    Naturaliste plateau: constraints on the timing and evolution of the Kerguelen Large Igneous Province and its role in Gondwana breakup

    Volcanism associated with the Kerguelen Large Igneous Province is found scattered in southwestern Australia (the ca 136 to ca 130 Ma Bunbury Basalts, and ca 124 Ma Wallaby Plateau), India (ca 118 Ma Rajmahal Traps and Cona Basalts), and Tibet (the ca 132 Ma Comei Basalts), but apart from the ∼70 000 km2 Wallaby Plateau, these examples are spatially and volumetrically minor. Here, we report dredge, geochronological and geochemical results from the ∼90 000 km2 Naturaliste Plateau, located ∼170 to ∼500 km southwest of Australia. Dredged lavas and intrusive rocks range from mafic to felsic compositions, and prior geophysical analyses indicate these units comprise much of the plateau substrate. 40Ar/39Ar plagioclase ages from mafic units and U–Pb zircon ages from silicic rocks indicate magmatic emplacement from 130.6 ± 1.2 to 129.4 ± 1.3 Ma for mafic rocks and 131.8 ± 3.9 to 128.2 ± 2.3 Ma for silicic rocks (2σ). These Cretaceous Naturaliste magmas incorporated a significant component of continental crust, with relatively high 87Sr/86Sr (up to 0.78), high 207Pb/204 Pb ratios (15.5–15.6), low 143Nd/144Nd (0.511–0.512) and primitive-mantle normalised Th/Nb of 11.3 and La/Nb of 3.97. These geochemical results are consistent with the plateau being underlain by continental basement, as indicated by prior interpretations of seismic and gravity data, corroborated by dredging of Mesoproterozoic granites and gneisses on the southern plateau flank. The Cretaceous Naturaliste Plateau igneous rocks have signatures indicative of extraction from a depleted mantle, with trace-element and isotopic values that overlap with Kerguelen Plateau lavas reflect crustal contamination. Our chemical and geochronological results therefore show the Naturaliste Plateau contains evidence of an extensive igneous event representing some of the earliest voluminous Kerguelen hotspot magmas. Prior work reports that contemporaneous correlative volcanic sequences underlie the nearby Mentelle Basin, and the Enderby Basin and Princess Elizabeth Trough in the Antarctic. When combined, the igneous rocks in the Naturaliste, Mentelle, Wallaby, Enderby, Princess Elizabeth, Bunbury and Comei-Cona areas form a 136–124 Ma Large Igneous Province covering >244 000 km2

    Le Leg 2 de la campagne GEODYNZ-SUD, menée au SW de la Nouvelle-Zélande, a permis de reconnaître les structures qui accompagnent du Nord au Sud le passage de la faille alpine à la subduction oblique sous la marge du Fiodland, puis à celle naissante, intra-océanique sous la ride de Macquarie. Au Nord et au-dessus de la plaque australienne subductée vers l'Est, un faisceau longitudinal de décrochements converge vers le système transpressif de la faille alpine en découpant la marge continentale.     Una Visión General del Sistema Financiero Colombiano

    Desde finales de los 80 el sistema financiero colombiano ha experimentado cambios sensibles. En efecto, la liberalización financiera, el fortalecimiento de la regulación prudencial, la conversión de un número importante de sociedades en establecimientos de crédito, el aumento en los requisitos de capital, etc. han determinado un cambio de perfil en el sistema. Adicionalmente, en el pasado reciente las autoridades han tomado medidas en cuanto a la estructura de los encajes, aumentos en los requisitos de capital, el acceso al crédito externo, etc. que afectan de manera importante a las entidades financieras. A raíz de estas medidas ha surgido un debate acerca del tipo de sistema financiero que resulta más deseable para Colombia. La discusión es de vital importancia puesto que la estructura de encajes, las formas de intervención del Banco de la República en los mercados cambiario y monetario, la supervisión y todo el aparato regulatorio deben ser consistentes con el tipo de sistema que se desee. Con el fin de contribuir al debate, en este documento se presenta una breve descripción del estado actual del sistema financiero y su evolución reciente, se plantea una reflexión normativa acerca del tipo de sistema financiero que puede resultar más deseable y, finalmente,se presentan algunas recomendaciones.

    Immediate antiviral therapy appears to restrict resting CD4 + cell HIV-1 infection without accelerating the decay of latent infection

