    An investigation of the application of programming techniques to farm management problems, with special reference to beef cattle feeding in the Lothians of Scotland and in Alberta

    The Lothians of Scotland are generally farmed intensive- ly, with arable farming predominating in the more easterly regions. Potatoes form an integral part of the cropping pat- tern with sugar beets and turnips somewhat less widely grown. Wheat and barley are the principal cereal crops. Grass is in- cluded in the rotation on practically every farm. The returns from cropping are usually complemented by returns from the production of beef cattle. Farmers of the area usually have one or more beef cattle enterprises, frequently with emphasis on fattening cattle for slaughter.The farming pattern, with emphasis on potato production, requires a substantial labour force, measured on an "acres per man" basis, as compared to less intensively farmed areas such as the unirrigated portions of Alberta. The availability of this labour force during non -critical seasons might be expect- ed to result in a level of mechanization differing from that developed for less intensively farmed areas. For field work, a larger number of smaller capacity machines might be expected, and for cattle feeding a lower level of mechanization might be expected. Despite these expectations there is a keen interest in highly mechanized processes. This is particularly evident in processes associated with cattle production.This intensive, well integrated pattern of agriculture offers a challenging application for linear programming as a management aid. In addition, the interaction between cropping practise, manpower availability, and level of mechanization, particularly with respect to beef cattle feeding, offers an excellent medium for the extension of linear programming into the fixed -cost sector of farm business.Tabulated, the objectives of the research leading to the preparation of this thesis were to:1) Investigate the application of linear programming as an aid to farm planning in the Lothians.2) Complementary to this objective, to assess the availability and suitability of Scottish agri- culture research data for the preparation of linear programmes.3) The development of extensions to linear programming to permit the analysis of any mechanized activity in full economic association with other farm activities.4) An investigation of the use of linear programming, with extensions, as a method for determining the optimum levels of mechanization for beef cattle feeding

    Follow-up of phase I trial of adalimumab and rosiglitazone in FSGS: III. Report of the FONT study group

    Abstract Background Patients with resistant primary focal segmental glomerulosclerosis (FSGS) are at high risk of progression to chronic kidney disease stage V. Antifibrotic agents may slow or halt this process. We present outcomes of follow-up after a Phase I trial of adalimumab and rosiglitazone, antifibrotic drugs tested in the Novel Therapies in Resistant FSGS (FONT) study. Methods 21 patients -- 12 males and 9 females, age 16.0 ± 7.5 yr, and estimated GFR (GFRe) 121 ± 56 mL/min/1.73 m2 -- received adalimumab (n = 10), 24 mg/m2 every 14 days or rosiglitazone (n = 11), 3 mg/m2 per day for 16 weeks. The change in GFRe per month prior to entry and after completion of the Phase I trial was compared. Results 19 patients completed the 16-week FONT treatment phase. The observation period pre-FONT was 18.3 ± 10.2 months and 16.1 ± 5.7 months after the study. A similar percentage of patients, 71% and 56%, in the rosiglitazone and adalimumab cohorts, respectively, had stabilization in GFRe, defined as a reduced negative slope of the line plotting GFRe versus time without requiring renal replacement therapy after completion of the FONT treatment period (P = 0.63). Conclusion Nearly 50% of patients with resistant FSGS who receive novel antifibrotic agents may have a legacy effect with delayed deterioration in kidney function after completion of therapy. Based on this proof-of-concept preliminary study, we recommend long-term follow-up of patients enrolled in clinical trials to ascertain a more comprehensive assessment of the efficacy of experimental treatments

    Promising insights into the health related quality of life for children with severe obesity

