    Development of procedures for calculating stiffness and damping properties of elastomers. Part 3: The effects of temperature, dissipation level and geometry

    Effects of temperature, dissipation level and geometry on the dynamic behavior of elastomer elements were investigated. Force displacement relationships in elastomer elements and the effects of frequency, geometry and temperature upon these relationships are reviewed. Based on this review, methods of reducing stiffness and damping data for shear and compression test elements to material properties (storage and loss moduli) and empirical geometric factors are developed and tested using previously generated experimental data. A prediction method which accounts for large amplitudes of deformation is developed on the assumption that their effect is to increase temperature through the elastomers, thereby modifying the local material properties. Various simple methods of predicting the radial stiffness of ring cartridge elements are developed and compared. Material properties were determined from the shear specimen tests as a function of frequency and temperature. Using these material properties, numerical predictions of stiffness and damping for cartridge and compression specimens were made and compared with corresponding measurements at different temperatures, with encouraging results

    A test rig designed to measure stiffness and damping of elastomer cartridges under a rotating load excitation is described. The test rig employs rotating unbalance in a rotor which runs to 60,000 RPM as the excitation mechanism. A variable resonant mass is supported on elastomer elements and the dynamic characteristics are determined from measurements of input and output acceleration. Five different cartridges are considered: three of these are buttons cartridges having buttons located in pairs, with 120 between each pair. Two of the cartridges consist of 360 elastomer rings with rectangular cross-sections. Dynamic stiffness and damping are measured for each cartridge and compared with predictions at different frequencies and different strains

    Tests were performed on elastomer specimens of the material polybutadiene to determine the performance limitations imposed by strain, temperature, and frequency. Three specimens were tested: a shear specimen, a compression specimen, and a second compression specimen in which thermocouples were embedded in the elastomer buttons. Stiffness and damping were determined from all tests, and internal temperatures were recorded for the instrumented compression specimen. Measured results are presented together with comparisons between predictions of a thermo-viscoelastic analysis and the measured results. Dampers of polybutadiene and Viton were designed, built, and tested. Vibration measurements were made and sensitivity of vibration to change in unbalance was also determined. Values for log decrement were extracted from the synchronous response curves. Comparisons were made between measured sensitivity to unbalance and log decrement and predicted values for these quantities

    Spray automated balancing of rotors: Methods and materials

    The work described consists of two parts. In the first part, a survey is performed to assess the state of the art in rotor balancing technology as it applies to Army gas turbine engines and associated power transmission hardware. The second part evaluates thermal spray processes for balancing weight addition in an automated balancing procedure. The industry survey reveals that: (1) computerized balancing equipment is valuable to reduce errors, improve balance quality, and provide documentation; (2) slow-speed balancing is used exclusively, with no forseeable need for production high-speed balancing; (3) automated procedures are desired; and (4) thermal spray balancing is viewed with cautious optimism whereas laser balancing is viewed with concern for flight propulsion hardware. The FARE method (Fuel/Air Repetitive Explosion) was selected for experimental evaluation of bond strength and fatigue strength. Material combinations tested were tungsten carbide on stainless steel (17-4), Inconel 718 on Inconel 718, and Triballoy 800 on Inconel 718. Bond strengths were entirely adequate for use in balancing. Material combinations have been identified for use in hot and cold sections of an engine, with fatigue strengths equivalent to those for hand-ground materials

    The design of 20-mm bore ball bearings is described for cryogenic turbo-machinery applications, operating up to speeds of 120,000 rpm. A special section is included on the design of hybrid bearings. Each hybrid bearing is composed of a ball bearing in series with a conventional pressurized fluid-film journal bearing. Full details are presented on the design of a test vehicle which possesses the capability of testing the above named bearings within the given speed range under externally applied radial and axial loads

    The loss of tumour phospho-extracellular responsive kinase (pERK) positivity is the major treatment biomarker for mitogen-activated protein kinase/extracellular responsive kinase (MEK) inhibitors. Here, we demonstrate that there is a poor correlation between pERK inhibition and the anti-proliferative effects of MEK inhibitors in melanoma cells. We suggest that Ki67 is a better biomarker for future clinical studies

