    Concentration and localization of zinc during seed development and germination in wheat

    In a field experiment, the effect of foliar Zn applications on the concentration of Zn in seeds of a bread wheat cultivar (Triticum aestivum L. cv. Balatilla) was studied during different stages of seed development. In addition, a staining method using dithizone (DTZ: diphenyl thiocarbazone) was applied to (1) study the localization of Zn in seeds, (2) follow the remobilization of Zn during germination, and (3) develop a rapid visual Zn screening method for seed and flour samples. In all seed development stages, foliar Zn treatments were effective in increasing seed Zn concentration. The highest Zn concentration in the seeds was found in the first stage of seed development (around the early milk stage); after this, seed Zn concentration gradually decreased until maturity. When reacting with Zn, DTZ forms a redcolored complex. The DTZ staining of seed samples revealed that Zn is predominantly located in the embryo and aleurone parts of the seeds. After 36 h of germination, the coleoptile and roots that emerged from seeds showed very intensive red color formation and had Zn concentrations up to 200 mg kg1, indicating a substantial remobilization of Zn from seed pools into the developing roots (radicle) and coleoptile. The DTZ staining method seems to be useful in ranking flour samples for their Zn concentrations. There was a close relationship between the seed Zn concentrations and spectral absorbance of the methanol extracts of the flour samples stained with DTZ. The results suggest that (1) accumulation of Zn in seeds is particularly high during early seed development, (2) Zn is concentrated in the embryo and aleurone parts, and (3) the DTZ staining method can be used as a rapid, semiquantitative method to estimate Zn concentrations of flour and seed samples and to screen genotypes for their Zn concentrations in seeds

    Introduction and evaluation of the wood preservative potentials of the poisonous Sternbergia candidum extracts

    The ability of Sternbergia candida (SB Candidum Mathew.) extract to suppress attack by Postia placenta (Fries) M. Larsen et Lombard (Mad 698), (a brown-rot) and Trametes versicolor (L.ex Fr.) Quel.(a white-rot) was investigated. The extract was dissolved from Sterinbergia Candidum’s bulb and leaves in 96% ethyl alcohol. Poisonous extract impregnated into wood blocks of Turkish oriental beech (Fagus orientalis L.) and Scots pine (Pinus sylvestris L.). Extract dosages were 0.25, 0.75, 1.50 and 3.00%. Treated blocks were exposed to P. placenta and T. versicolor attack for 12 weeks by the soil block method. Only extract dosages of 0.25 and 0.75% were found efficacious in suppressing attack. A tukey studentized test found both dosages significant at 5% level. In conclusion, the poisonous extracts could be used as effective wood preservative when concentration was adjusted

    Inferring Dynamic User Interests in Streams of Short Texts for User Clustering

    User clustering has been studied from different angles. In order to identify shared interests, behavior-based methods consider similar browsing or search patterns of users, whereas content-based methods use information from the contents of the documents visited by the users. So far, content-based user clustering has mostly focused on static sets of relatively long documents. Given the dynamic nature of social media, there is a need to dynamically cluster users in the context of streams of short texts. User clustering in this setting is more challenging than in the case of long documents, as it is difficult to capture the users’ dynamic topic distributions in sparse data settings. To address this problem, we propose a dynamic user clustering topic model (UCT). UCT adaptively tracks changes of each user’s time-varying topic distributions based both on the short texts the user posts during a given time period and on previously estimated distributions. To infer changes, we propose a Gibbs sampling algorithm where a set of word pairs from each user is constructed for sampling. UCT can be used in two ways: (1) as a short-term dependency model that infers a user’s current topic distribution based on the user’s topic distributions during the previous time period only, and (2) as a long-term dependency model that infers a user’s current topic distributions based on the user’s topic distributions during multiple time periods in the past. The clustering results are explainable and human-understandable, in contrast to many other clustering algorithms. For evaluation purposes, we work with a dataset consisting of users and tweets from each user. Experimental results demonstrate the effectiveness of our proposed short-term and long-term dependency user clustering models compared to state-of-the-art baselines

    Macrophage Subset Sensitivity to Endotoxin Tolerisation by Porphyromonas gingivalis

