    SARS-CoV-2 and the risk of Parkinson's disease: facts and fantasy.

    During the pandemic it has become clear that severe acure respiratory syndrome coronavirus (SARS-CoV-2) causes not just respiratory disease, but can affect multiple organs and tissues. Of note is the involvement of the CNS and PNS, and the fact that this involvement is independent from the severity of the respiratory disease. Acute and subacute neurological complications of SARS-CoV-2 infections are reported in up to 85% of patients, including those with severe COVID-19, but also in otherwise minimally symptomatic or asymptomatic people. As many as 65% of people with COVID-19 present with hyposmia, which is also a common premotor symptom in Parkinson’s disease. This symptom, added to the fact that parkinsonism has been reported following COVID-19, has drawn the attention of the medical community to the hypothetical link between SARS-CoV-2 infection and Parkinson’s disease.Fil: Merello, Marcelo Jorge.     Unilateral subthalamotomy in Parkinson's disease: Cognitive, psychiatric and, neuroimaging changes

    Unilateral subthalamotomy is an effective treatment for the cardinal motor features of Parkinson's disease (PD). However, non-motor changes possibly associated with right or left subthalamotomy remain unknown. Our aim was to assess cognitive, psychiatric and neuroimaging changes after treatment with unilateral subthalamotomy. Fourteen medicated patients with PD were evaluated before and after (mean 6 months after operation) unilateral subthalamotomy (5 right, 9 left). In addition to motor assessments, cognitive (global cognition and executive functions), psychiatric (apathy, depression, anxiety, mania, hypo- and hyperdopaminergic behaviours, impulsivity), quality of life evaluations and volume of lesions were obtained. After surgery, significant improvement of motor signs was observed. Unilateral subthalamotomy improved general cognitive status, but left subthalamotomy reduced semantic verbal fluency compared to the pre-operative state. Depression and quality of life were improved with both right and left subthalamotomy. However, hyper-emotionality was present after surgery and right subthalamotomy increased impulsivity and disinhibition (on NeuroPsychiatric Inventory and Ardouin Scale for Behaviour in PD), a result linked to larger lesion volumes. We conclude that unilateral subthalamotomy is effective for treating the cardinal motor features of PD and improves mood. Right subthalamotomy is associated with greater risk of impulsivity and disinhibition, while left subthalamotomy induces further impairment of semantic verbal fluency

    Se estudió el crecimiento anual y la distribución de recursos de clones control y de rebrotes en áreas incendiadas y en zonas control en Erica vagans y E. cinerea, encontrándose que no existen diferencias significativas entre ambas especies. Sin embargo, el crecimiento de los rebrotes tras el incendio fue superior al estimado en los controles. La asignación reproductiva en el rebrote después de incendios disminuyó en ambas especies, sobre todo en E. cinerea. Los patrones de asignación reproductiva dependientes del tamaño que aparecen en los controles no se encuentran en el rebrote, lo cual sugiere una asignación reproductiva plástica.Post-fire growth and annual growth of control branches were studied in Erica vagans and E. cinerea in two northem Spain localities. The dry weight of the resprouts was greater than that of the control branches. However, neither growth of the control branches nor resprouts differed among species. Reproductive allocation did not show differences among species, but significantly decreased in burned areas, specially in E. cinerea. Reproductive allocation was size-dependent in control branches, but this pattem did not appear in the resprouts. This variations suggest a flexible reproductive strategy

    Effects of Gestational Intermittent Hypoxia on Placental Morphology and Fetal Development in a Murine Model of Sleep Apnea

