    Effects of Vasopressin on Sweat Rate and Composition in Patients with Diabetes Insipidus and Normal Controls

    Baseline sweat rate and concentrations of sodium, chloride, and potassium, and the effect of exogenous vasopressin on these parameters were determined in 13 patients with acquired diabetes insipidus (ADI), four patients with nephrogenic diabetes insipidus (NDI), three subjects with cystic fibrosis, and age- and sex-matched controls. The four patients with NDI did not differ from the controls with respect to baseline sweat rate, but baseline sodium, chloride, and potassium concentrations were significantly elevated. In addition, parenteral vasopressin caused a significant decrease in sweat rate (p < .01) while the electrolyte concentrations remained unchanged. This indicates that vasopressin may also have an effect on electrolyte reabsorption in NDI patients. Alternatively, the amount of sweat precursor fluid may have been reduced. The patients with ADI did not differ from the controls with respect to baseline data, and parenteral vasopressin had no effect on their sweat rate and composition. Likewise, vasopressin had no effect in controls or patients with cystic fibrosis. We conclude that, except in patients with NDI, vasopressin does not play a significant role in the regulation of human eccrine sweating. Sweat gland physiology appears to be different in patients with NDI and in them vasopressin may have a significant effect on sweat

    ANTIGEN-SPECIFIC THYMUS CELL FACTORS IN THE GENETIC CONTROL OF THE IMMUNE RESPONSE TO POLY-(TYROSYL, GLUTAMYL)-POLY-D, L-ALANYL--POLY-LYSYL

    The genetic control of the antibody response to a synthetic polypeptide antigen designated poly-L(Tyr, Glu)-poly-D,L-Ala--poly-L-Lys [(T, G)-A--L] has been studied in congenic high responder C3H.SW (H-2b) and low responder C3H/HeJ (H-2k) strains of mice. This response is controlled by the Ir-1 gene and is H-2 linked. The method employed was to study the ability of specifically primed or "educated" T cells of each strain to produce cooperative factors for (T, G)-A--L in vitro. Such factors have been shown to be capable of replacing the requirement for T cells in the thymus-dependent antibody response to (T, G)-A--L in vivo. The T-cell factors produced were tested for their ability to cooperate with B cells of either high or low responder origin by transfer together with bone marrow cells and (T, G)-A--L into heavily irradiated, syngeneic (for bone marrow donor) recipients. Direct anti-(T, G)-A--L plaque-forming cells were measured later in the spleens of the recipients. The results showed that (a) educated T cells of both high and low responder origin produced active cooperative factors to (T, G)-A--L, and no differences between the strains in respect to production of T-cell factors could be demonstrated; and (b) such factors, whether of high or low responder origin, cooperated efficiently with B cells of high responder origin only, and hardly at all with B cells of low responder origin. The conclusion was drawn that the cellular difference between the two strains lies in the responsiveness of their B cells to specific signals or stimuli received from T cells. As far as could be discerned by the methods used, no T-cell defect existed in low responder mice and the expression of the controlling Ir-1 gene was solely at the level of the B cells in this case

    Overview study of Space Power Technologies for the advanced energetics program

    Space power technologies are reviewed to determine the state-of-the-art and to identify advanced or novel concepts which promise large increases in performance. The potential for incresed performance is judged relative to benchmarks based on technologies which have been flight tested. Space power technology concepts selected for their potentially high performance are prioritized in a list of R & D topical recommendations for the NASA program on Advanced Energetics. The technology categories studied are solar collection, nuclear power sources, energy conversion, energy storage, power transmission, and power processing. The emphasis is on electric power generation in space for satellite on board electric power, for electric propulsion, or for beamed power to spacecraft. Generic mission categories such as low Earth orbit missions and geosynchronous orbit missions are used to distinguish general requirements placed on the performance of power conversion technology. Each space power technology is judged on its own merits without reference to specific missions or power systems. Recommendations include 31 space power concepts which span the entire collection of technology categories studied and represent the critical technologies needed for higher power, lighter weight, more efficient power conversion in space

    ELABORATING INNOVATIVE SOLUTIONS WITH EXPERTS USING A MULTICRITERIA EVALUATION TOOL. THE CASE OF SOIL BORNE DISEASE CONTROL IN MARKETGARDENING CROPPING SYSTEMS

