    Age-related changes in arterial blood-gas variables in Holstein calves at moderate altitude

    Includes bibliographical references (pages 19-20).The goal of this study was to determine whether peripheral oxygen delivery and efficacy of alveolar-arterial oxygen (A-a O2) transfer, as estimated from the A-a O2 pressure gradient, are compromised in Holstein calves at moderate altitude. The primary objective was to evaluate age-related changes in arterial blood-gas variables, L-lactate, and hematocrit in healthy calves. The secondary objective was to determine if coughing and nasal discharge, commonly used indicators of respiratory disease, are associated with A-a O2 gradient. Arterial blood-gas tensions were evaluated in a cohort of 61 dairy calves on one farm at moderate altitude (1,601 m to 1,696 m). Sampling was performed on four occasions at approximately 10, 38, 150, and 261 days of age. Hyperventilation, as indicated by hypocapnia, was evident in calves of all ages. Increasing age was associated with a nonlinear increase in arterial oxygen tension (P<0.001) and a nonlinear decrease in A-a O2 gradient (P<0.001). The mean A-a O2 gradient at 10 and 38 days of age was over 18 mmHg, indicating poor efficacy of oxygen transfer. Cough score (P=0.02) but not nasal score (P=0.32) was associated with an in increase in A-a O2 pressure gradient. Mean hematocrit remained low (<27%) despite hypoxemia. From 38 days of age, median L-lactate concentration remained over 1.5 mmol/L, indicating substantial anaerobic respiration due to inadequate oxygen delivery. Twenty-five percent of calves were treated for respiratory disease. The maximum age at first treatment was 102 days. In conclusion, there was a nonlinear improvement in A-a O2 transfer efficacy with increasing age, but peripheral oxygen delivery remained compromised. Hyperventilation and impaired A-a O2 transfer due to functional immaturity of the pulmonary system may be risk factors for respiratory disease in dairy calves at moderate altitude.Published with support from the Colorado State University Libraries Open Access Research and Scholarship Fund

    Catecholamines and phosphate excretion by the remnant kidney. The remnant kidney (RK) exhibits an enhanced fractional excretion of phosphate (FEPi) even in the absence of parathyroid hormone (PTH). Thus, factors other than PTH contribute to this adaptive phosphaturia. Dopamine (DA) infusion is phosphaturic, whereas stimulation of adrenoreceptors is antiphosphaturic. Therefore, the hypothesis that alterations in catecholamines by the RK may be associated with the phosphaturia exhibited by this model was tested. Male Sprague-Dawley rats were subjected to right nephrectomy and surgical ablation of the left renal poles. Four weeks later rats with a RK (N = 10) and control rats with intact kidneys (N = 9) were anesthetized and thyroparathy-roidectomized (TPTX). Two hours after TPTX, urine samples were collected for measurements of urinary free DA excretion. Subsequently, 3% inulin in saline was infused for one hour and a 30 minute clearance was taken. The kidneys were then removed and frozen for determination of tissue norepinephrine (NE) and DA concentrations. Glomerular filtration rate was significantly lower in rats with a RK than in controls (0.57 ± 0.07 vs. 0.83 ± 0.08 ml/min/g kidney wt), whereas fractional excretion of phosphate (FEPi) was significantly higher (29.4 ± 4.7 vs. 8.3 ± 3.4%). Tissue NE concentration was significantly lower in the RK than in the control intact kidney (85.10 ± 4.95 vs. 129.60 ± 7.20 ng/g), whereas urinary DA excretion per nephron was significantly higher in the RK (0.12 ± 0.02 vs. 0.04 ± 0.006 pg/min). Infusion of the specific DA-1 receptor antagonist (SCH 23390) in rats with a RK significantly decreased the FEPi when compared to the saline infused control group (Δ-5.2 ± 4.3 vs. +9.5 ± 5.2%), however, urinary c-AMP excretions were not different. We conclude that decreased renal sympathetic nerve tone, unchanged tissue DA levels, and an increased DA synthesis per residual nephron are associated with the enhanced FEPi exhibited by the RK

    Generalidades do Trypanosoma cruzi, do Trypanosoma rangeli e do seu vetor (Rhodnius pallescens) no Panamá

