    Interactions between nutritional and opioidergic pathways in the control of LH secretion in male sheep

    Our aim was to determine the role of opioidergic processes in the effects of nutrition on the secretion of LH pulses in the mature male sheep. In the first of three experiments, adult Merino rams were acclimatised to a maintenance diet and then allocated to one of three dietary groups (n = 5): continuation of the maintenance diet (Group M); reduction to half of the maintenance allocation (Group HM); or supplementation of the maintenance diet with lupin grain (Group HD). An initial administration of naloxone (2 mg/kg body weight, i.v.) was followed at 40-min intervals by three further administrations (1 mg/kg). Blood was sampled every 20 min for 12 h before the initial naloxone administration and then for a further 6 h. LH pulse frequency after naloxone treatment was significantly higher in Group HD than in Group HM (P < 0.05). The second study tested whether the response to naloxone depended on calcium status. We used 22 adult Merino rams in two consecutive experiments, one in which the rams were fed a maintenance diet, and one in which the rams were fed with the maintenance diet plus 1 kg lupin grain for 5 weeks. In both experiments, rams were allocated to groups that received one of the following treatments: (a) 0.02 g/kg calcium borogluconate + 0.2 mg/kg naloxone hydrochloride (Nal + Ca2+; n = 6); (b) 0.2 mg/kg naloxone hydrochloride (Nal; n = 6); (c) 0.02 g/kg calcium borogluconate (Ca2+; n = 5); (d) 0.1 ml/kg NaCl 0.9% (Saline; n = 5). All treatments were given as a single i.v. administration daily for 5 days. Blood was sampled every 20 min for 24 h during the acclimatization period (Day 0) and on the last day (Day 5) of treatment. In the first study (under maintenance), none of the treatments affected LH pulse frequency. In the second study (the lupin-supplemented rams), LH pulse frequency was significantly increased (P < 0.05) by the administration of naloxone + Ca2+, naloxone alone and Ca2+ alone. Overall, rams on a low plane of nutrition showed the smallest response to naloxone, suggesting that an opioidergic mechanism is not involved in the suppressive effect of restricted nutrition on the gonadotrophic axis. Rather, because testosterone secretion was increased on the high plane of nutrition, the LH responses to naloxone are better explained by the effects of testosterone on opioidergic mechanisms. Finally, we failed to observe any interaction between opioids and calcium in the control of LH secretion

    Flour proteins linked to quality traits in an Australian doubled haploid wheat population

    The Cranbrook/Halberd doubled haploid population has provided a unique opportunity to examine in detail the contributions made by a number of different high molecular weight (HMW) and low molecular weight (LMW) glutenin alleles to the dough properties in a set of homogeneous lines of wheat. A range of different instruments was employed, including Farinograph, Extensograph, Do-Corder, Resistograph, and GRL/EasyMix, to study the dough rheology of the lines from 3 sites over 2 years. Correlation studies showed that 2 basic parameters (dough strength and extensibility) were measured by these different instruments. The results presented are mainly from the Extensograph, which is a major Australian standard for determining release and marketing classification of Australian wheats. Approaches to investigate the data include bulk segregant analysis, distribution of protein alleles in the population, and multiple linear regression. As expected, the HMW glutenin alleles made a major contribution to dough strength, with a minor, but not insignificant, contribution from the LMW glutenin alleles. From a knowledge of their glutenin alleles, a glutenin strength score (GSS) was devised to allow breeders to rank the dough strength of various lines. The GSS scoring system is based on both HMW and LMW glutenin alleles, adding to a total out of 10. Extensibility, on the other hand, was predominantly influenced by protein levels in the flour and environmental conditions such as site and season. However, the LMW glutenin alleles make a significant genetic contribution to the extensibility, which can be assessed by using a glutenin extensibility score. These two glutenin quality scores currently include only the alleles present in the parents, Cranbrook and Halberd, but this could be expanded to include a wider range of alleles by analysis of the quality data from other doubled haploid populations. These quality scores would then be an extremely useful tool for assessing the potential quality of parental and early generation germplasm in wheat breeding programs, by a knowledge of the allelic composition of their HMW and LMW glutenins