    Background Childhood obesity is a growing health concern known to adversely affect quality of life in children and adolescents. The Patient Reported Outcomes Measurement Information System (PROMIS) pediatric measures were developed to capture child self-reports across a variety of health conditions experienced by children and adolescents. The purpose of this study is to begin the process of validation of the PROMIS pediatric measures in children and adolescents affected by obesity. Methods The pediatric PROMIS instruments were administered to 138 children and adolescents in a cross-sectional study of patient reported outcomes in children aged 8–17 years with age-adjusted body mass index (BMI) greater than the 85th percentile in a design to establish known-group validity. The children completed the depressive symptoms, anxiety, anger, peer relationships, pain interference, fatigue, upper extremity, and mobility PROMIS domains utilizing a computer interface. PROMIS domains and individual items were administered in random order and included a total of 95 items. Patient responses were compared between patients with BMI 85 to < 99th percentile versus ≥ 99th percentile. Results 136 participants were recruited and had all necessary clinical data for analysis. Of the 136 participants, 5% ended the survey early resulting in missing domain scores at the end of survey administration. In multivariate analysis, patients with BMI ≥ 99th percentile had worse scores for depressive symptoms, anger, fatigue, and mobility (p < 0.05). Parent-reported exercise was associated with better scores for depressive symptoms, anxiety, and fatigue (p < 0.05). Conclusions Children and adolescents ranging from overweight to severely obese can complete multiple PROMIS pediatric measures using a computer interface in the outpatient setting. In the 5% with missing domain scores, the missing scores were consistently found in the domains administered last, suggesting the length of the assessment is important. The differences in domain scores found in this study are consistent with previous reports investigating the quality of life in children and adolescents with obesity. We show that the PROMIS instrument represents a feasible and potentially valuable instrument for the future study of the effect of pediatric obesity on quality of life

    Pioglitazone Enhances the Beneficial Effects of Glucocorticoids in Experimental Nephrotic Syndrome

    Glucocorticoids are the primary therapy for nephrotic syndrome (NS), but have serious side effects and are ineffective in ~20-50% of patients. Thiazolidinediones have recently been suggested to be renoprotective, and to modulate podocyte glucocorticoid-mediated nuclear receptor signaling. We hypothesized that thiazolidinediones could enhance glucocorticoid efficacy in NS. We found that puromycin aminonucleoside-induced proteinuria in rats was significantly reduced by both high-dose glucocorticoids (79%) and pioglitazone (61%), but not low-dose glucocorticoids (25%). Remarkably, pioglitazone + low-dose glucocorticoids also reduced proteinuria (63%) comparably to high-dose glucocorticoids, whereas pioglitazone + high-dose glucocorticoids reduced proteinuria to almost control levels (97%). Molecular analysis revealed that both glucocorticoids and pioglitazone enhanced glomerular synaptopodin and nephrin expression, and reduced COX-2 expression, after injury. Furthermore, the glomerular phosphorylation of glucocorticoid receptor and Akt, but not PPARγ, correlated with treatment-induced reductions in proteinuria. Notably, clinical translation of these findings to a child with refractory NS by the addition of pioglitazone to the treatment correlated with marked reductions in both proteinuria (80%) and overall immunosuppression (64%). These findings together suggest that repurposing pioglitazone could potentially enhance the proteinuria-reducing effects of glucocorticoids during NS treatment

    Genome-wide SNP identification by high-throughput sequencing and selective mapping allows sequence assembly positioning using a framework genetic linkage map

    <p>Abstract</p> <p>Background</p> <p>Determining the position and order of contigs and scaffolds from a genome assembly within an organism's genome remains a technical challenge in a majority of sequencing projects. In order to exploit contemporary technologies for DNA sequencing, we developed a strategy for whole genome single nucleotide polymorphism sequencing allowing the positioning of sequence contigs onto a linkage map using the bin mapping method.</p> <p>Results</p> <p>The strategy was tested on a draft genome of the fungal pathogen <it>Venturia inaequalis</it>, the causal agent of apple scab, and further validated using sequence contigs derived from the diploid plant genome <it>Fragaria vesca</it>. Using our novel method we were able to anchor 70% and 92% of sequences assemblies for <it>V. inaequalis </it>and <it>F. vesca</it>, respectively, to genetic linkage maps.</p> <p>Conclusions</p> <p>We demonstrated the utility of this approach by accurately determining the bin map positions of the majority of the large sequence contigs from each genome sequence and validated our method by mapping single sequence repeat markers derived from sequence contigs on a full mapping population.</p