    HIV type 1 (HIV-1) persists within resting CD4 + T cells despite anti-retroviral therapy (ART). To better understand the kinetics by which resting cell infection (RCI) is established, we developed a mathematical model that accurately predicts (r = 0.65, P = 2.5 × 10 -4) the initial frequency of RCI measured about 1 year postinfection, based on the time of ART initiation and the dynamic changes in viremia and CD4 + T cells. In the largest cohort of patients treated during acute seronegative HIV infection (AHI) in whom RCI has been stringently quantified, we found that early ART reduced the generation of latently infected cells. Although RCI declined after the first year of ART in most acutely infected patients, there was a striking absence of decline when initial RCI frequency was less than 0.5 per million. Notably, low-level viremia was observed more frequently as RCI increased. Together these observations suggest that (i) the degree of RCI is directly related to the availability of CD4 + T cells susceptible to HIV, whether viremia is controlled by the immune response and/or ART; and (ii) that two pools of infected resting CD4 + T cells exist, namely, less stable cells, observable in patients in whom viremia is not well controlled in early infection, and extremely stable cells that are established despite early ART. These findings reinforce and extend the concept that new approaches will be needed to eradicate HIV infection, and, in particular, highlight the need to target the extremely small but universal, long-lived latent reservoir

    Immunogenicity of AGS-004 Dendritic Cell Therapy in Patients Treated during Acute HIV Infection

    AGS-004 consists of matured autologous dendritic cells co-electroporated with in vitro transcribed RNA encoding autologous HIV antigens. In an open-label, single arm sub-study of AGS-004-003, AGS-004 was administered monthly to suppressed participants who started antiretroviral therapy (ART) during acute HIV infection. HIV-1 specific T cell responses were measured by multicolor flow cytometry after 3-4 doses. The frequency of resting CD4+ T-cell infection (RCI) was measured by quantitative viral outgrowth assay. Participants demonstrating increased immune response postvaccination were eligible for analytic treatment interruption (ATI). AGS-004 induced a positive immune response defined as ≥2-fold increase from baseline in the number of multifunctional HIV-1 specific CD28+/CD45RA- CD8+ effector/memory cytoxic T-lymphocytes (CTLs) in all six participants. All participants underwent ATI with rebound viremia at a median of 29 days. Immune correlates between time to viral rebound and the induction of effector CTLs were determined. Baseline RCI was low in most participants (0.043-0.767 IUPM). One participant had a >2-fold decrease (0.179-0.067 infectious units per million [IUPM]) in RCI at week 10. One participant with the lowest RCI had the longest ATI. AGS-004 dendritic cell administration increased multifunctional HIV-specific CD28+/CD45RA- CD8+ memory T cell responses in all participants, but did not permit sustained ART interruption. However, greater expansion of CD28-/CCR7-/CD45RA- CD8+ effector T cell responses correlated with a longer time to viral rebound. AGS-004 may be a useful tool to augment immune responses in the setting of latency reversal and eradication strategies

    Clones of infected cells arise early in HIV-infected individuals

    In HIV-infected individuals on long-term antiretroviral therapy (ART), more than 40% of the infected cells are in clones. Although most HIV proviruses present in individuals on long-term ART are defective, including those in clonally expanded cells, there is increasing evidence that clones carrying replication-competent proviruses are common in patients on long-term ART and form part of the HIV reservoir that makes it impossible to cure HIV infection with current ART alone. Given the importance of clonal expansion in HIV persistence, we determined how soon after HIV acquisition infected clones can grow large enough to be detected (clones larger than ca. 1 × 105 cells). We studied 12 individuals sampled in early HIV infection (Fiebig stage III-V/VI) and 5 who were chronically infected. The recently infected individuals were started on ART at or near the time of diagnosis. We isolated more than 6,500 independent integration sites from peripheral blood mononuclear cells before ART was initiated and after 0.5-18 years of suppressive ART. Some infected clones could be detected approximately 4 weeks after HIV infection and some of these clones persisted for years. The results help to explain how the reservoir is established early and persists for years

    Dynamic loading of human engineered heart tissue enhances contractile function and drives a desmosome-linked disease phenotype

    The role that mechanical forces play in shaping the structure and function of the heart is critical to understanding heart formation and the etiology of disease but is challenging to study in patients. Engineered heart tissues (EHTs) incorporating human induced pluripotent stem cell (hiPSC)-derived cardiomyocytes have the potential to provide insight into these adaptive and maladaptive changes. However, most EHT systems cannot model both preload (stretch during chamber filling) and afterload (pressure the heart must work against to eject blood). Here, we have developed a new dynamic EHT (dyn-EHT) model that enables us to tune preload and have unconstrained contractile shortening of >10%. To do this, three-dimensional (3D) EHTs were integrated with an elastic polydimethylsiloxane strip providing mechanical preload and afterload in addition to enabling contractile force measurements based on strip bending. Our results demonstrated that dynamic loading improves the function of wild-type EHTs on the basis of the magnitude of the applied force, leading to improved alignment, conduction velocity, and contractility. For disease modeling, we used hiPSC-derived cardiomyocytes from a patient with arrhythmogenic cardiomyopathy due to mutations in the desmoplakin gene. We demonstrated that manifestation of this desmosome-linked disease state required dyn-EHT conditioning and that it could not be induced using 2D or standard 3D EHT approaches. Thus, a dynamic loading strategy is necessary to provoke the disease phenotype of diastolic lengthening, reduction of desmosome counts, and reduced contractility, which are related to primary end points of clinical disease, such as chamber thinning and reduced cardiac output.Cardiolog
