    Examining Multiple Dimensions of Task Technology Fit

    Investigating the fit of an information technology to a user’s task, known as task-technology fit (TTF), is a frequent focus of HCI/MIS research. In such research, “fit as moderation”, one of Venkatraman’s (1989) six conceptualizations of fit, is common. This conceptualization assumes two variables, e.g., task and technology, and an outcome variable. Task and technology, however, have multiple dimensions that should be considered when investigating fit. We examine three methods HCI/MIS researchers have used for augmenting the fit as moderation conceptualization for multiple dimensions. We propose and test a new method involving a single PLS model capturing the multiple dimensions of fit, which is more consistent with Venkatraman’s (1989) original conceptualization and statistical model for fit as moderation than those currently in the literature. Our method as compared against one method in the literature, using a separate model for each dimension, works at least as well

    Observation of prompt single muons and of missing energy associated with ÎŒ+Ό−\mu^{+}\mu^{-} pairs produced in hadronic interactions

    In a study of interactions of 400 GeV protons in a totally absorbing iron calorimeter the authors report two observations indicating the hadronic production of heavy short-lived weakly decaying particles. First they have observed a prompt muon signal in the region .8<p/sub t /<2.5 GeV/c. The rate is comparable in magnitude to the prompt 2 mu rate in the same kinematic region. In addition to detecting mu /sup +/ mu /sup -/ events arising from electromagnetic sources (e.g. rho to mu /sup +/ mu /sup -/, psi to mu /sup +/ mu /sup -/ etc.) they have observed mu /sup +/ mu /sup -/ pairs associated with a significant amount of missing energy indicative of final state neutrinos. Interpreting these data as production of DD pairs followed by single or double muonic decays leads to a model dependent estimate of total production cross-section of order 15 mu b. (14 refs)

    Genomic complexity of the variable region-containing chitin-binding proteins in amphioxus

    <p>Abstract</p> <p>Background</p> <p>The variable region-containing chitin-binding proteins (VCBPs) are found in protochordates and consist of two tandem immunoglobulin variable (V)-type domains and a chitin-binding domain. We previously have shown that these polymorphic genes, which primarily are expressed in the gut, exhibit characteristics of immune genes. In this report, we describe VCBP genomic organization and characterize adjacent and intervening genetic features which may influence both their polymorphism and complex transcriptional repertoire.</p> <p>Results</p> <p>VCBP genes 1, 2, 4, and 5 are encoded in a single contiguous gene-rich chromosomal region and VCBP3 is encoded in a separate locus. The VCBPs exhibit extensive haplotype variation, including copy number variation (CNV), indel polymorphism and a markedly elevated variation in repeat type and density. In at least one haplotype, inverted repeats occur more frequently than elsewhere in the genome. Multi-animal cDNA screening, as well as transcriptional profilingusing a novel transfection system, suggests that haplotype-specific transcriptional variants may contribute to VCBP genetic diversity.</p> <p>Conclusion</p> <p>The availability of the <it>Branchiostoma floridae </it>genome (Joint Genome Institute, Brafl1), along with BAC and PAC screening and sequencing described here, reveal that the relatively limited number of VCBP genes present in the amphioxus genome exhibit exceptionally high haplotype variation. These VCBP haplotypes contribute a diverse pool of allelic variants, which includes gene copy number variation, pseudogenes, and other polymorphisms, while contributing secondary effects on gene transcription as well.</p

    Alternative Complement Activity in the Egg Cytosol of Amphioxus Branchiostoma belcheri: Evidence for the Defense Role of Maternal Complement Components

    Background: The eggs in most invertebrates are fertilized externally, and therefore their resulting embryos are exposed to an environment full of microbes, many of which are pathogens capable of killing other organisms. How the developing embryos of invertebrates defend themselves against pathogenic attacks is an intriguing question to biologists, and remains largely unknown. Methodology/Principal Findings: Here we clearly demonstrated that the egg cytosol prepared from the newly fertilized eggs of amphioxus Branchiostoma belcheri, an invertebrate chordate, was able to inhibit the growth of both the Gramnegative bacterium Vibrio anguillarum and the Gram-positive bacterium Staphylococcus aureus. All findings point to that it is the complement system operating via the alternative pathway that is attributable to the bacteriostatic activity. Conclusions/Significance: This appears to be the first report providing the evidence for the functional role of the maternal complement components in the eggs of invertebrate species, paving the way for the study of maternal immunity in other invertebrate organisms whose eggs are fertilized in vitro.
