    Cytotoxic activity of Agave lechuguilla Torr

    The cytotoxic activity of extract and isolated saponin from leaves of Agave lechuguilla was investigated. Ethanol extract from leaves of A. lechuguilla exhibited cytotoxic activity against HeLa cells in vitro (50% inhibitory concentration (IC50) = 89 μg/ml). Bioassay-guided fractionation of this extract had led to the isolation of 5-β steroidal saponin with an IC50 value against HeLa cells of 78 μg/ml. Neither the extract nor saponin showed significant cytotoxicity against MCF-7 cells (IC50 > 150 μg/ml). The extract and isolated saponins were tested for their cytotoxicity against Vero cells, where they showed IC50  values of 126, 171.6 and 172.2 μg/ml, respectively.Key words: Agave lechuguilla, cytotoxicity, plant extract, saponin

    Nitrogen-Doped Nanoporous Carbon Membranes with Co/CoP Janus-Type Nanocrystals as Hydrogen Evolution Electrode in Both Acidic and Alkaline Environments

    Self-supported electrocatalysts being generated and employed directly as electrodes for energy conversion has been intensively pursued in the fields of materials chemistry and energy. Herein, we report a synthetic strategy to prepare freestanding hierarchically structured, nitrogen-doped nanoporous graphitic carbon membranes functionalized with Janus-type Co/CoP nanocrystals (termed as HNDCM-Co/CoP), which were successfully applied as a highly efficient, binder-free electrode in the hydrogen evolution reaction (HER). Benefited from multiple structural merits, such as a high degree of graphitization, three-dimensionally interconnected micro/meso/macropores, uniform nitrogen doping, well-dispersed Co/CoP nanocrystals, as well as the confinement effect of the thin carbon layer on the nanocrystals, HNDCM-Co/CoP exhibited superior electrocatalytic activity and long-term operation stability for HER under both acidic and alkaline conditions. As a proof-of-concept of practical usage, a 5.6 cm x 4 cm x 60 μm macroscopic piece of HNDCM-Co/CoP was prepared in our laboratory. Driven by a solar cell, electroreduction of water in alkaline conditions (pH 14) was performed, and H2 was produced at a rate of 16 mL/min, demonstrating its potential as real-life energy conversion systems

    Clinical Manifestations and Case Management of Ebola Haemorrhagic Fever caused by a newly identified virus strain, Bundibugyo, Uganda, 2007-2008

    A confirmed Ebola haemorrhagic fever (EHF) outbreak in Bundibugyo, Uganda, November 2007-February 2008, was caused by a putative new species (Bundibugyo ebolavirus). It included 93 putative cases, 56 laboratory-confirmed cases, and 37 deaths (CFR = 25%). Study objectives are to describe clinical manifestations and case management for 26 hospitalised laboratory-confirmed EHF patients. Clinical findings are congruous with previously reported EHF infections. The most frequently experienced symptoms were non-bloody diarrhoea (81%), severe headache (81%), and asthenia (77%). Seven patients reported or were observed with haemorrhagic symptoms, six of whom died. Ebola care remains difficult due to the resource-poor setting of outbreaks and the infection-control procedures required. However, quality data collection is essential to evaluate case definitions and therapeutic interventions, and needs improvement in future epidemics. Organizations usually involved in EHF case management have a particular responsibility in this respect

    Race diversity in dryland maize (Zea mays L.) landraces from southern Nuevo León, Mexico

    This study was done for reviewing the diversity and distribution of native maize grown at southern Nuevo León, Mexico, in 40 accessions collected in the area. Information obtained was on the production unit and on the ears traits, according to the CONABIO guidelines. In this area maize is usually grown during the Spring-Summer period, and using little technology, without fertilization or pest control. The 40 accessions correspond to seven races and seven interracial crosses. The most frequent maize races were Ratón and Cónico Norteño, especially in dry areas with little rain. Races Celaya, Tablilla de Ocho and Elotes Cónicos found in this study, had not been previously reported; but the Tabloncillo and Olotillo races that had previously been reported were no longer found. Three collections stood out for producing large ears with large kernels, thus having a high yield potential. Native maize harvested in dryland areas might offer advantages to be grown under harsh environments.Objective: To analyze the race diversity and geographic distribution of the native maize landraces currently cropped at southern Nuevo León, México. Design/Methodology/Approach: Data was obtained from 41 accessions which represent the commercial production in the dry land area, where fertilization and pest control are scarcely used. Landraces were classified according to the CONABIO guidelines for ear traits. Results: The measured accessions correspond to seven maize races and to seven interracial crosses. The two most frequent maize races were Ratón and Cónico Norteño, mostly located in the dry areas with less rain. Study Limitations/Implications: Three races, Celaya, Tablilla de Ocho and Elotes Cónicos, had not been previously reported; while two formerly reported races Tabloncillo and Olotillo, were no longer found. This study did not include the grain-colored accessions. Findings/Conclusions: Three collections stood out for producing large ears with large kernels, thus showing a high yield potential. The maize landraces harvested in dryland areas might offer advantages to be grown under harsh environments or be used as gene donors for drought tolerance

