    New objects in old structures: The Iron Age hoard of the Palacio III megalithic funerary complex (Almadén de la Plata, Seville, Spain)

    Cultural contact, exchange and interaction feature high in the list of challenging topics of current research on European Prehistory. Not far off is the issue of the changing role of monuments in the making and maintaining of key cultural devices such as memory and identity. Addressing both these highly-debated issues from a science-based perspective, in this paper we look at an unusual case study set in southern Iberia and illustrate how these archaeological questions can benefit from robust materials-science approaches.We present the contextual, morphological and analytical study of an exceptional Early Iron Age hoard composed of a number of different (and mostly exotic) materials such as amber, quartz, silver and ceramic. This hoard, found under the fallen orthostat of a megalithic structure built at least 2000 years earlier, throws new light on long-distance exchange networks and the effect they could have had on the cultural identities and social relations of local Iberian Early Iron Age communities. Moreover, the archaeometric study reveals how diverse and distant the sources of these item are (Northern Europe to Eastern and Western Mediterranean raw materials, as well as local and eastern technologies), therefore raising questions concerning the social mechanisms used to establish change and resistance in contexts of colonial encounter

    Introduction. Brain ischemia and reperfusion produce alterations in the microenvironment of the parenchyma, including ATP depletion, ionic homeostasis alterations, inflammation, release of multiple cytokines and abnormal release of neurotransmitters. As a consequence, the induction of proliferation and migration of neural stem cells towards the peri-infarct region occurs. Development. The success of new neurorestorative treatments for damaged brain implies the need to know, with greater accuracy, the mechanisms in charge of regulating adult neurogenesis, both under physiological and pathological conditions. Recent evidence demonstrates that many neurotransmitters, glutamate in particular, control the subventricular zone, thus being part of the complex signalling network that influences the production of new neurons. Conclusion. Neurotransmitters provide a link between brain activity and subventricular zone neurogenesis. Therefore, a deeper knowledge of the role of neurotransmitters systems, such as glutamate and its transporters, in adult neurogenesis, may provide a valuable tool to be used as a neurorestorative therapy in this pathology.Peer Reviewe

    Systemic oncolytic adenovirus delivered in mesenchymal carrier cells modulate tumor infiltrating immune cells and tumor microenvironment in mice with neuroblastoma.

    Celyvir (autologous mesenchymal cells -MSCs- that carry an oncolytic adenovirus) is a new therapeutic strategy for metastatic tumors developed by our research group over the last decade. There are limitations for studying the immune effects of human oncolytic adenoviruses in murine models since these viruses do not replicate naturally in these animals. The use of xenografts in immunodeficient mice prevent assessing important clinical aspects of this therapy such as the antiadenoviral immune response or the possible intratumoral immune changes, both of tumor infiltrating leukocytes and of the microenvironment. In our strategy, the presence of MSCs in the medicinal product adds an extra level of complexity. We present here a murine model that overcomes many of these limitations. We found that carrier cells outcompeted intravenous administration of naked particles in delivering the oncolytic virus into the tumor masses. The protection that MSCs could provide to the oncolytic adenovirus did not preclude the development of an antiadenoviral immune response. However, the presence of circulating antiadenoviral antibodies did not prevent changes detected at the tumor masses: increased infiltration and changes in the quality of immune cells per unit of tumor volume, and a less protumoral and more inflammatory profile of the tumor microenvironment. We believe that the model described here will enable the study of crucial events related to the immune responses affecting both the medicinal product and the tumor

    All-in-one construct for genome engineering using Cre-lox technology

    Mycoplasma genitalium is an appealing model of a minimal cell and synthetic biology study, and it was one of the first organisms whose genome was fully sequenced and chemically synthesized. Despite its usefulness as a model organism, many genetic tools well established for other microorganisms are not currently available in mycoplasmas. We have developed several vectors to adapt the Cre-lox technology for genome engineering in M. genitalium, providing an all-in-one construct that could be also useful to obtain unmarked genetic modifications in many other slow growing microorganisms. This construct contains a modified promoter sequence based in TetR system that exhibits an enhanced control on Cre recombinase expression, virtually abolishing the presence of this recombinase in the absence of inducer. This allows to introduce the Cre recombinase gene and the desired genetic modification in a single transformation step. In addition, this inducible promoter may be a very promising tool for a wide range of molecular applications

