    Bats that walk: a new evolutionary hypothesis for the terrestrial behaviour of New Zealand's endemic mystacinids.

Mystacina tuberculata is also the last surviving member of Mystacinidae, the only mammalian family endemic to New Zealand (NZ) and a member of the Gondwanan bat superfamily Noctilionoidea. The capacity for true quadrupedal terrestrial locomotion in Mystacina is a secondarily derived condition, reflected in numerous skeletal and muscular specializations absent in other extant bats. The lack of ground-based predatory native NZ mammals has been assumed to have facilitated the evolution of terrestrial locomotion and the unique burrowing behaviour of Mystacina, just as flightlessness has arisen independently many times in island birds. New postcranial remains of an early Miocene mystacinid from continental Australia, Icarops aenae, offer an opportunity to test this hypothesis. RESULTS: Several distinctive derived features of the distal humerus are shared by the extant Mystacina tuberculata and the early Miocene Australian mystacinid Icarops aenae. Study of the myology of M. tuberculata indicates that these features are functionally correlated with terrestrial locomotion in this bat. Their presence in I. aenae suggests that this extinct mystacinid was also adapted for terrestrial locomotion, despite the existence of numerous ground-based mammalian predators in Australia during the early Miocene. Thus, it appears that mystacinids were already terrestrially-adapted prior to their isolation in NZ. In combination with recent molecular divergence dates, the new postcranial material of I. aenae constrains the timing of the evolution of terrestrial locomotion in mystacinids to between 51 and 26 million years ago (Ma). CONCLUSION: Contrary to existing hypotheses, our data suggest that bats are not overwhelmingly absent from the ground because of competition from, or predation by, other mammals. Rather, selective advantage appears to be the primary evolutionary driving force behind habitual terrestriality in the rare bats that walk. Unlike for birds, there is currently no evidence that any bat has evolved a reduced capacity for flight as a result of isolation on islands

    Delivery of Health Education in Adolescents with Behavioral Health Challenges

    BACKGROUND Adolescents with behavioral health issues tend to have inadequate access to health education, and are thus less aware of the importance of personal and dental hygiene, exercise, and healthy diet and lifestyle habits. Due to this disparity, this population has been known to harbor a higher prevalence of STI’s, drug and alcohol abuse, physical altercations, juvenile detention, and suicide attempts. PURPOSE The overall objective of this study was to examine the effect of integrating a health science curriculum in this population. METHODS Participants aged 5-17 years old were recruited and assigned to either control or science groups by Family and Children’s Services (FCS). We created an 8-week health science curriculum to teach everyday life skills. A questionnaire was utilized to assess participants’ comprehension of health information. The effect of the curriculum on participants’ behaviors was examined using a pre/post “Behavioral Insight” questionnaire. Non-parametric Mann-Whitney U test was utilized to determine whether there was a significant difference in behavioral goals and science based knowledge between the control and science groups. RESULTS We examined two measurements to evaluate the efficacy of delivering health science information to 23 participants in two science groups, compared to 20 participants in two control groups who did not receive the curriculum. The health knowledge assessment results demonstrated a marginally significant improvement of understanding and retaining health science, which was delivered to the science group in 8 independent sessions on a weekly basis (p=0.0669). We observed a statistically significant difference in participants’ understanding of their behavioral deficits and how to improve primary behavior (p=0.01). There was no significant difference in either group regarding the extent to which participants understand the particular behavior to improve (p\u3e0.05). CONCLUSION Findings from our study demonstrate that integration of an 8-week science curriculum into life skill training did not negatively affect participants’ understanding of their own behaviors. Compared to the control group, we observed a marginally significant improvement in health knowledge assessment scores among students in the science group. Finally, the 8-week science curriculum, both active and passive learning components, was determined to be an effective delivery method for the content involved. Further study in a larger sample may be necessary to detect significant effects of the curriculum

