    ANTISICKLING ACTIVITY EVALUATION OF FRACTIONS OBTAINED FROM WHOLE EXTRACTS OF NEWBOULDIA LAEVIS P. BEAUV (BIGNONIACEAE)

    Objective: This study aims to evaluate the fractions from whole extracts of roots and stem barks of Newbouldia laevis and their effect on sickling. Methods: Hydroethanolic extracts of stem barks and roots of Newbouldia laevis were fractionated by the technics of cold precipitation in ethanol. The fractions obtained after phytochemical screening were subjected to Emmel test to evaluate their anti-sickling activity. Active fractions were tested for DPPH and AAPH assay (AAPH induced membrane lipoperoxidation and evaluation of reduction of hemolysis). Results: Two fractions were obtained from each whole extract: supernatant and pellet fractions. Supernatants fractions obtained from whole roots barks extract and stem barks extract at a concentration of 30 mg/ml reduced sickling up respectively to 7% and 10% against 86% for the control. Pellets fractions obtained from the both extracts induced coagulation of SS blood at 30 mg/ml against 86% for the control. Conclusion: Supernatants fractions of hydroethanolic whole extract of roots and stem backs of Newbouldia laevis promise as the potential source of active molecules against sickle cell disease

    How do Spondias mombin L (Anacardiaceae) leaves extract increase uterine smooth muscle contractions to facilitate child birth in parturient women?

    Background: Spondias mombin L. (Anacardiaceae) leaves were used in Togolese folk to treat dystocia, expel placenta and manage post-partum hemorrhage during child birth.Objectives: This study aimed to establish how the extract of S. mombin leaves increase uterine smooth muscle contractions relevant to its traditional use to facilitate child birth.Methods: Tests were performed on uterus muscle strips from Sprague-Dawley rats. Central portion of uterine horns were dissected, cleaned of surrounding fat and loose connective tissue, and cut longitudinally into strips which were placed in the organ bath for isometric tension record in presence of different substances.Results: S. mombin leaves extract increased uterine spontaneous contractions. This effect was reduced by indomethacin (2 x 10-6 M), yohimbine (2 x 10-6 M) and 2- aminoethoxydiphenyl borate (2-APB) (5 x 10-5 M), but not by atropine (3.45 x 10-8 M) and cholesterol (2.5 mg/ml).Conclusion: The pharmacological justification for the traditional use of S. mombin leaves to treat dystocia and expel placenta was that its hydro-ethanolic extract induced prostaglandins release, α2-adrenoceptors stimulation, calcium release from internal stores and lifted inhibitory effect of cholesterol on uterine contractions in order to increase uterine smooth muscle contractions.Keywords: Spondias mombin, uterus, contractions, calcium

    Hepatoprotective and antioxidant effects of Acanthospermum hispidum (DC) leaves on carbon tetrachloride-induced acute liver damage in rat

    The leaves of Acanthospermum hispidum (DC) are used traditionally by Togolese to treat various hepatic disorders. The aim of the present study was to evaluate the hepatoprotective effect and antioxidant activities of the hydroethanolic extract of A. hispidum on carbon tetrachloride (CCl4)-induced liver damage in Wistar rats. The extract was also studied for its in vitro antioxidant activity using ferric thiocyanate (FTC) and 2,2,-azobis(2-amidinopropane hydrochloride) (AAPH) methods. The hydroethanolic extract of A. hispidum administratered oraly at 250 and 500 mg/kg showed a significant hepatoprotective effect (P < 0.001) by reducing levels of alanine amino transaminase (ALT), aspartate amino transaminase (AST) and alkaline phosphatase (ALP). The extract exhibited significant hepatoprotective and antioxidant activities. These data suggest that the hydroethanolic extracts of A. hispidum can prevent hepatic injuries from CCl4 -induced hepatotoxicity in rats and this is likely mediated through its antioxidant activities.© 2015 International Formulae Group.     Antihyperglycaemic and antioxidant activities of Crataeva adansonii DC. ssp. adansonii leaves extract on ICR mice

