innovative activity as follows: There is a relationship between innovation activity and corporate cooperation. We use descriptive statistical methods, i.e., unweighted and weighted arithmetic means, as well as a structural analysis method to test our hypothesis. To verify our subhypothesis H (1a), we had to examine the innovative activity of the domestic SMEs participating in the research. According to our second subhypothesis H (1b), we stated that innovative SMEs are more likely to be members of cooperation networks than noninnovative businesses are. The second subhypothesis H(1b) is accepted. However, we could not detect a significant difference or hostile behaviour among the innovative and noninnovative SMEs since both groups of companies participated in co-operation networks at a similar rate, as they were not members of formal co-operation networks. Since our third subhypothesis—the formal network co-operations influence the form of assistance used during innovation activity—supporting the main hypothesis was rejected, the main hypothesis was also rejected. Therefore, a significant relationship can be detected between innovation activity and corporate cooperation among domestic SMEs. Based on data analysis from research carried out at the beginning of 2021, Slovakia was ranked among moderate innovators based on the innovation performance of EU countries in 2019. If more companies were to participate in future research, it might be possible that a connection between corporate cooperation and innovation activity could also be discovered in Slovakia. A proposal was formulated based on the results of the primary data analysis. Since no significant relationship could be detected for these two factors, the obtained research results did not match, and the results were opposite to the results obtained during the Netnes epidemic

    Synthetic calcium carbonate improves the effectiveness of treatments with nanolime to contrast decay in highly porous limestone

    Three synthetized polymorphs of calcium carbonate have been tested in combination with the suspension of nanolime particles as potential consolidating agents for contrasting stone decay and overcome some of the limitations of nanolime agents when applied to substrates with large porosity. The modifications induced in the pore network of the Maastricht limestone were analyzed with microscopy and in a non-invasive fashion with small angle neutron scattering and synchrotron radiation micro-computed tomography. A reduction in porosity and pore accessibility at the micrometric scale was detected with the latter technique, and ascribed to the improved pore-filling capacity of the consolidation agent containing CaCO3 particles. These were found to be effectively bound to the carbonated nanolime, strengthening the pore-matrix microstructure. Penetration depth and positive effect on porosity were found to depend on the particle size and shape. Absence of significant changes in the fractal nature of the pore surface at the nanoscale, was interpreted as indication of the negligible contribution of nanolime-based materials in the consolidation of stones with large porosity. However, the results indicate that in such cases, their effectiveness may be enhanced when used in combination with CaCO3 particles, owing to the synergic effect of chemical/structural compatibility and particle size distribution

    Immunomodulatory Potential of Differently-Terminated Ultra-Small Silicon Carbide Nanoparticles

    Ultra-small nanoparticles with sizes comparable to those of pores in the cellular membrane possess significant potential for application in the field of biomedicine. Silicon carbide ultra-small nanoparticles with varying surface termination were tested for the biological system represented by different human cells (using a human osteoblastic cell line as the reference system and a monocyte/macrophage cell line as immune cells). The three tested nanoparticle surface terminations resulted in the observation of different effects on cell metabolic activity. These effects were mostly noticeable in cases of monocytic cells, where each type of particle caused a completely different response (‘as-prepared’ particles, i.e., were highly cytotoxic, –OH terminated particles slightly increased the metabolic activity, while –NH2 terminated particles caused an almost doubled metabolic activity) after 24 h of incubation. Subsequently, the release of cytokines from such treated monocytes and their differentiation into activated cells was determined. The results revealed the potential modulation of immune cell behavior following stimulation with particular ultra-small nanoparticles, thus opening up new fields for novel silicon carbide nanoparticle biomedical applications

    Listeria monocytogenes Infection in Macrophages Induces Vacuolar-Dependent Host miRNA Response

    Listeria monocytogenes is a Gram-positive facultative intracellular pathogen, causing serious illness in immunocompromised individuals and pregnant women. Upon detection by macrophages, which are key players of the innate immune response against infection, L. monocytogenes induces specific host cell responses which need to be tightly controlled at transcriptional and post-transcriptional levels. Here, we ask whether and how host miRNAs, which represent an important mechanism of post-transcriptional regulation in a wide array of biological processes, are altered by a model pathogen upon live infection of murine bone marrow derived macrophages. We first report that L. monocytogenes subverts the host genome-wide miRNA profile of macrophages in vitro. Specifically, we show that miR-155, miR-146a, miR-125a-3p/5p and miR-149 were amongst the most significantly regulated miRNAs in infected macrophages. Strikingly, these miRNAs were highly upregulated upon infection with the Listeriolysin-deficient L. monocytogenes mutant Δhly, that cannot escape from the phagosome thus representing a vacuolar-contained infection. The vacuolar miRNA response was significantly reduced in macrophages deficient for MyD88. In addition, miR-146a and miR-125a-3p/5p were regulated at transcriptional levels upon infection, and miR-125a-3p/5p were found to be TLR2 responsive. Furthermore, miR-155 transactivation in infection was regulated by NF-κB p65, while miR-146a and miR-125a-3p/5p expression was unaffected in p65-deficient primary macrophages upon L. monocytogenes infection. Our results demonstrate that L. monocytogenes promotes significant changes in the miRNA expression profile in macrophages, and reveal a vacuolar-dependent miRNA signature, listeriolysin-independent and MyD88-dependent. These miRNAs are predicted to target immune genes and are therefore most likely involved in regulation of the macrophage innate immune response against infection at post-transcriptional levels

    Quantitative analysis of mutant subclones in chronic myeloid leukemia : comparison of different methodological approaches

    Identification and quantitative monitoring of mutant BCR-ABL1 subclones displaying resistance to tyrosine kinase inhibitors (TKIs) have become important tasks in patients with Ph-positive leukemias. Different technologies have been established for patient screening. Various next-generation sequencing (NGS) platforms facilitating sensitive detection and quantitative monitoring of mutations in the ABL1-kinase domain (KD) have been introduced recently, and are expected to become the preferred technology in the future. However, broad clinical implementation of NGS methods has been hampered by the limited accessibility at different centers and the current costs of analysis which may not be regarded as readily affordable for routine diagnostic monitoring. It is therefore of interest to determine whether NGS platforms can be adequately substituted by other methodological approaches. We have tested three different techniques including pyrosequencing, LD (ligation-dependent)-PCR and NGS in a series of peripheral blood specimens from chronic myeloid leukemia (CML) patients carrying single or multiple mutations in the BCR-ABL1 KD. The proliferation kinetics of mutant subclones in serial specimens obtained during the course of TKI-treatment revealed similar profiles via all technical approaches, but individual specimens showed statistically significant differences between NGS and the other methods tested. The observations indicate that different approaches to detection and quantification of mutant subclones may be applicable for the monitoring of clonal kinetics, but careful calibration of each method is required for accurate size assessment of mutant subclones at individual time points
