Sheehan’s syndrome is the name given to postpartum hypopituitarism, usually precipitated by massive uterine haemorrhage and hypovolemic shock during or after childbirth resulting into pituitary injury in varying degrees. Most cases are diagnosed very late in life and the first presentation may be a life-threatening situation like shock or cardiorespiratory collape. This hidden nemesis in women compromises their quality of life and may result into mortality if not suspected early. PENGARUH LAYANAN ORIENTASI BIDANG PENGEMBANGAN HUBUNGAN SOSIAL TERHADAP PENYESUAIAN DIRI SISWA KELAS VII MTs NURUL JADID BENGKALIS

ABSTRAK M.Ibnu Sina Alpa Roby, (2020) : Pengaruh Layanan Orientasi Bidang Pengembangan Hubungan Sosial Terhadap Penyesuaian Diri pada Siswa Kelas VII di Madrasah Tsanawiyah Nurul Jadid Bengkalis Penelitian ini bertujuan untuk mengetahui pengaruh yang signifikan antara layanan orientasi bidang pengembangan hubungan sosial terhadap penyesuaian diri siswa kelas VII di Madrasah Tsanawiyah Nurul Jadid bengkalis. Jenis penelitian ini adalah korelasi. Subjek penelitian ini adalah seluruh siswa kelas VII berjumlah 65 siswa, sedangkan objek penelitian ini adalah pengaruh layanan orientasi bidang pengembangan hubungan sosial terhadap penyesuaian diri siswa. Populasinya adalah seluruh siswa di Madrasah Tsanawiyah Nurul Jadid bengkalis yang berjumlah 95 orang. Pengambilan sampel penelitian ini menggunakan teknik purposive sampling yaitu kelas VII yang berjumlah 65 orang siswa.Teknik pengumpulan data yang digunakan adalah angket dan dokumentasi. Teknik analisis yang digunakan untuk menguji hipotesis dalam penelitian ini adalah menggunakan rumus regresi linier sederhana. Berdasarkan hasil analisis terbukti bahwa ada pengaruh yang cukup signifikan antara layanan orientasi bidang pengembangan hubungan sosial terhadapa penyesuaian diri siswa kelas VII di Madrasah Tsanawiyah Nurul Jadid bengkalis yakni sebesar 0,539 dengan tingkat probabilitas 0,000. Hal ini diperoleh dari r >r pada taraf signifikan 5% dan 1% (0,250 0,325). Besar persentase pengaruh layanan orientasi bidang pengembangan hubungan sosial terhadap penyesuaian diri siswa sebesar 29,0% sedangkan sisanya sebesar 71,0% dipengaruhi oleh variabel lain yang tidak dimasukkan dalam penelitian ini. Guru bimbingan Konseling hendaknya meningkatkan intensitas pelaksanaan layanan orientasi bidang pengembangan hubungan sosial agar penyesuaiain diri siswa dapat berkembang dengan baik dan optimal. Suatu organisasi tidak akan berjalan tanpa adanya komunikasi. Pada dasarnya, komunikasi digunakan untuk menyampaikan kebutuhan organisasi. Manusia melakukan komunikasi sebagai bentuk penyampaian ilmu dan pengalaman. Komunikasi menjadi instrumen yang sangat berguna untuk menyampaikan atau menerima informasi dan pesan dari dan kepada orang lain. Kelalaian dalam melakukan kegiatan komunikasi dapat memberi dampak buruk yang cukup parah. Komunikasi yang buruk merupakan asal perselisihan dan konflik antar individu atau organisasi. Jenis penelitian ini adalah penelitian pustaka (library research). Data yang diperoleh dari hasil referensi-referensi penulis didapatkan dalam berbagai buku-buku dan jurnal-jurnal yang diolah dan dianalisa berdasarkan hasil referensi yang didapatkan, kemudian dikembangkan dengan imajinasi dari penulis agar dapat menghasilkan sebuah tujuan dari penulisan ini. Dengan mempelajari teori-teori komunikasi seperti cultural approach to organization, strukturisasi model weberian, critical theory of communication kiranya organisasi itu akan dapat berjalan dengan lancar dan lebih efektif, dimana komunikasi yang efektif itu dapat memperlancar komunikasi antara komunikator dan komunikan

Pathogenic variants in EP300 and ANKRD11 in patients with phenotypes overlapping Cornelia de Lange syndrome

Cornelia de Lange syndrome (CdLS), Rubinstein–Taybi syndrome (RSTS), and KBG syndrome are three distinct developmental human disorders. Variants in seven genes belonging to the cohesin pathway, NIPBL, SMC1A, SMC3, HDAC8, RAD21, ANKRD11, and BRD4, were identified in about 80% of patients with CdLS, suggesting that additional causative genes remain to be discovered. Two genes, CREBBP and EP300, have been associated with RSTS, whereas KBG results from variants in ANKRD11. By exome sequencing, a genetic cause was elucidated in two patients with clinical diagnosis of CdLS but without variants in known CdLS genes. In particular, genetic variants in EP300 and ANKRD11 were identified in the two patients with CdLS. EP300 and ANKRD11 pathogenic variants caused the reduction of the respective proteins suggesting that their low levels contribute to CdLS-like phenotype. These findings highlight the clinical overlap between CdLS, RSTS, and KBG and support the notion that these rare disorders are linked to abnormal chromatin remodeling, which in turn affects the transcriptional machinery

Characterization and functionality of proliferative human sertoli cells

It has long been thought that mammalian Sertoli cells are terminally differentiated and nondividing postpuberty. For most previous in vitro studies immature rodent testes have been the source of Sertoli cells and these have shown little proliferative ability when cultured. We have isolated and characterized Sertoli cells from human cadaveric testes from seven donors ranging from 12 to 36 years of age. The cells proliferated readily in vitro under the optimized conditions used with a doubling time of approximately 4 days. Nuclear 5-ethynyl-2´-deoxyuridine (EdU) incorporation confirmed that dividing cells represented the majority of the population. Classical Sertoli cell ultrastructural features, lipid droplet accumulation, and immunoexpression of GATA-4, Sox9, and the FSH receptor (FSHr) were observed by electron and fluorescence microscopy, respectively. Flow cytometry revealed the expression of GATA-4 and Sox9 by more than 99% of the cells, and abundant expression of a number of markers indicative of multipotent mesenchymal cells. Low detection of endogenous alkaline phosphatase activity after passaging showed that few peritubular myoid cells were present. GATA-4 and SOX9 expression were confirmed by reverse transcription polymerase chain reaction (RT-PCR), along with expression of stem cell factor (SCF), glial cell line-derived neurotrophic factor (GDNF), and bone morphogenic protein 4 (BMP4). Tight junctions were formed by Sertoli cells plated on transwell inserts coated with fibronectin as revealed by increased transepithelial electrical resistance (TER) and polarized secretion of the immunoregulatory protein, galectin-1. These primary Sertoli cell populations could be expanded dramatically in vitro and could be cryopreserved. The results show that functional human Sertoli cells can be propagated in vitro from testicular cells isolated from adult testis. The proliferative human Sertoli cells should have important applications in studying infertility, reproductive toxicology, testicular cancer, and spermatogenesis, and due to their unique biological properties potentially could be useful in cell therapy.Fil: Chui, Kitty.
