    Determination of the neighboring molecule to the FC receptor on human macrophages

    Chemical cross-linking studies have been carried out to investigate near neighbors to the receptor for immunoglobulin G[lgG] on U937 cells both before and after solubilization of the receptor. A cross-linked product with a molecular weight of 350.000 daltons was achieved with. DMS on intact and lysed U937 cells, DTBP on intact cells and DSS on cell lysates, with anti-FcR as the immunoprecipitation reagent. Following cross-linking of the U937 cells with DTBP and analysis by two-dimensional gel electrophoresis the p350 molecule appeared to be a p 1 70 dimer. The pl70 molecular has been related to some nonspecificity of the anti-FcR. These procedures were thus unsuccessful in identifying a neighboring molecule to the U937 cell FcR. DSS cross-linking of Fc fragments of IgG and Fab fragments of anti-FcR to the FcR on intact U937 cells was unsuccessful. Lack of success in this regard is more likely attributed to difficulties in DSS usage then to inability to cross-link close-lying molecules to the FcR. The investigations presented clarify the problems that must be overcome before successful cross-linking can be achieved between the U937 cell FcR and its neighboring molecule [s]

The enzymatic hydrolysis creates the H antigen. This modification transforms the immune response to the blood group from Type A to O. The product of the enzyme treatment produces Type O blood a universally transfusable product for potential medical use. Purified recombinant α-NAGAs from E. coli and S. linguale were characterized in vitro for molecular mass, substrate specificity, pH and temperature optima, and product inhibition. A mutant of the S. linguale α-NAGA, H225A, was evaluated for activity in vitro and in vivo. S. linguale enzyme was characterized in vivo using Type A1 and A2 RBCs for pH, temperature, and buffer optima. In vitro each enzyme appeared to function as a dimer under the conditions tested, was highly active at a neutral pH, and over a range of temperatures. Turnover rate, kcat, 33 s-1 (E. coli) and 173 s-1 (S. linguale) were derived from the kinetic assays. Product competitive inhibition, Ki 0.18 mM, was evident with the S. linguale α-NAGA while the E. coli α-NAGA demonstrated uncompetitive inhibition, Ki 2.6 mM. The E. coli α-NAGA did not appear to covert Type A RBCs. S. linguale α-NAGA appeared to convert Type A RBCs to blood group O with efficiency in glycine or alanine buffers at neutral pH at 4°C or 25°C. Pretreating RBCs with glycine solution 24 hours before enzyme conversion appeared to facilitate higher enzyme efficiency. The S. linguale α-NAGA H225A mutant was inactive both in vitro and in vivo. The S. linguale α-NAGA appeared to possess desirable attributes to enzymatically convert RBCs. The use of Type A converted RBCs by S. linguale α-NAGA could potentially increase the universal blood supply by 40%.Includes bibliographical reference

Neste artigo propõe-se sondar acerca de como sua orientação sexual e condição sorológica podem ter influenciado na maneira dele compor figuras masculinas. Observa-se que a atitude de Raul estava em alinhamento à de outros artistas da época, que ficaram notórios por expressar o confronto com a aids. Mas, em que medida isso estaria indicado na postura do artista e em suas figurações masculinas? Para investigar sobre Raul Cruz e parte de sua produção naquele conturbado momento histórico, ampara-se nos conceitos foucaultianos de resistência e contraconduta, somado aos escritos de Susan Sontag referentes à crise da aids.
