    Connexin43 regulates joint location in zebrafish fins

    AbstractJoints are essential for skeletal form and function, yet their development remains poorly understood. In zebrafish fins, joints form between the bony fin ray segments providing essentially unlimited opportunities to evaluate joint morphogenesis. Mutations in cx43 cause the short segment phenotype of short fin (sofb123) mutants, suggesting that direct cell–cell communication may regulate joint location. Interestingly, increased cx43 expression in the another long fin (alfdty86) mutant appears to cause joint failure typical of that mutant. Indeed, knockdown of cx43 in alfdty86 mutant fins rescues joint formation. Together, these data reveal a correlation between the level of Cx43 expression in the fin ray mesenchyme and the location of joints. Cx43 was also observed laterally in cells associated with developing joints. Confocal microscopy revealed that the Cx43 protein initially surrounds the membranes of ZNS5-positive joint cells, but at later stages becomes polarized toward the underlying Cx43-positive mesenchymal cells. One possibility is that communication between the Cx43-positive mesenchyme and the overlying ZNS5-positive cells regulates joint location, and upregulation of Cx43 in joint-forming cells contributes to joint morphogenesis

    An investigation on the impact of natural language on conversational recommendations

    In this paper, we investigate the combination of Virtual Assistants and Conversational Recommender Systems (CoRSs) by designing and implementing a framework named ConveRSE, for building chatbots that can recommend items from different domains and interact with the user through natural language. An user experiment was carried out to understand how natural language influences both the cost of interaction and recommendation accuracy of a CoRS. Experimental results show that natural language can indeed improve user experience, but some critical aspects of the interaction should be mitigated appropriately

    A comparison of services for intent and entity recognition for conversational recommender systems

    Conversational Recommender Systems (CoRSs) are becoming increasingly popular. However, designing and developing a CoRS is a challenging task since it requires multi-disciplinary skills. Even though several third-party services are available for supporting the creation of a CoRS, a comparative study of these platforms for the specific recommendation task is not available yet. In this work, we focus our attention on two crucial steps of the Conversational Recommendation (CoR) process, namely Intent and Entity Recognition. We compared four of the most popular services, both commercial and open source. Furthermore, we proposed two custom-made solutions for Entity Recognition, whose aim is to overcome the limitations of the other services. Results are very interesting and give a clear picture of the strengths and weaknesses of each solution

    A domain-independent framework for building conversational recommender systems

    Conversational Recommender Systems (CoRSs) implement a paradigm where users can interact with the system for defining their preferences and discovering items that best fit their needs. A CoRS can be straightforwardly implemented as a chatbot. Chatbots are becoming more and more popular for several applications like customer care, health care, medical diagnoses. In the most complex form, the implementation of a chatbot is a challenging task since it requires knowledge about natural language processing, human-computer interaction, and so on. In this paper, we propose a general framework for making easy the generation of conversational recommender systems. The framework, based on a content-based recommendation algorithm, is independent from the domain. Indeed, it allows to build a conversational recommender system with different interaction modes (natural language, buttons, hybrid) for any domain. The framework has been evaluated on two state-of-the-art datasets with the aim of identifying the components that mainly influence the final recommendation accuracy

    A Comparative Analysis of the Photo-Protective Effects of Soy Isoflavones in Their Aglycone and Glucoside Forms

    Isoflavones exist in nature predominantly as glucosides such as daidzin or genistin and are rarely found in their corresponding aglycone forms daidzein and genistein. The metabolism and absorption of isoflavones ingested with food is well documented, but little is known about their use as topical photo-protective agents. The aim of this study was to investigate in a comparative analysis the photo-protective effects of isoflavones in both their aglycone and glucoside forms. In human skin fibroblasts irradiated with 60 mJ/cm(2) ultraviolet B (UVB), we measured the expression levels of COX-2 and Gadd45, which are involved in inflammation and DNA repair, respectively. We also determined the cellular response to UVB-induced DNA damage using the comet assay. Our findings suggest that both the isoflavone glucosides at a specific concentration and combination with an aglycone mixture exerted an anti-inflammatory and photo-protective effect that prevented 41% and 71% of UVB-induced DNA damage, respectively. The advantages of using either isoflavone glucosides or an aglycone mixture in applications in the field of dermatology will depend on their properties and their different potential uses

    The isoflavones daidzein and genistein are natural compounds which have anti-inflammatory and photoprotective activities, and may be effective in the repair of ultraviolet (UV)-induced photodamage. In this study, an alcoholic solution of aglycone isoflavones with a genistein:daidzein ratio of 1:4 [Rottapharm (RPH)-aglycone] was examined for its effects on the repair of DNA damage induced by a single dose of UVB irradiation (20 mJ/cm2). For this purpose, human skin cells were first UVB-irradiated and then treated with RPH-aglycone. Comet assay analysis was used to estimate the UVB-induced DNA damage at different time points after treatment by measuring the tail moment parameter. We found that treatment with 10 μmol/L RPH-aglycone solution resulted in a significantly reduced tail moment at 1 h after treatment, and 34-35% enhancement of damage repair at 4 h after treatment. These results suggest that isoflavone aglycones are protective against UVB-induced DNA damage. © 2014 British Association of Dermatologists

    A virtual customer assistant for the wealth management domain in the UWMP project

    The Universal Wealth Management Platform (UWMP) project has the objective of creating a new service model in the financial domain. An integral part of this service model is the creation of a new Virtual Customer Assistant, that is able to assist customers via natural language dialogues. This paper is a report of the activities performed to develop this assistant. It illustrates a general architecture of the system, and describes the most important decisions made for its implementation. It also describes the main financial operations that it is able to assist customers with. Finally, it delineates some avenues for future work

    New constraints on the origin of the ophiolitic rocks within sinorogenic turbiditic sequences at Cilento region (southern Italy)

    Mafic igneous rocks (pillow lavas and gabbros) embedded as olistoliths within Miocene turbiditic sequences crop out in the Cilento area at the Mount Centaurino (Campania region, Southern Italy). The concentration of major oxides, as well as trace element ratios (Nb/Yb, Nb/Ta, Th/Nb) and the chondrite-normalized Rare Earth Elements (REE) patterns suggest a tholeiitic character with Mid Oceanic Ridge Basalts (MORB) affinity. The chemical composition of pillow lavas is consistent with magmas generated by 10% degrees of non-modal fractional partial melting, of a spinel-bearing MORB-type asthenospheric mantle. Regarding gabbros, the calculated composition of parental melts in equilibrium with the clinopyroxenes show a wide compositional range, and there are very different from the pillow basalts of the Mount Centaurino, suggesting that the clinopyroxenes might have derived from more evolved melts compared to those that produced the basalts. The origin of these olistoliths is not yet understood. Here we suggest that these rocks represent fragment of a dismantled accretionary wedge embedded during the deposition of the Cilento group sedimentary successions in a thrust top basin
