An Application of Analytic Hierarchy Process in Vehicle Routing Problem
Vehicle Routing Problem (VRP) facilitates on finding a set of trips, one for each vehicle and to deliver known quantities of goods from a single depot to a set of geographically dispersed customers. This paper proposes an effective hybrid approach that combines customer prioritization with the Clarke and Wright's savings algorithm to solve the capacitated vehicle routing problem. In this model, in addition to traditional objective of resolving vehicle routing problem, the customer satisfaction have been taken into account. Initially, all the customers have been clustered with the help of Clarke and Wright's saving algorithm and later the customers have been prioritized on assigning optimal route using Analytic Hierarchy Process (AHP) as a Multi Criteria Decision Making (MCDM) tool. The highlight of this research is to diminish the total transportation cost without violating the vehicle capacity and ultimately improve the customer satisfaction
Cancellation of Towing Ship Interference in Passive SONAR in a Shallow Ocean Environment
Towed array sonars are preferred for detecting stealthy underwater targets that emit faint acoustic signals in the ocean, especially in shallow waters. However, the towing ship being near to the array behaves as a loud target, introducing additional interfering signals to the array, severely affecting the detection and classification of potential targets. Canceling this underlying interference signal is a challenging task and is investigated in this paper for a shallow ocean operational scenario where the problem is more critical due to the multipath phenomenon. A method exploiting the eigenvector analysis of spatio-temporal covariance matrix based on space time adaptive processing is proposed for suppressing tow ship interference and thus improving target detection. The developed algorithm learns the interference patterns in the presence of target signals to mitigate the interference across azimuth and to remove the spectral leakage of own-ship. The algorithm is statistically analyzed through a set of relevant metrics and is tested on simulated data that are equivalent to the data received by a towed linear array of acoustic sensors in a shallow ocean. The results indicate a reduction of 20-25dB in the tow ship interference power while the detection of long-range low SNR targets remain largely unaffected with minimal power-loss. In addition, it is demonstrated that the spectral leakage of tow ship, on multiple beams across the azimuth, due to multipath, is also alleviated leading to superior classification capabilities. The robustness of the proposed algorithm is validated by the open ocean experiment in the coastal shallow region of the Arabian Sea at Off-Kochi area of India, which produced results in close agreement with the simulations. A comparison of the simulation and experimental results with the existing PCI and ECA methods is also carried out, suggesting the proposed method is quite effective in suppressing the tow ship interference and is immensely beneficial for the detection and classification of long-range targets

The APOB insertion/deletion polymorphism (rs17240441) influences postprandial lipaemia in healthy adults
BACKGROUND:
Apolipoprotein (apo)B is the structural apoprotein of intestinally- and liver- derived lipoproteins and plays an important role in the transport of triacylglycerol (TAG) and cholesterol. Previous studies have examined the association between the APOB insertion/deletion (ins/del) polymorphism (rs17240441) and postprandial lipaemia in response to a single meal; however the findings have been inconsistent with studies often underpowered to detect genotype-lipaemia associations, focused mainly on men, or with limited postprandial characterisation of participants. In the present study, using a novel sequential test meal protocol which more closely mimics habitual eating patterns, we investigated the impact of APOB ins/del polymorphism on postprandial TAG, non-esterified fatty acids, glucose and insulin levels in healthy adults.
FINDINGS:
Healthy participants (n = 147) consumed a standard test breakfast (0 min; 49 g fat) and lunch (330 min; 29 g fat), with blood samples collected before (fasting) and on 11 subsequent occasions until 480 min after the test breakfast. The ins/ins homozygotes had higher fasting total cholesterol, LDL-cholesterol, TAG, insulin and HOMA-IR and lower HDL-cholesterol than del/del homozygotes (P < 0.017). A higher area under the time response curve (AUC) was evident for the postprandial TAG (P < 0.001) and insulin (P = 0.032) responses in the ins/ins homozygotes relative to the del/del homozygotes, where the genotype explained 35% and 7% of the variation in the TAG and insulin AUCs, respectively.
