    Smoking-induced long-lasting modifications of human platelet serotonin catabolism through a MAO epigenetic regulation

    Postulating that serotonin, secreted from smoking-activated platelets, could be involved in smoking-induced vascular modifications, we studied 115 men distributed in smokers (S), former smokers (FS) and never smokers (NS). The platelet serotonin content was similar in S and NS but lower in FS. This was unexpected because the monoamine oxidase (MAO) activity, which catabolizes serotonin, was inhibited during smoking. However, the amount of platelet MAO was higher in S and FS than in NS. The persistent elevated MAO amount in FS prompted us to study the methylation of its gene promoter in an additional series of patients: it was markedly lower for S and FS vs. NS due to cigarette smoke-induced increase of nucleic acid demethylase activity. This smoking-induced demethylation of the MAO gene promoter, resulting in high MAO amount persisting long after quitting smoking, has cardiovascular consequences and could impact fields such as behavior, mental health, and cancer

    Polynucleotide phosphorylase has an impact on cell biology of Campylobacter jejuni

    Polynucleotide phosphorylase (PNPase), encoded by the pnp gene, is known to degrade mRNA, mediating post-transcriptional regulation and may affect cellular functions. The role of PNPase is pleiotropic. As orthologs of the two major ribonucleases (RNase E and RNase II) of Escherichia coli are missing in the Campylobacter jejuni genome, in the current study the focus has been on the C. jejuni ortholog of PNPase. The effect of PNPase mutation on C. jejuni phenotypes and proteome was investigated. The inactivation of the pnp gene reduced significantly the ability of C. jejuni to adhere and to invade Ht-29 cells. Moreover, the pnp mutant strain exhibited a decrease in C. jejuni swimming ability and chick colonization. To explain effects of PNPase on C. jejuni 81-176 phenotype, the proteome of the pnp mutant and parental strains were compared. Overall, little variation in protein production was observed. Despite the predicted role of PNPase in mRNA regulation, the pnp mutation did not induce profound proteomic changes suggesting that other ribonucleases in C. jejuni might ensure this biological function in the absence of PNPase. Nevertheless, synthesis of proteins which are involved in virulence (LuxS, PEB3), motility (N-acetylneuraminic acid synthetase), stress-response (KatA, DnaK, Hsp90), and translation system (EF-Tu, EF-G) were modified in the pnp mutant strain suggesting a more specific role of PNPase in C. jejuni. In conclusion, PNPase deficiency induces limited but important consequences on C. jejuni biology that could explain swimming limitation, chick colonization delay, and the decrease of cell adhesion/invasion ability

    Future socio-political scenarios for aquatic resources in Europe: An operationalized framework for aquaculture projections

    Climate-driven changes in aquatic environments have already started to affect the European aquaculture sector’s most commercially important finfish and shellfish species. In addition to changes in water quality and temperature that can directly influence fish production by altering health status, growth performance and/or feed conversion, the aquaculture sector also faces an uncertain future in terms of production costs and returns. For example, the availability of key ingredients for fish feeds (proteins, omega-3 fatty acids) will not only depend on future changes in climate, but also on social and political factors, thereby influencing feed costs. The future cost of energy, another main expenditure for fish farms, will also depend on various factors. Finally, marketing options and subsidies will have major impacts on future aquaculture profitability. Based on the framework of four socio-political scenarios developed in the EU H2020 project climate change and European aquatic resources (CERES), we defined how these key factors for the aquaculture sector could change in the future. We then apply these scenarios to make projections of how climate change and societal and economic trends influence the mid-century (2050) profitability of European aquaculture. We used an established benchmarking approach to contrast present-day and future economic performance of “typical farms” in selected European production regions under each of the scenarios termed “World Markets,” “National Enterprise,” “Global Sustainability” and “Local Stewardship.” These scenarios were based partly on the IPCC Special Report on Emissions Scenarios framework and their representative concentration pathways (RCPs) and the widely used shared socio-economic pathways (SSPs). Together, these scenarios contrast local versus international emphasis on decision making, more versus less severe environmental change, and different consequences for producers due to future commodity prices, cash returns, and costs. The mid-century profitability of the typical farms was most sensitive to the future development of feed costs, price trends of returns, and marketing options as opposed to the direct effect of climate-driven changes in the environment. These results can inform adaptation planning by the European aquaculture sector. Moreover, applying consistent scenarios including societal and economic dimensions, facilitates regional to global comparisons of adaptation advice both within and across Blue Growth sectors