    Crataeva adansonii DC. ssp. adansonii is a medicinal plant belong to Capparidaceae family, used traditionally by Togolese to treat diabetes mellitus. The aim of this study was to evaluate the antihyperglycaemic and antioxidant activities of the plant leaves. The effect of hydroethanolic extract of C. adansonii ssp. adansonii leaves was assessed in vivo on blood glucose levels in fasting normal and glucose loaded hyperglycemic (oral glucose tolerance test) mice. The phytochemical screening has been done by coloring tests in chemistry. In vitro antioxidants assays had been performed by 2, 2'-azobis 2 amidinopropane dihydrochloride (AAPH) free radical scavenging assay, Ferric reducing antioxidant power (FRAP) assay, Fe2+ chelating assay and total antioxidant capacity (TAC) quantification. After 30 min glucose loading (4 g/kg), the extract 500 mg/kg was showed a significant (p < 0.001) decreased of the peak of blood glucose compared to the hyperglycemic control. The two doses of extract (250 and 500 mg/kg) did not act on the basic blood glucose compared to the normoglycemic control. Phytochemical screening revealed a presence of flavonoids, tannins, alkaloids and sterols. AAPH free radical scavenging activity, reducing power, Fe2+ chelating activity and total antioxidant capacity have shown dose-dependent antioxidant capacity and reducing power of extract compared to ascorbic acid and Butylated hydroxyl Toluene used as reference drug. These finding prove that C. adansonii ssp. adansonii leaves can be used in the treatment of diabetes mellitus and its complications related to oxidative stress and support the use of the plant in traditional medicine in diabetes treatment.  Keywords: Crataeva adansonii, antihyperglycaemic, phytochemical, antioxidant

    Burn wound healing effects of the root hydroethanolic extract of Cochlospermum planchonii in mice

    Cochlospermum planchonii is a plant traditionally used in Togo to treat burn wounds. The present study aimed at evaluating the effect of the root hydroethanolic extract of Cochlospermum planchonii on the skin burn wounds. Burns were induced in ICR mice by applying on the skin of the dorsal region with a metal stick (0.7 cm diameter) preheated at 100 °C for 10 seconds. Four (4) groups of 8 mice were formed, Control Group (I); Group II, III and IV treated respectively with C. planchonii gel 2.5%, 5%, and Brulex® (Zinc oxide 15% cream). The surfaces of the wounds were evaluated daily by the ImageJ software. Biopsies were performed on day six and twelve for hydroxyproline assays and histological examinations. On the 12th day, wound contraction rates were 44.12%, 67.82%, 66.68% and 69.32% respectively for Group I, Group II, Group III and Group IV. Hydroxyproline levels on day 12 were significantly higher in samples of animals treated with C. planchonii 2.5% and 5% (+114,93 ± 1,61 and +120,54 ± 7,22 of augmentation respectively, P<0.01 when compared to the control group). Histological examination confirmed these observations. C. planchonii root hydroethanolic extract could be a potentially natural remedy for burn wound

    Fetal Toxicity and Cytotoxicity of Lannea kerstingii Engl and Krause Stem Bark (Anacardiaceae)