    The eco-epidemiology of T. cruzi infection was investigated in the Eastern border of the Panama Canal in Central Panama. Between 1999 and 2000, 1110 triatomines were collected: 1050 triatomines (94.6%) from palm trees, 27 (2.4%) from periurban habitats and 33 (3.0%) inside houses. All specimens were identified as R. pallescens. There was no evidence of vector domiciliation. Salivary glands from 380 R. pallescens revealed a trypanosome natural infection rate of 7.6%, while rectal ampoule content from 373 triatomines was 45%. Isoenzyme profiles on isolated trypanosomes demonstrated that 85.4% (n = 88) were T. cruzi and 14.6% (n = 15) were T. rangeli. Blood meal analysis from 829 R. pallescens demonstrated a zoophilic vector behavior, with opossums as the preferential blood source. Seroprevalence in human samples from both study sites was less than 2%. Our results demonstrate that T. cruzi survives in the area in balanced association with R. pallescens, and with several different species of mammals in their natural niches. However, the area is an imminent risk of infection for its population, consequently it is important to implement a community educational program regarding disease knowledge and control measures.A epidemiologia da infecção do T. cruzi foi investigada na margem oriental do canal do Panamá, na região central da Republica do Panamá. A informação obtida durante o estudo avaliou fatores de risco da doença de Chagas nesta área. Entre 1999 e 2000, 1110 triatomíneos foram coletados: 1050 triatomíneos (94,6%) em palmeiras, 27 (2,4%) em habitats periurbanos e 33 (3,0%) no interior de casas. Todos os espécimens foram identificados como R. pallescens. Não havia nenhuma evidência de domiciliação do vetor. O exame de glândulas salivares de 380 R. pallescens revelaram taxa de infecção natural por Trypanosoma de 7,6%, mas o conteúdo da ampola rectal de 373 triatomíneos mostrou 45% de positividade. Os perfis de isoenzimas em Trypanosomas isolados demonstraram que 85,4% (n = 88) eram T. cruzi e 14,6% (n = 15) eram T. rangeli. A análise da refeição de sangue de 829 R. pallescens demonstrou comportamento zoofílico do vetor, sendo os gambás a fonte preferencial de sangue. Soroprevalência nos seres humanos de ambos locais de estudo foi menos que 2%. Nossos resultados demonstram que T. cruzi sobrevive na área em associação equilibrada com R. pallescens e com diversas espécies diferentes de mamíferos em seus nichos naturais. Entretanto, a área é um risco eminente de infecção para sua população, pelo que é importante executar um programa educacional na comunidade a respeito das medidas, do conhecimento e do controle da doença

    Ultralow phase noise microwave generation with an Er:fiber-based optical frequency divider

    We present an optical frequency divider based on a 200 MHz repetition rate Er:fiber mode-locked laser that, when locked to a stable optical frequency reference, generates microwave signals with absolute phase noise that is equal to or better than cryogenic microwave oscillators. At 1 Hz offset from a 10 GHz carrier, the phase noise is below -100 dBc/Hz, limited by the optical reference. For offset frequencies > 10 kHz, the phase noise is shot noise limited at -145 dBc/Hz. An analysis of the contribution of the residual noise from the Er:fiber optical frequency divider is also presented.Comment: 4 pages, 3 figure

    Ex-vivo HRMAS of adult brain tumours: metabolite quantification and assignment of tumour biomarkers

    Background: High-resolution magic angle spinning (HRMAS) NMR spectroscopy allows detailed metabolic analysis of whole biopsy samples for investigating tumour biology and tumour classification. Accurate biochemical assignment of small molecule metabolites that are “NMR visible” will improve our interpretation of HRMAS data and the translation of NMR tumour biomarkers to in-vivo studies. Results: 1D and 2D 1H HRMAS NMR was used to determine that 29 small molecule metabolites, along with 8 macromolecule signals, account for the majority of the HRMAS spectrum of the main types of brain tumour(astrocytoma grade II, grade III gliomas, glioblastomas, metastases, meningiomas and also lymphomas). Differences in concentration of 20 of these metabolites were statistically significant between these brain tumour types. During the course of an extended 2D data acquisition the HRMAS technique itself affects sample analysis: glycine, glutathione and glycerophosphocholine all showed small concentration changes; analysis of the sample after HRMAS indicated structural damage that may affect subsequent histopathological analysis. Conclusions: A number of small molecule metabolites have been identified as potential biomarkers of tumour type that may enable development of more selective in-vivo 1H NMR acquisition methods for diagnosis and prognosis of brain tumours

    Pulmonary arterial pressures, arterial blood-gas tensions, and serum biochemistry of beef calves born and raised at high altitude

    Includes bibliographical references (page 8).High-altitude exposure is physiologically challenging. This is particularly true for animals native to low-altitude environments, such as British breeds of cattle. The objective of this study was to document the effect of high altitude on select physiological parameters of healthy beef calves (Bos taurus) born and raised on a high-altitude ranch typical of the Rocky Mountain region. Pulmonary arterial pressures, arterial blood-gas tensions, serum biochemistry, and hematocrit were evaluated. The calves studied were a composite of British (50%-75%) and Continental (25-50%) breeds born on one ranch at an altitude of 2410 m. Calves were sampled at an altitude of 2410 m when 1 month old and again at an altitude of 2730 m when 3 and 6 months old. Between 3 and 6 months of age, calves had access to grazing from 2730 m to approximately 3500 m above sea level. On each occasion, 16 to 50 calves were sampled. Only calves that remained healthy throughout all three testing periods were included in the dataset. Calves with the highest pulmonary arterial pressures at 1 month of age tended to have the highest pressures at 6 months of age (r = 0.43, P = 0.16, n = 12). Respiratory alkalosis was greatest at 6 months of age (pH 7.48 ± 0.06). Mean alveolar-arterial oxygen pressure gradients were 11.7and 11.6 mmHg at 3 and 6 months of age, indicating poor transfer of oxygen from the alveoli into the pulmonary blood. Median values for blood lactate ranged from 1.4 to 3.4 mmol/L indicating substantial anaerobic respiration at all ages. Mean hematocrits were ≤ 35.7%, only slightly higher than values obtained from age-matched calves at sea level. These results suggest that the provision of oxygen to the peripheral tissues of beef calves may be compromised at altitudes over 2410 m.
