    Jack bean (Canavalia ensiformis) urease induces eicosanoid-modulated hemocyte aggregation in the Chagas' disease vector Rhodnius prolixus

    AbstractUreases are multifunctional proteins that display biological activities independently of their enzymatic function, such as induction of exocytosis and insecticidal effects. Rhodnius prolixus, a major vector of Chagas' disease, is a model for studies on the entomotoxicity of jack bean urease (JBU). We have previously shown that JBU induces the production of eicosanoids in isolated tissues of R. prolixus. In insects, the immune response comprises cellular and humoral reactions, and is centrally modulated by eicosanoids. Cyclooxygenase products signal immunity in insects, mainly cellular reactions, such as hemocyte aggregation. In searching for a link between JBU's toxic effects and immune reactions in insects, we have studied the effects of this toxin on R. prolixus hemocytes. JBU triggers aggregation of hemocytes after injection into the hemocoel and when applied to isolated cells. On in vitro assays, the eicosanoid synthesis inhibitors dexamethasone (phospholipase A2 indirect inhibitor) and indomethacin (cyclooxygenase inhibitor) counteracted JBU's effect, indicating that eicosanoids, more specifically cyclooxygenase products, are likely to mediate the aggregation response. Contrarily, the inhibitors esculetin and baicalein were inactive, suggesting that lipoxygenase products are not involved in JBU's effect. Extracellular calcium was also necessary for JBU's effect, in agreement to other cell models responsive to ureases. A progressive darkening of the medium of JBU-treated hemocytes was observed, suggestive of a humoral response. JBU was immunolocalized in the cultured cells upon treatment along with cytoskeleton damage. The highest concentration of JBU tested on cultured cells also led to nuclei aggregation of adherent hemocytes. This is the first time urease has been shown to affect insect hemocytes, contributing to our understanding of the entomotoxic mechanisms of action of this protein

    Stability Of Gluten Free Sweet Biscuit Elaborated With Rice Bran, Broken Rice And Okara

    A challenge to the food sector has been the development of new products incorporating co-products from the food processing industry with minimal impact on their pre-determined structures and adding nutritional quality. In order to add value and develop alternatives for the use of co-products generated during the agroindustrial processing, this work aimed to study the stability of gluten-free sweet biscuits developed with soybean okara, rice bran and broken rice. The formulations were elaborated with increasing percentages of these ingredients and compared with the standard (commercial sweet biscuit) for ten months. The analyses were: weight, diameters (internal and external), thickness, specific volume, instrumental parameters of color, texture, scanning electron microscopy, water activity, proximal composition and isoflavones. The experimental sweet biscuits had characteristics of color, weight, volume and diameters (internal and external) very similar to the commercial, whereas texture, lipids and energy value decreased, and aw, moisture and protein increased during storage.     Synthesis and luminescence properties of analogues of the green fluorescent protein chromophore

    The green fluorescent protein (GFP) is extensively used as a biomarker for fluorescence biological imaging. The chromophore in GFP is only fluorescent when confined into the β–barrel of the protein. Similarly, synthetic analogues of the fluorophore of GFP are usually non-emissive in solution, due to free rotation around the aryl-alkene bond and (Z/E)-isomerization of the double bond. Here, the synthesis and characterization of three analogues of the fluorophore of GFP are reported. The introduction of more electron donating substituents induces a red-shift in the absorption and emission. The fluorophores are more emissive in the solid state than in solution, and a study of their crystal structure reveals that the (Z/E)-isomerization is efficiently blocked in the crystals.publishe

