    Non-suicidal self-injury (Nssi) in adolescent inpatients: assessing personality features and attitude toward death

    <p>Abstract</p> <p>Background</p> <p>Non-suicidal self-injury (NSSI) is a common concern among hospitalized adolescents, and can have significant implications for short and long-term prognosis. Little research has been devoted on how personality features in severely ill adolescents interact with NSSI and "attitude toward life and death" as a dimension of suicidality. Developing more specific assessment methodologies for adolescents who engage in self-harm without suicidal intent is relevant given the recent proposal of a non-suicidal self-injury (NSSI) disorder and may be useful in predicting risk in psychiatrically impaired subjects.</p> <p>Methods</p> <p>Consecutively hospitalized adolescents in a psychiatric unit (N = 52; 71% females; age 12-19 years), reporting at least one recent episode of self-harm according to the <it>Deliberate Self-harm Inventory</it>, were administered the <it>Structured Clinical Interview for DSM Mental Disorders and Personality Disorders (SCID I and II)</it>, the <it>Children's Depression Inventory </it>and the <it>Multi-Attitude Suicide Tendency Scale (MAST)</it>.</p> <p>Results</p> <p>Mean age onset of NSSI in the sample was 12.3 years. All patients showed "repetitive" NSSI (high frequency of self-harm), covering different modalities. Results revealed that 63.5% of adolescents met criteria for Borderline Personality Disorder (BPD) and that the rest of the sample also met criteria for personality disorders with dysregulated traits. History of suicide attempts was present in 46.1% of cases. Elevated depressive traits were found in 53.8%. Results show a statistically significant negative correlation between the score on the "Attraction to Life" subscale of the MAST and the <it>frequency </it>and <it>diversification </it>of self-harming behaviors.</p> <p>Conclusions</p> <p>Most adolescent inpatients with NSSI met criteria for emotionally dysregulated personality disorders, and showed a reduced "attraction to life" disposition and significant depressive symptoms. This peculiar psychopathological configuration must be addressed in the treatment of adolescent inpatients engaging in NSSI and taken into account for the prevention of suicidal behavior in self-injuring adolescents who do not exhibit an explicit intent to die.</p

    Numerical and Experimental Characterization of the Seismic Parameters of a Soft Soil Reinforced with Rigid Inclusions

    International audienceThe characterization of the properties of soft soils reinforced with periodically placed cylindrical rigid inclusions is essential to improve the lifetime of railway tracks submitted to the critical speed issue. This work is about the use of numerical and experimental resources to characterize the velocity of surface waves of these reinforced media. The numerical approach is presented, using the spectral elements method (SEM3D) coupled with non-periodic homogenization. The homogenized medium is used to modify a discontinuous media into a medium with smoother contrasts of properties. Into the context of numerical simulations, it allows to suppress the sharp discontinuities, and thus reduces the cost of the computation linked to the mesh refinement. A cross-validation is made with a reduced scale experimental model, performed with the high-quality measurement bench MUSC. Two different configurations are studied, the first one with three inclusions is used to cross-validate the use of the homogenization method with the experimental measurements and the second is made on more complex case containing a high number of inclusions on the same type of resins

    Improved generation of human induced pluripotent stem cell-derived cardiac pacemaker cells using novel differentiation protocols

    Current protocols for the differentiation of human-induced pluripotent stem cells (hiPSC) into cardiomyocytes only generate a small amount of cardiac pacemaker cells. In previous work, we reported the generation of high amounts of cardiac pacemaker cells by co-culturing hiPSC with mouse visceral endoderm-like (END2) cells. However, potential medical applications of cardiac pacemaker cells generated according to this protocol, comprise an incalculable xenogeneic risk. We thus aimed to establish novel protocols maintaining the differentiation efficiency of the END2 cell-based protocol, yet eliminating the use of END2 cells. Three protocols were based on the activation and inhibition of the Wingless/Integrated (Wnt) signaling pathway, supplemented either with retinoic acid and the Wnt activator CHIR99021 (protocol B) or with the NODAL inhibitor SB431542 (protocol C) or with a combination of all three components (protocol D). An additional fourth protocol (protocol E) was used, which was originally developed by the manufacturer STEMCELL Technologies for the differentiation of hiPSC or hESC into atrial cardiomyocytes. All protocols (B, C, D, E) were compared to the END2 cell-based protocol A, serving as reference, in terms of their ability to differentiate hiPSC into cardiac pacemaker cells. Our analysis revealed that protocol E induced upregulation of 12 out of 15 cardiac pacemaker-specific genes. For comparison, reference protocol A upregulated 11, while protocols B, C and D upregulated 9, 10 and 8 cardiac pacemaker-specific genes, respectively. Cells differentiated according to protocol E displayed intense fluorescence signals of cardiac pacemaker-specific markers and showed excellent rate responsiveness to adrenergic and cholinergic stimulation. In conclusion, we characterized four novel and END2 cell-independent protocols for the differentiation of hiPSC into cardiac pacemaker cells, of which protocol E was the most efficient