    Herpesvirus Telomerase RNA(vTR)-Dependent Lymphoma Formation Does Not Require Interaction of vTR with Telomerase Reverse Transcriptase (TERT)

    Telomerase is a ribonucleoprotein complex involved in the maintenance of telomeres, a protective structure at the distal ends of chromosomes. The enzyme complex contains two main components, telomerase reverse transcriptase (TERT), the catalytic subunit, and telomerase RNA (TR), which serves as a template for the addition of telomeric repeats (TTAGGG)n. Marek's disease virus (MDV), an oncogenic herpesvirus inducing fatal lymphoma in chickens, encodes a TR homologue, viral TR (vTR), which significantly contributes to MDV-induced lymphomagenesis. As recent studies have suggested that TRs possess functions independently of telomerase activity, we investigated if the tumor-promoting properties of MDV vTR are dependent on formation of a functional telomerase complex. The P6.1 stem-loop of TR is known to mediate TR-TERT complex formation and we show here that interaction of vTR with TERT and, consequently, telomerase activity was efficiently abrogated by the disruption of the vTR P6.1 stem-loop (P6.1mut). Recombinant MDV carrying the P6.1mut stem-loop mutation were generated and tested for their behavior in the natural host in vivo. In contrast to viruses lacking vTR, all animals infected with the P6.1mut viruses developed MDV-induced lymphomas, but onset of tumor formation was significantly delayed. P6.1mut viruses induced enhanced metastasis, indicating functionality of non-complexed vTR in tumor dissemination. We discovered that RPL22, a cellular factor involved in T-cell development and virus-induced transformation, directly interacts with wild-type and mutant vTR and is, consequently, relocalized to the nucleoplasm. Our study provides the first evidence that expression of TR, in this case encoded by a herpesvirus, is pro-oncogenic in the absence of telomerase activity

    Internationalisation speed and MNE performance: A study of the market-seeking expansion of retail MNEs

    Existing research is divided on whether firms that rapidly expand their overseas operations perform better than firms that internationalize slowly. Drawing on Penrose’s theory of the growth of the firm we argue that the positive effects of rapid internationalization give way to negative effects with increasing internationalization speed, leading to an inverted U-shaped association between internationalization speed and firm performance. We analyse the market-seeking expansion of 110 retailers over a 10-year period (2003–2012) and find support for a curvilinear relationship between internationalization speed and firm performance that is moderated by the geographic scope of firms’ internationalization path and firms’ international experience. Our study contributes to resolving conflicting views on the link between internationalization speed and firm performance

    Ring-Like Distribution of Constitutive Heterochromatin in Bovine Senescent Cells

    Background: Cells that reach ‘‘Hayflick limit’ ’ of proliferation, known as senescent cells, possess a particular type of nuclear architecture. Human senescent cells are characterized by the presence of highly condensed senescent associated heterochromatin foci (SAHF) that can be detected both by immunostaining for histone H3 three-methylated at lysine 9 (H3K9me3) and by DAPI counterstaining. Methods: We have studied nuclear architecture in bovine senescent cells using a combination of immunofluorescence and 3D fluorescent in-situ hybridization (FISH). Results: Analysis of heterochromatin distribution in bovine senescent cells using fluorescent in situ hybridization for pericentric chromosomal regions, immunostaining of H3K9me3, centromeric proteins CENP A/B and DNA methylation showed a lower level of heterochromatin condensation as compared to young cells. No SAHF foci were observed. Instead, we observed fibrous ring-like or ribbon-like heterochromatin patterns that were undetectable with DAPI counterstaining.
