Near-infrared photoimmunotherapy targeting EGFR-Shedding new light on glioblastoma treatment

Glioblastomas (GBMs) are high-grade brain tumors, differentially driven by alterations (amplification, deletion or missense mutations) in the epidermal growth factor receptor (EGFR), that carry a poor prognosis of just 12–15 months following standard therapy. A combination of interventions targeting tumor-specific cell surface regulators along with convergent downstream signaling pathways may enhance treatment efficacy. Against this background, we investigated a novel photoimmunotherapy approach combining the cytotoxicity of photodynamic therapy with the specificity of immunotherapy. An EGFR-specific affibody (ZEGFR:03115) was conjugated to the phthalocyanine dye, IR700DX, which when excited with near-infrared light produces a cytotoxic response. ZEGFR:03115–IR700DX EGFR-specific binding was confirmed by flow cytometry and confocal microscopy. The conjugate showed effective targeting of EGFR positive GBM cells in the brain. The therapeutic potential of the conjugate was assessed both in vitro, in GBM cell lines and spheroids by the CellTiter-Glo® assay, and in vivo using subcutaneous U87-MGvIII xenografts. In addition, mice were imaged pre- and post-PIT using the IVIS/Spectrum/CT to monitor treatment response. Binding of the conjugate correlated to the level of EGFR expression in GBM cell lines. The cell proliferation assay revealed a receptor-dependent response between the tested cell lines. Inhibition of EGFRvIII+ve tumor growth was observed following administration of the immunoconjugate and irradiation. Importantly, this response was not seen in control tumors. In conclusion, the ZEGFR:03115–IR700DX showed specific uptake in vitro and enabled imaging of EGFR expression in the orthotopic brain tumor model. Moreover, the proof-of-concept in vivo PIT study demonstrated therapeutic efficacy of the conjugate in subcutaneous glioma xenografts

Agnete Lothʼs edition of the longer version of Jarlmanns saga og Hermanns included an accompanying English paraphrase (by Gillian Fellows Jensen), but there has never been a full translation into English, much less of the shorter version as edited by Hugo Rydberg. We rectify that omission here, providing a normalized text of Rydbergʼs edition with an English translation alongside in the hopes of making this entertaining saga more accessible to a wider audience

Analiza przydatności przedwojennych map scaleniowych w wybranych procedurach określających przebieg granic na przykładzie obrębu Matyniów oraz Czarniecka Góra

In the areas of the Russian partition, materials defining the course of the borders created during the interwar period are found. Such documents are prewar consolidations. Certified surveyors use similar materials in the work to determine the course of borders in fields. On the basis of the pre-war consolidations, they perform the activities of resuming the border marks in accordance with Art. 39 of the Geodetic and Cartographic Law or activities to establish the course of borders based on §39 of the Regulation on the Land and Building Register. In this article, land consolidation maps of the villages of Matyniów and Czarniecka Góra (located in the Konecki of Świętokrzyskie Voivodeship) were analyzed in order to check whether similar materials could form the basis for the resumption of the border marks.Na terenach zaboru rosyjskiego odnajduje się materiały określające przebieg granic powstałe w okresie międzywojennym. Dokumentami taki są scalenia przedwojenne. Geodeci uprawnieni wykorzystują podobne materiały w pracach mających na celu określenie przebiegu granicy w terenie. Na podstawie przedwojennych scaleń wykonawcy dokonują czynności wznowienia znaków granicznych zgodnie z art. 39 ustawy Prawo geodezyjne i kartograficzne lub czynności ustalenia przebiegu granic na podstawie §39 rozporządzenia w sprawie ewidencji gruntów i budynków. W pracy przeanalizowano mapy scaleniowe gruntów wsi Matyniów oraz Czarniecka Góra położonych w powiecie koneckim województwa świętokrzyskiego w celu sprawdzenia, czy podobne materiały mogą stanowić podstawę do przeprowadzenia czynności wznowienia znaków granicznych

Praktyczne problemy realizacji wybranych procedur określających przebieg granic

Dużą część prac geodezyjnych zgłaszanych do ośrodków dokumentacji geodezyjnej i kartograficznej stanowią opracowania, których efektem końcowym jest określenie w terenie przebiegu granicy. Do momentu nowelizacji rozporządzenia w sprawie ewidencji gruntów i budynków ustalenie przebiegu granic pojedynczych działek ewidencyjnych możliwe było tylko w trybie ustawy Prawo geodezyjne i kartograficzne. Obecnie możliwe jest zastosowanie procedury ustalenia przebiegu granic działek ewidencyjnych wykonywane na zlecenie podmiotów prywatnych na potrzeby aktualizacji operatu ewidencji gruntów i budynków. Przebieg granic na gruncie można określić także po przeprowadzeniu czynności wyznaczenia punktów granicznych ujawnionych uprzednio w ewidencji gruntów lub wznowienia znaków granicznych. W artykule przeanalizowano opracowania geodezyjne sporządzone zgodnie z art. 39 ustawy Prawo geodezyjne i kartograficzne oraz wykonywane zgodnie z § 39 rozporządzenia w sprawie ewidencji gruntów i budynków na potrzeby aktualizacji operatu ewidencji gruntów i budynków. Na podstawie dokonanej analizy wskazano problemy geodezyjno-prawne spowodowane nieprecyzyjnymi regulacjami, z którymi spotykają się wykonawcy podczas określania w terenie przebiegu granicy. W pracy przeanalizowano także materiały, które w świetle obowiązujących przepisów prawa mogą stanowić podstawę do przeprowadzenia czynności wznowienia znaków granicznych, wyznaczenia punktów granicznych oraz ustalenia przebiegu granic.A large part of the surveying work reported to geodetic and cartography documentation centers are those that, in the end, result in the determination of a border in a field. Until the amendment of the regulation on the Land and Buildings Register, the determination of the borders of individual land parcels was possible only due to acting on the basis of the Geodetic and Cartographic Law. Currently, it is possible to use the procedure for determining the course of the borders of plots performed on behalf of private entities for the purpose of updating the Land and Buildings Register. Determination of the course of the boundaries in a field is also possible on the basis of the performed tasks of determining the border points previously disclosed in the land register or the resumption of border marks. This article analyzes geodetic studies based on Art. 39 of the Geodetic and Cartographic Law, and in accordance with § 39 of the Regulation on the Land and Buildings Register made for the purpose of updating the Land and Buildings Register. On the basis of the analysis, geodetic and legal problems were identified, which are caused by the imprecise regulations encountered by the contractors while carrying out activities aimed at defining the borderline of the field. The author also analyzes the materials that, in light of the binding legal provisions, may serve as a basis for carrying out the renewal of border marks, setting border points, and determining the course of borders

The paper aims to present logistics solutions in Poland’s e-commerce market using the example of the rapidly growing InPost SA Group. The group’s development strategy focuses on the e-commerce market and the expansion of services including innovative solutions. The subject literature, and documents and information provided by the research subject, the InPost SA, are the sources used in this study. The paper presents the specificities of the e-commerce sector, especially courier services, express and parcel deliveries. Receivers of shipments in the e-commerce market are a demanding group of customers, who continually pressure the courier market operators to expand their range of services.
