Are endocannabinoid type 1 receptor gene (CNR1) polymorphisms associated with obesity and metabolic syndrome in postmenopausal Polish women?
Objective: The aim of this study was to determine whether genetic variation at the cannabinoid receptor-1 (CNR1) locus could have an effect on adiposity, fat distribution and obesity-related metabolic disorders in Polish postmenopausal women.Design and Subjects: The A3813G, G1422A and A4895G single nucleotide polymorphisms of CNR1 were genotyped in 348 randomly selected postmenopausal women aged 50-60 years recruited from the Wroclaw city population. Measurements: CNR1 genotypes, anthropometric measures (BMI, WC, body fat distribution by DEXA) and metabolic parameters (glucose, lipid profile, insulin FIRI) were determined.Results: The 3813G allele was not significantly associated with higher body mass, BMI, WC, total fat, or fat percentage, but was associated with higher android fat deposit (2971.78 ± 1655.08 ± 2472.64 ± 1300.53, p = 0.007) and percentage of android fat (37.59 ± 8.45 vs. 35.66 ± 7.63, p = 0.062). The 1422A allele was associated with higher total fat (31587.72 ± 9161.28 g vs. 26078.26 ± 7552.14 g, p = 0.019), fat percentage (40.51 ± 5.66% vs. 37.51 ± 4.99%, p = 0.052), and percentage of android fat (40.86 ± 9.73% vs. 36.09 ± 7.70%, p = 0.047). No associations were observed for the A4895G variant.Conclusions: There is an association of variants of CNR1 with obesity-related phenotypes in Polish postmenopausal women. As CB1 is a drug target for obesity, pharmacogenetic receptor gene analysis of obesity treatment by endocannabinoid blockade may be of interest to identify the best responders
Anorexia and bulimia risk factors among 18-years-old girls
WSTĘP. Zaburzenia odżywiania coraz częściej występują wśród nastolatek
i młodych kobiet. Wczesna identyfikacja osób ze zwiększonym
ryzykiem zaburzeń odżywiania umożliwia odpowiednie postępowanie
profilaktyczne i może zapobiec wystąpieniu choroby. Celem
pracy była ocena częstości występowania konstytucjonalnych, behawioralnych
i psychologicznych czynników ryzyka anoreksji i bulimii
u 18-letnich dziewcząt.
MATERIAŁ I METODY. Badania przeprowadzono wśród 100 uczennic
liceum ogólnokształcącego, które wyraziły zgodę na udzielenie odpowiedzi
w anonimowej ankiecie. Pytania ankietowe dotyczyły stylu życia,
w tym sposobu odżywiania, stosowanych używek i aktywności fizycznej
oraz psychologicznych czynników ryzyka jadłowstrętu psychicznego
i bulimii.
WYNIKI. Wskaźnik masy ciała (BMI, body mass index) dziewcząt mieścił
się w granicach 15-33 kg/m2 (średnio 20,6 ± 3,1). Większość badanych
(67%) miała prawidłową masę ciała (BMI 18,5-24,9), 9% miało
nadwagę lub otyłość (BMI > 25), natomiast 24% badanych miało niedowagę
(BMI < 18,5), z czego 6 dziewcząt miało BMI niższe niż 17,5.
Ponad połowa (53%) respondentek nie akceptowała swojego wyglądu,
a problem z utrzymaniem prawidłowej masy ciała zgłosiła co trzecia

dziewcząt deklarowała aktywność fizyczną w czasie wolnym. Duża
grupa dziewcząt paliła tytoń (46% paliło regularnie, a 12% - incydentalnie)
i piła alkohol przynajmniej raz w tygodniu (30%).
WNIOSEK. U 18-letnich dziewcząt istnieje duża częstość występowania
konstytucjonalnych, behawioralnych i psychologicznych czynników
ryzyka anoreksji i bulimii, do których najczęściej należą: brak akceptacji
własnego ciała i nieprawidłowe odżywianie.INTRODUCTION. Eating disorders are an increasing problem among
teenagers and young women. Early identification of people with risk
factors of eating disorders allows a suitable prophylactic proceeding
and can prevent from the disease. The aim of our study was evaluation
of frequency of constitutional, behavioral and psychological risk
factors of anorexia and bulimia among 18-years old girls.
