    Bone marrow cells contribute to tissue regeneration in the intestine and skin.

    Adult bone marrow contains progenitor cells that can extricate themselves from their bone marrow cavity niche, and engraft within foreign tissues, whereupon they produce specific differentiated adult lineages. Bone marrow engraftment is upregulated with increasing regenerative pressure, which has triggered speculation as to the therapeutic potential of bone marrow cells. In this thesis, I describe for the first time, that transplanted adult bone marrow cells engraft within the intestines of mice and humans and give rise to a substantial proportion of the mesenchymal cells in the lamina propria, namely, the intestinal subepithelial myofibroblasts. I show that bone marrow contribution to intestinal myofibroblasts is significantly enhanced in a mouse model of colitis. Furthermore, I show that bone marrow cells form multiple vascular lineages in the mouse colon, with an enhanced propensity in colitis. These results have implications for the use of bone marrow in the treatment of inflammatory bowel disease, either by directly producing mesenchymal and vascular lineages, and/or possibly via the indirect regulation of intestinal epithelial cells. I also report that transplanted bone marrow cells engraft into the skin and contribute to multiple epidermal lineages. These bone marrow-derived cells are commonly located within postulated epidermal stem cell niches, and can express putative epidermal stem cell markers. Bone marrow cell engraftment was significantly enhanced in the wounded epidermis, and bone marrow-derived epidermal cells can proliferate in vivo, and form colonies in vitro. I found no evidence that bone marrow cells fuse with indigenous epidermal cells to form heterokaryons. These results highlight the potential of bone marrow cells in the treatment of diseases of the gastrointestinal tract and skin

    A new encoding scheme for visible light communications with applications to mobile connections

    A new, novel and unconventional encoding scheme called concurrent coding, has recently been demonstrated and shown to offer interesting features and benefits in comparison to conventional techniques, such as robustness against burst errors and improved efficiency of transmitted power. Free space optical communications can suffer particularly from issues of alignment which requires stable, fixed links to be established and beam wander which can interrupt communications. Concurrent coding has the potential to help ease these difficulties and enable mobile, flexible optical communications to be implemented through the use of a source encoding technique. This concept has been applied for the first time to optical communications where standard light emitting diodes (LEDs) have been used to transmit information encoded with concurrent coding. The technique successfully transmits and decodes data despite unpredictable interruptions to the transmission causing significant drop-outs to the detected signal. The technique also shows how it is possible to send a single block of data in isolation with no pre-synchronisation required between transmitter and receiver, and no specific synchronisation sequence appended to the transmission. Such systems are robust against interference -- intentional or otherwise -- as well as intermittent beam blockage

    Transcriptome analysis of <i>Streptococcus gordonii </i>Challis DL1 indicates a role for the biofilm-associated <i>fruRBA </i>operon in response to <i>Candida albicans</i>

    Multiple levels of interkingdom signaling have been implicated in maintaining the ecological balance between Candida albicans and commensal streptococci to assure a state of oral health. To better understand the molecular mechanisms involved in the initial streptococcal response to the presence of C. albicans that can initiate oral surface colonization and biofilm formation, hypha-forming cells were incubated with Streptococcus gordonii cells for 30 minutes to assess the streptococcal transcriptome response. A genome wide microarray analysis and quantitative PCR validation of S. gordonii transcripts identified a number of genes, the majority of which were involved in metabolic functions that were differentially expressed in the presence of hyphae. The fruR, fruB and fruA genes encoding the transcriptional regulator, fructose-1-phosphate kinase, and fructose-specific permease, respectively, of the phosphoenolpyruvate-dependent fructose phosphotransferase system, were consistently up-regulated. An S. gordonii mutant in which these genes were deleted by allelic replacement, formed an architecturally-distinct, less robust biofilm with C. albicans than did parental strain cells. Complementing the mutant with plasmid borne fruR, fruB and fruA genes caused phenotype reversion, indicating that the genes in this operon played a role in dual species biofilm formation. This genome wide analysis of the S. gordonii transcriptional response to C. albicans has identified several genes that have potential roles in interkingdom signaling and responses

    Mathematical modeling of cell population dynamics in the colonic crypt and in colorectal cancer

    Colorectal cancer is initiated in colonic crypts. A succession of genetic mutations or epigenetic changes can lead to homeostasis in the crypt being overcome, and subsequent unbounded growth. We consider the dynamics of a single colorectal crypt by using a compartmental approach [Tomlinson IPM, Bodmer WF (1995) Proc Natl Acad Sci USA 92: 11130-11134], which accounts for populations of stem cells, differential cells, and transit cells. That original model made the simplifying assumptions that each cell popuation divides synchronously, but we relax these assumptions by adopting an age-structured approach that models asynchronous cell division, and by using a continuum model. We discuss two mechanims that could regulate the growth of cell numbers and maintain the equilibrium that is normally observed in the crypt. The first will always maintain an equilibrium for all parameter values, whereas the second can allow unbounded proliferation if the net per capita growth rates are large enough. Results show that an increase in cell renewal, which is equivalent to a failure of programmed cell death or of differentiation, can lead to the growth of cancers. The second model can be used to explain the long lag phases in tumor growth, during which news, higher equilibria are reached, before unlimited growth in cell number ensues