CONCLUSIONS:
In summary, our findings indicate that the APOB ins/del polymorphism is likely to be an important genetic determinant of the large inter-individual variability in the postprandial TAG and insulin responses to dietary fat intake
Optimum Small Optical Beam Displacement Measurement
We derive the quantum noise limit for the optical beam displacement of a
TEM00 mode. Using a multimodal analysis, we show that the conventional split
detection scheme for measuring beam displacement is non-optimal with 80%
efficiency. We propose a new displacement measurement scheme that is optimal
for small beam displacement. This scheme utilises a homodyne detection setup
that has a TEM10 mode local oscillator. We show that although the quantum noise
limit to displacement measurement can be surpassed using squeezed light in
appropriate spatial modes for both schemes, the TEM10 homodyning scheme
out-performs split detection for all values of squeezing.Comment: 13 pages, 7 figure

A systematic review of the gene-lifestyle interactions on metabolic disease-related outcomes in Arab populations
The increased prevalence of metabolic diseases in the Arab countries is mainly associated with genetic susceptibility, lifestyle behaviours, such as physical inactivity, and an unhealthy diet. The objective of this review was to investigate and summarise the findings of the gene-lifestyle interaction studies on metabolic diseases such as obesity and type 2 diabetes in Arab populations. Relevant articles were retrieved from a literature search on PubMed, Web of Science, and Google Scholar starting at the earliest indexing date through to January 2024. Articles that reported an interaction between gene variants and diet or physical activity were included and excluded if no interaction was investigated or if they were conducted among a non-Arab population. In total, five articles were included in this review. To date, among three out of twenty-two Arab populations, fourteen interactions have been found between the FTO rs9939609, TCF7L2 rs7903146, MC4R rs17782313, and MTHFR rs1801133 polymorphisms and diet or physical activity on obesity and type 2 diabetes outcomes. The majority of the reported gene-diet/ gene-physical activity interactions (twelve) appeared only once in the review. Consequently, replication, comparisons, and generalisation of the findings are limited due to the sample size, study designs, dietary assessment tools, statistical analysis, and genetic heterogeneity of the studied sample
Battle of GLP-1 delivery technologies
Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) belong to an important therapeutic class for treatment of type 2 diabetes. Six GLP-1 RAs, each utilizing a unique drug delivery strategy, are now approved by the Food and Drug Administration (FDA) and additional, novel GLP-1 RAs are still under development, making for a crowded marketplace and fierce competition among the manufacturers of these products. As rapid elimination is a major challenge for clinical application of GLP-1 RAs, various half-life extension strategies have been successfully employed including sequential modification, attachment of fatty-acid to peptide, fusion with human serum albu- min, fusion with the fragment crystallizable (Fc) region of a monoclonal antibody, sustained drug delivery sys- tems, and PEGylation. In this review, we discuss the scientific rationale of the various half-life extension strategies used for GLP-1 RA development. By analyzing and comparing different approved GLP-1 RAs and those in development, we focus on assessing how half-life extending strategies impact the pharmacokinetics, pharmacodynamics, safety, patient usability and ultimately, the commercial success of GLP-1 RA products. We also anticipate future GLP-1 RA development trends. Since similar drug delivery strategies are also applied for de- veloping other therapeutic peptides, we expect this case study of GLP-1 RAs will provide generalizable concepts for the rational design of therapeutic peptides products with extended duration of action
Donor to recipient age matching in lung transplantation: A European experience
\ua9 2024 The AuthorsBackground: The age profile of organ donors and patients on lung transplantation (LT) waiting lists have changed over time. In Europe, the donor population has aged much more rapidly than the recipient population, making allocation decisions on lungs from older donors common. In this study we assessed the impact of donor and recipient age discrepancy on LT outcomes in the UK and France. Methods: A retrospective analysis of all adult single or bilateral LT in France and the UK between 2010 and 2021. Recipients were stratified into 3 age author groups: young (≤30 years), middle-aged (30–60) and older (≥60). Their donors were also stratified into 2 groups <60, ≥60. Primary graft dysfunction (PGD) rates and recipient