    Get PDF
Bromelain, a cysteine protease with various therapeutic and industrial applications, was expressed in Escherichia coli, BL21-AI clone, under different cultivation conditions (post-induction temperature, L-arabinose concentration and post-induction period). The optimized conditions by response surface methodology using face centered central composite design were 0.2% (w/v) L-arabinose, 8 hr and 25°C. The analysis of variance coupled with larger value of R2 (0.989) showed that the quadratic model used for the prediction was highly significant (p < 0.05). Under the optimized conditions, the model produced bromelain activity of 9.2 U/mg while validation experiments gave bromelain activity of 9.6 ± 0.02 U/mg at 0.15% (w/v) L-arabinose, 8 hr and 27°C. This study had innovatively developed cultivation conditions for better production of recombinant bromelain in shake flask culture

    Get PDF
This study examined effects of adolescent males\u27 perceptions of being bullied because of verbal taunts related to gender nonconformity (i.e., They say I\u27m gay ). Participants included 251 ninth- (n = 77), tenth- (n = 96), and eleventh- (n = 78) grade students in a private, all-male college preparatory school. Participants were divided into two groups based on whether they were bullied by being called gay. Out of the 251 participants, 121 (48%) reported having been bullied and 127 (50%) stated that they had not been bullied during the past year (2% did not report). Of the 121 participants who had been bullied, 32 (26%) reported that they had been bullied because others called them gay (Group 1) and 89 (74%) reported that they had been bullied for other reasons, exclusive of being called gay (Group 2). Consistent with predictions, the boys who were bullied because they were called gay experienced greater psychological distress, greater verbal and physical bullying, and more negative perceptions of their school experiences than boys who were bullied for other reasons. Implications for school-based intervention services for bullying are discussed

Screening of electrocoagulation process parameters for treated palm oil mill effluent using minimum-runs resolution IV design

    Get PDF
The present study aimed at the screening of parameters for electrocoagulation treatment of treated palm oil mill efuent using minimum-runs resolution IV design. The responses examined include: chemical oxygen demand removal (%), total suspended solids removal (%) and turbidity reduction (%), and the varied dependent factors comprise: electrical current density (mA/cm2), time (min), pH, electrolyte concentration (g/L), stirring speed (rpm), electrode spacing (mm) and electrode confguration (monopolar or dipolar). The statistical results revealed that the current density has a signifcant infuence on the treatment performance at two-level interactions with pH, electrode spacing and electrode concentration and at three-level correlations with time and pH. Thus, the most important factors afecting the removal efciency of the organic compounds were found to be pH, time, electrode spacing, electrolyte concentration and electrode confguration at a P value less than 0.05, respectively, in the descending order of signifcance. Therefore, the optimized electrocoagulation process could be reached with current density equal to 5 mA/cm2, electrolysis time of 5 min, electrode spacing of 5 mm and using monopolar electrode confguration. This combination provided the maximum ability of the process for chemical oxygen demand (68.84%), total suspended solids (93.27%) and turbidity reduction (92.88%) predictions, with the corresponding experimental values of 69.27, 97.59 and 96.91%, respectively

Cancer is considered as the most lethal disease for human beings, and up to now many attempts were failed for prevention and treatment of this tremendous health problem. Consequently, this study purpose was to investigate novel therapeutic methods for cancer. The bacterial toxin can result in cell death throughout the induction of apoptosis in cancer cell lines. We evaluated apoptosis and the expression levels of long non-coding RNAs (lncRNAs) in PC3, ACHN and HDF cell lines that were transfected with pCDNA3.1(+)-seh and empty plasmid. pCDNA3.1(+)-seh treatment showed overexpression of GAS5 (p = 0.0033 and p = 0.0033) in PC3 and ACHN cells, down regulation of PCA3 and NEAT1 (p = 0.0092 and p = 0.0097) in the PC3 cells, and down regulation of PVT1 and MALAT1 (p = 0.0239 and p = 0.0133) in the ACHN cells in comparison with the empty plasmid, but there was no significant effect on HDF normal cells. Additionally, this study data demonstrated that the cell adhesion was down regulated. The flow cytometry data showed transfection by pCDNA3.1 (+)-seh could elevate PC3 and ACHN cell apoptosis levels in comparison with empty plasmid. This study findings propose that SEH toxin of S. aureus could be a useful candidate for therapeutic researches in cancer vaccine development

Objective: To evaluate whether evidence-based depression prevention programs can be optimized by matching youths to interventions that address their psychosocial vulnerabilities. Method: This randomized controlled trial included 204 adolescents (mean [SD] age ¼ 14.26 [1.65] years; 56.4% female). Youths were categorized as high or low on cognitive and interpersonal risks for depression and randomly assigned to Coping With Stress (CWS), a cognitive-behavioral program, or Interpersonal Psychotherapy–Adolescent Skills Training (IPT-AST), an interpersonal program. Some participants received a match between risk and prevention (eg, high cognitive–low interpersonal risk teen in CWS, low cognitive–high interpersonal risk teen in IPT-AST), others received a mismatch (eg, low cognitive-high interpersonal risk teen in CWS). Outcomes were depression diagnoses and symptoms through 18 months postintervention (21 months total). Results: Matched adolescents showed significantly greater decreases in depressive symptoms than mismatched adolescents from postintervention through 18-month follow-up and across the entire 21-month study period (effect size [d] ¼ 0.44, 95% CI ¼ 0.02, 0.86). There was no significant difference in rates of depressive disorders among matched adolescents compared with mismatched adolescents (12.0% versus 18.3%, t193 ¼ .78, p ¼ .44). Conclusion: This study illustrates one approach to personalizing depression prevention as a form of precision mental health. Findings suggest that risk-informed personalization may enhance effects beyond a one-size-fits-all approach. Boiler-Feed and Process Water Reclamation from Biotreated Palm Oil Mill Effluent (BPOME): A Developmental Review

    Get PDF
Increased water shortages and new environmental policies and regulations have stimulated significant development in reuse programs in the past 20 years.1 Reclaimed water has been recognized as a vital and drought-proof water source to ensure economic and agricultural activities. According to the conclusions of various water reuse surveys, the best water reuse projects, in terms of economic viability and public acceptance, are those that substitute reclaimed water in lieu of potable water for use in industrial, irrigation, environmental restoration, cleaning and toilet flushing. The main benefits of using reclaimed water in these situations are conservation of water resources and pollution reduction. 1 Low Pressure Membranes (LPMs) such as Microfiltration (MF) and Ultrafiltration (UF) integrated with adsorption system could be a viable option for separation of contaminants in wastewater for reuse as boiler-feed or process water in industry. This article technically discusses the main constituents of concern in boiler-feed and process water requirements and finally suggests potentially efficient and low-energy-low-cost hybridized LPM /adsorption treatment facilities to achieve this paradigm shift towards new water sources