    The northern region of Piauí state is an important center of irrigated organic fruit farming, wherein sandy soils with low organic matter content are predominant, besides climate with high temperatures, long periods of water deficit and constant wind. Given this situation, it becomes necessary to investigate alternative management options able to meet the challenges of agricultural productivity. In this context, green manuring becomes an important practice, mainly when using fast-growing species and with great potential for biomass production. Thus, a study was carried out at Embrapa Meio-Norte to measure Crotalaria juncea biomass yield after thinning and at various sowing densities. The evaluated parameters were plant height, stem diameter, number of branches per plant, fresh and dry weight of shoots and roots. The results showed that sowing density had no influence on plant height; however, unthinned plants reached greater heights, which might have been due to reduced competition for light. Thinning had no effect on diameters of plant stem base; on the other hand, lower planting densities promoted larger diameters also due to less competition. Likewise, the number of branches per plant was not affected by thinning, being greater at lower planting densities on account of less competition. Thinned plants achieved higher averages of fresh shoot weight. Nevertheless, shoot dry biomass was greater at higher planting densities. Furthermore, both fresh and dry weight of roots were not influenced by any of the adopted treatments. © 2016, Universidade Federal Rural do Semi-Arid. All rights reserved.29487888

Molecules which mediate the transfer of light to another molecule (photosensitizers) are the cornerstone of exploitation of light. The current state of research and recent progress in applications of photosensitizers in diverse fields are documented in twelve chapters written by scientists from around the world. Chapter 1 gives a general overview on the applications of photosensitizers in synthetic organic chemistry. This review is followed by Chapters 2-7, which deal with the various applications of photosensitizers in medicine, in particular for photodynamic cancer therapy.     Every year thousands of workers from the Brazilian Northeastern states move to the sugarcane plantations in the state of São Paulo to work in sugarcane mills. Most of them are young with little schooling and come from peasant families. Among other things, researchers call attention to the risks to which these workers are submitted, related to transportation conditions, housing and work environment. The organization of work is always focused on increasing the surplus value, which has resulted in the deterioration of the labor force, expressed by several diseases and countless deaths of migrant workers. In the scope of this article, it is our intention to define “risk” concept and relate it to the processes of subjectivities and to understand the meanings that migrant workers attribute to the risks to which they are exposed during the trip and also in workplaces (sugarcane plantations). Regarding methodology, we used a qualitative approach. Data were collected through semi-structured interviews conducted with a sample of 12 migrant workers from the city of Santa Cruz da Baixa Verde (Pernambuco), in the Pajeú microregion, and also of cities located in the microregion of Serra do Teixeira (Paraíba), especially the surrounding areas of Princesa Isabel (state of Paraíba). The data allow us to assert the existence of an intrinsic relationship between the risks they run and the affirmation of the gender identity of the subjects investigated. © 2016, UNIV SAOPAULO.     Effects of effortful swallowing on cardiac autonomic control in individuals with neurogenic dysphagia: A prospective observational analytical study

    Considering that neurogenic oropharyngeal dysphagia is a prevalent condition with or without cardiac disease we should contemplate issues surrounding cardiovascular difficulties during rehabilitation. This study aims to evaluate the effects of effortful swallowing maneuver (ESM) on heart rate variability (HRV) in subjects with neurogenic oropharyngeal dysphagia. We studied 22 individuals [8 Stroke and 14 Parkinson Disease (PD) subjects aged between 41 and 75 years old] with neurogenic oropharyngeal dysphagia regardless of gender. HRV was assessed under two circumstances: spontaneous swallowing versus ESM. Surface electromyography of the suprahyoid muscles was undertaken to measure the swallowing muscle excitation, which then confirmed higher muscle activity during ESM. We attained no changes in HRV between the two swallowing events [HR: spontaneous swallowing 78.68 ± 13.91 bpm vs. ESM 102.57 ± 107.81 bpm, p = 0.201; RMSSD (root-mean square of differences between adjacent normal RR intervals in a time interval): spontaneous swallowing 16.99 ± 15.65 ms vs. ESM 44.74 ± 138.85 ms, p = 0.312; HF (high frequency): spontaneous swallowing 119.35 ± 273 ms2 vs. ESM 99.83 ± 194.58 ms2, p = 0.301; SD1 (standard deviation of the instantaneous variability of the beat-to-beat heart rate): spontaneous swallowing 12.02 ± 1.07 ms vs. ESM 31.66 ± 98.25 ms, p = 0.301]. The effortful swallowing maneuver did not cause clinically significant changes in autonomic control of HR in this group of subjects with oropharyngeal dysphagia

