    Microfluidic chromatography for early stage evaluation of biopharmaceutical binding and separation conditions

    Optimization of separation conditions for biopharmaceuticals requires evaluation of a large number of process variables. To miniaturize this evaluation a microfluidic column (1.5 mu L volume and 1cm height) was fabricated and packed with a typical process scale resin. The device was assessed by comparison to a protein separation at conventional laboratory scale. This was based upon measurement of the quality of packing and generation of breakthrough and elution curves. Dynamic binding capacities from the microfluidic column compared well with the laboratory scale. Microfluidic scale gradient elution separations also equated to the laboratory column three orders of magnitude larger in scale

    A COMMUNITY-BASED VULNERABILITY ASSESSMENT OF FLOODS IN URBAN AREAS OF KAMPUNG MELAYU, JAKARTA

    Flooding has become a serious problem in Jakarta. During floods .of 2007, Kampung Melayu, East Jakarta was the worst hit.by the floods. Community have different perceptions on disaster and have different effort to overcome the hazards. Therefore, local government and relevant institution should investigate this situation and make this information a valuable input in developing and implementing response plans in flood mitigation. This research is to explore the vulnerability of floods based on local people\u27s perception. There were 83 households interviewed using questionnaire. Certain elements at risk related with physical and socio:economic aspects were identified. Physical information concerned the building structure and building contents. Several socio-economic characteristics were used as key indicators to analyze the vulnerability of people. Generally, the result of this research shows that the ability of people to cope with the flooding i$ linked with the capacity of the people itself. The capability of people to deal withflooding was influenced by several indicators.based on their socio- . economic characteristics. For example, lower income people will experience more suffering than the wealthier, because they cannot afford the\u27 costs of repair, reconstruction. Although the wealthier are likely to experience a higher degree of economic damage due to possessions of higher value. Base on the analysis, all coping strategies and flood measures are not enough to cope with flooding in the study area

    The Atmospheric Effects of Stratospheric Aircraft: a First Program Report

    Studies have indicated that, with sufficient technology development, high speed civil transport aircraft could be economically competitive with long haul subsonic aircraft. However, uncertainty about atmospheric pollution, along with community noise and sonic boom, continues to be a major concern; and this is addressed in the planned 6 yr HSRP begun in 1990. Building on NASA's research in atmospheric science and emissions reduction, the AESA studies particularly emphasizing stratospheric ozone effects. Because it will not be possible to directly measure the impact of an HSCT aircraft fleet on the atmosphere, the only means of assessment will be prediction. The process of establishing credibility for the predicted effects will likely be complex and involve continued model development and testing against climatological patterns. Lab simulation of heterogeneous chemistry and other effects will continue to be used to improve the current models

    Knowledge, attitude and preventive practices regarding tuberculosis and its predictors among HIV patients in General Hospital, Minna, North-Central, Nigeria

    Introduction: Tuberculosis is the commonest cause of death among HIV patients in Nigeria. Though tuberculosis is common among HIV patients, their knowledge about transmission and prevention is poor. Objective: To determine knowledge, attitude and preventive practices regarding tuberculosis and its predictors among HIV patients receiving treatment. Methods: A cross sectional study was carried out involving 226 randomly selected HIV patients receiving care at General Hospital, Minna. Study duration was from June 2015 to July 2017. Data on socio-demographic characteristics, psychosocial factors, knowledge, attitude and preventive practices regarding tuberculosis was collected using a self-administered, WHO modified, pretested and validated questionnaires. Data collected was analysed using SPSS version 22. Results: Out of 226 respondents, 60(26.5%) had poor knowledge, 123(54.4%) had negative attitude and 48(21.2%) had poor preventive practices regarding tuberculosis. The predictors of knowledge were age (B=0.087, 95%CI=0.031, 0.143, p=0.002), depression (B=-0.444, 95%CI=-0.673,-0.214, p<0.001) monthly income (B=-0.058, 95%CI=-0.095, -0.021, p=0.002) and marital status (B=-1.276, 95%CI=-2.525,-0.025, P=0.046). The predictors of attitude were anxiety (B=-0.395, 95%CI=-0.584, -0.206, p<0.001), cigarette smoking (B=4.473, 95%CI=0.992, 7.954, p=0.012) and duration of HIV infection (B=0.371, 95%CI=0.110, 0.631, p=0.005). The predictors of practice were depression (B=-0.176, 95%CI=-0.276, -0.076, p=0.001), cigarette smoking (B=2.200, 95%CI=0.926, 3.474, p=0.001) and duration of HIV infection (B=0.098, 95%CI=0.003, 0.194, p=0.043). Conclusion: Overall, knowledge and practices regarding tuberculosis prevention were good, but attitude was poor. The predictors of knowledge, attitude and preventive practices were age, depression, anxiety, cigarette smoking, monthly income, marital status and duration of HIV infection

    Application of a Coupled Vegetation Competition and Groundwater Simulation Model to Study Effects of Sea Level Rise and Storm Surges on Coastal Vegetation

