    Evaluation of techniques to break seed dormancy in Redroot pigweed (Amaranthus retroflexus)

    If we understand factors that trigger seed dormancy release, then we can accurately predict whether the seed will remain dormant or germinate out of the seed bank. With regards to annual weed species, detailed understanding of what breaks seed dormancy is therefore critical for determining how many weed seeds will germinate into problematic weeds. To investigate the breaking of dormancy in weed seeds, we conducted an experiment with Redroot pigweed (Amaranthus retroflexus). Dormant seeds were treated with cold stratification (4 °C for 30 days), application of gibberellic acid (at 500, 1000, 1500, and 2000 parts per million) or ultrasound (for 10, 20, 30, and 40 minutes), soaking in hot water (90 °C for 3, 5, 7 and 10 minutes) or 98% sulfuric acid (for 1, 2 and 3 minutes) to determine which treatment most effectively broke dormancy. The results showed that Redroot pigweed seed dormancy was effectively broken by cold stratification, gibberellic acid, or ultrasound. Short treatments with hot water had minimal effect while longer times or treatment with sulfuric acid eliminated seed germination. In addition to germination percentage, germination rate, plumule length, radicle length, seedling length, seedling dry weight, and seed vigor index were also measured; similarly application of gibberellic acid had the most significant effect on these parameters. The results of this study add to our understanding of what processes effectively or ineffectively break Redroot pigweed seed dormancy and promote growth

    Holding up a mirror: researching symmetrically to explore exclusion, othering and whiteness in local environmental governance

    The positioning of minoritised and racialised groups such as Global South immigrants as deficient in knowledge, language or motivation is a form of othering prevalent in all spheres of Global North societies, and the environmental sector is no exception. In both academic literature and policy, there is an assumption that minoritised groups are “hard to reach” and less interested in sustainability than the usual white middle-class environmental subjects. But, what might the picture look like if it focused on those who have the power to make choices about whose interests matter and whose can be ignored? By holding up a mirror to the powerful and asking them to reflect on their own practices and assumptions, we centre the operation of structural inequality to show that the lack of inclusivity in sustainability policy is very often a product of structural whiteness rather than the deficiencies of racialised communities. This article draws on interviews with key informants involved in local environmental governance to explore how people in positions of power serve to obstruct, erase or facilitate the engagement of racialised communities in activities relevant for environmental change. We argue that a symmetrical approach to research and analysis is needed to avoid othering immigrants while surfacing whiteness as the context in which othering occurs. We call for greater attention to how white structural advantage shapes the design and implementation of local green agendas in order to develop just and transformative approaches to environmental policy-making

    A VEL3 histone deacetylase complex establishes a maternal epigenetic state controlling progeny seed dormancy

    Mother plants play an important role in the control of dormancy and dispersal characters of their progeny. In Arabidopsis seed dormancy is imposed by the embryo-surrounding tissues of the endosperm and seed coat. Here we show that VERNALIZATION5/VIN3-LIKE 3 (VEL3) maintains maternal control over progeny seed dormancy. Acting early in endosperm development, it establishes an epigenetic state that primes the depth of primary seed dormancy that will be reached later during seed maturation. VEL3 relocates MSI1 to the nucleolus and associates with other components of the histone deacetylase complex (HDAC). Furthermore VEL3 preferentially associates with pericentromeric chromatin and is required for deacetylation and H3K27me3 deposition in the central cell. Interestingly, the epigenetic state established by the maternal VEL3 is retained in mature seeds, and controls seed dormancy in part through the repression of programmed cell death-associated gene ORE1. Our data demonstrates a novel mechanism by which maternal control of progeny seed physiology persists post-shedding, maintaining parental control of seed behaviour

    A broad excited state was observed in 6-He with energy E_x = 5 +/- 1 MeV and width Gamma = 3 +/- 1 MeV, following the reaction Li-7(gamma,p)He-6. The state is consistent with a number of broad resonances predicted by recent cluster model calculations.     The reverse mortgage market in New Zealand: key drivers of loan determination