MATERIALS AND METHODS. Our study was performed among 100
female pupils of secondary school, who fulfill the anonymous questionnaire.
The questions regarded to elements of lifestyles such as
diet, drugs use, physical activity and psychological risk factors of
anorexia and bulimia.
RESULTS. BMI value of investigated subjects varied from 15 to
33 kg/m2 (mean 20.6 ± 3.1). Most of subjects (67%) have a normal
BMI (18.5-24.9), 9% was overweight or obese (BMI > 25), 24%
was underweight (BMI < 18.5) among which 6% had a BMI < 17.5.
More than half of the girls (53%) didn’t accept their body image and
33% reported subjective overweight. Improper diet habits, irregular meals or too small daily meals number, were reported by 76% of
subjects. Physical activity in leisure time declared only 33% of girls.
High number of pupils smoked cigarettes (46% - regularly and
12% - incidentally). One third of subjects drunk alcohol at least
once a week.
CONCLUSION. There is high frequency of constitutional, behavioral
and psychological risk factors of anorexia and bulimia among
18-years old girls. The most frequent risk factors are body image
dissatisfaction and improper diet habits
Associations between overweight, health behaviors and Figure Rating Scale by Stunkard at 18-year-old secondary school students from Wroclaw
Introduction: Overweight in adolescence are associated with unfavourable consequences throughout the life-span, especially in chronic disorders such metabolic syndrome, type 2 diabetes.. Diet, physical activity and psychosocial factors are important risk factors of obesity in this group.
The aim of the study was to find the relationship between the body mass index (BMI) and diet, physical activity and perception of body mass according to Figure Rating Scale by Stunkard.
Material and methods: The study involved 104 secondary school students from Wrocław aged 18 years old (84 girls and 20 boys). Physical activity was assessed by the International Physical Activity Questionnaire (IPAQ) (http://www.ipaq.ki.se). Original survey questionnaire, based on the health programme “Keep straight”, was used to assess selected behaviors in the field of nutrition. Perception of body mass was checked with the Figure Rating Scale, according to Stunkard. The anthropometric research was based on the body weight, height, waist and hip circumference, BMI and WHR were calculated.
Results: Most of the students (82%) represented a proper BMI, respectively 83% girls and 75% boys and correctly percepted their body mass index. We have shown adverse relationship between BMI and the number of dietary errors (r=-0,23,
Jakość nasienia u młodych, zdrowych mężczyzn, mieszkańców Dolnego Śląska (AndroLS)
Introduction: Contrary to other parts of the continent, little information is available regarding semen quality among subjects from central and eastern Europe.
Material and methods: We evaluated semen profiles among a sample of men from an industrialised region of Poland. We directly invited 5000 healthy inhabitants of the region (aged 18–35 years; with unchecked fecundity) to participate in the study. Among the 500 who were eligible and willing to participate, we acquired detailed information and semen and blood samples from 177 subjects.
Results: Semen volume, sperm concentration, and total sperm count were, respectively, (mean ± SD): 3.1 ± 1.5 ml, 60 ± 44 x 106/ml and 170 ± 137 x 106/ml. Percentage of normal forms was 14.7 ± 6.5%.
Conclusions: Due to the relatively low sperm motility (mean ± SD: 54 ± 16%) and vitality (mean ± SD: 60 ± 15%) values, these variables require special attention during routine evaluations. The WHO 2010 criteria for these two parameters were met in only 60% and 66% of the samples, respectively. Further studies on men with different educational levels, social environments, or living conditions are needed to confirm our results. Wstęp: Odmiennie niż w przypadku innych części kontynentu dysponujemy niewielką ilością informacji dotyczących jakości nasienia w populacjach centralnej i wschodniej Europy.
