    A TaqMan qPCR method for detecting kdr resistance in Aphis gossypii demonstrates improved sensitivity compared to conventional PCR–RFLP

Cotton aphid, Aphis gossypii Glover, has emerged as a prominent pest in Australian cotton production, and monitoring pesticide resistance including pyrethroids in field populations is crucial for its sustainable management. We examined the distribution of kdr resistance in 35 field-collected A. gossypii populations and used TaqMan qPCR assays with pooled samples. The study demonstrated proof of concept that pooled insect qPCR methodology provided effective detection with better sensitivity than individual PCR–RFLP genotyping techniques for the kdr resistance allele. The practical outcome is that routine resistance monitoring can examine more sites while increasing the likelihood of detecting incipient resistance at those sites. More importantly, the method is adaptable to any genetically caused resistance and so not limited to A. gossypii or even insect control. It cannot be overstressed that the ability to detected resistance at very low frequencies is critical to all sustainable resistance management. Early detection of resistance provides critical time for the modification of chemical use prior to potential insecticide control failure

    The Effect of Consumers and Mutualists of Vaccinium membranaceum at Mount St. Helens: Dependence on Successional Context

    In contrast to secondary succession, studies of terrestrial primary succession largely ignore the role of biotic interactions, other than plant facilitation and competition, despite the expectation that simplified interaction webs and propagule-dependent demographics may amplify the effects of consumers and mutualists. We investigated whether successional context determined the impact of consumers and mutualists by quantifying their effects on reproduction by the shrub Vaccinium membranaceum in primary and secondary successional sites at Mount St. Helens (Washington, USA), and used simulations to explore the effects of these interactions on colonization. Species interactions differed substantially between sites, and the combined effect of consumers and mutualists was much more strongly negative for primary successional plants. Because greater local control of propagule pressure is expected to increase successional rates, we evaluated the role of dispersal in the context of these interactions. Our simulations showed that even a small local seed source greatly increases population growth rates, thereby balancing strong consumer pressure. The prevalence of strong negative interactions in the primary successional site is a reminder that successional communities will not exhibit the distribution of interaction strengths characteristic of stable communities, and suggests the potential utility of modeling succession as the consequence of interaction strengths

    At the 15th ICRAV in Dubai, evidence was presented that certain 19-norsteroids typically observed in acid solvolysed stallion urine extracts may actually be chemical artefacts formed through the decarboxylation of endogenous steroid-19-oic acids. In the case of the nandroloone frequently detected in such samples, this explanation provides an alternative to the traditional assumption that nandrolone sulphate and/or nandrolone sulphate and /or nandrolone glucurornide are normal components of stallion urine. This, in turn, raises the possibility of using intact phase II conjugates of nandrolone as markers for exogenous nandrolone administration

    Direct detection of boldenone sulfate and glucuronide conjugates in horse urine by ion trap liquid chromatography-mass spectrometry

    A study of the equine phase II metabolism of the anabolic agent boldenone is reported. Boldenone sulfate, boldenone glucuronide and their C17-epimers were synthesised as reference standards in our lab and a method was developed for their detection in a horse urine matrix. Solid phase extraction was used to purify the analytes, which were then detected by ion trap LC/MS. Negative and positive ionisation mode MS2 were used for the detection of sulfate and glucuronide conjugates, respectively. Boldenone sulfate and 17-epiboldenone glucuronide were detected as the major and minor phase II metabolites, respectively, in horse urine samples collected following the administration of boldenone undecylenate by intramuscular injection

    Knowing is half the battle: Assessments of both student perception and performance are necessary to successfully evaluate curricular transformation.