    Spitzer 3.6 micron and 4.5 micron full-orbit lightcurves of WASP-18

    We present new lightcurves of the massive hot Jupiter system WASP-18 obtained with the Spitzer spacecraft covering the entire orbit at 3.6 micron and 4.5 micron. These lightcurves are used to measure the amplitude, shape and phase of the thermal phase effect for WASP-18b. We find that our results for the thermal phase effect are limited to an accuracy of about 0.01% by systematic noise sources of unknown origin. At this level of accuracy we find that the thermal phase effect has a peak-to-peak amplitude approximately equal to the secondary eclipse depth, has a sinusoidal shape and that the maximum brightness occurs at the same phase as mid-occultation to within about 5 degrees at 3.6 micron and to within about 10 degrees at 4.5 micron. The shape and amplitude of the thermal phase curve imply very low levels of heat redistribution within the atmosphere of the planet. We also perform a separate analysis to determine the system geometry by fitting a lightcurve model to the data covering the occultation and the transit. The secondary eclipse depths we measure at 3.6 micron and 4.5 micron are in good agreement with previous measurements and imply a very low albedo for WASP-18b. The parameters of the system (masses, radii, etc.) derived from our analysis are in also good agreement with those from previous studies, but with improved precision. We use new high-resolution imaging and published limits on the rate of change of the mean radial velocity to check for the presence of any faint companion stars that may affect our results. We find that there is unlikely to be any significant contribution to the flux at Spitzer wavelengths from a stellar companion to WASP-18. We find that there is no evidence for variations in the times of eclipse from a linear ephemeris greater than about 100 seconds over 3 years.Comment: 17 pages, 10 figures.     Surface reactions of metal clusters. II. Reactivity surveys with D2, N2, and CO

    Reactions on the surface of a variety of transition metal clusters have been studied in the gas phase at near room temperature using a newly developed fast-flow reaction device. Initial examples of the use of this device are provided by survey studies of the reactivity of iron, cobalt, nickel, copper, and niobium clusters in contact with low concentrations of D2, N2 and CO. Dissociative chemisorption of D2 is found to occur with dramatic sensitivity to cluster size in the cases of iron, cobalt, and niobium clusters, the detailed pattern of reactivity differing markedly for each metal. The corresponding reaction is also observed with nickel clusters, but here the reactivity shows only a slow, steady increase with cluster size. Copper clusters are found to be completely unreactive to H2 chemisorption under these conditions. Molecular nitrogen is found to chemisorb readily to clusters of cobalt and niobium, with a reactivity pattern very similar to that observed with D2. Iron clusters are found to show slight reactivity with N2; only a small amount of chemisorption is observed on the most reactive clusters at high N2 concentration, but the pattern of this reactivity with cluster size is consistent with that observed in D2 chemisorption. In contrast to these highly structured reactivity patterns of D2 and N2, carbon monoxide is found to show only a slow, monotonic increase in reactivity with cluster size. It is suggested that these dramatic reactivity patterns for chemisorption on metal clusters provide stringent tests for future theories as to the nature of chemisorption on metal surfaces at a detailed, molecular level

    The receptor protein tyrosine phosphatase PTPRB negatively regulates FGF2-dependent branching morphogenesis

    PTPRB is a transmembrane protein tyrosine phosphatase known to regulate blood vessel remodelling and angiogenesis. Here we demonstrate that PTPRB negatively regulates branching morphogenesis in the mammary epithelium. We show that Ptprb is highly expressed in adult mammary stem cells and also, although at lower levels, in estrogen receptor positive luminal cells. During mammary development Ptprb expression is down-regulated during puberty, a period of extensive of ductal outgrowth and branching. In vivo shRNA knockdown of Ptprb in the cleared mammary fat pad transplant assay resulted in smaller epithelial outgrowths with an increased branching density and also increased branching in an in vitro organoid assay. Organoid branching was dependent on stimulation by FGF2, and Ptprb knockdown in mammary epithelial cells resulted in a higher level of FGFR activation and ERK1/2 phosphorylation, both at baseline and following FGF2 stimulation. Therefore, PTPRB regulates branching morphogenesis in the mammary epithelium by modulating the response of the FGFR signalling pathway to FGF stimulation. Considering the importance of branching morphogenesis in multiple taxa, our findings have general importance outside mammary developmental biology

    HmuY Haemophore and Gingipain Proteases Constitute a Unique Syntrophic System of Haem Acquisition by Porphyromonas gingivalis

    Haem (iron protoporphyrin IX) is both an essential growth factor and virulence regulator for the periodontal pathogen Porphyromonas gingivalis, which acquires it mainly from haemoglobin via the sequential actions of the R- and K-specific gingipain proteases. The haem-binding lipoprotein haemophore HmuY and its cognate receptor HmuR of P. gingivalis, are responsible for capture and internalisation of haem. This study examined the role of the HmuY in acquisition of haem from haemoglobin and the cooperation between HmuY and gingipain proteases in this process. Using UV-visible spectroscopy and polyacrylamide gel electrophoresis, HmuY was demonstrated to wrest haem from immobilised methaemoglobin and deoxyhaemoglobin. Haem extraction from oxyhaemoglobin was facilitated after oxidation to methaemoglobin by pre-treatment with the P. gingivalis R-gingipain A (HRgpA). HmuY was also capable of scavenging haem from oxyhaemoglobin pre-treated with the K-gingipain (Kgp). This is the first demonstration of a haemophore working in conjunction with proteases to acquire haem from haemoglobin. In addition, HmuY was able to extract haem from methaemalbumin, and could bind haem, either free in solution or from methaemoglobin, even in the presence of serum albumin