Materiał i metody: W badaniu ocenialiśmy próbki nasienia młodych, zdrowych mężczyzn. Zaproszenia skierowaliśmy do 5000 mieszkańców uprzemysłowionego regionu Polski w wieku 18–35 lat, o nieznanym stanie płodności. Spośród 500 mężczyzn, którzy wstępnie zgodzili się na udział w badaniu i spełniali kryteria włączenia, pełne informacje (próbki nasienia i krwi) uzyskaliśmy w 177 przypadkach.
Wyniki: Objętość nasienia, koncentracja plemników i liczba plemników u badanych mężczyzn wynosiły odpowiednio (średnia ± odchylenie standardowe): 3.1 ± 1.5 ml, 60 ± 44 x 106/ml i 170 ± 137 x 106/ml. Odsetek form o prawidłowej budowie wynosił 14.7 ± 6.5%.
Wnioski: Ze względu na relatywnie małą ruchliwość (średnia ± odchylenie standardowe: 54 ± 16%) i żywotność plemników (średnia ± odchylenie standardowe: 60 ± 15%) wydaje się, że cechy te wymagają szczególnej uwagi podczas rutynowej diagnostyki. Kryteria normy wg WHO (2010 r.) w zakresie dwóch wspomnianych wyżej parametrów spełniało w naszym badaniu odpowiednio tylko 60% i 66% próbek. Uzyskane wyniki wymagają potwierdzenia u mężczyzn o różnym: stopniu wykształcenia, uwarunkowaniach socjalnych i miejscu zamieszkania.
Relationship of β3-adrenergic receptor polymorphism with metabolic syndrome and oxidative stress parameters in postmenopausal women
Introduction: Some studies indicate, that the Trp64/Arg64 polymorphism of β3-adrenergic receptor (ADRB3) is associated
with obesity, insulin resistance and earlier onset of type 2 diabetes mellitus.The aim of our study was evaluation of frequency of this ADRB3 polymorphism and his association with metabolic syndrome
parameters and oxidative stress in postmenopausal women.
Material and methods: We performed the study among 94 women, aged 50-60, selected randomly from Wroclaw city
population. Estimation of anthropometric parameters, densitometry (total body fat, android and gynoid deposits - using
DPX(+) Lunar, USA device) and biochemical estimations such as lipid profile, glucose, insulin, estradiol and FSH serum
level (using commercial kits) were carried out. Oxidative stress was estimated by measurement of thiobarbituric-reactive
substances (TBARS) serum concentration, using Yagi method, on spectrofluorimeter Perkin-Elmer LS55. Blood for analysis
was collected before, direct after and 6 h after the 30-minutes physical test using cycloergometer. ADRB3 genotyping was
performed by PCR and minisequencing using ABI 310 sequencer (Applied Biosystems).
Results: The frequency of Trp64/Arg64 genotype in investigated population was 15.8%. The Arg64/Arg64 genotype had only
one woman. Women bearing Trp64/Arg64 genotype showed higher mean serum level of triglycerides and lower serum level
of HDL-cholesterol in comparison to women bearing Trp64/Trp64 genotype, however without statistical significance (p > 0.05)
(respectively, means ± SD for triglycerides: 140.3 ± 64.1 vs. 113.9 ± 56.2 mg/dl; and for HDL-cholesterol: 60.9 ± 11.9 vs.
67.0 ± 16.9 mg/dl). Both groups did not differ in any other investigated anthropometric nor biochemic parameter.
Conclusions: 1. The Trp64/Arg64 polymorphism of β3-adrenergic receptor could be associated with lipid profile disoders
observed in metabolic syndrome in postmenopausal women, however it should be explaned basing on the study with
more included subjects. 2. The Trp64/Arg64 polymorphism of β3-adrenergic receptor has no influence on oxidative stress
intensification after standardized physical effort in postmenopausal women.

otyłości, insulinooporności i wcześniejszym rozwojem cukrzycy typu 2. Celem niniejszych badań była ocena
częstości występowania i związku wspomnianego polimorfizmu ADRB3 z zespołem metabolicznym i parametrami stresu
oksydacyjnego u kobiet po menopauzie.