    General-Purpose Coarse-Grained Toughened Thermoset Model for 44DDS/DGEBA/PES

    The objective of this work is to predict the morphology and material properties of crosslinking polymers used in aerospace applications. We extend the open-source dybond plugin for HOOMD-Blue to implement a new coarse-grained model of reacting epoxy thermosets and use the 44DDS/DGEBA/PES system as a case study for calibration and validation. We parameterize the coarse-grained model from atomistic solubility data, calibrate reaction dynamics against experiments, and check for size-dependent artifacts. We validate model predictions by comparing glass transition temperatures measurements at arbitrary degree of cure, gel-points, and morphology predictions against experiments. We demonstrate for the first time in molecular simulations the cure-path dependence of toughened thermoset morphologies

    In vivo model for microbial invasion of tooth root dentinal tubules

    ABSTRACT Objective Bacterial penetration of dentinal tubules via exposed dentine can lead to root caries and promote infections of the pulp and root canal system. The aim of this work was to develop a new experimental model for studying bacterial invasion of dentinal tubules within the human oral cavity. Material and Methods Sections of human root dentine were mounted into lower oral appliances that were worn by four human subjects for 15 d. Roots were then fixed, sectioned, stained and examined microscopically for evidence of bacterial invasion. Levels of invasion were expressed as Tubule Invasion Factor (TIF). DNA was extracted from root samples, subjected to polymerase chain reaction amplification of 16S rRNA genes, and invading bacteria were identified by comparison of sequences with GenBank database. Results All root dentine samples with patent tubules showed evidence of bacterial cell invasion (TIF value range from 5.7 to 9.0) to depths of 200 mm or more. A spectrum of Gram-positive and Gram-negative cell morphotypes were visualized, and molecular typing identified species of Granulicatella, Streptococcus, Klebsiella, Enterobacter, Acinetobacter, and Pseudomonas as dentinal tubule residents. Conclusion A novel in vivo model is described, which provides for human root dentine to be efficiently infected by oral microorganisms. A range of bacteria were able to initially invade dentinal tubules within exposed dentine. The model will be useful for testing the effectiveness of antiseptics, irrigants, and potential tubule occluding agents in preventing bacterial invasion of dentine

    Globalization, the ambivalence of European integration and the possibilities for a post-disciplinary EU studies

    Using the work of Manuel Castells as a starting point, this article explores the ambivalent relationship between globalization and European integration and the variety of ways in which the mainstream political science of the EU has attempted to deal with this issue. The analysis here suggests that various 'mainstreaming' disciplinary norms induce types of work that fail to address fully the somewhat paradoxical and counter-intuitive range of possible relationships between globalization and European integration. The article explores critically four possible analytical ways out of this paradox—abandonment of the concept of globalization, the development of definition precision in globalization studies, the reorientation of work to focus on globalization as discourse, and inter- and post-disciplinarity. The argument suggests that orthodox discussions of the relationship require a notion of social geography that sits at odds with much of the literature on globalization and while greater dialogue between disciplines is to be welcomed, a series of profound epistemological questions need to be confronted if studies of the interplay between global and social process are to be liberated from their disciplinary chains

    Hierarchical model for the scale-dependent velocity of seismic waves

    Elastic waves of short wavelength propagating through the upper layer of the Earth appear to move faster at large separations of source and receiver than at short separations. This scale dependent velocity is a manifestation of Fermat's principle of least time in a medium with random velocity fluctuations. Existing perturbation theories predict a linear increase of the velocity shift with increasing separation, and cannot describe the saturation of the velocity shift at large separations that is seen in computer simulations. Here we show that this long-standing problem in seismology can be solved using a model developed originally in the context of polymer physics. We find that the saturation velocity scales with the four-third power of the root-mean-square amplitude of the velocity fluctuations, in good agreement with the computer simulations.Comment: 7 pages including 3 figure

    Jekyll and Hyde: men's constructions of feminism and feminists

    Research and commentary on men's responses to feminism has demonstrated the range of ways in which men have mobilised both against and for feminist principles. This paper argues that further analyses of men's responses require a sophisticated theory of discourse acknowledging the fragmented and contradictory nature of representation. A corpus of men's talk on feminism and feminists was studied to identify the pervasive patterns in men's accounting and regularities in rhetorical organisation. Material from two samples of men was included: a sample of white middle-class 17-18 year old school students and a sample of 60 interviews with a more diverse sample of older men aged 20 to 64. Two interpretative repertoires of feminism and feminists were identified. These set up a 'Jekyll and Hyde' binary and positioned feminism along with feminists very differently as reasonable versus extreme and monstrous. Both repertoires tended to be deployed together and the paper explores the ideological and interactional consequences of typical deployments along with the identity work accomplished by the men as they positioned themselves in relation to these
