    New Applications of Radio Frequency Identification Stations for Monitoring Fish Passage through Headwater Road Crossings and Natural Reaches

    Within the Ouachita National Forest, roads and streams intersect each other thousands of times. Many of these road crossings alter stream hydrology and potentially limit longitudinal fish movement. To investigate the potential impacts of these road crossings on fish passage, we monitored movements of 3 native fish species (n = 2,171) individually tagged with radio frequency identification (RFID) tags in 2012 and 2013. We installed solar-powered RFID stations in 2 streams with road crossings and 2 reference streams without road crossings. Each of the 4 monitoring stations included a pair of antennas bracketing a road crossing (or similarly-sized natural reach) to continuously detect upstream or downstream passage. To monitor natural reference streams, we avoided full-duplex RFID technology, which would have required rigid in-stream structures. Alternatively, we utilized new applications of RFID technology such as direct in-stream installation of half-duplex wire antennas and figure-eight crossover antenna designs. These techniques appear promising, but technical difficulties limited the consistency of fish passage detection and consequently limited the strength of ecological conclusions. Even so, we report evidence that fish passed at significantly higher rates across reference reaches than reaches with road crossings. Furthermore, Creek Chub (Semotilus atromaculatus) passed reference reaches at significantly higher rates than Highland Stonerollers (Campostoma spadiceum), which passed at higher rates than Longear Sunfish (Lepomis megalotis). Stream intermittency appeared to exacerbate reduced passage rates associated with the road crossings

    Relations Between Consultant Characteristics and Measures of Consultation Quality and Outcome

    The purpose of this study was to explore school-based consultation effectiveness (as perceived by classroom teachers) and important indices of consultation quality and student outcome. Specifically, the research explored 1) important characteristics of school-based consultants using the Consultant Effectiveness Scale (CES), 2) the relations between quality of the consultation process and student outcome, and 3) the measurement similarities between the CES and the measure of consultation process quality. Questionnaires were completed by 85 teachers working in grades K-12 in a suburban area of Chicago. To participate, teachers must have engaged in a consultative relationship within the past 12 months. Consultants characteristics perceived by teachers as most effective are identified and discussed. Some indices of quality were found to be more important in effecting child outcome than others. This finding, which partially supports and contradicts previous literature, is detailed. Finally, positive relations were found between the CES and the measure of consultation quality, which may significantly contribute to existing CES research. Implications of these findings for school psychologists are provided in depth. The researcher also suggests ways in which the information may be used to improve consultative relationships with teachers and thereby, services to children

    A Sterile 20 Family Kinase and Its Co-factor CCM-3 Regulate Contractile Ring Proteins on Germline Intercellular Bridges

    Germ cells in most animals are connected by intercellular bridges, actin-based rings that form stable cytoplasmic connections between cells promoting communication and coordination [1]. Moreover, these connections are required for fertility [1, 2]. Intercellular bridges are proposed to arise from stabilization of the cytokinetic ring during incomplete cytokinesis [1]. Paradoxically, proteins that promote closure of cytokinetic rings are enriched on stably open intercellular bridges [1, 3, 4]. Given this inconsistency, the mechanism of intercellular bridge stabilization is unclear. Here, we used the C. elegans germline as a model for identifying molecular mechanisms regulating intercellular bridges. We report that bridges are actually highly dynamic, changing size at precise times during germ cell development. We focused on the regulation of bridge stability by anillins, key regulators of cytokinetic rings and cytoplasmic bridges [1, 4-7]. We identified GCK-1, a conserved serine/threonine kinase [8], as a putative novel anillin interactor. GCK-1 works together with CCM-3, a known binding partner [9], to promote intercellular bridge stability and limit localization of both canonical anillin and non-muscle myosin II (NMM-II) to intercellular bridges. Additionally, we found that a shorter anillin, known to stabilize bridges [4, 7], also regulates NMM-II levels at bridges. Consistent with these results, negative regulators of NMM-II stabilize intercellular bridges in the Drosophila egg chamber [10, 11]. Together with our findings, this suggests that tuning of myosin levels is a conserved mechanism for the stabilization of intercellular bridges that can occur by diverse molecular mechanisms

    A re-investigation of the path of carbon in photosynthesis utilizing GC/MS methodology. Unequivocal verification of the participation of octulose phosphates in the pathway

    A GC/EIMS/SIM methodology has been developed to re-examine the path of carbon in photosynthesis. Exposing isolated spinach chloroplasts to (13)CO(2 )on a solid support for a defined period followed by quenching and work-up provided a mixture of labelled sugar phosphates. After enzymatic dephosphorylation and derivatization, the Mox-TMS sugars were analysed using the above method. The purpose of the study was to try to calculate the atom% enrichment of (13)C in as many of the individual carbons in each of the derivatized sugars as was practical using diagnostic fragment ions. In the event, only one 45 s experiment provided sufficient data to enable a range of enrichment values to be calculated. This confirmed that D-glycero-D-altro-octulose phosphate was present in the chloroplasts and was heavily labelled in the C4, C5 and C6 positions, in keeping with the hypothesis that it had an inclusive role and a labelling pattern consistent with a new modified pathway of carbon in photosynthesis

    Factors Affecting Trypanosome Maturation in Tsetse Flies

    Trypanosoma brucei brucei infections which establish successfully in the tsetse fly midgut may subsequently mature into mammalian infective trypanosomes in the salivary glands. This maturation is not automatic and the control of these events is complex. Utilising direct in vivo feeding experiments, we report maturation of T. b. brucei infections in tsetse is regulated by antioxidants as well as environmental stimuli. Dissection of the maturation process provides opportunities to develop transmission blocking vaccines for trypanosomiasis. The present work suggests L-cysteine and/or nitric oxide are necessary for the differentiation of trypanosome midgut infections in tsetse