Materiał i metody: Do badania zakwalifikowano 94 kobiety w wieku 50-60 lat, wybrane losowo spośród mieszkanek
Wrocławia. U wszystkich badanych wykonano pomiary antropometryczne, densytometrię całego ciała z oceną depozytu
andro- i gynoidalnego (z użyciem aparatu DPX(+) Lunar, USA) oraz oznaczenia biochemiczne: profil lipidowy, stężenie
glukozy, insuliny, estradiolu i hormonu folikulotropowego (FSH, follicle-stimulating hormone) z użyciem komercyjnych zestawów
odczynników. Wskaźnikiem stresu oksydacyjnego było stężenie substancji reagujących z kwasem tiobarbiturowym
(TBARS) oznaczane w surowicy metodą Yagi, na spektrofluorymetrze Perkin-Elmer LS55. Krew do oznaczenia stresu
oksydacyjnego pobierano przed, bezpośrednio po i 6 godzin po 30-minutowym wysiłku fizycznym na cykloergometrze.
Genotypowanie ADRB3 przeprowadzono metodą łańcuchowej reakcji polimerazy (PCR, polymerase chain reaction) i minisekwencjonowania
z użyciem aparatu ABI 310 (Applied Biosystems).
Wyniki: Częstość występowania genotypu Trp64/Arg64 w badanej populacji wyniosła 15,8%. Genotyp Arg64/Arg64 wystąpił
tylko u jednej osoby. Kobiety z genotypem Trp64/Arg64 miały średnio wyższe stężenie triglicerydów i niższe stężenie cholesterolu
frakcji HDL w surowicy w porównaniu z kobietami z genotypem Trp64/Trp64, chociaż różnica ta nie była istotna statystycznie
(p > 0,05) (odpowiednio: średnia ± odchylenie standardowe dla triglicerydów: 140,3 ± 64,1 vs. 113,9 ± 56,2 mg/dl;
dla cholesterolu frakcji HDL: 60,9 ± 11,9 vs. 67,0 ± 16,9 mg/dl). Poza tym obie grupy nie różniły się żadnym innym badanym
parametrem antropometrycznym i biochemicznym.
Wnioski: 1. Polimorfizm Trp64/Arg64 receptora β3-adrenergicznego może wiązać się z występowaniem zaburzeń lipidowych
charakterystycznych dla zespołu metabolicznego u kobiet po menopauzie, jednak problem ten wymaga przeprowadzenia
badań wśród większej grupy badanych. 2. Polimorfizm Trp64/Arg64 receptora β3-adrenergicznego nie wpływa na
nasilenie stresu oksydacyjnego po standaryzowanym wysiłku fizycznym u kobiet po menopauzie.

Regular yoga exercises and quality of life in women with low back pain - a pilot study
Back pain symptoms are one of prevailing disfunctions that determine lowering quality of life and functioning in work environment becoming not only a health problem, but also a social problem in developed countries. In available literature yoga exercises are more and more indicated as a method of leveling low back pain. The aim of this pilot study was to assess the effectiveness of 8-week yoga exercises on quality of life and back pain symptoms by it’s location and intensity in women with normal and excessive body weight. Twenty seven women completed both surveys (preliminary and evaluation), and systematically took part in yoga exercises for 8 weeks. The duration of a single yoga class was 45-60 minutes long. Yoga exercise program required attending to a conducted yoga classes (ashtanga yoga, yin yoga, hatha yoga, vinyasa yoga and yoga for hips and spine), at least twice a week for two months. After 8 weeks 92,59% of participants of program declared noticeable reduce in back pain intensity related to yoga exercises (including 22,22% of complete pain relief). Pain in the lumbar region of the spine was still dominating after 8 weeks (55,56%). It was observed that the majority of the SF-36 health domains scores increased values after 8-week yoga program, what could be related to the reduce of back pain intensity. After 8-week yoga program decrease in mean values of body mass were observed, however these changes were not statistically significant. Two-month yoga intervention may be recommended as a complementary physiotherapeutic method in low back pain treatment as well as to improve the quality of life for women. The relationship between yoga and prevention of obesity requires further research on larger group and the impact of obesity phenotypes is needed to be investigated
Influence of standardized physical effort on adponectin level and insulin sensitivity in postmenopausal women
WSTĘP. Insulinooporność i niskie stężenie adiponektyny to znane
czynniki chorób układu krążenia. Pojawia się pytanie, czy aktywność
fizyczna wpływa na te czynniki. Celem niniejszego badania
była ocena wpływu standaryzowanego wysiłku fizycznego na
stężenie insuliny i adiponektyny u kobiet po menopauzie.