    Burnout menjadi salah satu gangguan psikologis yang rentan dihadapi oleh perawat dalam perannya memberikan perawatan pasien di ruang isolasi Coronavirus Disease 2019. Tujuan penelitian untuk mengetahui gambaran burnout perawat di ruang isolasi Coronavirus Disease 2019. Jenis penelitian ini adalah kuantitatif dengan desain deskriptif survei pada 60 perawat di Unit Pelaksana Teknis Rumah Sakit Umum Daerah Bali Mandara melalui purposive sampling. Penelitian ini menggunakan Maslach Burnout Inventori sebagai alat pengumpulan data yang terdiri dari 21 pernyataan. Data dianalisis secara univariat dan disajikan dalam distribusi frekuensi. Hasil analisis mendapatkan burnout perawat di ruang isolasi Coronavirus Disease 201ada pada kategori rendah yaitu 75%, sedang 20%, dan cukup tinggi 5%. Mayoritas burnout perawat di ruang isolasi Coronavirus Disease 2019 berada pada kategori rendah. Berdasarkan dimensi burnout, sebagian besar perawat mengalami kelelahan emosional rendah, depersonalisasi rendah, dan penurunan pencapaian diri rendah.     Thrombin Induces Macrophage Migration Inhibitory Factor Release and Upregulation in Urothelium: A Possible Contribution to Bladder Inflammation

    Macrophage migration inhibitory factor (MIF) is a pro-inflammatory cytokine expressed by urothelial cells that mediates bladder inflammation. We investigated the effect of stimulation with thrombin, a Protease Activated Receptor-1 (PAR1) agonist, on MIF release and MIF mRNA upregulation in urothelial cells.MIF and PAR1 expression was examined in normal human immortalized urothelial cells (UROtsa) using real-time RT-PCR, Western blotting and dual immunostaining. MIF and PAR1 immunostaining was also examined in rat urothelium. The effect of thrombin stimulation (100 nM) on urothelial MIF release was examined in UROtsa cells (in vitro) and in rats (in vivo). UROtsa cells were stimulated with thrombin, culture media were collected at different time points and MIF amounts were determined by ELISA. Pentobarbital anesthetized rats received intravesical saline (control), thrombin, or thrombin +2% lidocaine (to block nerve activity) for 1 hr, intraluminal fluid was collected and MIF amounts determined by ELISA. Bladder or UROtsa MIF mRNA was measured using real time RT-PCR.UROtsa cells constitutively express MIF and PAR1 and immunostaining for both was observed in these cells and in the basal and intermediate layers of rat urothelium. Thrombin stimulation of urothelial cells resulted in a concentration- and time-dependent increase in MIF release both in vitro (UROtsa; 2.8-fold increase at 1 hr) and in vivo (rat; 4.5-fold) while heat-inactivated thrombin had no effect. In rats, thrombin-induced MIF release was reduced but not abolished by intravesical lidocaine treatment. Thrombin also upregulated MIF mRNA in UROtsa cells (3.3-fold increase) and in the rat bladder (2-fold increase) where the effect was reduced (1.4-fold) by lidocaine treatment.Urothelial cells express both MIF and PAR1. Activation of urothelial PAR1 receptors, either by locally generated thrombin or proteases present in the urine, may mediate bladder inflammation by inducing urothelial MIF release and upregulating urothelial MIF expression

    Quantification of Cell Signaling Networks Using Kinase Activity Chemosensors

    The ability to directly determine endogenous kinase activity in tissue homogenates provides valuable insights into signaling aberrations that underlie disease phenotypes. When activity data is collected across a panel of kinases, a unique “signaling fingerprint” is generated that allows for discrimination between diseased and normal tissue. Here we describe the use of peptide-based kinase activity sensors to fingerprint the signaling changes associated with disease states. This approach leverages the phosphorylation-sensitive sulfonamido-oxine (Sox) fluorophore to provide a direct readout of kinase enzymatic activity in unfractionated tissue homogenates from animal models or clinical samples. To demonstrate the application of this technology, we focus on a rat model of nonalcoholic fatty liver disease (NAFLD). Sox-based activity probes allow for the rapid and straightforward analysis of changes in kinase enzymatic activity associated with disease states, providing leads for further investigation using traditional biochemical approaches

    Lipoteichoic acid is involved in the ability of the immunobiotic strain lactobacillus plantarum CRL1506 to modulate the intestinal antiviral innate immunity triggered by TLR3 activation

