    Fathers today: design of a randomized controlled trial examining the role of oxytocin and vasopressin in behavioral and neural responses to infant signals

    BACKGROUND: Previous research has mostly focused on the hormonal, behavioral and neural correlates of maternal caregiving. We present a randomized, double-blind, placebo-controlled within-subject design to examine the effects of intranasal administration of oxytocin and vasopressin on parenting behavior and the neural and behavioral responses to infant cry sounds and infant threat. In addition, we will test whether effects of oxytocin and vasopressin administration are moderated by fathers' early childhood experiences. METHODS: Fifty-five first-time fathers of a child between two and seven months old will participate in three experimental sessions with intervening periods of one to two weeks. Participants self-administer oxytocin, vasopressin or a placebo. Infant-father interactions and protective parenting responses are observed during play. Functional Magnetic Resonance Imaging (fMRI) is used to examine the neural processing of infant cry sounds and infant threat. A handgrip dynamometer is used to measure use of handgrip force when listening to infant cry sounds. Participants report on their childhood experiences of parental love-withdrawal and abuse and neglect. DISCUSSION: The results of this study will provide important insights into the hormonal, behavioral and neural correlates of fathers' parenting behavior during the early phase of fatherhood.     Oxytocin Receptor Gene Associated with the Efficiency of Social Auditory Processing

    Oxytocin has been shown to facilitate social aspects of sensory processing, thereby enhancing social communicative behaviors and empathy. Here we report that compared to the AA/AG genotypes, the presumably more efficient GG genotype of an oxytocin receptor gene polymorphism (OXTR rs53576) that has previously been associated with increased sensitivity of social processing is related to less self-reported difficulty in hearing and understanding people when there is background noise. The present result extends associations between oxytocin and social processing to the auditory and vocal domain. We discuss the relevance of our findings for autistic spectrum disorders (ASD), as ASD seems related to specific impairments in the orienting to, and selection of speech sounds from background noise, and some social processing impairments in patients with ASD have been found responsive to oxytocin treatment

    The Significance of Early Temperamental Reactivity for Children's Social Competence With Peers: A Meta-Analytic Review and Comparison With the Role of Early Attachment

    Early temperamental reactivity and attachment security are key predictors of children's social competence with peers. Leveraging meta-analytic evaluation of the significance of early attachment for social competence already available (Groh et al., 2014), this quantitative review examined the significance of early temperamental reactivity for social competence with peers and compared the strength of this association with that for attachment. Based on 140 independent samples (u = 382; N = 49,891), the meta-analytic association between early difficult temperament and (lower) social competence was significant (r = 0.13, z = 0.13; 95% CI [0.11, 0.16]), but decreased as time between assessments increased. Findings were similar for negative and positive emotionality. Greater negative emotionality was associated with lower social competence (r = 0.14, z = 0.14; 95% CI [0.11, 0.17], k = 93, u = 172), and greater positive emotionality was associated with better social competence (r = 0.18, z = 0.18; 95% CI [0.12, 0.24], k = 43, u = 54). Meta-analytic associations were reduced when overlapping informants and overlapping items in temperament and social competence assessments were excluded (difficult temperament: r = 0.10, z = 0.10; 95% CI [0.06, 0.13]; negative emotionality: r = 0.10, z = 0.10; 95% CI [0.05, 0.15]; positive emotionality: r = 0.10, z = 0.10; 95% CI [0.06, 0.14]). Meta-analytic associations between these broadband temperament dimensions and social competence were smaller than the meta-analytic association between attachment security and social competence. Discussion focuses on the developmental significance of early temperament for social competence and ways to reconcile literatures on early temperament and attachment in future research on the developmental antecedents of children's social competence.     The role of supportive parenting and stress reactivity in the development of self-regulation in early childhood

    Maternal sensitivity and supportive discipline are important determinants of child self-regulation. Some evidence suggests that specific genetic or temperamental markers determine children’s susceptibility to the impact of maternal parenting on child self-regulation. Cortisol reactivity as a susceptibility marker moderating the relation between maternal parenting and child self-regulation has not yet been studied. In this longitudinal population-based study (N = 258), the moderating role of infant cortisol stress response to the Strange Situation Procedure at age 1 was examined in the association between parenting (sensitivity and supportive discipline) at age 3 and child self-regulation at age 3 and 4. Maternal sensitivity and supportive discipline were related to child immediate and prolonged delay of gratification at age 3, and maternal sensitivity was related to working memory skills at age 4. No evidence of differential susceptibility to maternal parenting was found, based on differences in infant cortisol stress response