MATERIAŁ I METODY. Badania przeprowadzono wśród 33 kobiet
po menopauzie w wieku 50-60 lat wybranych losowo spośród
mieszkańców Wrocławia. U wszystkich badanych wykonano pomiary
antropometryczne i densytometrię całego ciała z oceną depozytu
andro- i gynoidalnego, a ponadto przeprowadzono 30-minutowy
wysiłek fizyczny z użyciem ergometru rowerowego. Krew do
oznaczeń biochemicznych była pobierana bezpośrednio przed
i po wysiłku fizycznym oraz 6 godzin po wysiłku fizycznym. W surowicy oznaczono profil lipidowy oraz stężenie glukozy, insuliny
i adiponektyny z użyciem gotowych zestawów odczynników.
WYNIKI. Spośród wszystkich badanych wyodrębniono i porównano
2 grupy kobiet: 1. - kobiety otyłe (wskaźnik masy ciała [BMI, body
mass index] > 30, obwód talii > 82 cm, n = 19) i 2. - kobiety
szczupłe (BMI < 25, obwód talii < 82 cm, n = 14). Obie grupy
różniły się (p < 0,05) nie tylko parametrami antropometrycznymi,
ale również metabolicznymi: otyłe kobiety miały wyższe stężenie
glukozy (odpowiednio w grupie 1. i 2.: 88,5 ± 9,2 mg/dl vs. 81,2 ±
± 7,5 mg/dl), triglicerydów (148,1 ± 73,8 mg/dl vs. 100,1 ± 40,7 mg/dl),
insuliny (8,5 ± 5,85 µjm./ml vs. 4,9 ± 2,1 µjm./ml) i wskaźnik HOMA
(homeostasis model assessment method) (1,87 ± 1,45 vs. 0,99 ±
± 0,48), a niższe stężenie cholesterolu frakcji HDL (60,6 ± 14,6 mg/dl
vs. 76,3 ± 23,0 mg/dl), adiponektyny (12,7 ± 4,4 vs. 16,6 ± 4,4 µmg/ml)
i QUICKI (quantitative insulin sensitivity check index) (0,16 ± 0,02
vs. 0,17 ± 0,02). Badane grupy nie różniły się ani średnią wieku, ani
stężeniem cholesterolu całkowitego czy frakcji LDL. Nie stwierdzono
różnic w stężeniu adiponektyny i stężeniu insuliny oznaczanej przed
wysiłkiem fizycznym, bezpośrednio po i 6 godzin po wysiłku fizycznym.
WNIOSEK. Stężenie adiponektyny i insuliny nie ulega zmianie po
1-razowym standaryzowanym wysiłku fizycznym.INTRODUCTION. Insulin resistance and low adiponectin serum level,
are known cardiovascular risk factors. The question appears, does
physical activity influence these factors.
The aim of our study was evaluation of the effect of standardized
physical effort on insulin and adiponectin serum levels in postmenopausal
women.
MATERIAL AND METHODS. We performed the study in 33 postmenopausal
women, aged 50-60, selected randomly from Wroclaw
city population. Estimation of anthropometric parameters and
densitometry (total body fat, android and gynoid deposits) as well
as the 30-minutes endurance test using cycloergometer was carried
out in all subjects. Blood for analysis was collected before,
direct after and 6 h after the test. Biochemical estimations such as
lipid profile, glucose, insulin and adiponectin level were carried
out using commercial kits.