    Photo--assisted current and shot noise in the fractional quantum Hall effect

    The effect of an AC perturbation on the shot noise of a fractional quantum Hall fluid is studied both in the weak and the strong backscattering regimes. It is known that the zero-frequency current is linear in the bias voltage, while the noise derivative exhibits steps as a function of bias. In contrast, at Laughlin fractions, the backscattering current and the backscattering noise both exhibit evenly spaced singularities, which are reminiscent of the tunneling density of states singularities for quasiparticles. The spacing is determined by the quasiparticle charge νe\nu e and the ratio of the DC bias with respect to the drive frequency. Photo--assisted transport can thus be considered as a probe for effective charges at such filling factors, and could be used in the study of more complicated fractions of the Hall effect. A non-perturbative method for studying photo--assisted transport at ν=1/2\nu=1/2 is developed, using a refermionization procedure.Comment: 14 pages, 6 figure

    The optimum slaughter weight for different ewe mature sizes

    Lamb producers have the option to market lambs at a range of slaughter weights. However, there are limited price premiums for heavier carcasses on a per kilogram basis. Any economic advantage of heavy lambs is realised by extra weight and not price. Both genetic and on-farm factors contribute to extra weight gain. Firstly, lamb weight and growth is correlated to its mature size and lambs from larger parents grow faster and reach heavier weights, but also have greater feed requirements. Secondly, stocking and reproductive rate account for the majority of variation in whole-farm profit, but increasing these also increases feed requirements. The production of heavy lambs is therefore a trade-off with maximising stocking and reproductive rate within the pool of available feed resources. We hypothesise that slaughter weight does not increase with mature size, due to the priority to increase stocking and reproductive rate for profit maximisation

    Far-infrared vibrational properties of high-pressure-high-temperature C60 polymers and the C60 dimer

    We report high-resolution far-infrared transmission measurements of the 2 + 2 cycloaddition C-60 dimer and two-dimensional rhombohedral and one-dimensional orthorhombic high-pressure high-temperature C60 polymers. In the spectral region investigated(20-650 cm(-1)), we see no low-energy interball modes, but symmetry breaking of the linked C-60 balls is evident in the complex spectrum of intramolecular modes. Experimental features suggest large splittings or frequency shifts of some IhC60-derived modes that are activated by symmetry reduction, implying that the balls are strongly distorted in these structures. We have calculated the vibrations of all three systems by first-principles quantum molecular dynamics and use them to assign the predominant IhC60 symmetries of observed modes. Pur calculations show unprecedentedly large downshifts of T-1u(2)-derived modes and extremely large splittings of other modes, both of which are consistent with the experimental spectra. For the rhombohedral and orthorhombic polymers, the T-1u(2)-derived mode that is polarized along the bonding direction is calculated to downshift below any T-1u(1)-derived modes. We also identify a previously unassigned feature near 610 cm(-1) in all three systems as a widely split or shifted mode derived from various silent IhC60 vibrations, confirming a strong perturbation model for these linked fullerene structures

    The direct Cu NQR Study of the Stripe Phase in the Lanthanum Cuprates

    Using Cu NQR in Eu-doped La_(2-x)Sr_xCuO_4 we find the evidence of the pinned stripe phase at 1.3K for 0.08<x<0.18. The pinned fraction increases by one order of magnitude near hole doping x=1/8. The NQR lineshape reveals three inequivalent Cu positions.     Distortion in a 7xxx aluminum alloy during liquid phase sintering

    The distortion in a sintered 7xxx aluminum alloy, Al-7Zn-2.5Mg-1Cu (wt. pct), has been investigated by sintering three rectangular bars in each batch at 893 K (620 °C) for 0 to 40 minutes in nitrogen, followed by air or furnace cooling. They were placed parallel to each other, equally spaced apart at 2 mm, with their long axes being perpendicular to the incoming nitrogen flow. Pore evolution in each sample during isothermal sintering was examined metallographically. The compositional changes across sample mid-cross section and surface layers were analyzed using energy dispersive X-ray spectroscopy and X-ray photoelectron spectroscopy depth profiling, respectively. The two outer samples bent toward the middle one, while the middle sample was essentially distortion free after sintering. The distortion in the outer samples was a result of differential shrinkage between their outer and inner surfaces during isothermal sintering. The porous outer surface showed an enrichment of oxygen around the large pores as well as lower magnesium and zinc contents than the interior and inner surface of the same sample, while the inner surface was distinguished by the presence of AlN. The differential shrinkage was caused by different oxygen contents in local sintering atmosphere and unbalanced loss of magnesium and zinc between the outer and inner surfaces