    Obstructive sleep apnea (OSA) during pregnancy is characterized by episodes of intermittent hypoxia (IH) during sleep, resulting in adverse health outcomes for mother and offspring. Despite a prevalence of 8-20% in pregnant women, this disorder is often underdiagnosed.We have developed a murine model of gestational OSA to study IH effects on pregnant mothers, placentas, fetuses, and offspring. One group of pregnant rats was exposed to IH during the last 2 weeks of gestation (GIH). One day before the delivery date, a cesarean section was performed. Other group of pregnant rats was allowed to give birth at term to study offspring's evolution.Preliminary results showed no significant weight differences in mothers and fetuses. However, the weight of GIH male offspring was significantly lower than the controls at 14 days (p < 0.01). The morphological study of the placentas showed an increase in fetal capillary branching, expansion of maternal blood spaces, and number of cells of the external trophectoderm in the tissues from GIH-exposed mothers. Additionally, the placentas from the experimental males were enlarged (p < 0.05). Further studies are needed to follow the long-term evolution of these changes to relate the histological findings of the placentas with functional development of the offspring in adulthood.Ayudas para la realización de proyectos de investigación UVa 2021 (PROYEMER 57-E.O.

    Entacapone potentiates the long-duration response but does not normalize levodopa-induced molecular changes

The mechanisms underlying entacapone effects are unknown. We investigated the effect of entacapone, on: long-duration response (LDR) to levodopa, levodopa-induced postsynaptic pharmacodynamic mechanisms and molecular changes in hemiparkinsonian rats. 6-Hydroxydopamine-unilaterally lesioned rats were treated with levodopa (25 mg/kg) + vehicle; levodopa + entacapone (30 mg/kg) or saline, twice daily for 22 days. The LDR and the apomorphine-induced rotations were measured. In situ hybridization was performed measuring the expression of striatal preproenkephalin, preprodynorphin and dopamine D-3 receptor mRNAs, subthalamic cytochrome oxidase mRNA and nigral glutamic acid decarboxylase mRNA. Entacapone potentiated the LDR but did not modify either the apomorphine-induced rotational behavior or the molecular changes. Our results suggest that the effects of entacapone on levodopa-induced motor response are not mediated by postsynaptic mechanisms and that administration of entacapone is not able to normalize the molecular alterations induced by levodopa in the basal ganglia.This work was supported by an unrestricted grant from Novartis-Orion Pharma (Barcelona, Spain).     A strategy for implementing non-perturbative renormalisation of heavy-light four-quark operators in the static approximation

    We discuss the renormalisation properties of the complete set of ΔB=2\Delta B = 2 four-quark operators with the heavy quark treated in the static approximation. We elucidate the role of heavy quark symmetry and other symmetry transformations in constraining their mixing under renormalisation. By employing the Schroedinger functional, a set of non-perturbative renormalisation conditions can be defined in terms of suitable correlation functions. As a first step in a fully non-perturbative determination of the scale-dependent renormalisation factors, we evaluate these conditions in lattice perturbation theory at one loop. Thereby we verify the expected mixing patterns and determine the anomalous dimensions of the operators at NLO in the Schroedinger functional scheme. Finally, by employing twisted-mass QCD it is shown how finite subtractions arising from explicit chiral symmetry breaking can be avoided completely.Comment: 41 pages, 6 figure

    Thalamic innervation of the direct and indirect basal ganglia pathways in the rat: Ipsi- and contralateral projections

    The present study describes the thalamic innervation coming from the rat parafascicular nucleus (PF) onto striatal and subthalamic efferent neurons projecting either to the globus pallidus (GP) or to the substantia nigra pars reticulata (SNr) by using a protocol for multiple neuroanatomical tracing. Both striatofugal neurons targeting the ipsilateral SNr (direct pathway) as well as striatal efferent neurons projecting to the ipsilateral GP (indirect pathway) were located within the terminal fields of the thalamostriatal afferents. In the subthalamic nucleus (STN), both neurons projecting to ipsilateral GP as well as neurons projecting to ipsilateral SNr also appear to receive thalamic afferents. Although the projections linking the caudal intralaminar nuclei with the ipsilateral striatum and STN are far more prominent, we also noticed that thalamic axons could gain access to the contralateral STN. Furthermore, a small number of STN neurons were seen to project to both the contralateral GP and PF nuclei. These ipsi- and contralateral projections enable the caudal intralaminar nuclei to modulate the activity of both the direct and the indirect pathway