    Global climate change poses challenges to areas such as low-lying coastal zones, where sea level rise (SLR) and storm-surge overwash events can have long-term effects on vegetation and on soil and groundwater salinities, posing risks of habitat loss critical to native species. An early warning system is urgently needed to predict and prepare for the consequences of these climate-related impacts on both the short-term dynamics of salinity in the soil and groundwater and the long-term effects on vegetation. For this purpose, the U.S. Geological Survey’s spatially explicit model of vegetation community dynamics along coastal salinity gradients (MANHAM) is integrated into the USGS groundwater model (SUTRA) to create a coupled hydrology–salinity–vegetation model, MANTRA. In MANTRA, the uptake of water by plants is modeled as a fluid mass sink term. Groundwater salinity, water saturation and vegetation biomass determine the water available for plant transpiration. Formulations and assumptions used in the coupled model are presented. MANTRA is calibrated with salinity data and vegetation pattern for a coastal area of Florida Everglades vulnerable to storm surges. A possible regime shift at that site is investigated by simulating the vegetation responses to climate variability and disturbances, including SLR and storm surges based on empirical information

    Efficacy of temsirolimus in metastatic chromophobe renal cell carcinoma

    &lt;p&gt;Background: Renal cell carcinoma (RCC) is a histopathologically and molecularly heterogeneous disease with the chromophobe subtype (chRCC) accounting for approximately 5% of all cases. The median overall survival of advanced RCC has improved significantly since the advent of tyrosine kinase inhibitors and mammalian target of rapamycin (mTOR) inhibitors. However, high-quality evidence for the use of new generation tyrosine kinase inhibitors in patients with advanced chRCC is lacking. Few published case reports have highlighted the use of temsirolimus in chRCC.&lt;/p&gt; &lt;p&gt;Case presentation: Here, we report the case of a 36-year-old Caucasian woman with metastatic chRCC with predominantly skeletal metastases who was refractory to sunitinib who demonstrated a durable clinical response to temsirolimus lasting 20 months. We review the available evidence pertaining to the use of new generation molecularly targeted agents, in particular mTOR inhibitors in chRCC and discuss their emerging role in the management of this disease which would aid the oncologists faced with the challenge of treating this rare type of RCC.&lt;/p&gt; &lt;p&gt;Conclusion: Conducting randomised clinical trials in this rarer sub-group of patients would be challenging and our case report and the evidence reviewed would guide the physicians to make informed decision regarding the management of these patients.&lt;/p&gt

    The Src Homology 3 Domain Is Required for Junctional Adhesion Molecule Binding to the Third PDZ Domain of the Scaffolding Protein ZO-1

    Tight junctions are cell-cell contacts that regulate the paracellular flux of solutes and prevent pathogen entry across cell layers. The assembly and permeability of this barrier are dependent on the zonula occludens (ZO) membrane-associated guanylate kinase (MAGUK) proteins ZO-1, -2, and -3. MAGUK proteins are characterized by a core motif of protein-binding domains that include a PDZ domain, a Src homology 3 (SH3) domain, and a region of homology to guanylate kinase (GUK); the structure of this core motif has never been determined for any MAGUK. To better understand how ZO proteins organize the assembly of protein complexes we have crystallized the entire PDZ3-SH3-GUK core motif of ZO-1. We have also crystallized this core motif in complex with the cytoplasmic tail of the ZO-1 PDZ3 ligand, junctional adhesion molecule A (JAM-A) to determine how the activity of different domains is coordinated. Our study shows a new feature for PDZ class II ligand binding that implicates the two highly conserved Phe−2 and Ser−3 residues of JAM. Our x-ray structures and NMR experiments also show for the first time a role for adjacent domains in the binding of ligands to PDZ domains in the MAGUK proteins family

    Aircraft Engine Particulate Matter Emissions from Sustainable Aviation Fuels: Results from Ground-Based Measurements during the NASA/DLR Campaign ECLIF2/ND-MAX

    The use of alternative jet fuels by commercial aviation has increased substantially in recent years. Beside the reduction of carbon dioxide emission, the use of sustainable aviation fuels (SAF) may have a positive impact on the reduction of particulate emissions. This study summarizes the results from a ground-based measurement activity conducted in January 2018 as part of the ECLIF2/ND-MAX campaign in Ramstein, Germany. Two fossil reference kerosenes and three different blends with the renewable fuel component HEFA-SPK (Hydroprocessed Esters and Fatty Acids Synthetic Paraffinic Kerosene) were burned in an A320 with V2527-A5 engines to investigate the effect of fuel naphthalene/aromatic content and the corresponding fuel hydrogen content on non-volatile particle number and mass emissions. Reductions up to 70% in non-volatile particle mass emission compared to the fossil reference fuel were observed at low power settings. The reduction trends to decrease with increasing power settings. The fuels showed a decrease in particle emission with increasing fuel hydrogen content. Consequently, a second fossil fuel with similar hydrogen content as one of the HEFA blends featured similar reduction factors in particle mass and number. Changes in the fuel naphthalene content had significant impact on the particle number emission. A comparison to in-flight emission data shows similar trends at cruise altitudes. The measurements highlight the importance of individual fuel components in regulating engine emissions, particularly at the low thrust settings typically employed during ground operations (e.g. during idle and taxi). Therefore, when selecting and mixing SAF blends to meet present fuel-certification standards, attention should be paid to minimizing complex aromatic content to achieve the greatest possible air quality and climate benefits

    Batek people describe their many laughter taboos with utmost seriousness, and in ethical terms of good and bad. Despite this, people often get it wrong—sometimes laughing all the more when the taboos forbid it. Because laughter can be ambiguous and impossible to control, being wrong can be accepted without the need for discussion or reflection. People thus act autonomously while holding deeply shared ethical orientations. Here, ethics can be both culturally predefined and shaped by individuals, as when it comes to laughter people draw on individual and shared concerns in an ad hoc, flexible manner. Laughter's tangled contradictions thus demonstrate that people's understandings of being “good” are mutually implicated with their understandings of what it means to be a person in relation to others