    Macrophages (MΦs) determine oral mucosal responses; mediating tolerance to commensal microbes and food whilst maintaining the capacity to activate immune defences to pathogens. MΦ responses are determined by both differentiation and activation stimuli, giving rise to two distinct subsets; pro-inflammatory M1- and anti-inflammatory/regulatory M2- MΦs. M2-like subsets predominate tolerance induction whereas M1 MΦs predominate in inflammatory pathologies, mediating destructive inflammatory mechanisms, such as those in chronic P.gingivalis (PG) periodontal infection. MΦ responses can be suppressed to benefit either the host or the pathogen. Chronic stimulation by bacterial pathogen associated molecular patterns (PAMPs), such as LPS, is well established to induce tolerance. The aim of this study was to investigate the susceptibility of MΦ subsets to suppression by P. gingivalis. CD14hi and CD14lo M1- and M2-like MΦs were generated in vitro from the THP-1 monocyte cell line by differentiation with PMA and vitamin D3, respectively. MΦ subsets were pre-treated with heat-killed PG (HKPG) and PG-LPS prior to stimulation by bacterial PAMPs. Modulation of inflammation was measured by TNFα, IL-1β, IL-6, IL-10 ELISA and NFκB activation by reporter gene assay. HKPG and PG-LPS differentially suppress PAMP-induced TNFα, IL-6 and IL-10 but fail to suppress IL-1β expression in M1 and M2 MΦs. In addition, P.gingivalis suppressed NFκB activation in CD14lo and CD14hi M2 regulatory MΦs and CD14lo M1 MΦs whereas CD14hi M1 pro-inflammatory MΦs were refractory to suppression. In conclusion, P.gingivalis selectively tolerises regulatory M2 MΦs with little effect on pro-inflammatory CD14hi M1 MΦs; differential suppression facilitating immunopathology at the expense of immunity

    Crystals of β-Zn3(BO3)(PO4) have been grown by the Kyropoulos method. The asymmetric unit contains three Zn sites, three B-atom sites (all with symmetry 3), two P sites (both with m symmetry) and nine O-atom sites (four with m symmetry). The fundamental building units of the title structure are isolated BO3 triangles and PO4 tetra­hedra, which are bridged by ZnO4 tetra­hedra or ZnO5 trigonal bipyramids through common O atoms, leading to a three-dimensional framework structure. Some significant structural differences between the β-polymorph and the α-polymorph are discussed

    Hard Occlusions in Visual Object Tracking

    Visual object tracking is among the hardest problems in computer vision, as trackers have to deal with many challenging circumstances such as illumination changes, fast motion, occlusion, among others. A tracker is assessed to be good or not based on its performance on the recent tracking datasets, e.g., VOT2019, and LaSOT. We argue that while the recent datasets contain large sets of annotated videos that to some extent provide a large bandwidth for training data, the hard scenarios such as occlusion and in-plane rotation are still underrepresented. For trackers to be brought closer to the real-world scenarios and deployed in safety-critical devices, even the rarest hard scenarios must be properly addressed. In this paper, we particularly focus on hard occlusion cases and benchmark the performance of recent state-of-the-art trackers (SOTA) on them. We created a small-scale dataset containing different categories within hard occlusions, on which the selected trackers are evaluated. Results show that hard occlusions remain a very challenging problem for SOTA trackers. Furthermore, it is observed that tracker performance varies wildly between different categories of hard occlusions, where a top-performing tracker on one category performs significantly worse on a different category. The varying nature of tracker performance based on specific categories suggests that the common tracker rankings using averaged single performance scores are not adequate to gauge tracker performance in real-world scenarios.Comment: Accepted at ECCV 2020 Workshop RLQ-TO

    MicroRNAs: shortcuts in dealing with molecular complexity?

    Recent studies from Clarke's group published in the journal Cell indicate that miRNAs may be the elusive universal stem cell markers that the field of cancer stem cell biology has been seeking. Distinct profiles of miRNAs appear to reflect the state of cell differentiation not only in breast cancer cells, but also in normal mammary epithelial cells. Moreover, they are conserved across tissues and species. The authors of this work also show evidence that downregulation of miRNA-200c in normal and malignant breast stem cells and in embryonal carcinoma cells has functional relevance, being responsible for the proliferative potential of these cells in vitro and in vivo

    Meeting Report: Hackathon-Workshop on Darwin Core and MIxS Standards Alignment

    The Global Biodiversity Information Facility and the Genomic Standards Consortium convened a joint workshop at the University of Oxford, 27–29 February 2012, with a small group of experts from Europe, USA, China and Japan, to continue the alignment of the Darwin Core with the MIxS and related genomics standards. Several reference mappings were produced as well as test expressions of MIxS in RDF. The use and management of controlled vocabulary terms was considered in relation to both GBIF and the GSC, and tools for working with terms were reviewed. Extensions for publishing genomic biodiversity data to the GBIF network via a Darwin Core Archive were prototyped and work begun on preparing translations of the Darwin Core to Japanese and Chinese. Five genomic repositories were identified for engagement to begin the process of testing the publishing of genomic data to the GBIF network commencing with the SILVA rRNA database