    Purpose: To evaluate the fetal toxicity and cytotoxicity of L. kerstingii in pregnant rats exposed in the organogenic period.Methods: Mated female rats were randomly assigned to three experimental groups of 8 animals each. Pregnant rats received orally 500 or 1000 mg/kg of 50 % hydroalcohol extract of L. kerstingii, daily from the 17th to the 20th day of gestation. On day 21 of pregnancy, the females were sacrificed. Laparotomy was performed and uterine horns were removed. The number of implants, resorptions, dead and live fetuses were then recorded. The ovaries were also observed and corpora lutea were counted. The cytotoxic effect of L. kerstingii hydroalcohol extract was evaluated on Caco-2 cell lines using MTT (3-(4, 5-dimetylthiazol-2-yl)-2, 5- diphenyltetrazolium bromide) and neutral red uptake assay.Results: No visible signs of toxicity were observed in the female rats and their pups through-out the study period. However, L. kerstingii (500 and 1000 mg/kg) caused a significant decrease (p < 0.01) in fetal weight compared with control. With regard to implantation, resorption and mortality, there was no significant difference between groups. L. kerstingii hydroal-cohol extract (IC50, 29 μg/mL) was more cytotoxic than the aqueous extract (IC50, 141 μg/mL).Conclusion: The administration of hydroalcohol extract of L. kerstingii to female rats in late pregnancy is toxic to the fetus.Keywords: Lannea kerstingii, Fetal toxicity, Cytotoxicity, Mortality, Oxidative stress, Anorexi

    Antidiabetic Activity of Plumeria Alba Linn (Apocynaceae) Root Extract and Fractions in Streptozotocin-Induced Diabetic Rats

    Purpose: To investigate the effect of total hydroalcohol root extract of Plumeria alba (Apocynaceae) and its active fraction on diabetes in rats.Methods: The effect of total hydroalcohol extract at 250 mg/kg and different fractions was evaluated on oral glucose tolerance test (OGTT) in mice. The effect of the active fractions on OGTT was measured on a model of diabetic rats (fructose-enriched fat diet and streptozotocin-induced).Results: The results show that the total extract (250 mg/kg), ethyl acetate fraction and supernatant fraction (obtained after centrifugation of total extract) at a dose of 100 mg/kg significantly (p < 0.01) reduced hyperglycemia induced by glucose overload in mice. Fructose-enriched fat diet increased blood cholesterol, triglycerides and high density lipoprotein (HDL) levels in hyperlipidemic untreated rats compared to normal control rats. Administration of total extract (250 mg/kg/day) and supernatant fraction of P. alba (100 mg/kg/day) during 14 days significantly reduced lipid parameters (total cholesterol, p < 0.001; triglycerides, p < 0.01; HDL, p < 0.05). Analysis of oxidative stress markers shows that the supernatant fraction and total extract significantly increased serum glutathione level (p < 0.01) but significantly lowered malondialdehyde (MDA) concentration in liver (p < 0.05).Conclusion: These results suggest that the total extract and fractions of P. alba exhibit significant antidiabetic and hypolipidemic properties in streptozotocin induced diabetic animals.     PRELIMINARY EVALUATION OF THE WOUND HEALING EFFECT OF VITEX DONIANA SWEET (VERBENACEAE) IN MICE.

    Vitex doniana is traditionally used in Togo to treat various diseases including wounds. The aim of this work was to evaluate the efficiency of Vitex doniana on cutaneous wound healing. Wounds were induced in ICR mice divided into four groups as following: Group I received carbopol 974P NF empty gel, Groups II and III were treated topically with carbopol gel containing 2.5% and 5% of Vitex doniana extract. Group IV received Betadine® 10% as standard drug. The efficacy of treatment was evaluated by planimetry and histological analysis. We secondary used the gel containing Vitex doniana at 2.5% and the pure extract at 10 mg/ml on the model of ear edema induced by xylene. Skin toxicity test was performed with the gel containing Vitex doniana at 5% and the pure extract at 30 mg/ml. Vitex doniana at 5% and 2.5% provided better wound contraction (91.14% and 86.38%) at day 12 post-excision when compared to control (51.15%). The results of histological evaluation supported the outcome of excision wound model. Moreover Vitex doniana inhibited significantly edema induced by xylene when compared to control (p< 0.05). In skin toxicity test, no abnormal symptoms were developed over 14 day-time period. Vitex doniana inhibits the topical inflammation and accelerate cutaneous wound repair