    Face Mis-ID: An Interactive Pedagogical Tool Demonstrating Disparate Accuracy Rates in Facial Recognition

    This paper reports on the making of an interactive demo to illustrate algorithmic bias in facial recognition. Facial recognition technology has been demonstrated to be more likely to misidentify women and minoritized people. This risk, among others, has elevated facial recognition into policy discussions across the country, where many jurisdictions have already passed bans on its use. Whereas scholarship on the disparate impacts of algorithmic systems is growing, general public awareness of this set of problems is limited in part by the illegibility of machine learning systems to non-specialists. Inspired by discussions with community organizers advocating for tech fairness issues, we created the Face Mis-ID Demo to reveal the algorithmic functions behind facial recognition technology and to demonstrate its risks to policymakers and members of the community. In this paper, we share the design process behind this interactive demo, its form and function, and the design decisions that honed its accessibility, toward its use for improving legibility of algorithmic systems and awareness of the sources of their disparate impacts

    Cerebrospinal fluid biomarker supported diagnosis of Creutzfeldt-Jakob disease and rapid dementias: a longitudinal multicentre study over 10 years

    To date, cerebrospinal fluid analysis, particularly protein 14-3-3 testing, presents an important approach in the identification of Creutzfeldt-Jakob disease cases. However, one special point of criticism of 14-3-3 testing is the specificity in the differential diagnosis of rapid dementia. The constant observation of increased cerebrospinal fluid referrals in the national surveillance centres over the last years raises the concern of declining specificity due to higher number of cerebrospinal fluid tests performed in various neurological conditions. Within the framework of a European Community supported longitudinal multicentre study (‘cerebrospinal fluid markers') we analysed the spectrum of rapid progressive dementia diagnoses, their potential influence on 14-3-3 specificity as well as results of other dementia markers (tau, phosphorylated tau and amyloid-β1-42) and evaluated the specificity of 14-3-3 in Creutzfeldt-Jakob disease diagnosis for the years 1998-2008. A total of 29 022 cerebrospinal fluid samples were analysed for 14-3-3 protein and other cerebrospinal fluid dementia markers in patients with rapid dementia and suspected Creutzfeldt-Jakob disease in the participating centres. In 10 731 patients a definite diagnosis could be obtained. Protein 14-3-3 specificity was analysed for Creutzfeldt-Jakob disease with respect to increasing cerebrospinal fluid tests per year and spectrum of differential diagnosis. Ring trials were performed to ensure the comparability between centres during the reported time period. Protein 14-3-3 test specificity remained high and stable in the diagnosis of Creutzfeldt-Jakob disease during the observed time period across centres (total specificity 92%; when compared with patients with definite diagnoses only: specificity 90%). However, test specificity varied with respect to differential diagnosis. A high 14-3-3 specificity was obtained in differentiation to other neurodegenerative diseases (95-97%) and non-neurological conditions (91-97%). We observed lower specificity in the differential diagnoses of acute neurological diseases (82-87%). A marked and constant increase in cerebrospinal fluid test referrals per year in all centres did not influence 14-3-3 test specificity and no change in spectrum of differential diagnosis was observed. Cerebrospinal fluid protein 14-3-3 detection remains an important test in the diagnosis of Creutzfeldt-Jakob disease. Due to a loss in specificity in acute neurological events, the interpretation of positive 14-3-3 results needs to be performed in the clinical context.
