Debugging experiment machinery through time-course event sequence analysis

    Get PDF
This application note describes an open-source web application software package for viewing and analysing time-course event sequences in the form of log files containing timestamps. Web pages allow the visualisation of time-course event sequences as time curves and the comparison of sequences against each other to visualise deviations between the timings of the sequences. A feature allows the analysis of the sequences by parsing selected sections with a support vector machine model that heuristically calculates a value for the likelihood of an error occurring based on the textual output in the log files. This allows quick analysis for errors in files with large numbers of log events. The software is written in ASP.NET with Visual Basic code-behind to allow it to be hosted on servers and integrated into web application frameworks

[RuCl2(η6 ‑p‑cymene)(P*)] and [RuCl2(κ-P*‑η6 ‑arene)] Complexes Containing P‑Stereogenic Phosphines. Activity in Transfer Hydrogenation and Interactions with DNA

The preparation of a series of half-sandwich ruthenium complexes, [RuCl2(η6-p-cymene)(P*)] (P* = SPMeRR′) and [RuCl2(κ-P*-η6-arene)], containing P-stereogenic phosphines is reported. The borane-protected Pstereogenic phosphines have been obtained by addition of the (H3B)PMe2R (R = t-Bu (1), Cy (2), Fc (3))/sec-BuLi/ (−)-sparteine adduct to benzyl halides, carbonyl functions, and epoxides with yields between 40 and 90% and ee values in the 70−99% range. Those containing an aryl secondary function have been used in the preparation of [RuCl2(η6 -p-cymene)-(P*)] complexes. Borane deprotection has been performed using HBF4, except for (H3B)PRMe(CH2SiMe2Ph) phosphines, where DABCO was used to avoid partial cleavage of the CH2− Si bond. In the case of (H3B)P(t-Bu)Me(CH2C(OH)Ph2) (1l) the dehydrated phosphine was obtained. The tethered complexes were obtained by p-cymene substitution in chlorobenzene at 120 °C, except for ferrocenyl-containing complexes, which decomposed upon heating. The presence of substituents in the aryl arm of some of the phosphines introduces new chiral elements in the tethered [RuCl2(κ-P*-η6-arene)] compounds. Full characterization of all compounds both in solution and in the solid state has been carried out. Crystal structure determinations of four phosphine−borane molecules confirm the S configuration at the phosphorus atom (1a,e,l and 2d). Moreover, the crystal structure of one p-cymene complex (5i) and four tethered complexes reveal the strain of the compounds with two atoms in the tether (7c,g,l and 8i). Tethering has a marked effect on the catalytic performance transfer hydrogenation of acetophenone and on the nature of hydridic species originating during the activation period. The chiral induction attains 58% ee with complexes with the bulkiest substituents in the pendant arm of the phosphine. Three of the prepared complexes can interact with DNA and present a reasonable cytotoxicity toward cancer cells. Intercalation of the free aromatic pendant arm of the phosphines seems to be fundamental for such interactions

    Get PDF
This study aimed to determine the impact of sex on the differences and relationships of the one repetition maximum (1RM) performance between 3 overhead pressing exercises (push press [PP], push jerk [PJ] and split jerk [SJ]). 15 men (body mass [BM]: 82.3 ± 9.9 kg; weightlifting training experience: 2.6 ± 1.6 years) and 15 women (BM: 64.4 ± 7.0 kg; weightlifting training experience: 2.2 ± 1.4 years) participated in this study. A ratio-scaled (1RM·BM) was used for the comparison between them. The 1RM of the 3 exercises were evaluated within the same testing session using a combined 1RM assessment method. The interaction effect of exercise and sex did not reach statistical significance (p = 0.671; η = 0.001). In contrast, there were significant main effects of exercise (p ≤ 0.01) and sex (p < 0.001) with medium (η = 0.096) and large effect sizes (η = 0.306), respectively. A similar main effect of exercise was reported for both men (PP [1.0 ± 0.1 kg·kg] < PJ [1.1 ± 0.2 kg·kg] ∼ SJ [1.2 ± 0.2 kg·kg]) and women (PP [0.8 ± 0.1 kg·kg] < PJ [0.9 ± 0.2 kg·kg] ∼ SJ [0.9 ± 0.2 kg·kg]). The 1RM performance of the 3 exercises was significantly correlated for men (r [range] = 0.856-0.963) and women (r [range] = 0.636-0.925). Sex does not affect the differences in the 1RM performance across weightlifting overhead pressing exercises. However, stronger correlations and lower variation in the 1RM performance during the PP, PJ, and SJ are expected for men compared with women