    Epigenomics of being bullied:changes in DNA methylation following bullying exposure

    Bullying among children is ubiquitous and associated with pervasive mental health problems. However, little is known about the biological pathways that change after exposure to bullying. Epigenome-wide changes in DNA methylation in peripheral blood were studied from pre- to post measurement of bullying exposure, in a longitudinal study of the population-based Generation R Study and Avon Longitudinal Study of Parents and Children (combined n = 1,352). Linear mixed-model results were meta-analysed to estimate how DNA methylation changed as a function of exposure to bullying. Sensitivity analyses including co-occurring child characteristics and risks were performed, as well as a Gene Ontology analysis. A candidate follow-up was employed for CpG (cytosine-phosphate-guanine) sites annotated to 5-HTT and NR3C1. One site, cg17312179, showed small changes in DNA methylation associated to bullying exposure (b = −2.67e-03, SE = 4.97e-04, p = 7.17e-08). This site is annotated to RAB14, an oncogene related to Golgi apparatus functioning, and its methylation levels decreased for exposed but increased for non-exposed. This result was consistent across sensitivity analyses. Enriched Gene Ontology pathways for differentially methylated sites included cardiac function and neurodevelopmental processes. Top CpG sites tended to have overall low levels of DNA methylation, decreasing in exposed, increasing in non-exposed individuals. There were no gene-wide corrected findings for 5-HTT and NR3C1. This is the first study to identify changes in DNA methylation associated with bullying exposure at the epigenome-wide significance level. Consistent with other population-based studies, we do not find evidence for strong associations between bullying exposure and DNA methylation

    The significance of attachment security for children’s social competence with peers: a meta-analytic study

    This meta-analytic review examines the association between attachment during the early life course and social competence with peers during childhood, and compares the strength of this association with those for externalizing and internalizing symptomatology. Based on eighty independent samples (N = 4,441), the association between security and peer competence was significant (d = 0.39, CI 0.32; 0.47) and not moderated by the age at which peer competence was assessed. Avoidance (d = 0.17, CI 0.05; 0.30), resistance (d = 0.29, CI 0.09; 0.48), and disorganization (d = 0.25, CI 0.10; 0.40) were significantly associated with lower peer competence. Attachment security was significantly more strongly associated with peer competence than internalizing (but not externalizing) symptomatology. Discussion focuses on the significance of early attachment for the development of peer competence versus externalizing and internalizing psychopathology

    Not the Root of the Problem-Hair Cortisol and Cortisone Do Not Mediate the Effect of Child Maltreatment on Body Mass Index.

    BACKGROUND: Experiencing maltreatment during childhood exerts substantial stress on the child and increases the risk for overweight and obesity later in life. The current study tests whether hair cortisol-a measure of chronic stress-and its metabolite cortisone mediate the relation between abuse and neglect on the one hand, and body mass index (BMI) on the other. METHOD: The sample consisted of 249 participants aged 8 to 87 years (M = 36.13, SD = 19.33). We collected data on child abuse and neglect using questionnaires, measured cortisol and cortisone concentrations in hair, and BMI. In a structural model, the effects of abuse and neglect on hair cortisol, hair cortisone, and BMI were tested, as well as the covariance between hair cortisol and BMI, and hair cortisone and BMI. RESULTS: Within the sample, 23% were overweight but not obese and 14% were obese. Higher levels of experienced abuse were related to higher cortisone concentrations in hair (β = 0.24, p < .001) and higher BMI (β = 0.17, p =.04). Neglect was not related to hair cortisol, hair cortisone, or BMI. Hair cortisol and cortisone did not mediate the association between maltreatment, and BMI. Sensitivity analyses demonstrate the same pattern of results in a subsample of adult participants currently not living with their parents. However, in younger participants who were still living with their parents, the associations between abuse and cortisone (β = 0.14, p =.35) and abuse and BMI (β = 0.02, p =.92) were no longer significant. CONCLUSION: These findings confirm that experiencing abuse is related to higher BMI but suggest that hair cortisol and cortisone are not the mechanism underlying the association between child maltreatment and BMI. This is the first study to show abuse may be associated to elevated concentrations of hair cortisone-evidence of long-term alterations in chronic stress levels. Future research may benefit from exploring the effects of maltreatment on weight gain in longitudinal designs, including measures of other potential mediators such as eating as a coping mechanism, and more direct indicators of metabolic health