    The ceratopogonid Culicoides paradoxalis Ramilo & Delécolle, 2013 is recorded for the first time in Spain based on reliable morphological evidence according to the previous descriptions of other authors. A total of 438 females (349 nulliparous and 89 parous) and a single male were collected with CDC miniature light traps at three different livestock-associated locations in Extremadura Autonomous Community (Spain) in 2014. Most specimens were captured between June and August, suggesting a univoltine pattern for this species extended over summer and early autumn. Although the number of collections of C. paradoxalis is low in comparison with the dominant species, the occurrence of this species in monitoring surveillance programs should deserve specific attention in order to estimate the accurate ratio of potential vectors unmistakably. Interesting information about the period of flight and illustrated morphological features are presented for C. paradoxalis in the current paper.Se cita por primera vez en España el ceratopogónido Culicoides paradoxalis Ramilo & Delécolle, 2013, basándose en evidencias morfológicas de acuerdo a las descripciones previas de otros autores. Un total de 438 hembras (349 nulíparas y 89 paras) y un macho se recolectaron con minitrampas de luz CDC en tres localidades ganaderas en la Comunidad Autónoma de Extremadura (España) en 2014. La mayor parte de los especímenes fueron capturados entre junio y agosto, mostrando un único período de vuelo que se extendió durante todo el verano y principios del otoño. Aunque el número de capturas de C. paradoxalis es reducido en comparación con los Culicoides dominantes, la aparición de esta nueva especie merece especial atención en los programas de vigilancia entomológica con el fin de estimar inequívocamente la proporción exacta de vectores potenciales.     Psychometric characteristics of a commuting-to-school behaviour questionnaire for families

    The purposes of this study were: (a) to describe the patterns of modes of commuting to school (children) and to work (parents) separated by gender and age, (b) to validate the questions on children’s mode of commuting to and from school according to their parents, and (c) to analyse the reliability of a family questionnaire focused on commuting to school behaviours. A total of 611 parents (mean age: 43.28 ± 6.25 years old) from Granada (Spain) completed “Family commuting-to-school behaviour” questionnaire in two sessions separated by 14 days, (2016 and 2018). The validation between family and children’s questions was assessed using the Kappa and Spearman correlation coefficients, and the test–retest reliability within the family questions was assessed using the Kappa and the weighted Kappa. The children’s modes of commuting to school (mean age: 11.44 ± 2.77 years old) were mainly passive (57.7% to school) while parents’ modes of commuting to work were mainly active (71.6%). The validity of the mode of commuting was significant with high Kappa and Spearman coefficients. The test–retest reliability presented a good agreement for the mode of commuting to school in children, distance and time to school, and the mode of commuting to work in parents, while the questions on acceptable distance to walk or cycle to school showed a moderate to good agreement. The “Family commuting-to-school behaviour” questionnaire could be a useful tool to assess the mode of commuting of children, distance and time to school for researchers and practitioners

    Análisis del compromiso deportivo de jóvenes waterpolistas: Un estudio basado en la Teoría de la AutoDeterminación

    Fundamentado en la Teoría de la Autodeterminación (TAD), el objetivo del estudio fue analizar la relación entre la percepción de jóvenes waterpolistas sobre el estilo interpersonal del entrenador, la satisfacción y frustración de sus necesidades psicológicas básicas (NPB), la motivación autodeterminada y su compromiso deportivo. Participaron 77 jugadores de waterpolo con una edad comprendida entre los 13 y los 20 años (M = 15.00, DT = 1.70). Los resultados mostraron que la percepción de los jugadores de un estilo de apoyo a la autonomía del entrenador, la satisfacción de las NPB, la motivación intrínseca y el compromiso deportivo se relacionaba positiva y significativamente, siguiendo la secuencia establecida en la TAD. En sentido contrario, el estilo controlador percibido por los jugadores se relacionaba positiva y significativamente con la frustración de las NPB y la desmotivación y negativamente con el compromiso deportivo. Los resultados destacan la importancia del estilo interpersonal del entrenador sobre los procesos motivacionales y el compromiso deportivo de los jóvenes waterpolistas. Grounded in Self-Determination Theory (SDT), the aim of this study was to analyze the relationship between waterpolo players perception on coach interpersonal style, satisfaction and frustration of their basic psychological needs (BPN), self-determined motivation and sport commitment. 77 male water polo players, with ages between 13 and 20 years old (M = 15.00, SD = 1.70), took part in the study. Results showed that players' perception of autonomy support coaching style was related positively and significantly with the satisfaction of BPN, intrinsic motivation and sport commitment, following established sequence in SDT. However, controlling coaching style was related positively and significantly with the frustration of BPN, amotivation and negatively with sport commitment. The results highlight the importance of coach interpersonal style on motivational processes and the sports commitment of young water polo players

    Feasibility and Reliability of a Questionnaire to Assess the Mode, Frequency, Distance and Time of Commuting to and from School: The PACO Study

    Active commuting to and from school has several health implications. Self-reporting is the most common assessment tool, but there is a high heterogeneity of questionnaires in the scientific literature. The purpose of this study was to analyse the feasibility and reliability of the Spanish “New Version of Mode and Frequency of Commuting To and From School” questionnaire in children and adolescents. A total of 635 children (5–12 years old) and 362 adolescents (12–18 years old) filled out the questionnaire twice (14 days apart). Feasibility was evaluated using an observational checklist. The test-retest reliability of the “New Version of Mode and Frequency of Commuting To and From School” questionnaire and the distance and time to school were examined using the kappa and weight kappa coefficient (κ). No misunderstanding of questions was reported. The time to complete the questionnaire was 15 ± 3.62 and 9 ± 2.26 min for children and adolescents, respectively. The questionnaire showed substantial and almost perfect kappa coefficients for the overall six items (k = 0.61–0.94) in children and adolescents.