Mechanical power production assessment during weightlifting exercises. A systematic review

    Get PDF
The assessment of the mechanical power production is of great importance for researchers and practitioners. The purpose of this review was to compare the differences in ground reaction force (GRF), kinematic, and combined (bar velocity x GRF) methods to assess mechanical power production during weightlifting exercises. A search of electronic databases was conducted to identify all publications up to 31 May 2019. The peak power output (PPO) was selected as the key variable. The exercises included in this review were clean variations, which includes the hang power clean (HPC), power clean (PC) and clean. A total of 26 articles met the inclusion criteria with 53.9% using the GRF, 38.5% combined, and 30.8% the kinematic method. Articles were evaluated and descriptively analysed to enable comparison between methods. The three methods have inherent methodological differences in the data analysis and measurement systems, which suggests that these methods should not be used interchangeably to assess PPO in Watts during weightlifting exercises. In addition, this review provides evidence and rationale for the use of the GRF to assess power production applied to the system mass while the kinematic method may be more appropriate when looking to assess only the power applied to the barbell

Effects of Combined CCR5/Integrase Inhibitors-Based Regimen on Mucosal Immunity in HIV-Infected Patients Naïve to Antiretroviral Therapy: A Pilot Randomized Trial

Whether initiation of antiretroviral therapy (ART) regimens aimed at achieving greater concentrations within gut associated lymphoid tissue (GALT) impacts the level of mucosal immune reconstitution, inflammatory markers and the viral reservoir remains unknown. We included 12 HIV- controls and 32 ART-naïve HIV patients who were randomized to efavirenz, maraviroc or maraviroc+raltegravir, each with fixed-dose tenofovir disoproxil fumarate/emtricitabine. Rectal and duodenal biopsies were obtained at baseline and at 9 months of ART. We performed a comprehensive assay of T-cell subsets by flow cytometry, T-cell density in intestinal biopsies, plasma and tissue concentrations of antiretroviral drugs by high-performance liquid chromatography/mass spectroscopy, and plasma interleukin-6 (IL-6), lipoteichoic acid (LTA), soluble CD14 (sCD14) and zonulin-1 each measured by ELISA. Total cell-associated HIV DNA was measured in PBMC and rectal and duodenal mononuclear cells. Twenty-six HIV-infected patients completed the follow-up. In the duodenum, the quadruple regimen resulted in greater CD8+ T-cell density decline, greater normalization of mucosal CCR5+CD4+ T-cells and increase of the naïve/memory CD8+ T-cell ratio, and a greater decline of sCD14 levels and duodenal HIV DNA levels (P = 0.004 and P = 0.067, respectively), with no changes in HIV RNA in plasma or tissue. Maraviroc showed the highest drug distribution to the gut tissue, and duodenal concentrations correlated well with other T-cell markers in duodenum, i.e., the CD4/CD8 ratio, %CD4+ and %CD8+ HLA-DR+CD38+ T-cells. Maraviroc use elicited greater activation of the mucosal naïve CD8+ T-cell subset, ameliorated the distribution of the CD8+ T-cell maturational subsets and induced higher improvement of zonulin-1 levels. These data suggest that combined CCR5 and integrase inhibitor based combination therapy in ART treatment naïve patients might more effectively reconstitute duodenal immunity, decrease inflammatory markers and impact on HIV persistence by cell-dependent mechanisms, and show unique effects of MVC in duodenal immunity driven by higher drug tissue penetration and possibly by class-dependent effects