    This paper examines the drivers of loan principals in the reverse mortgage and equity release market in New Zealand using a hedonic price model (HPM) approach. Our analysis using reverse mortgages data between 2004–2021, sourced from one major reverse mortgage bank, provides four key findings. First, the term of payment for repaid reverse mortgages is positively associated with loan principals, implying that longer repayment terms allow applicants who were able to repay mortgages to borrow more. Second, there is partial evidence to suggest the presence of a positive linear impact of the value of the current property on its loan principal, in line with previous house price modelling studies. Third, older applicants (age 75+) borrow less than younger applicants, which may be due to their repaying ability. Fourth, we confirm a positive effect of interest rates on reverse mortgage amounts but reject the positive association between wider loan-to-value policy restrictions and equity release lending amounts. The results broadly highlight that the house price is more relevant than any individual characteristic of a property in determining loan principals, and that all drivers are relevant in the early stage of the development of the reverse mortgages market in New Zealand

    Hydrodynamic parameters of air-bubble stimulated underwater spark discharges

    Considerable amount of research work has been focused on investigation and optimization of strong acoustic waves generated by spark discharges in water. There are several methods to achieve and to stimulate underwater spark breakdowns, including free-discharges, wire-guided and gas-bubble stimulated discharges. In the present work, air bubbles are injected into water in order to achieve spark discharges in relatively long inter-electrode gaps. This paper reports on hydrodynamic and acoustic characteristics of spark discharges stimulated by air bubbles and presents the functional relationships between the hydrodynamic and electrical parameters of such discharges, including breakdown voltage, spark plasma resistance and energy available in the discharge. A hydrodynamic analytical model has been developed and used to calculate the acoustic efficiency of the underwater spark discharges

    Functional consequence of the MET-T1010I polymorphism in breast cancer.

    Major breast cancer predisposition genes, only account for approximately 30% of high-risk breast cancer families and only explain 15% of breast cancer familial relative risk. The HGF growth factor receptor MET is potentially functionally altered due to an uncommon germline single nucleotide polymorphism (SNP), MET-T1010I, in many cancer lineages including breast cancer where the MET-T1010I SNP is present in 2% of patients with metastatic breast cancer. Expression of MET-T1010I in the context of mammary epithelium increases colony formation, cell migration and invasion in-vitro and tumor growth and invasion in-vivo. A selective effect of MET-T1010I as compared to wild type MET on cell invasion both in-vitro and in-vivo suggests that the MET-T1010I SNP may alter tumor pathophysiology and should be considered as a potential biomarker when implementing MET targeted clinical trials

    Dietary garlic and hip osteoarthritis: evidence of a protective effect and putative mechanism of action

    Background Patterns of food intake and prevalent osteoarthritis of the hand, hip, and knee were studied using the twin design to limit the effect of confounding factors. Compounds found in associated food groups were further studied in vitro. Methods Cross-sectional study conducted in a large population-based volunteer cohort of twins. Food intake was evaluated using the Food Frequency Questionnaire; OA was determined using plain radiographs. Analyses were adjusted for age, BMI and physical activity. Subsequent in vitro studies examined the effects of allium-derived compounds on the expression of matrix-degrading proteases in SW1353 chondrosarcoma cells. Results Data were available, depending on phenotype, for 654-1082 of 1086 female twins (median age 58.9 years; range 46-77). Trends in dietary analysis revealed a specific pattern of dietary intake, that high in fruit and vegetables, showed an inverse association with hip OA (p = 0.022). Consumption of 'non-citrus fruit' (p = 0.015) and 'alliums' (p = 0.029) had the strongest protective effect. Alliums contain diallyl disulphide which was shown to abrogate cytokine-induced matrix metalloproteinase expression. Conclusions Studies of diet are notorious for their confounding by lifestyle effects. While taking account of BMI, the data show an independent effect of a diet high in fruit and vegetables, suggesting it to be protective against radiographic hip OA. Furthermore, diallyl disulphide, a compound found in garlic and other alliums, represses the expression of matrix-degrading proteases in chondrocyte-like cells, providing a potential mechanism of action