    Bedload and Suspended Sediment of a Watershed Impacted by Dams

    Sediment transport relates to suspended sediment and bedload. The suspended sediment plays the most important role on the land-ocen sediment flux. On the other hand, the bedload should be considered in order to assess the impacts of dams on sediment transport and sediment yield. Recent effects of dam construction have been widely reported. The sediment load has been reduced by more than 75% for major rives, such as Nilo, Orange, Volta, Indus, Ebro, Kizil Irmark, Colorado, and Rio Grande and more than 40% of its lux is trapped within large dams. In addition, the multiple trapping through sequential dams has impacted the sediment transfer from terrestrial to coastal zone, triggering the coastal erosion. In terms of sediment retention and transport, China stands out the most impacted country by dams, followed by United States, and continents such as Europe, Africa, and South America. Based on the foregoing, the impact of dams on sediment transport and yield of an important Brazilian watershed with multiple dams will be the focus of this chapter. Thus, a three years field sampling (2009-2011) was carried out to measure the sediment yield of Capibaribe Watershed, and also its contribution to coastal erosion. The ratio between QB and SSQ ranged from 0.12% to 27.3% with 76% of all values lower than 5%. Usually, the bedload transport rate of a river is about 5–25% of the suspended sediment transport. This ranging sheds light on the lack of bedload reaching the coastal zone and it is likely one of the reasons to yield coastal erosion. The low rates can be attributed to the presence of dams which have been admitted to have a strong effect on sediment transport. The sediment yield was equal to 3.69, 4.36, and 6.7 t km-2 ano-1 in 2009, 2010, and 2011, respectively. In comparison with bedload yield, the suspended sediment yield was higher than 95% for all studied years. Therefore, the limited bedload supply – mainly responsible for construction of coastal landform – is likely contributing to the coastal erosion along part of the northeast region, Brazil. The multiple dams along the Capibaribe River watershed produce a deficit in sediment flux to coastal zone of Pernambuco State, Brazil, which relies on the low ratios between bedload and suspended sediment.     Sugestão de modelo de prontuário médico pediátrico para os Hospitais Universitários de Ensino

    O objetivo deste artigo foi apresentar os conceitos de prontuário manual e eletrônico, para apontar a necessidade da confecção de um Manual sobre o Prontuário Médico Pediátrico, mais completo, coerente e fidedigno, dirigido aos acadêmicos de diferentes áreas de saúde que atuam na rede do SUS. Apesar de se reconhecer a necessidade do Prontuário Eletrônico do Paciente (PEP), as dificuldades para sua implementação ainda não foram resolvidas. O atual estágio de desconforto dos acadêmicos e dos professores diante da constatação de que os registros realizados nos prontuários manuais das Unidades de Saúde do Município não satisfazem às exigências mínimas para acompanhamento dos pacientes, aponta para a necessidade de implantação de um novo modelo de Prontuário Médico nos ambulatórios dos hospitais universitários de ensino que fazem parte do nível de maior complexidade do Sistema Único de Saúde (SUS). O levantamento de dados mostrou que tais prontuários apresentam grande número de diagnósticos ilegíveis, falta de coerência e lógica na escrita, além de, muitas vezes, não possuírem os dados básicos relativos aos pacientes. Os modelos em formato tradicional, ou seja, em papel, assumem importância fundamental, porque, além de assistência, há atividades de ensino e pesquisa, cursos de graduação e de pós-graduação das ciências da saúde, educação permanente e educação dirigida aos pacientes e à comunidade, assim como o treinamento de pessoal de nível médio e auxiliar, utilizando-se de tal documentação. Conclui-se que este Manual, mesmo sem a sofisticação do PEP, surge como importante instrumento facilitador da comunicação entre profissionais da área de saúde e acadêmicos das equipes interdisciplinares, neste momento, oferecendo a oportunidade de qualificar a atenção à clientela pediátrica atendida na rede do SUS, preenchendo uma lacuna, considerando que, nos municípios, em geral, não existe um prontuário específico, único, para o registro das ocorrências e atendimento de criança