    Student-centered pedagogies increase learning and retention. Quantifying change in both student learning gains and student perception of their experience allows faculty to evaluate curricular transformation more fully. Student buy-in, particularly how much students value and enjoy the active learning process, has been positively associated with engagement in active learning and increased learning gains. We hypothesize that as the frequency of students who have successfully completed the course increases in the student population, current students may be more likely to buy-in to the curriculum because this common experience could create a sense of community. We measured learning gains and attitudes during the transformation of an introductory biology course at a small, liberal arts college using our novel curriculum, Integrating Biology and Inquiry Skills (IBIS). Students perceived substantial learning gains in response to this curriculum, and concept assessments confirmed these gains. Over time, buy-in increased with each successive cohort, as demonstrated by the results of multiple assessment instruments, and students increasingly attributed specific components of the curriculum to their learning. These findings support our hypothesis and should encourage the adoption of curricular transformation using IBIS or other student-centered approaches

    Dissecting the contributions of dispersal and host properties to the local abundance of a tropical mistletoe

    The interplay between dispersal and adaptation to local environments ultimately determines the distribution of plant species, but their relative contribution remains little understood. Tropical mistletoes provide the opportunity to dissect these contributions of dispersal and adaptation, because as hemiparasitic plants, they are typically adapted to grow on a handful of species within diverse tropical communities and are non-randomly dispersed by mutualistic frugivorous birds. Here we hypothesized that the primary determinant of the abundance of a tropical mistletoe (Dendropemon caribaeus, Loranthaceae) in Puerto Rico will be the compatibility between the mistletoe and plant species in a community. Alternatively, the mistletoe's abundance could be primarily shaped by other factors such as the availability of potential host plants, or factors that determine how mistletoe seeds are dispersed by avian frugivores. We conducted surveys and experiments to assess the capacity of this mistletoe to grow on trees available in the local community, and measured the monthly phenology and seed dispersal patterns of the mistletoe and other bird-dispersed plants in the community over a period of 4 years. A path model was used to evaluate how the abundance of the mistletoe was shaped by host abundance, fruiting phenology, bird dispersal and compatibilities with host plants. Our analyses show that the compatibility between mistletoe and host tree species, measured by mistletoe survival and growth rate, was the most important factor for mistletoe abundance. The next most important factor was the phenological characteristics of the hosts; this outcome likely arose because frugivory and seed dispersal services for mistletoes and hosts are performed by the same birds. Synthesis. Mistletoes often parasitize only a subset of the suitable plant species that are available in a given community. Our results indicate that such patterns are not only determined by host quality and abundance but also by the phenological patterns of trees that influence the probabilities of mistletoe seeds being deposited on them by shared avian seed dispersers.This research was supported by NSF grants DEB‐1028174 and DEB‐1145994 to T.A.C., the Alfred P.     Evaluation of three serological tests for the diagnosis of Brucella suis in dogs using Bayesian latent class analysis

    Brucella suis infection of dogs is an emerging issue worldwide requiring specific management to address zoonotic risks and animal welfare concerns. Diagnosis in dogs is routinely based on serological testing, but to date these tests have only been validated for use in production animal species and humans. This study aimed to assess the diagnostic performance of three commonly used serological tests in dogs. Canine sera (n = 989) were tested with the Rose Bengal rapid plate agglutination test (RBRPT), the complement fixation test (CFT) and a competitive ELISA (C-ELISA). Diagnostic test performance was evaluated using a three test, two population Bayesian latent class analysis accounting for conditional dependence between the three tests. Positive and negative predictive values (PPV, NPV) were calculated for a range of expected prevalence estimates for the individual tests and test combinations interpreted in series and parallel. The RBRPT showed the highest individual Se of 0.902 (95 % posterior credible interval [PCI] 0.759–0.978) and the CFT the highest individual diagnostic specificity (Sp) of 0.914 (95 % PCI 0.886–0.946). The C-ELISA had marginally the best overall diagnostic performance (Youden's index = 0.807). The CFT and the C-ELISA interpreted in parallel returned the highest combined Se and Sp (0.988 and 0.885, respectively). All tests returned NPVs of > 0.982 in 2–8 % prevalence settings. Series interpretation of the three-test combination as well as the two-test combinations of the RBRPT and the C-ELISA and the CFT and the C-ELISA produced a PPV of 0.502 when the estimated prevalence was 8 %. While all tests are suitable for the detection of B. suis antibodies in dogs, they should not be interpreted in isolation as their diagnostic value is dependent on the pre-test probability of the disease. As such they are useful tools for the diagnosis of B. suis in dogs when exposure, history and clinical presentation indicate a risk of brucellosis