survival was compared between matched and mismatched donor and recipient age groups. Propensity matching was employed to minimize covariate imbalances and to improve the internal validity of our results. Results: Our study cohort was 4,696 lung transplant recipients (LTRs). In young and older LTRs, there was no significant difference in 1 and 5-year post-transplant survival dependent on the age category of the donor. Young LTRs who received older donor grafts had a higher risk of severe grade 3 PGD. Conclusion: Our findings show that clinically usable organs from older donors can be utilized safely in LT, even for younger recipients. Further research is needed to assess if the higher rate of PGD3 associated with use of older donors has an effect on long-term outcomes

Association between FTO variant and change in body weight and its interaction with dietary factors: the DiOGenes study
Although FTO is an established obesity-susceptibility locus, it remains unknown whether it influences weight change in adult life and whether diet attenuates this association. Therefore, we investigated the association of FTO-rs9939609 with changes in weight and waist circumference (WC) during 6.8 years follow-up in a large-scale prospective study and examined whether these associations were modified by dietary energy percentage from fat, protein, carbohydrate, or glycemic index (GI). This study comprised data from five countries of European Prospective Investigation into Cancer and Nutrition (EPIC) and was designed as a case-cohort study for weight gain. Analyses included 11,091 individuals, of whom 5,584 were cases (age (SD), 47.6 (7.5) years), defined as those with the greatest unexplained annual weight gain during follow-up and 5,507 were noncases (48.0 (7.3) years), who were compared in our case-noncase (CNC) analyses. Furthermore, 6,566 individuals (47.9 (7.3) years) selected from the total sample (all noncases and 1,059 cases) formed the random subcohort (RSC), used for continuous trait analyses. Interactions were tested by including interaction terms in the models. In the RSC-analyses, FTO-rs9939609 was associated with BMI (β (SE), 0.17 (0.08) kg·m(-2)/allele; P = 0.034) and WC (0.47 (0.21) cm/allele; P = 0.026) at baseline, but not with weight change (5.55 (12.5) g·year(-1)/allele; P = 0.66) during follow up. In the CNC-analysis, FTO-rs9939609 was associated with increased risk of being a weight-gainer (OR: 1.1; P = 0.045). We observed no interaction between FTO-rs9939609 and dietary fat, protein and carbohydrate, and GI on BMI and WC at baseline or on change in weight and WC. FTO-rs9939609 is associated with BMI and WC at baseline, but association with weight gain is weak and only observed for extreme gain. Dietary factors did not influence the associations

A systematic review of the effect of gene–lifestyle interactions on metabolic-disease-related traits in South Asian populations
Context Recent data from the South Asian subregion have raised concern about the dramatic increase in the prevalence of metabolic diseases, which are influenced by genetic and lifestyle factors. Objective The aim of this systematic review was to summarize the contemporary evidence for the effect of gene–lifestyle interactions on metabolic outcomes in this population. Data sources PubMed, Web of Science, and SCOPUS databases were searched up until March 2023 for observational and intervention studies investigating the interaction between genetic variants and lifestyle factors such as diet and physical activity on obesity and type 2 diabetes traits. Data extraction Of the 14 783 publications extracted, 15 were deemed eligible for inclusion in this study. Data extraction was carried out independently by 3 investigators. The quality of the included studies was assessed using the Appraisal Tool for Cross-Sectional Studies (AXIS), the Risk Of Bias In Non-randomized Studies—of Interventions (ROBINS-I), and the methodological quality score for nutrigenetics studies. Data analysis Using a narrative synthesis approach, the findings were presented in textual and tabular format. Together, studies from India (n = 8), Pakistan (n = 3), Sri Lanka (n = 1), and the South Asian diaspora in Singapore and Canada (n = 3) reported 543 gene–lifestyle interactions, of which 132 (∼24%) were statistically significant. These results were related to the effects of the interaction of genetic factors with physical inactivity, poor sleep habits, smoking, and dietary intake of carbohydrates, protein, and fat on the risk of metabolic disease in this population. Conclusions The findings of this systematic review provide evidence of gene–lifestyle interactions impacting metabolic traits within the South Asian population. However, the lack of replication and correction for multiple testing and the small sample size of the included studies may limit the conclusiveness of the evidence.