    International audienceMarket-gardening cropping systems in protected cultivation are very sensitive to soil-borne pests and diseases. Their productivity used to rely on pesticides, but alternative systems have now to be found for environmental, societal and health reasons. Many cultural techniques are known to provide some control of soilborne diseases, but are only partially efficient. The aim of the project is to design alternative systems with professionals, by improving the efficiency of the present techniques and/or imagining more innovative systems. The research project takes place in two steps. The first one consists in building a tool to assess the resistance or resilience of a given cropping system to soil-borne pests; the second one consists in using the tool with professionals in order to build alternative cropping and farming systems in cooperation. The model built for evaluation is a qualitative multicriteria tool. As scientific knowledge is not available for each technique or combination of techniques, empirical knowledge collected from growers and technical advisers is used to fill the gaps. The model is already built for root-knot nematodes and under construction for the other fungi. The second step will consist in using the tool with technical advisers and growers to redesign cropping systems and select the promising ones that should be put into trial in R&D stations. Co-building farming systems with stakeholders appears as an absolute necessity, to imagine solutions both efficient and acceptable for growers. The presentation will enable to discuss how combining expert and scientific knowledge may not only fill the knowledge gap, but also enable to build innovative solutions thanks to the diversity of experts' standpoints

    ANTIGEN-SPECIFIC T-CELL FACTOR IN CELL COOPERATION: PHYSICAL PROPERTIES AND MAPPING IN THE LEFT-HAND (K) HALF OF H-2

    Mouse thymus cells, educated to poly(tyrosyl,glutamyl)-polyDLalanyl--polylysyl [(T,G)-A--L], release an antigen-specific factor on brief culture in vitro. The factor cooperates with bone marrow cells in the antibody response to (T,G)-A--L in irradiated recipients. Its mol wt determined from Sephadex G100 chromatography is in the region of 50,000. The factor is removed by specific antigen-coated columns, but not by anti-immunoglobulin (anti-Fab, anti-µ, anti-Fv) adsorbents. The factor is removed by alloantisera directed against the H-2 haplotype of the strain in which it is produced. Moreover, only antisera with specificity for the K side of H-2 were successful in removing the factor activity

    Use of a cAMP BRET Sensor to Characterize a Novel Regulation of cAMP by the Sphingosine 1-Phosphate/G13 Pathway

    Regulation of intracellular cyclic adenosine 3',5'-monophosphate (cAMP) is integral in mediating cell growth, cell differentiation, and immune responses in hematopoietic cells. To facilitate studies of cAMP regulation we developed a BRET (bioluminescence resonance energy transfer) sensor for cAMP, CAMYEL (cAMP sensor using YFP-Epac-RLuc), which can quantitatively and rapidly monitor intracellular concentrations of cAMP in vivo. This sensor was used to characterize three distinct pathways for modulation of cAMP synthesis stimulated by presumed Gs-dependent receptors for isoproterenol and prostaglandin E2. Whereas two ligands, uridine 5'-diphosphate and complement C5a, appear to use known mechanisms for augmentation of cAMP via Gq/calcium and Gi, the action of sphingosine 1-phosphate (S1P) is novel. In these cells, S1P, a biologically active lysophospholipid, greatly enhances increases in intracellular cAMP triggered by the ligands for Gs-coupled receptors while having only a minimal effect by itself. The enhancement of cAMP by S1P is resistant to pertussis toxin and independent of intracellular calcium. Studies with RNAi and chemical perturbations demonstrate that the effect of S1P is mediated by the S1P2 receptor and the heterotrimeric G13 protein. Thus in these macrophage cells, all four major classes of G proteins can regulate intracellular cAMP

    'An Apotheosis of Well-Being': Durkheim on austerity and double-dip recessions

    This article is an attempt to contribute a view on the economic crisis from classical sociology, a voice often missing from the sociological response to the crisis. The work of Émile Durkheim provides a unique perspective here centred on morality and inequality produced in a historical context akin to our neoliberal times. It is argued there are four key points to take from Durkheim’s work. Firstly, that the initial credit crunch can be more fully understood with reference to the economic anomie which Durkheim sees as ‘chronic’ in a time of marketization. Secondly, that this creates an antagonistic relationship between a supposedly self-dependent rich and lazy poor. Thirdly, this conception of self-dependency and individual initiative makes any attempt to regulate the economy akin to sacrilege. Finally, the state is unwilling to intervene due to the emergence of ‘pseudo-democracies’. Therefore, Durkheim’s theory accounts for the initial crisis, austerity and double-dip recessions in a sociological framework. The article concludes by returning to the centrality of morality to the crisis for Durkheim and highlighting the omission of this in contemporary debates