    Studies have demonstrated that lipoteichoic acid (LTA) is involved in the immunomodulatory properties of some immunobiotic lactobacilli. The aim of this work was to evaluate whether LTA contributes to the capacity of Lactobacillus plantarum CRL1506 in modulating the intestinal innate antiviral immune response. A D-alanyl-lipoteichoic acid biosynthesis protein (dltD) knockout CRL1506 strain (L. plantarumΔdltD) was obtained, and its ability to modulate Toll-like receptor (TLR)-3-mediated immune response was evaluated in vitro in porcine intestinal epithelial (PIE) cells and in vivo in Balb/c mice. Wild-type (WT) CRL1506 (L. plantarum WT) was used as positive control. The challenge of PIE cells with the TLR3 agonist poly(I:C) significantly increased interferon (IFN)-β, interleukin (IL)-6, and monocyte chemoattractant protein (MCP)-1 expressions. PIE cells pretreated with L. plantarumΔdltD or L. plantarum WT showed higher levels of IFN-β while only L. plantarum WT significantly reduced the expression of IL-6 and MCP-1 when compared with poly(I:C)-treated control cells. The oral administration of L. plantarum WT to mice prior the intraperitoneal injection of poly(I:C) significantly increased IFN-β and IL-10 and reduced intraepithelial lymphocytes (CD3+NK1.1+CD8αα+) and pro-inflammatory mediators (TNF-α, IL-6, and IL-15) in the intestinal mucosa. Similar to the WT strain, L. plantarumΔdltD-treated mice showed enhanced levels of IFN-β after poly(I:C) challenge. However, treatment of mice with L. plantarumΔdltD was not able to increase IL-10 or reduce CD3+NK1.1+CD8αα+ cells, TNF-α, IL-6, or IL-15 in the intestine. These results indicate that LTA would be a key molecule in the anti-inflammatory effect induced by the CRL1506 strain in the context of TLR3-mediated inflammation.Fil: Mizuno, Hiroya.     Nuevas fuentes de antioxidantes naturales: caracterización de compuestos bioactivos en cinco frutos nativos de Chile.

    Diferentes berries de la zona centro y sur de Chile fueron analizados con el fin de buscar fuentes promisorias de polifenoles con clara actividad sobre la salud humana. Se estudiaron cinco bayas nativas: arrayan, frutilla blanca, murtilla y calafate, y un berry tradicional (uva tintorera). Se determinó in vitro sus propiedades antioxidantes según el ensayo de polifenoles totales de Folin Ciocalteu, antocianinas por pH diferencial, capacidad antioxidante por medición de capacidad de reducción del radical libre 2.2-difenil-1picrilhidracilo (DPPH) y poder de reducción férrica (FRAP) y perfil químico por HPLC-DAD. De los cinco berries, calafate registró el valor más alto (1066,4 ± 24,9 mg GAE/100g de muestra) para polifenoles totales y antocianinas (1031,9 ± 48,1 mg de cianidina-3-glucósido/100g de muestra) seguido por uva tintorera. Calafate presentó excelente poder reductor (11279,2 ± 2027,4 umol Trolox/100g ensayo FRAP y 5235,0 ± 445,9, umol/100g en PPH). El perfil químico de antocianinas mostró delfinidina, cianidina, malvidina, petunidina, peonidina y pelargonidina en los cinco berries. Alrededor de 30 flavonoles derivados de quercetina, myricetina e isorhamnetina fueron identificados así cómo elagitaninos presentes en frutilla blanca, compuestos muy interesantes para estudios posteriores. Estos resultados contribuyen a destacar el uso potencial de estos berries como alimentos funcionales.Berries from central and southern Chile were analyzed in order to find promising sources of polyphenols with clear activity on human health. Five native fruits like arrayan, white strawberry, murtilla and calafate, and a traditional berry (grape called “tintorera”) were studied. Antioxidant properties were determined in vitro according to the total polyphenol assay of Folin Ciocalteu, anthocyanins by differential pH, antioxidant capacity by measuring the capacity of reduction of the free radical 2.2-diphenyl-1-picrylhydracil (DPPH) and iron reducing capacity (FRAP) and chemical profile by HPLC-DAD. Calafate showed the highest value (1066.4 ± 24.9 mg gallic acid / 100g sample) for total polyphenols and anthocyanins (1031.9 ± 48.1 mg of cyanidin-3-glucoside / 100g of sample) followed by blue grape. Calafate displayed excellent reducing power (11279.2 ± 2027.4 umol Trolox / 100g FRAP assay and 5235.0 ± 445.9, umol / 100g in DPPH), followed by grape. The chemical profile ofanthocyanins showed delphinidin, cyanidin, malvidin, petunidin, peonidin and pelargonidin in the five berries. Around 30 flavonols derived from quercetin, yricetin and sorhamnetin were identified as well as ellagitannins present in white trawberry, very interesting compounds for further studies. These results contribute to highlight the potential use of these berries as functional foods