    Suppression of static stripe formation by next-neighbor hopping

    We show from real-space Hartree-Fock calculations within the extended Hubbard model that next-nearest neighbor (t') hopping processes act to suppress the formation of static charge stripes. This result is confirmed by investigating the evolution of charge-inhomogeneous corral and stripe phases with increasing t' of both signs. We propose that large t' values in YBCO prevent static stripe formation, while anomalously small t' in LSCO provides an additional reason for the appearance of static stripes only in these systems.Comment: 4 pages, 5 figure

    Positive cross-correlations induced by ferromagnetic contacts

    Due to the Fermionic nature of carriers, correlations between electric currents flowing through two different contacts attached to a conductor present a negative sign. Possibility for positive cross-correlations has been demonstrated in hybrid normal/superconductor structures under certain conditions. In this paper we show that positive cross-correlations can be induced, if not already present, in such structures by employing ferromagnetic leads with magnetizations aligned anti-parallel to each other.     A genetic management toolkit: hormonal and behavioural research towards the development of artificial insemination technology in the African wild dog (Lycaon pictus)

    African wild dogs have a complex pack structure with separate male and female dominance hierarchies in which reproduction is typically exclusive to the alpha male and female. Current efforts to maintain genetic diversity involve translocation of live animals in captivity, a process that involves the combination of male and female single-sex groups to form a new social pack. Due to their complex social structure, such introductions are difficult: regularly leading to aggression and injuries to the animals. Moreover, this may not result in breeding among genetically valuable individuals if they do not become dominant. To overcome these translocation-associated problems of intra-pack aggression our team has studied a number of facets of basic reproduction as well as stress and behavioural management in African wild dogs with the goal of developing strategies for semen cryopreservation and artificial insemination as a key to the introduction of new genes into existing stable packs (Van den Berghe et al. 2012, Anim Reprod Sci 133, 1-9). We began by establishing detailed reproductive and adrenal hormone profiles from multiple females in several reproductive states in both captive and free-ranging populations. Faecal samples from captive females in four European institutions and from wild males and females in South Africa were analysed for oestradiol, progestagens, testosterone and glucocorticoids. Captive data demonstrated that female wild dogs have reproductive characteristics similar to other canids, such as spontaneous ovulation and obligate pseudopregnancy. Almost all adult females became pseudopregnant, implying that the mechanism of reproductive suppression in this sex is likely to be behavioural rather than physiological. This was supported by data from free-ranging populations. Adrenal activity in captive and free-ranging animals was measured by faecal cortisol metabolites. Captive females had higher concentrations than free-ranging females. Glucocorticoid concentrations were more variable in captive individuals, whereas in both wild males and females, they were highest during gestation and denning. Adrenal activity in males, but not females, was also affected by age, with the highest glucocorticoid concentrations measured in yearlings. We have also investigated the use of dog appeasing pheromone (DAP) collars to down-regulate aggression associated with the formation of new packs in captivity. Our initial findings indicate that DAP may reduce baseline stress levels in females and, when applied during regrouping of same-sex individuals, results in a relative reduction in observed levels of aggression. Building on these findings, work is underway to develop a semen bank and artificial insemination techniques. Although reproductive suppression in females is likely to be behavioural, the effect of dominance on male fertility is still unresolved. We have begun investigating the relationship between hierarchy and sperm quality across the seasons. This will guide our development of a robust semen collection and freezing protocol that will permit the establishment of a high-quality semen bank. We are also progressing with non-invasive techniques to predict ovulation for timed artificial insemination of females. These techniques must first be established in captivity before our ultimate goal to implement them as tools for genetic meta-population management in semi-captive and free-ranging populations