RESULTS. We compared 2 distinguished groups of women:
I - obese (BMI > 30, waist > 82 cm, n = 19) and II - lean (BMI
< 25, waist < 82 cm, n = 14). Both groups differed (p < 0.05) not
only in anthropometric but also in metabolic parameters: obese
women had higher serum level of glucose (respectivelly in I and II
group: (88.5 ± 9.2 mg/dl vs. 81.2 ± 7.5 mg/dl), triglycerides (148.1 ± 73.8 mg/dl vs. 100.1 ± 40.7 mg/dl), insulin (8.5 ± 5.85 µIU/ml
vs. 4.9 ± 2.1 µIU/ml) and HOMA (1.87 ± 1.45 vs. 0.99 ± 0.48), and
lower level of HDL cholesterol (60.6 ± 14.6 mg/dl vs. 76.3 ± 23.0 mg/dl),
adiponectin (12.7 ± 4.4 vs. 16.6 ± 4.4 ìg/ml) and QUICKI (0.16 ±
± 0.02 vs. 0.17 ± 0.02). Investigated groups did not differ in mean
age and total and LDL cholesterol. We did not show any differences
in insulin and adiponectin levels measured before and after
physical test, either.
CONCLUSION. Serum level of adiponectin and insulin do not
change after single standardized physical effort
Influence of standardized physical effort on reduced glutathione level in postmenopausal women - the role of β3-adrenergic receptor polymorphism
WSTĘP. Istnieją doniesienia, że polimorfizm Trp64/Arg64 receptora
b3-adrenergicznego (ADRB3) wiąże się z częstszym występowaniem
otyłości. Udowodniono także, że otyłość wiąże się z nasilonym
stresem oksydacyjnym. Celem naszych badań była ocena
związku wspomnianego polimorfizmu ADRB3 z parametrami
powysiłkowego stresu oksydacyjnego u kobiet po menopauzie.
MATERIAŁ I METODY. Badania przeprowadzono u 80 kobiet
w wieku 50–60 lat, wybranych losowo spośród mieszkanek Wrocławia.
U wszystkich badanych wykonano pomiary antropometryczne,
badanie na zawartość tkanki tłuszczowej (total body) metodą
DXA z użyciem aparatu DPX(+) Lunar (Stany Zjednoczone) oraz
oznaczenia biochemiczne: profil lipidowy, stężenie glukozy, estradiolu
i FSH z użyciem komercyjnych zestawów odczynników. Wskaźnikiem stresu oksydacyjnego było stężenie zredukowanego
glutationu w krwinkach oznaczane w heparynizowanej krwi
pełnej kolorymetryczną metodą Beutlera. Krew do oznaczenia stresu
oksydacyjnego pobierano przed, bezpośrednio po i 6 godzin
po 30-minutowym wysiłku fizycznym na cykloergometrze. Genotypowanie
ADRB3 przeprowadzono metodą PCR i minisekwencjonowania
z użyciem aparatu ABI 310 (Applied Biosystems).
WYNIKI. Częstość występowania genotypu Trp64/Arg64 w badanej
populacji wyniosła 11%. Genotyp Arg64/Arg64 wystąpił tylko u jednej
osoby. U pozostałych 88% kobiet stwierdzono genotyp Trp64/
/Trp64. Podobne w obu grupach średnie wyjściowe stężenie zredukowanego
glutationu w krwinkach po 6 godzinach od wysiłku obniżało
się istotnie (p < 0,05) jedynie w grupie z genotypem Trp64/Trp64.
WNIOSKI. Polimorfizm Trp64/Arg64 receptora b3-adrenergicznego
ma najprawdopodobniej wpływ na stężenie zredukowanego glutationu
po standaryzowanym wysiłku fizycznym u kobiet po menopauzie.INTRODUCTION. Some, but not all studies indicate, that the Trp64/
/Arg64 polymorphism of β3-adrenergic receptor (ADRB3) is associated
with obesity. Furthermore, obesity is connected with enhanced
oxidative stress.
The aim of our study was evaluation of frequency of this ADRB3
polymorphism and its association with metabolic syndrome parameters
and oxidative stress in postmenopausal women.
MATERIAL AND METHODS. We performed the study in 80 women,
aged 50-60, selected randomly from Wroclaw city population.
Estimation of anthropometric parameters, total body fat - using
DPX(+) Lunar, USA device and biochemical estimations such as
lipid profile, glucose, estradiol and FSH serum level (using commercial
kits) were carried out. Oxidative stress was estimated by
measurement of reduced glutathione concentration, using Beutler
colorimetric method. Blood for analysis was collected before, direct
after and 6 h after the 30-minutes physical test using cycloergometer.
ADRB3 genotyping was performed by PCR and minisequencing
using ABI 310 sequencer (Applied Biosystems).
RESULTS. The frequency of Trp64/Arg64 genotype in investigated
population was 11%. The Arg64/Arg64 genotype had only one woman.
Trp64/Trp64 genotype had 88% of women. The reduced glutathione
level similar in both groups before effort, 6 h after standardized
physical effort decreased statistically significant (p < 0.05)
only in Trp64/Trp64 group.
CONCLUSIONS. The Trp64/Arg64 polymorphism of β3-adrenergic
receptor seems to have an influence on reduced glutathione level
after standardized physical effort in postmenopausal women
Differences in Chronic Low-Grade Inflammation and Metabolic Disturbances between <i>VDR</i> Genotypes in an Ethnically Homogenous Postmenopausal Female Population from Poland
(1) Vitamin D deficiency and changes in the endocrine system may stimulate systemic inflammation. VDR expression and the vitamin D concentration decrease with age, which is important in postmenopausal women for whom estrogen deficiency causes rapid bone loss. This group is, moreover, particularly at risk of developing atherosclerosis and its adverse consequences, such as chronic inflammation. The aim of this study was to assess the differentiation by the VDR genotype of the risk factors for so-called chronic low-grade inflammation and metabolic disorders. (2) We studied the differences between the anthropometric, metabolic, and inflammation parameters of VDR genotypes for Apa-I, Bsm-I, Fok-I, and Taq-I in a sample of 321 women aged 50–60 from an ethnically homogeneous urban population in Poland. (3) The TT Taq-I genotype presented a significantly higher rate of insulin resistance (HOMA) and lower serum levels of adiponectin than the other two genotypes. The AA genotype of the Bsm-I polymorphism was associated with a more atherogenic serum profile and significantly higher LDL and LDL/HDL values and Castelli Index. (4) Chronic low-grade inflammation was associated with the TT Taq-I genotype and presented a higher rate of insulin resistance. The AA genotype of the Bsm-I polymorphism presented a more atherogenic serum lipid profile and, therefore, a higher risk of developing cardiovascular disease
Exploratory data analysis of a clinical study group: Development of a procedure for exploring multidimensional data.
Thorough knowledge of the structure of analyzed data allows to form detailed scientific hypotheses and research questions. The structure of data can be revealed with methods for exploratory data analysis. Due to multitude of available methods, selecting those which will work together well and facilitate data interpretation is not an easy task. In this work we present a well fitted set of tools for a complete exploratory analysis of a clinical dataset and perform a case study analysis on a set of 515 patients. The proposed procedure comprises several steps: 1) robust data normalization, 2) outlier detection with Mahalanobis (MD) and robust Mahalanobis distances (rMD), 3) hierarchical clustering with Ward's algorithm, 4) Principal Component Analysis with biplot vectors. The analyzed set comprised elderly patients that participated in the PolSenior project. Each patient was characterized by over 40 biochemical and socio-geographical attributes. Introductory analysis showed that the case-study dataset comprises two clusters separated along the axis of sex hormone attributes. Further analysis was carried out separately for male and female patients. The most optimal partitioning in the male set resulted in five subgroups. Two of them were related to diseased patients: 1) diabetes and 2) hypogonadism patients. Analysis of the female set suggested that it was more homogeneous than the male dataset. No evidence of pathological patient subgroups was found. In the study we showed that outlier detection with MD and rMD allows not only to identify outliers, but can also assess the heterogeneity of a dataset. The case study proved that our procedure is well suited for identification and visualization of biologically meaningful patient subgroups
